Abstract
Description
本発明は、標的レセプターを活性化し得るマルチマー融合タンパク質、このような融合ポリペプチドを産生する方法、およびこの標的レセプターの活性化が所望される疾患または状態の処置、診断またはモニタリングするための方法に関する。
可溶性Ephリガンドドメインをクラスター化して、それらの同族レセプターを活性化し得るマルチマーを作製することは、特許文献1に記載されている。特許文献2は、抗体を用いてエリスロポエチンレセプターを活性化する方法を記載する。
本発明は、活性化されるためにマルチマー化を必要とする標的レセプターを活性化し得るマルチマー融合ポリペプチドを提供する。本発明のポリペプチドは、標的レセプターの活性化が望ましい状態を処置するために有用であり、種々のインビトロおよびインビボでの診断用途および予後診断用途を有する。本発明のポリペプチドは、例えば、標的特異的抗体または天然のリガンドと比較して改善された標的レセプター活性化能力を示す、単一特異性(monospecific)または二重特異性(bispecific)のテトラマーであり得る。
本方法を記載する前に、特定の方法および記載された実験条件は変化し得るので、本発明がこのような特定の方法、および記載された実験条件に限定されないことが理解されるべきである。本明細書中で用いられる用語が、特定の実施形態を記載することのみを目的とし、限定することを意図しないこともまた理解されるべきである。なぜなら、本発明の範囲は、添付の特許請求の範囲によってのみ限定されるからである。
本明細書中で用いられる場合、用語「標的レセプター関連状態または標的レセプター関連疾患」とは、一般に、特定の標的レセプターと関連した哺乳動物宿主(特に、ヒト宿主)の状態を包含する。従って、標的レセプター関連状態を処置することは、標的レセプター活性の減少を反映する症状を有するか、または疾患、状態もしくは処置のレジメンに応じてこのようなレベルの低下を有することが予想される、哺乳動物(特に、ヒト)の処置を包含する。標的レセプター関連状態または標的レセプター関連疾患を処置することは、ヒト被験体の処置を包含し、ここで、本発明の活性化ダイマーを用いた標的レセプターの活性化の促進は、標的レセプター関連状態または標的レセプター関連疾患から生じる望ましくない症状の改善をもたらす。本明細書中で用いられる場合、「標的レセプター関連状態」はまた、特定の標的レセプターの活性化を一時的に、または長期にわたって変更することが望ましい状態を包含する。
標的レセプターの例は、Ephファミリーのメンバー(例えば、EphA1、EphA2、EphA3、EphA4、EphA5、EphA6、EphA7、EphA8、EphB1、EphB2、EphB3、EphB4、EphB5、EphB6)、Tieレセプター(例えば、Tie−1またはTie−2)である。適切なリガンドまたはそのフラグメントとしては、エフリン(ephrin)(例えば、エフリン−A1、エフリン−A2、エフリン−A3、エフリン−A4、エフリン−A5、エフリン−B1、エフリン−B2、エフリン−B3)の可溶性ドメイン、およびアンジオポエチン(angiopoietin)(例えば、アンジオポエチン−1(ang−1)、ang−2、ang−3、および/またはang−4)のフィブリノゲンドメインが挙げられる。
特定の実施形態では、本発明の活性化ダイマーは、選択された標的レセプターに対して作製された抗体から単離された1以上の免疫グロブリン結合ドメインを含む。用語「免疫グロブリン」または「抗体」とは、本明細書中で用いられる場合、抗原(本発明の場合、標的レセプターまたはその一部である)に特異的に結合して抗原を認識する、免疫グロブリン遺伝子由来のフレームワーク領域またはそのフラグメントを含む、哺乳動物(ヒトを包含する)ポリペプチドをいう。意図される活性化ダイマーが哺乳動物治療剤として用いられる場合、免疫グロブリン結合領域は、対応する哺乳動物免疫グロブリンに由来すべきである。活性化ダイマーが非治療用途(例えば、診断およびELISA)を意図される場合、免疫グロブリン結合領域は、ヒトまたは非ヒト哺乳動物(例えば、マウス)のいずれかに由来し得る。ヒト免疫グロブリン遺伝子または遺伝子フラグメントとしては、κ定常領域、λ定常領域、α定常領域、γ定常領域、δ定常領域、ε定常領域、およびμ定常領域、ならびに無数の免疫グロブリン可変領域遺伝子が挙げられる。軽鎖は、κまたはλのいずれかであると分類される。重鎖は、γ、μ、α、δまたはεと分類され、次いで、それぞれ、免疫グロブリンのクラスIgG、IgM、IgA、IgD、およびIgEを規定する。各IgGのクラス内には、異なるアイソタイプ(例えば、IgG1、IgG2など)が存在する。代表的には、抗体の抗原結合領域は、結合の特異性および親和性を決定する際に最も重要である。
本発明の融合ポリペプチドの個々の成分は、当該分野で公知の分子生物学的方法を用いて、核酸分子から産生され得る。核酸分子は、適切な宿主細胞中に導入された場合に融合ポリペプチドを発現し得るベクター中に挿入される。適切な宿主細胞としては、細菌細胞、酵母細胞、昆虫細胞および哺乳動物細胞が挙げられるがこれらに限定されない。DNAフラグメントをベクターに挿入するための当業者に公知の任意の方法を用いて、転写/翻訳制御シグナルの制御下で本発明の融合ポリペプチドをコードする発現ベクターを構築し得る。これらの方法は、インビトロ組換えDNAおよび合成技術およびインビボ組換えを包含し得る(Sambrookら,Molecular Cloning,A Laboratory Manual,Cold Spring Harbor Laboratory;Current Protocols in Molecular Biology,Ausubelら編,Greene Publ.Assoc.,Wilwy−Interscience,NYを参照のこと)。
核酸構築物は、抗標的レセプター抗体由来の結合ドメインをコードする領域を含む。一般に、このような結合ドメインは、VH鎖可変領域またはVL鎖可変領域に由来する。所望の結合特性を示す抗体の同定および選択の後、各抗体の重鎖および/または軽鎖の可変領域が単離され、増幅され、クローニングされ、そして配列決定される。アミノ酸をコードするおよび/もしくは制限部位を保有するヌクレオチド配列の付加、アミノ酸をコードするヌクレオチド配列の欠失、またはアミノ酸をコードするヌクレオチド配列の置換を含め、改変は、VHヌクレオチド配列およびVLヌクレオチド配列に行われ得る。
本発明に従って、核酸構築物は、標的レセプターリガンド由来の結合ドメインをコードする成分を含む。所望の結合特性を示す、リガンドの標的レセプター結合ドメインの同定後、このようなドメインをコードする核酸が、核酸構築物において用いられる。このような核酸は、アミノ酸をコードするかおよび/もしくは制限部位を保有するヌクレオチド配列の付加、アミノ酸をコードするヌクレオチド配列の欠失、またはアミノ酸をコードするヌクレオチド配列の置換を含め、改変され得る。
本発明の活性化ダイマーはまた、標的レセプターを検出することおよび/または標的レセプターのレベルを測定することが所望される場合、インビトロまたはインビボでのスクリーニング方法において用いられ得る。スクリーニング方法は、当該分野で周知であり、そして無細胞アッセイ、細胞ベースのアッセイおよび動物アッセイを包含する。インビトロアッセイは、固体状態または可溶性のいずれかであり得る。標的レセプター検出は、標的レセプターに結合した活性化ダイマーを同定し得る標識または検出可能な基の使用を含め、当該分野で公知の多数の方法で達成され得る。検出可能な標識は、免疫アッセイの分野において十分に開発されており、そして一般に、本発明の活性化ダイマーを用いるアッセイに関連して用いられ得る。
本発明の活性化ダイマーがそれらのレセプターへの高親和性結合を示す能力によって、これらは、それらのレセプターを効率的に活性化するために治療的に有用となる。従って、特定の例では、標的レセプター(例えば、エフリン)についての内因性リガンドの効果を増加させることが所望され得る。例えば、神経系外傷の領域では、特定の状態は、エフリン応答性の増加から利益を獲得し得る。それゆえ、本明細書中に記載される組成物の使用によって、このような状態を罹患する患者におけるエフリンの結合親和性を増大させることが有益であり得る。
因子(例えば、タンパク質または核酸)の治療送達について当該分野で公知の方法(例えば、細胞トランスフェクション、遺伝子治療、送達ビヒクルもしくは薬学的に受容可能なキャリアを用いた直接投与、本発明の活性化ダイマーをコードする核酸を含む組換え細胞を提供することによる間接的送達)は、被験体における標的レセプターを活性化するために、本発明の活性化ダイマーまたは活性化ダイマーをコードする核酸の治療送達のために用いられ得る。
本発明はまた、本発明の活性化ダイマーおよび薬学的に受容可能なキャリアを含む、薬学的組成物を提供する。用語「薬学的に受容可能な」とは、連邦の統制機関もしくは州政府によって承認されるかまたは米国薬局方もしくは動物、より特定にはヒトにおいて使用するための他の一般的に認識される薬局方に列挙されることを意味する。用語「キャリア」とは、治療剤とともに投与される、希釈剤、佐剤、賦形剤、またはビヒクルをいう。このような薬学的キャリアは、無菌の液体(例えば、水および油(石油起源、動物起源、植物起源もしくは合成起源の油(例えば、落花生油、大豆油、鉱油、ゴマ油など)を含む))であり得る。適切な薬学的賦形剤としては、デンプン、グルコース、ラクトース、スクロース、ゼラチン、麦芽、米粉(rice,flour)、白亜、シリカゲル、ステアリン酸ナトリウム、モノステアリン酸グリセロール、滑石、塩化ナトリウム、脱脂粉乳、グリセロール、プロピレングリコール(propylene,glycol)、水、エタノールなどが挙げられる。この組成物はまた、所望の場合、微量の湿潤剤もしくは乳化剤、またはpH緩衝剤を含み得る。これらの組成物は、液剤、懸濁剤、乳剤、錠剤、丸剤、カプセル剤、散剤、徐放処方物などの形態を採り得る。この組成物は、トリグリセリドのような伝統的結合剤およびキャリアを用いて、坐剤としても処方され得る。経口処方物は、標準的キャリア(例えば、薬学的グレードのマンニトール、ラクトース、デンプン、ステアリン酸マグネシウム、サッカリンナトリウム、セルロース、炭酸マグネシウムなど)を含み得る。適切な薬学的キャリアの例は、E.W.Martinによる「Remington’s Pharmaceutical Sciences」に記載される。
本発明はまた、本発明の少なくとも1つの活性化ダイマーが充填された1以上の容器を備える、薬学的パックまたは薬学的キットを提供する。必要に応じて、このような容器に関連して、医薬品または生物製剤の製造、使用または販売を規制する政府機関によって規定される形式の通知が存在し得、この通知は、(a)ヒトへの投与のための製造、使用もしくは販売の、当該機関による認可、(b)指示書、またはその両方を反映する。
本発明は、本発明の融合ポリペプチドを発現するトランスジェニック非ヒト動物を包含する。トランスジェニック動物は、核酸を、(例えば、マイクロインジェクション、レトロウイルス感染によって)受精した卵母細胞の雄性前核に導入し、偽妊娠した雌性養母動物においてこの卵母細胞を発生させることによって生成され得る。発現ベクターにおいて有用な調節配列または他の配列のいずれかは、トランスジェニック配列の一部を形成し得る。組織特異的調節配列は、導入遺伝子に作動可能に連結されて、特定の細胞への導入遺伝子の発現を導き得る。本発明の融合ポリペプチドを発現するトランスジェニック非ヒト動物は、このような融合タンパク質を産生する手段としてを含め、種々の適用において有用である。
5匹の8週齢Balb/cマウスに、精製ヒトTie−2−Fc(hTie2−Fc)を最初に免疫した;各マウスに、200μlの乳濁液(100μgの精製hTie2−Fcタンパク質および100μlのフロイント完全アジュバントを含む)を皮下注射した。最初の注射の15日後、各マウスは、200μlの乳濁液(100μlのPBSおよび100μlのフロイント不完全アジュバント中に100μgの精製hTie2−Fcを含む)の皮下注射を受けた。この注射を、この5匹のマウスについて、7日後に繰り返した。1匹のマウスを、hTie−2に対するハイブリドーマの作製のために用いた。4匹の残りのマウスの各々に、hTie2−Fcの最初の注射の6ヵ月後、200μlの乳濁液(各々、100μlのPBSおよび100μlのフロイント不完全アジュバント中に100μgの精製ラットTie−2−Fc(rTie2−Fc)を含む)の皮下注射を行った。11日後、rTie2−Fcに対するマウスの免疫応答を、各マウスについて、200μlの乳濁液(100μlのPBSおよび100μlのフロイント不完全アジュバント中に100μgの精製rTie2−Fcを含む)の皮下注射によって追加免疫(boost)した。マウス血清を、注射の3日後、尾静脈から収集し、次いで、rTie2−Fcに対する抗体力価をELISAにより決定した。最大の力価を有する2匹のマウスに、100μl PBS中の100μgの精製rTie2−Fcの尾静脈注射により、最後の追加免疫を与えた。このマウスを3日後に屠殺し、そしてそれらの脾細胞を、Sp2/0−Ag14細胞との融合のために収集した。
一般に、rTie−2に特異的な抗体を発現するハイブリドーマ由来の抗体可変領域を、マウス抗体可変領域遺伝子に特異的なプライマーを用いてRT−PCR産物のDNA配列を最初に決定し、次いでこの決定した配列に基づいて特異的プライマーを用いて、ScFvをコードするDNAフラグメントを増幅することによってクローニングした。これらのScFv DNAフラグメントをクローニングし、その結果、これらは、複数のプラスミドとカセット交換されて、活性化ダイマーの全ての組合せを生じ得た。例えば、1つの増幅されたScFvフラグメントは、N末端においてシグナル配列に、そしてC末端においてIgG Fcドメインについてのコード配列に融合され得るか、または、このフラグメントは、ScFvコード配列の3’末端が翻訳停止コドンを含むように、IgG Fcコード配列のC末端に融合され得る。
単一特異性および二重特異性の四価活性化ダイマーを構築するための一般的スキームは、B2またはA12AのいずれかのScFv遺伝子が、1セットの制限酵素で切断した場合はマウスROR1シグナル配列(配列番号27)とhFcをコードする遺伝子(GenBank登録番号X70421のヌクレオチド85〜765)との間に、または異なるセットの酵素で切断した場合はhFc遺伝子の後に、挿入される能力に基づいた。ScFv遺伝子のこの設計により、プラスミド内のScFvカセットの交換が可能になり、標準的な公知の方法を用いて、ScFvとhFcとの異なる組合せが得られた。全ての構築物は、ScFv遺伝子の5’末端への制限部位の操作の結果、シグナルペプチドの切断部位とScFv遺伝子の開始部との間に、必要に応じて3つのアミノ酸リンカー(スペーサー)を有する。同様に、hFc遺伝子のアミノ末端に対する融合は、3アミノ酸配列(Gly−Pro−Gly)によって促進され、そしてhFc遺伝子のカルボキシ末端への融合は、残基Gly4−Ser−Gly−Ala−Pro(配列番号32)からなる8アミノ酸配列によって促進された。ScFv遺伝子における末端制限部位リンカーの結果、カルボキシ末端ScFvを有する全ての構築物は、アミノ酸Gly−Pro−Glyで終わる。
rTie−2に対する抗体、およびこれらの抗体に関連するキメラ分子を、培養ラット大動脈内皮細胞においてそれらがTie−2のリン酸化を誘導する能力について評価した。継代3回目と6回目との間のコンフルエントなRAEC(Vec Technologies)を、0.2%ゼラチンでコーティングしたT−75フラスコにおいて、MCDB−131培地(Vec Technologies)中で増殖させた。細胞を、無血清DME−Hiグルコース培地(Irvine Scientific)中で2時間飢餓させ、その後、0.1% BSAおよびチャレンジ分子を含む1.5ml無血清DME−Hiグルコース培地中で37℃にて5分間インキュベーションした。次いで、チャレンジ培地を除去し、そして細胞を、10μg/mlロイペプチン、10μg/mlアプロチニン、および1mM PMSFを含む、20mM Tris(pH7.6)、150mM NaCl、50mM NaF、1mMオルトバナジン酸Na、5mMベンズアミジン、1mM EDTA、1% NP−40、0.5%デオキシコール酸ナトリウム、0.1% SDS中で溶解した。溶解産物を、5μg抗Tie−2マウスモノクローナル抗体KP−m33、10μgビオチン化抗マウスIgG(Jackson Laboratories)、および100μlのニュートラアビジン(neutravidin)ビーズ(Pierce)とともに4℃にて16時間インキュベートすることにより、Tie−2を免疫沈降させた。ビーズを遠心分離により収集し、RIPA緩衝液で3回洗浄し、そして結合したタンパク質を、100℃にて5分間加熱することにより、10% β−メルカプトエタノールを含む40μlの5×Laemmli緩衝液を用いて溶出させた。4〜12%のTris/グリシンポリアクリルアミドゲル(Novex)でのSDS−ゲル電気泳動後、タンパク質をPVDFメンブレンに転写し、そしてマウス抗ホスホチロシン(anti−phophotyrosine)モノクローナル抗体4G10(Upstate)でプローブし、次いでヤギ抗マウスIgG−HRP結合体(Pierce)、続いてECL試薬(Amersham)を用いて検出した。RAECにおいてTie−2リン酸化を誘導する能力を、各活性化ダイマーについて決定した。活性を、FD1−Fc−FD1(BA1)(Tie−2の結合および活性化においてAng1と同等に活性であることが示されたキメラタンパク質(Davisら(2003)Nature Struct.Biol.10:38−44))を用いて得られる刺激のレベルとの比較により評価した(FD1またはFD2=それぞれ、Ang1またはAng2のヒトフィブリノゲンドメイン)。BA1を約0.5〜1.0μg/mlにおいて用いた場合、RAECにおけるTie−2の最大刺激(ECmax)が観察され、そして偽処理細胞におけるリン酸化レベルは低かった。同様に、ScFvB2−Fc−ScFvB2、ScFVA12A−Fc−ScFvB2、ScFvB2−Fc−FD1、およびScFvB2−Fc−FD2のECmaxは、約0.5〜1.0μg/mlであった。全ての場合、ScFvベースの分子は、ハイブリドーマ馴化培地から単離された関連のネイティブ抗体について観察されたよりも多くのリン酸化シグナルを誘導し得た。
Tie−2特異的ScFvおよびFD1またはFD2の両方を含むように、二重特異性四価分子を構築した。このキメラ分子を、ScFvB2をコードする遺伝子をhFcのN末端に融合し、そしてAng1 FD(GenBank登録番号Q15389のPhe283〜Phe498)またはAng2 FD(GenBank登録番号O15123のPhe281〜Phe496)をコードする遺伝子をC末端に融合することにより作製した。プラスミドpTE514は、ScFvB2−Fc−FD1(配列番号30)についての遺伝子をコードする。そしてプラスミドpTE514は、mROR1シグナルペプチドおよびCMV−MIEプロモーターを含んでいた。プラスミドpTE614は、ScFvB2−Fc−FD2(配列番号31)についての遺伝子をコードする。そしてプラスミドpTE614は、mROR1シグナルペプチドおよびCMV−MIEプロモーターを含んでいた。ScFvB2−Fc−ScFvB2およびScFvA12A−Fc−ScFvB2と同様に、pTE514から発現されるタンパク質およびpTE614から発現されるタンパク質は、Gly−Ala−ProリンカーをmROR1シグナルペプチドとScFvB2との間に有し、Gly−Pro−GlyリンカーをN末端ScFvB2とhFcとの間に有し、そしてGly4−Ser−Gly−Ala−Proリンカー(配列番号32)をhFcのC末端とAng FDのN末端との間に有した。ScFvB2−Fc−FD1およびScFvB2−Fc−FD2を両方とも、上記の通りに発現させて精製した。
二重特異性四価分子を、hTie−2に特異的なヒト抗体由来の2つのScFv、またはhTie−2に特異的なヒト抗体由来の1つのScFvおよびヒトFD1もしくはヒトFD2のいずれかを含む本発明のダイマー化融合構築物から形成する。hTie−2に特異的なヒトScFvを、当該分野で公知の方法により、そして上記の通りに、入手する。1つの実施形態では、ヒトScFvを、Reiterら(1999)J.Mol.Biol.290:685−698およびGillilandら(1996)Tissue Antigens 47(1):1−20に記載の通りに、組換えにより入手する。
抗rTie−2ハイブリドーマの産生に関する上記の手順に従って、抗rTie−1ハイブリドーマを生成した。手短に述べると、マウスを、精製したラットTie−1−Fcタンパク質およびフロイントアジュバントで3回免疫した。最大の抗Tie−1抗体力価を有するマウス由来の脾細胞を、ポリエチレングリコール(PEG)を用いて、Sp2/0−Ag14骨髄腫細胞と融合させた。融合後、これらの細胞を、2本のT225フラスコ中で培養した。翌日、HATをこれらの培養物に添加した。融合の9日後、これらの培養物に、新鮮な培地を補充した。ヒトIgGを、1mg/mlでこれらの培養物に添加した。融合の10日後、HAT耐性細胞を、室温にて増殖培地中で、1μg/mlビオチン−ラットTie−1−Fcで1時間にわたって、そして2.5μg/mlフィコエリトリン(PE)結合体化ストレプトアビジンで45分間にわたって、逐次染色した。染色後、これらの細胞をフローサイトメトリーによって分析した。rTie1−Fcに結合した細胞を、単一細胞を96ウェルプレート中に選別することによりクローニングした。96ウェルプレート培養物を、選別の10日後に2セットに分けた。マウスIgG重鎖可変領域のRT−PCR、続いて配列決定を、1セットの96ウェルプレート培養物について行った。独特のIgG重鎖可変領域配列を有するクローンを同定し、そして抗rTie−1抗体の産生のために増幅(expand)した。抗体をrTie−1タンパク質の結合について試験し、2つのクローン1−1F11および1−2G3を、さらなる詳細な研究のために選択した。
2種類のScFvベースのキメラ分子を、ScFvベースの分子がrTie−1レセプターを活性化する能力を評価するために構築した。1種類の分子は、hFcのN末端およびC末端の両方に融合した1つのScFvを用い、その結果は、rTie−1を結合し得る単一特異性四価分子であった。この分子は、4つのrTie−1分子を同時に結合し得るはずである。プラスミドpTE778は、ScFv1−1F11−Fc−ScFv1−1F11(配列番号50)についての遺伝子をコードし、mROR1シグナルペプチドおよびCMV−MIEプロモーターを含む。このタンパク質を、上記の通りに発現させて精製した。
Claims (16)
- (A)x−M−(A’)yを含む融合ポリペプチドであって、ここで、成分Aおよび成分A’は、標的レセプターを結合し得るポリペプチドであり、成分Mは、マルチマー化成分であり、そしてxおよびyは、独立して、1〜10の間の数である、融合ポリペプチド。
- AおよびA’が、各々、前記標的レセプターに特異的な抗体由来の、抗体またはフラグメントである、請求項1に記載の融合ポリペプチド。
- AおよびA’が、同じかまたは異なる、抗体または抗体フラグメントである、請求項2に記載の融合ポリペプチド。
- AおよびA’が、単鎖Fv(ScFv)フラグメントである、請求項3に記載の融合ポリペプチド。
- Aが、前記標的レセプターに特異的な抗体由来の、抗体またはフラグメントであり、そしてA’が、同じ標的レセプターを結合し得るリガンドまたはリガンドフラグメントである、請求項1に記載の融合ポリペプチド。
- Aが、Tieレセプターに対する抗体または抗体フラグメントであり、そしてA’が、Tieレセプターリガンドのフィブリノゲンドメインである、請求項5に記載の融合ポリペプチド。
- 前記TieレセプターがヒトTieレセプターであり、Aがヒト化抗体またはヒト化抗体フラグメントであり、そしてA’がヒトAng−1またはAng−2のフィブリノゲンである、請求項6に記載の融合ポリペプチド。
- AおよびA’が、各々、同じ標的レセプターに結合し得る、リガンドまたはリガンドフラグメントである、請求項1に記載の融合ポリペプチド。
- 前記マルチマー化成分が、IgGのFcドメイン、IgGの重鎖のFcドメイン、ならびに重鎖のCH2定常領域およびCH3定常領域からなる群より選択される、請求項1〜8のいずれか1項に記載の融合ポリペプチド。
- 前記標的レセプターが、活性化されるためにマルチマー化を必要とする、請求項1に記載の融合ポリペプチド。
- 前記標的レセプターが、EPOレセプター、G−CSFレセプター、GM−CSFレセプター、GHレセプター、EGFレセプター、FGFレセプター、VEGFレセプター、PDGFレセプター、Ephファミリーレセプター、TGF−βレセプターファミリー、Tie−1およびTie−2からなる群より選択される、請求項10に記載の融合ポリペプチド。
- 前記融合ポリペプチドの成分が、(A)x−M−(A’)y、(A)x−(A’)y−M、M−(A)x−(A’)y、(A’)y−M−(A)x、(A’)y−(A)x−M、M−(A’)y−(A)x、(A)x−M−(A’)y、(A)x−(A’)y−M、およびM−(A)x−(A’)yからなる群より選択される配置で配置される、請求項1〜11のいずれかに記載の融合ポリペプチド。
- 請求項1〜12のいずれか1項に記載の融合ポリペプチドをコードする、単離された核酸。
- 請求項13に記載の融合ポリペプチドのうちの2つによって形成される、ダイマー。
- 融合ポリペプチドを産生する方法であって、請求項13に記載の核酸を含むベクターでトランスフェクトした宿主細胞を、該宿主細胞からのタンパク質の発現に適切な条件下で培養する工程、およびこのようにして産生された融合タンパク質を回収する工程を包含する、方法。
- 請求項14に記載のダイマーおよび薬学的に受容可能なキャリアを含む、薬学的組成物。
