Description
本出願は、"META-SUBSTITUTED ARYL AMIDE SPHINGOSINE 1-PHOSPHATE ANALOGS AS S1P RECEPTOR ANTAGONISTS" という表題の仮出願(2004年12月06日出願、出願番号60/633,587)の優先権を主張する。また、この出願は参照することによって本明細書に含まれる。
本明細書に記載された発明は、the National Institutes of Healthから与えられた助成金番号GM067958 および GM064101による政府の補助の下になされたものである。米国政府は本発明についての一定の権利を有する。

メタ置換された化合物であるVPC23031、VPC25239およびVPC23019の合成ルートは、3-ヨード-1-ニトロベンゼンと適切な末端アルキンとの薗頭カップリング(Sonogashira coupling)(6)から開始した。その後、得られた付加体のニトロ基と三重結合を同時に水素化してメタ置換アニリン類を生成した。次にこのアニリン類を、保護されているセリンに結合させて、得られたアミド類を遊離のアルコールを得るために水素化分解した。このアルコール類を続いて、リン酸化し、過酸化水素で酸化し、その後、酸触媒の下で広範囲に脱保護を行って、最終生成物であるVPC23031、VPC25239およびVPC23019を得た。オルト置換された化合物であるVPC23089は薗頭カップリングによる2-ヨードアニリンと1-オクチンの結合から開始した。得られたアニリンを、その後、保護されているセリンとPyBOP試薬を用いて結合させた。得られたアミドをその後、ベンジルエーテル保護基を取り除くと同時にアリール三重結合を還元するために、水素化/水素化分解ステップに進めた。遊離のアルコールを次にリン酸化し、過酸化水素で酸化し、その後、酸触媒の下で広範囲に脱保護を行って、最終生成物であるVPC23089を得た。NMRおよび質量分析を全ての構造を確かめるために用いた。VPC23019はAvanti Polar Lipidsから得ることができる。
化合物VPC44116は、図6Aおよび図6Bに説明したとおりに調製した。N-BOC保護基は既知の方法によって除去される。得られた化合物の1Hスペクトルと13Cスペクトルは図7および図8に示したとおりである。質量分析のデータは以下のとおりである。MS m/z = 371.9、370.8(基準ピーク)、348.8、270.5、178.6。
適切なレセプタープラスミドDNA(ヒトS1P1レセプター、ヒトS1P2レセプター、ヒトS1P3レセプター、ヒトS1P4レセプター、ヒトS1P5レセプター、ヒトLPA1レセプター、ヒトLPA2レセプター、もしくはヒトLPA3レセプターをコードしたもの)を当量のヒトGi2α(S1P3には、変異型(C352F)ラットGi2αを使用)、ウシβ1タンパク質、およびウシγ2タンパク質をコードした発現プラスミドと混合する。その後、これらのDNAを、リン酸カルシウム沈殿法 (7) を用いてHEK293T細胞(ここで「T」はSV-40ウイルスのラージT抗原の発現を示す)の単層(monolayer)を導入するのに用いる。60時間後に細胞を集め、膜を調製し、等分して、使用するまでの間-70℃で保存される(8)。レセプターおよびGタンパク質の形質移入(transfection)は、[γ-35S]GTP結合アッセイ([γ-35S]GTP binding assay)(後述)によって確認される。Gタンパク質のみで形質移入されたHEK293T細胞の分析では、アゴニストによる刺激に応答しなかった(図2Bを参照)。
24細胞単層は、Lipofectamine 2000 (Invitrogen, Carlsbad, CA) か FuGENE 6 (Roche Applied Science, Indianapolis, IN) を用いて、ヒトS1P1レセプター、ヒトS1P2レセプター、ヒトS1P3レセプターをコードしたDNAと、pIREスプロ2プラスミドDNA(pIRESpuro2 plasmid DNA)(Clontech, San Jose, CA,)とを同時形質移入する。ピューロマイシン(Sigma- Aldrich, St. Louis, MO)を培地に加えることによって、ピューロマイシンアセチルトランスフェラーゼ遺伝子(puromycin acetyltransferase gene)を発現したクローン群(Clonal population)を選択できる。T24細胞は単層中で37℃、5% CO2/95% 空気の雰囲気下、95% DMEM/F-12媒体、および、10%の木炭/デキストラン処理したFBSからなる培養基(growth media)中で培養される。
前述のとおり[γ-35S]GTP結合アッセイを行う(8)。膜は1〜5μgのタンパク質を含み、5μgのサポニン、10μgのGDP、0.1nMの[γ-35S]GTP(1200 Ci/mmol)を含む0.1mlの結合バッファで(mM単位で含む:HEPES/50、NaCl/100、MgCl2/10、pH 7.5)インキュベートし、30分間、30℃で脂質標識し(indicated lipid(s))、Brandel Cell Harvester (Gaithersburg, MD)を用いて回収される。その後、試料を結合した放射性核種で分析する。
細胞移動アッセイ(cell migration assay)は、0.1%ゼラチンで下側を覆ったポリカーボネート膜を含む修正したボイデンチャンバー(Boyden chamber)(処置される培養組織、直径24mm、厚み10μm、孔8μm、Transwell(登録商標); Costar Corp., Cambridge, MA)を用いて行う。ポリカーボネート膜の下側は、一度、移動媒体(migration media)(フェノールレッドを含まないDMEM/F12、および、脂肪酸を含まない0.1% BSA)ですすぎ、2ミリリットルの移動媒体を含んでいる低いチャンバー(lower chamber)に浸す。ヒトS1P1レセプターDNAと安定的に形質移動したT24細胞は、木炭/デキストラン処理したFBSと10μg/mlピューロマイシンを含むDMEM/F12媒体中で、150x25mm の組織培養プレートに100%コンフルエンス(confluence)まで培養し、12時間以上、血清を欠乏させた。血清欠乏細胞(Serum-starved cells)は培養皿から10Xトリプシン-EDTA(5 mM EDTA および 25 mM HEPESを含むハンクス液(Hanks' balanced salt solution)、ならびに0.1%トリプシン;Invitrogen, San Diego, CA)で取り除き、移動バッファ(migration buffer)中で懸濁させ、遠心分離(130 x gで5分間)で集めた。上澄みを吸引によって除去し、cell pelletを移動媒体中に再懸濁させた(106 cells/ml)。1ミリリットルのセル懸濁液を各移動チャンバーの上から加え、S1PアゴニストVPC22277(10nM)を低いチャンバーに加えた。低いチャンバーに加えたアンタゴニストの存在下、もしくは、存在しない状態で(VPC23019 (0 nM〜1000 nM)、VPC23019、VPC23031、VPC23089、およびVPC25239 (全て 50 nM))、細胞の膜の下側への移動が許容された(4時間、37℃)。上部チャンバー(upper chamber)中の非遊走細胞(non-migrating cell)は吸引によって除去され、膜の底面に付着している遊走細胞(migratory cells)は、10X Trypsin-EDTA中で1分間室温においてインキュベーションし、細胞を膜から除去するためにプレートを軽くたたくことにより単離された。膜ごとの遊走細胞の塊は100μlの細胞懸濁液と同じ体積のCyQuant染料溶液(3.0mlの2X溶解バッファおよび15μl CyQuant染料)とを混合し、得られた溶液の蛍光強度をFlexStation(登録商標) fluorimeter (Molecular Devices, Menlo Park, CA)を用いて定量することにより評価した。それぞれの測定は2つの個々の移動チャンバーでの平均を表す。VPC23019に付随する拮抗作用の可逆性を測定するために、細胞を10μM VPC23019と共に37℃で30分間インキュベートした。単層はリン酸緩衝生理食塩水で3回洗浄し、直ちに前述の細胞移動アッセイのための処理をした。
未変性のT24細胞、および、ヒトS1P2レセプターDNAかヒトS1P3レセプターDNAかのどちらかを安定に形質移入したT24細胞の細胞内のカルシウムを測定するのに、FlexStation(登録商標) fluorimeter (Molecular Devices, Menlo Park, CA)を用いた。細胞を96ウェル(96-well)の透明な底面黒色マイクロプレート(clear bottom black microplate)(Corning Costar Corp.)に播き(〜50000 細胞/ウェル)、37℃で一晩放置した。添加液(20 mM HEPES、脂肪酸を含まない0.1% BSA、および2.5mMプロベネシド(probenecid)を含むハンクス液pH 6.4)中の4 μM Fluo-4AMエステルによって、30分間37℃で染料を添加した。細胞単層をリン酸緩衝生理食塩水で3回洗浄した後、添加液を加え、FlexStation(登録商標)中において、細胞を一連の化合物に25℃で3分間接触させた。全ての場合において、個々の化合物のそれぞれの濃度について、少なくとも3回の実験を行った。VPC23019(10μl)に付随する拮抗作用の可逆性を測定するために、化合物を添加染料と共に細胞に加えて37℃で30分間インキュベートした。細胞はリン酸緩衝生理食塩水で3回洗浄し、直ちに前述のように化合物に接触させた。
VPC23019のS1P1レセプターDNAおよびS1P3レセプターとの結合定数(Kb)はLewとAngusによって論じられた非線形回帰法(9)を用いてフィッテイングした曲線からSchild分析して決定した。つまり、アゴニストが存在しないときと様々な濃度のアゴニストが存在する場合でのアゴニスト投与量依存曲線から得られたEC50値のlogの値を負にしたものをプロットすることによって生じた最もフィットする曲線の非線形分析を、Kb値を得るためにアンタゴニストの濃度に対してプロットした。アンタゴニストが単純な競合的相互作用の判断基準にあっているかどうかを確立するためにF-テスト分析も行った。
[32P]-S1Pは、スフィンゴシンをインキュベートすることによって調製し、[γ-32P]ATPをHEK293T細胞からの細胞可溶化物とともに、ヒトスフィンゴシンキナーゼ2型DNA(human sphingosine kinase type 2 DNA)と一過性に形質移入した。200μlの反応物には、0.025mMスフィンゴシン、1mCi [γ-32P]ATP (7000 Ci/mmol)およびキナーゼバッファ(mM単位で含む: Mg(C2H3O2) (50mM Tris中、pH 7.5)/10、NaF/10、および、セミカルバジド(semicarbizide)/2)が含まれる。反応は、20μgの細胞可溶化物を加え、37℃で30分以上インキュベーションすることから開始した。[32P]-S1Pは、1.0N HCl、2.0M KCl、メタノール、および、クロロホルムを反応混合物に加えて抽出し、ボルテックスし(vortexed)、1000 x gで5分〜10分間遠心分離した。有機相を分離し、水相部分を残したまま、さらに2回抽出した。混合した有機相部分を窒素気流下で乾燥させ、脂肪酸を含まない0.1%BSA水溶液(aqueous 0.1% fatty acid free BSA)に再懸濁させた。得られたもの([32P]-S1P)の比放射能は、放射標識した[γ-32P]ATPの比放射能(すなわち、7000 Ci/mmol)として見積もった。
レセプターおよびGタンパク質DNAの両方と一過性に形質移入したHEK293T細胞からのタンパク質5μgを含む膜を、0.5mlの結合バッファ中(mM単位で含む:HEPES/50、NaCl/100、MgCl2/10、pH 7.5)でインキュベートし、50pM [32P]S1P、室温で1時間脂質標識した。結合したリガンドは遊離のリガンドと急速ろ過によって分離し、液体シンチレーションカウンターで分析した。非特異的結合は、過剰のS1P存在下での膜への、レセプターおよびGタンパク質DNAと一過性に形質移入したHEK293T細胞(熱変性させたものと熱変性させていないものの両方)からの放射性リガンドの残余結合(residual binding)として測定し、それは通常は全体の結合の60%であった。リガンド-レセプター相互作用に関する結合定数(Ki)は、Chang-Prusoffの式(Chang-Prusoff equation、Ki = IC50/(1+[L]/Kd)を用いて、IC50値から決定した。この式を適用するにあたって、放射性リガンドの濃度[L]は0.05nMでKd値はS1P-S1P相互作用で報告された値(すなわち、8.1nM(12))を用いた。
全ての投与量依存性曲線についてのEC50値およびIC50値は、Graphpad Prism(コピーライト)プログラムを用いた全てのデータの非線形回帰分析によって決定した。集めたデータについての誤差は標準誤差(S.E.M.)で報告している。
〈 VPC23019はS1P1レセプターおよびS1P3レセプターへの受容体活性化作用を有さない。 〉
S1PアナログSARについての我々の実験中に、我々はアリール-アミド化合物VPC23019(図1)が、破砕細胞[γ-35S]GTP結合アッセイ(broken-cell [γ-35S]GTP binding assay)においてS1P1レセプター(図2A)およびS1P3レセプター(図2C)へのアゴニスト活性を有さないことを発見した。実際には、VPC23019の濃度が100nM以上のときにこのアッセイにおいて逆作動を示唆する特性が観察された。しかしながら、中性アンタゴニストの存在下でのこの活性により、VPC23019についての観察結果をこれらの濃度での逆作動薬活性として特徴付ける必要があると考えられる。これらの化合物が利用(available)できないため、内在性の膜-関連アゴニスト活性についてのVPC23019の変化によって、これらの濃度で観測された[γ-35S]GTP結合の減少が起きたのだと考えることができる。このアゴニスト活性の欠落は、T24細胞(膀胱癌に由来)を用いた全細胞アッセイによって確かめられた。ここで、S1P1レセプター(T24-S1P1)かS1P3レセプター(T24-S1P3)は安定に発現した。RT-PCR分析では、T24細胞中の内在的S1P2レセプターmRNAのみが検出された。しかしながら、応答の有効性は同じであったが、S1P1、S1P2、もしくはS1P3(図2E)の組み換え型が安定に発現しているときは、S1Pは少なくとも100倍以上強力である。したがって、T24細胞系によって形質移入によって導入された個々のS1Pレセプターを調べる機会が提供された。多くの研究によってS1Pが細胞移動を推進することが示されている。しかしながら、最近、S1Pは移動を阻害する(S1P2レセプターの刺激による可能性がある)可能性もあることが示された(10)。この阻害効果を避けるために、我々はVPC22277(図1)を用いた。VPC22277はS1P1レセプターおよびS1P3レセプターのアゴニストであるがS1P2レセプターのアゴニストではないS1Pアナログである(表1)―(アリールアミド類の全ての化合物(11)はS1P2レセプターへの活性を全く有さない)。pEC50値は、最大の[γ-35S]GTP結合の50%となる化合物のモル濃度の-log(-log molar concentration)である。pEC50値は、pEC50値±S.E.M.で報告する。
VPC23019がS1P1レセプターもしくはS1P3レセプターに対しての逆作動薬活性を示すという知見により、我々はこの化合物がアゴニスト活性を阻害するかについて調べた。[γ-35S]GTP結合アッセイを用いて、我々はS1P1レセプター(図3A)かそれともS1P3レセプター(図3B)について、VPC23019濃度が増加するにつれてS1Pのインキュベーションが投与量依存性(S1P濃度効果曲線の右側への平行移動)を示すことを見出した。アゴニストに媒介された応答についてのこのシフトは2種類の細胞全体のセルアッセイ(セル移動アッセイ(図3C)およびカルシウム動員)でも見られた。VPC23019は、30μMまでの濃度のLPA1-3EDGファミリーレセプターに活性を示さなかったし、これらのサイトでのLPAの活性の阻害もしなかった。
[γ-35S]GTP結合アッセイでのVPC23019に関係するアンタゴニスト活性についてのSchild分析により、S1P1レセプター(図3A)およびS1P3レセプター(図3B)についてpKb値、7.49 ±0.15および5.98 ±0.08がそれぞれ得られた。さらに、化合物が競合的アゴニストとして作用するかを予測するLewとAngusによって論じられた非線形回帰法(9)によって、VPC23019が両方のレセプターに対して競合的アゴニストとして作用することが示唆された。T24-S1P3細胞でのカルシウム動員のSchild分析によりVPC23019のKb値が得られ、そのKb値は[γ-35S]GTP結合アッセイで観測されたもののおよそ10倍以下であった。しかしながら、非線形回帰分析により、T24-S1P3細胞でのカルシウム動員アッセイにおいて、VPC23019が競合的アゴニストとして作用しないことが示唆された。重要なことには、両方の細胞全体のS1Pレセプターアッセイ(Ca2+動員(S1P3、図3D)および細胞移動(S1P1、開示なし))では、アンタゴニストを洗い流した後、完全に回復した。したがって、VPC23019の拮抗作用は可逆的であり、さらには十分に克服可能(surmountable)であって、競合的アンタゴニストに不可欠な基準を満たしている。
