Abstract
Description
インプラント可能な薬物送達系に使用される懸濁ベヒクルの性質のために、懸濁配合物バルクの混合、およびその後のインプラント系への懸濁配合物のローディングはしばしば高温で行われる。高温で種々の懸濁ベヒクル中に懸濁されたPACAP類似体(または単に“PACAP”)の安定性を評価するために、凍結乾燥したPACAPを4種の異なる懸濁ベヒクル(LL/GML/PVP、BA/PVP、EHL/PVP、およびPEG/PVP)に懸濁した。PACAPを懸濁ベヒクルに懸濁するために、3.3mg PACAPアセテートを異なる懸濁液のそれぞれに手動で混合し、PACAP含量約3%とした。次に、PACAP懸濁配合物の安定性を65℃で4時間インキュベーション後に評価した。製造されたPACAP懸濁液の安定性は、RP−HPLCおよびSECにより評価し、テーブル1(図10)を参照することにより理解できるように、結果は、4時間のインキュベーション期間中PACAPが安定で、したがって、一般に懸濁配合物製造工程に伴う温度条件に耐えられるはずであることを示した。
プロセッシング(processing)研究の予備的な所見が、PACAP凍結乾燥物および懸濁液に対する糖(ショ糖)および非イオン性界面活性剤(Tween 80)の安定化作用評価のきっかけとなった。評価を行うために、3種のレベルのTween 80(0、0.05、0.2wt%)およびショ糖(0、0.5/1、1/1、w/w)において、10mM NH4OAc(pH6.4)中のPACAPにより試料を作製した。10mM ヒスチジン(pH6.4)およびコハク酸ナトリウム(pH6.4)中のPACAP溶液も調製した。PACAP溶液のそれぞれを凍結乾燥し、再構成した固体PACAP粒子を得て、個々のバイアルにそれぞれの溶液由来の再構成材料3.3mgの試料を入れ、試料は以下のものを含有した:(1)添加剤無し、(2)0.2wt% Tween 80、(3)0.2wt% Tween 80+0.5:1、w/w ショ糖、および(4)1:1 w/w ショ糖。種々の3.3mg 試料をLL/GML/PVPおよびBA/PVPと手動で混合し、PACAP含量約3%を有する懸濁配合物を提供し、懸濁配合物は65℃で4時間インキュベーションした。初めのインキュベーション期間後、種々の懸濁配合物を以下に記載のように貯蔵し、RP−HPLCおよびSECにより評価した。
固体PACAP粒子は、NH4OAc緩衝剤、pH6で作製された3種の異なるポリペプチド溶液から凍結乾燥工程(FTS Duro stop)により製造された。3種の異なる溶液は、それぞれ異なる量のショ糖を含有し、第1はショ糖を含まず、第2はショ糖対PACAPの重量比0.5/1で含有し、そして第3はショ糖対PACAPの重量比1/1で含有した。3種の溶液のそれぞれから製造された固体PACAP粒子は2℃〜8℃の範囲の制御温度、ほぼ生理的温度(40℃)、および生理的温度を超える温度(60℃)で貯蔵した。それぞれの群の固体PACAP粒子内に含有されるPACAPの安定性は、24日間および3ヶ月間において評価された。それぞれの粒子群内のPACAP安定性を評価するために、粒子は水で再構成し、逆相RP−HPLC(0.1% トリフルオロ酢酸を含有する移動相によるアセトニトリルグラジエント溶出、および214nmでのUV検出)およびサイズ排除HPLC(SEC、アイソクラティック溶出、0.1% トリフルオロ酢酸および200mM 塩化ナトリウムを含有する70%水溶液と混合した30% アセトニトリル、220nmでのUV検出)により分析した。
長期にわたるPACAP分子の安定性に対するpHの効果を評価した。そのような評価を行うために、PACAP原料は初めに3種の異なる溶液、pH2を有する第1の溶液、pH4を有する第2の溶液、そしてpH6を有する第3の溶液において水に溶かした。それぞれの溶液のpHは希HClを使用して所望する値に調整した。3種の溶液のそれぞれを凍結乾燥バイアルに入れ、固体PACAP粒子は本明細書にすでに記載の凍結乾燥工程により、3種の溶液のそれぞれから製造した。次に3種の溶液のそれぞれ由来の粒子を60℃で2ヶ月間貯蔵し、その後固体粒子に含有されるPACAPの安定性をRP−HPLCおよびSECを使用して評価した。図9は2ヶ月間の安定性の結果を示し、そしてそれは酸性pHで製造された固体PACAP粒子の著しく改善された安定性を証明する。
ある量の潜在的な安定化糖と共に、種々のpHで製造された固体PACAP粒子に包含されるPACAPの安定性を評価した。それぞれの例において、固体PACAP粒子は3種の異なる糖:ショ糖、トレハロース、およびメチルマンノピラノシド(すべてSigma製)の中の1種の7.3モル当量から構成された。初めの2種の水溶液は、2に調整されたpHを有する第1の溶液、および6に調整されたpHを有する第2の溶液にPACAPを溶解することにより作製した(先のように、この実施例に記載の種々の溶液のpHは希HClを使用して調整した)。第3および第4の水溶液は、2に調整されたpHを有する溶液、および6に調整されたpHを有する溶液にPACAPおよびメチル‐MPを共に溶解することにより作製した。第5および第6の水溶液は、2に調整されたpHを有する溶液、および6に調整されたpHを有する溶液にPACAPおよびトレハロースを共に溶解することにより作製した。第7および第8の水溶液は、2に調整されたpHを有する溶液、および6に調整されたpHを有する溶液にPACAPおよびショ糖を共に溶解することにより作製した。
ヒスチジン緩衝剤(Sigma)を含有するPACAP粒子、ならびに塩化カルシウム(CaCl2、JT Baker)、ショ糖およびドデシル硫酸ナトリウム(SDS、Piece)と組み合わせたヒスチジン緩衝剤を含有するPACAP粒子を製造し、そのような粒子によって提供されるPACAPの安定化を評価した。評価するPACAP粒子を製造するために、6種の異なる水溶液を作製した。それぞれの溶液はその中に溶解した所望する量のPACAPを含有し、6種の溶液それぞれのpHは希HClの添加により6に調整した。初めの水溶液は添加剤(ヒスチジン緩衝剤、CaCl2、ショ糖またはSDS)なしに作製した。第2の水溶液は10mM濃度のヒスチジン緩衝剤と共に作製した。第3の水溶液は10mM濃度のヒスチジン緩衝剤および10mM濃度のCaCl2と共に作製した。第4の水溶液は10mM濃度のヒスチジン緩衝剤と共に作製し、PACAPに関して0.5/1重量比(ショ糖/PACAP)でショ糖を含有した。第5の水溶液は10mM濃度のヒスチジン緩衝剤、10mM濃度のCaCl2、およびPACAPに関して0.5/1重量比(ショ糖/PACAP)を提供する量のショ糖と共に作製した。第6の水溶液は、10mM濃度のヒスチジン緩衝剤および0.02wt% SDSと共に作製した。次にすでに本明細書に記載の凍結乾燥工程を使用して、固体PACAP粒子を6種の溶液のそれぞれから製造した。その後、それぞれの溶液から製造したPACAP粒子は60℃で2ヶ月間貯蔵し、そのような貯蔵後に種々の異なる粒子内に含有されるPACAPの安定性をRP−HPLCおよびSECを使用して評価した。テーブル7から分かるように、ヒスチジン緩衝剤の存在はPACAP凝集物形成を著しく抑制する(凝集物形成はおよそ1/6に減少)。その上、ショ糖、CaCl2およびSDSはさらにペプチドを安定化し、凝集物形成をおよそ1/14〜1/20に減少させた。
酸性およびほぼ中性pHで製造されたPACAP粒子におけるCaCl2の安定化作用を評価するために、4種の異なる型の粒子を配合物し、60℃で2ヶ月間貯蔵し、その後PACAP分解を分析した。4種の異なる粒子配合物は4種の異なる水溶液から製造した。4種の溶液のそれぞれはその中に溶解した所望する量のPACAPを含有し、それぞれの溶液のpHは希HClを使用して所望するレベルに調整した。初めの水溶液は添加剤(ヒスチジンまたはCaCl2)なしに作製し、初めの溶液のpHは2に調整した。第2の水溶液は10mM濃度のCaCl2と共に作製し、第2の溶液のpHは2に調整した。第3の水溶液は10mM濃度のヒスチジン緩衝剤と共に作製し、第4の水溶液は10mM濃度のヒスチジン緩衝剤および10mM濃度のCaCl2と共に作製した。固体PACAP粒子は、すでに本明細書に記載の凍結乾燥工程を使用して、4種の水溶液のそれぞれから製造し(異なる粒子のそれぞれの組成はテーブル8に概説する)、それぞれの溶液から製造されたPACAP粒子は60℃で2ヶ月間貯蔵した。60℃で2ヶ月間貯蔵後の粒子に含有されるPACAPの安定性は、RP−HPLCおよびSECを使用して評価した。テーブル8から分かるように、そのような評価の結果は、CaCl2がpH2および6の両方におけるPACAP粒子の安定性をさらに改善することを証明した(テーブル8)。
Claims (35)
- ポリペプチド;ならびに、金属イオンおよび酸性条件下において生理的条件までまたはそれを超える温度で安定である糖からなる群から選択される安定化物質;を含む安定化されたポリペプチド粒子であって、酸性の再構成pHを示すように配合されているポリペプチド粒子。
- 該ポリペプチド粒子が、ポリペプチド、金属イオンおよび、生理的条件まで、およびそれらを超える温度において酸性pHで安定である糖を含む、請求項1に記載のポリペプチド粒子。
- 該安定化物質が、生理的条件までおよびそれらを超える温度において酸性条件下で安定である二糖および単糖から選択される、請求項1に記載のポリペプチド粒子。
- 該安定化物質が、トレハロースおよびメチル‐マンノピラノシドから選択される、請求項3に記載のポリペプチド粒子。
- 該安定化物質が、酸性条件下において生理的条件までおよびそれを超える温度で安定である二糖および単糖から選択される安定化物質を含み、そして安定化物質対ポリペプチドのwt/wt比、約0.1/1〜約1/1の間の範囲で安定化物質がポリペプチド粒子中に含まれる、請求項1に記載のポリペプチド粒子。
- 該安定化物質が、酸性条件下において生理的条件までおよびそれを超える温度で安定である二糖および単糖から選択される安定化物質を含み、そして安定化物質対ポリペプチドのwt/wt比、約0.1/1〜約0.5/1の間の範囲で安定化物質がポリペプチド粒子中に含まれる、請求項1に記載のポリペプチド粒子。
- 該安定化物質が、酸性条件下において生理的条件までおよびそれらを超える温度で安定である二糖および単糖から選択される安定化物質を含み、そして安定化物質対ポリペプチドのwt/wt比、約0.1/1〜約0.25/1の間の範囲で安定化物質がポリペプチド粒子中に含まれる、請求項1に記載のポリペプチド粒子。
- 該安定化物質が、二価金属イオン塩に由来する金属イオンを含む、請求項1に記載のポリペプチド粒子。
- 該安定化物質が、CaCl2、MgCl2、およびZnCl2からなる群から選択される二価金属イオン塩に由来する金属イオンを含む、請求項1に記載のポリペプチド粒子。
- 該安定化物質が金属イオンを含み、ポリペプチド粒子に含有される安定化物質対ポリペプチドのモル比が約1/1〜約10/1の範囲である、請求項1に記載のポリペプチド粒子。
- 該安定化物質が金属イオンを含み、ポリペプチド粒子に含有される安定化物質対ポリペプチドのモル比が約2/1〜約6/1の範囲である、請求項1に記載のポリペプチド粒子。
- 該安定化物質が金属イオンを含み、ポリペプチド粒子に含まれる安定化物質対ポリペプチドのモル比が約4/1である、請求項1に記載のポリペプチド粒子。
- 該ポリペプチドが下垂体アデニル酸シクラーゼポリペプチド/グルカゴンスーパーファミリーから選択される、請求項1に記載のポリペプチド粒子。
- 該ポリペプチドが下垂体アデニル酸シクラーゼポリペプチド、グルカゴン、グルカゴン様ペプチド、成長ホルモン放出因子、血管作用性腸ポリペプチド、ペプチドヒスチジンメチオニン、セクレチン、およびグルコース‐依存的インスリン刺激ポリペプチドからなる群から選択される、請求項1に記載のポリペプチド粒子。
- 下垂体アデニル酸シクラーゼポリペプチドならびに、トレハロースおよびメチル‐マンノピラノシドから選択される安定化糖を含む安定化されたポリペプチド粒子であって、該ポリペプチド粒子に含有される安定化糖対下垂体アデニル酸シクラーゼポリペプチドのwt/wt比が約0.55/1であり、そしてポリペプチド粒子が酸性の再構成pHを示すように配合される、前記ポリペプチド粒子。
- 下垂体アデニル酸シクラーゼポリペプチド、ならびにCa2+、Mg2+、およびZn2+から選択される安定化金属イオンを含む安定化されたポリペプチド粒子であって、該ポリペプチド粒子に含有される金属イオン対下垂体アデニル酸シクラーゼポリペプチドのモル比が約4/1であり、そしてポリペプチド粒子が酸性の再構成pHを示すように配合される、前記ポリペプチド粒子。
- ポリペプチド;ならびに、金属イオン、界面活性剤、緩衝剤、および生理的条件までおよびそれを超える温度においてほぼ中性のpH環境中で安定である糖からなる群から選択される2以上の安定化物質;を含むポリペプチド粒子であって、ほぼ中性の再構成pHを示すように配合されているポリペプチド粒子。
- 該ポリペプチドが下垂体アデニル酸シクラーゼポリペプチド/グルカゴンスーパーファミリーから選択される、請求項18に記載のポリペプチド粒子。
- 該ポリペプチドが下垂体アデニル酸シクラーゼポリペプチド、グルカゴン、グルカゴン様ペプチド、成長ホルモン放出因子、血管作用性腸ポリペプチド、ペプチドヒスチジンメチオニン、セクレチン、およびグルコース‐依存的インスリン刺激ポリペプチドからなる群から選択される、請求項18に記載のポリペプチド粒子。
- 該ポリペプチド粒子が安定化糖ならびにアミノ酸緩衝剤、ペプチド緩衝剤および無機緩衝剤から選択される緩衝剤を含む、請求項18に記載のポリペプチド粒子。
- 安定化糖対ポリペプチドのwt/wt比が約0.25/1〜約1/1の間の範囲である、請求項21に記載のポリペプチド粒子。
- 該ポリペプチド粒子がアミノ酸緩衝剤およびペプチド緩衝剤から選択される緩衝剤ならびに安定化金属イオンを含む、請求項18に記載のポリペプチド粒子。
- 金属イオン対ポリペプチドのモル比が約2/1〜約10/1の間の範囲である、請求項23に記載のポリペプチド粒子。
- 該ポリペプチド粒子がアミノ酸緩衝剤およびペプチド緩衝剤から選択される緩衝剤ならびに界面活性剤を含む、請求項18に記載のポリペプチド粒子。
- 該界面活性剤がポリペプチド粒子の約0.02wt%〜約0.2wt%を占める、請求項25に記載のポリペプチド粒子。
- 該ポリペプチド粒子がアミノ酸緩衝剤およびペプチド緩衝剤から選択される緩衝剤、界面活性剤、金属イオンならびに安定化糖を含む、請求項18に記載のポリペプチド粒子。
- 該界面活性剤がドデシル硫酸ナトリウムを含む、請求項18に記載のポリペプチド粒子。
- 該界面活性剤がドデシル硫酸ナトリウムを含み、ポリペプチド粒子の約0.02wt%〜約0.2wt%を占める、請求項25に記載のポリペプチド粒子。
- 該ポリペプチドが下垂体アデニル酸シクラーゼポリペプチドであり、そして該ポリペプチド粒子が、アミノ酸緩衝剤およびペプチド緩衝剤から選択される緩衝剤、ならびに約0.25/1〜約1/1の間の範囲の安定化糖対ポリペプチドのwt/wt比で含まれる安定化糖、約2/1〜約10/1の範囲の金属イオン対ポリペプチドのモル比で含有される金属イオン、および、該ポリペプチド粒子の約0.02wt%〜約0.2wt%を占める界面活性剤からなる群から選択される1以上の付加的な安定化物質を含む、請求項18に記載のポリペプチド粒子。
- 酸性条件下で安定であるポリペプチドを含む安定化されたポリペプチド粒子であって、酸性の再構成pHを示すように配合される、前記ポリペプチド粒子。
- 安定化されたポリペプチド粒子がpH5許容の再構成pHを示すように配合される、請求項30に記載の安定化されたポリペプチド粒子。
- 安定化されたポリペプチド粒子が約pH2〜pH4の再構成pHを示すように配合される、請求項30に記載の安定化されたポリペプチド粒子。
- 該ポリペプチドが下垂体アデニル酸シクラーゼポリペプチド、グルカゴン、グルカゴン様ペプチド、成長ホルモン放出因子、血管作用性腸ポリペプチド、ペプチドヒスチジンメチオニン、セクレチン、およびグルコース‐依存的インスリン刺激ポリペプチドからなる群から選択されるポリペプチドを含み、そして該粒子がpH5より低い再構成pHを示すように配合される、請求項30に記載の安定化されたポリペプチド粒子。
- 該ポリペプチドが下垂体アデニル酸シクラーゼポリペプチド、グルカゴン、グルカゴン様ペプチド、成長ホルモン放出因子、血管作用性腸ポリペプチド、ペプチドヒスチジンメチオニン、セクレチン、およびグルコース‐依存的インスリン刺激ポリペプチドからなる群から選択されるポリペプチドを含み、そして該粒子がpH2〜pH4の再構成pHを示すように配合される、請求項30に記載の安定化されたポリペプチド粒子。
- 該ポリペプチドが下垂体アデニル酸シクラーゼポリペプチド類似体を含み、そして初期下垂体アデニル酸シクラーゼポリペプチド類似体の90%以上の回収率を可能にし、60℃で2ヶ月間保存後の二量体形成が2%未満であるように粒子が配合される、請求項30に記載の安定化されたポリペプチド粒子。
