Abstract
Description
R1は、ハロゲン、C1-C6アルキル、C2-C6アルケニル、C1-C6アルコキシ、C3-C6シクロアルキル、C3-C6シクロアルキルC1-C6アルキルまたはアリールであり、
R2およびR3は、アリールであり、
Halは、フッ素または塩素であり、
mは1または2であり、
nは1または2である]
500mLの4つ口フラスコに、機械的攪拌器、ガラス栓、ゴム弁および不活性ガス流入口を備えた凝縮器を装着した。前記フラスコに、トリス(ジベンジリデンアセトン)ジパラジウム(0)(Pd2(dba)3)(25mg、0.01mol%)、1,1'-ビス-(ジフェニルホスフィノ)フェロセン(dppf)(46mg、0.03mol%)、ナトリウムtert-ブトキシド(37g、1.4eq)、キシレン(90mL)、1-クロロ-3,5-ジフルオロベンゼン(49.2g、0.33mol)、およびベンゾフェノンイミン(50g、0.28mol)を順に加えた。該混合物を8時間還流し、この時点で前記ベンゾフェノンイミンが消費されたことがGC解析により確認された。該反応混合物を若干冷却し、該反応混合物に150mLの2M HClをゆっくりと加え(このとき、いくぶんかの気体の発生が認められた)、さらに該混合物を十分にかき混ぜながら45分間還流した。まだ温かいうちに該反応混合物を分液漏斗に加え、水層を回収した。分液漏斗にさらに90mLのキシレンを加え、つぎに有機層を4×100mLの1M HClにて洗浄した。酸性画分を合わせて、濃水酸化アンモニウム水溶液でアルカリ性にした。遊離アミンを250mLのメチルtert-ブチルエーテルにて抽出した。有機層を硫酸マグネシウムで乾燥し、真空下で濃縮すると、目的生成物が収率89%で得られた。
50mLの3つ口丸底フラスコをオーブン乾燥させ、窒素下で冷却し、該フラスコに14.50gのキシレンを投入した。その後、該フラスコに1,3,5-トリクロロベンゼン(9.760g、53.49mmol)、ベンゾフェノンイミン(7.620g、42.04mmol)、(dba)3Pd2触媒(0.023g、0.03mmol(キシレン2g中))、dppf配位子(0.042g、0.08mmol(キシレン2g中))およびナトリウムt-ブトキシド(5.550g、57.70mmol)を投入した。該反応混合物を3時間還流下に加熱(136℃)し、その後室温まで冷却した。芳香族イミン中間体をその後22mLの2N HCl(水溶液)を用いて70℃で45分間加水分解した。該溶液を室温に冷却すると、ジクロロアニリンの塩酸塩が溶液から沈殿した。粗製の塩酸塩をブフナー漏斗を用いて濾過することにより回収し、その後メチルtert-ブチルエーテルで洗浄した。さらに該ジクロロアニリン塩に30mLのメチルtert-ブチルエーテルを加え、50% NaOHでpHを12に調整した。メチルtert-ブチルエーテル層を回収し、その後真空下でストリッピングすると、3,5-ジクロロアニリンが収率86%で得られた。
50mLの3つ口丸底フラスコをオーブン乾燥させ、窒素下で冷却し、該フラスコに18.6gのキシレンを投入した。その後、該フラスコに2,6-ジクロロトルエン(7.995g、49.65mmol)、ベンゾフェノンイミン(7.612g、42.0mmol)、(dba)3Pd2触媒(0.023g、0.025mmol(キシレン2.27g中))、dppf配位子(0.042g、0.08mmol(キシレン2.53g中))およびナトリウムt-ブトキシド(5.689g、59.19mmol)を投入した。該反応混合物を6時間、125℃に加熱し、その後室温まで冷却した。芳香族イミン中間体をその後65℃にて2N HCl(水溶液)で加水分解し、室温まで冷却し、その後分液漏斗に移した。水層を回収し、有機層をさらに2回、2N HClで抽出した。前記水層をその後20mLのメチルtert-ブチルエーテルと合わせ、pHが11になるように50% NaOH(水溶液)で処理した。該水層をその後20mLのメチルtert-ブチルエーテルでさらに2回抽出した。メチルtert-ブチルエーテル層を合わせて、真空下でストリッピングすると、3-クロロ-2-メチルアニリンが収率75.5%、GC-MSでの純度97.7%にて得られた。
Claims (15)
- 置換されたハロゲン化1-クロロベンゼンから置換されたハロゲン化アニリンを製造する方法であって、
a) 遷移金属触媒錯体および塩基の存在下で、置換されたハロゲン化1-クロロベンゼンをイミンと選択的に反応させ、N-アリールイミンを形成させ、
b) N-アリールイミンを加水分解し、
c) 置換されたハロゲン化アニリンを単離する、
ことを含んでなる方法。 - R1は、ハロゲンまたはC1-C6アルキルであり、m は1であり、n は1または2である、請求項2または3に記載の方法。
- 前記置換されたハロゲン化1-クロロベンゼンが1-クロロ-3,5-ジフルオロベンゼンである、請求項2〜4のいずれか1項に記載の方法。
- 前記置換されたハロゲン化1-クロロベンゼンが1,3,5-トリクロロベンゼンである、請求項2〜4のいずれか1項に記載の方法。
- 前記置換されたハロゲン化1-クロロベンゼンが2,6-ジクロロトルエンである、請求項2〜4のいずれか1項に記載の方法。
- 前記塩基がアルコキシド塩である、請求項1〜7のいずれか1項に記載の方法。
- 前記アルコキシド塩がナトリウムtert-ブトキシドである、請求項8に記載の方法。
- 前記遷移金属触媒錯体が白金、パラジウムまたはニッケル錯体である、請求項1〜9のいずれか1項に記載の方法。
- 前記遷移金属触媒錯体がキレート配位子を含む、請求項10に記載の方法。
- 前記キレート配位子がホスフィンまたはビスホスフィンのアルキルまたはアリール誘導体である、請求項11に記載の方法。
- 前記遷移金属触媒錯体がPd2(dba)3/dppfまたはPd2(dba)3/dppbより選択される、請求項11に記載の方法。
- 前記遷移金属触媒錯体がPd2(dba)3/dppfである、請求項13に記載の方法。
- 前記遷移金属触媒錯体がPd2(dba)3/dppbである、請求項13に記載の方法。
