で表される光学活性金属錯体触媒、および塩基の存在下で反応させて光学活性β―ニトロアルコール類を製造する。

一般式(c):
で表される光学活性金属錯体触媒、および塩基の存在下で反応を行うことを特徴とする光学活性β―ニトロアルコールの製造方法。
前記ニトロアルカン類が一般式(b):
以上の反応によって得られる反応混合物から、目的の光学活性ニトロアルコール類の回収、精製は、公知の方法、例えば、蒸留、吸着による方法、抽出、再結晶等の方法を組み合わせて行うことができる。
(1R)−1−(2−ナフチル)−2−ニトロエタノールの合成:
反応容器に、窒素雰囲気下、N,N'−ビス[2−(2,4,6−トリメチルベンゾイル)−3−オキソブチリデン]−(1S,2S)−ビス(3,5−ジメチルフェニル)エチレン−1,2−ジアミナトコバルト(II)錯体(7.53 mg, 0.01 mmol, 2 mol%;式(c−4))のジクロロメタン溶液(2.0 mL)を室温下で加え、-70℃に降温した後に、2−ナフトアルデヒド(78.1 mg, 0.5 mmol)のジクロロメタン溶液(1.0 mL)、ニトロメタン(1.0 mL, 18.5 mmol)とジクロロメタン(1.0 mL)の混合溶液、ジイソプロピルエチルアミン(64.7 mg, 0.5 mmol)を順に加えた。この反応混合物を76時間撹拌した後、含水THFを加えて反応を停止させた。通常の後処理を行った後、得られた粗生成物をシリカゲルカラムクロマトグラフィー(ヘキサン:酢酸エチル=4:1)で分離精製したところ、1−(2−ナフチル)−2−ニトロエタノールが得られた(89.7 mg, 収率 83%)。この生成物の光学純度を高速液体クロマトグラフィー(光学活性化カラム:ダイセル化学工業社製、Chiralpak AD-H; EtOH 10

<実施例2、3>
反応温度を-70℃から表1に示す温度で行った以外は、実施例1と同様にして反応を行った。反応時間、得られた(1R)−1−(2−ナフチル)−2−ニトロエタノールの収率、光学純度を表1に示す。
塩基としてジイソプロピルエチルアミンの代わりに、表2に示すアミン化合物を使用した以外は実施例3と同様にして反応を行った。反応時間、得られた(1R)−1−(2−ナフチル)−2−ニトロエタノールの収率、光学純度を表2に示す。
(1R)−1−(2−クロロフェニル)−2−ニトロエタノールの合成:
2−ナフトアルデヒドの代わりに2−クロロベンズアルデヒドを使用し、塩基として用いるジイソプロピルエチルアミンの、アルデヒドに対する添加量(倍モル)を表3に示した以外は実施例2と同様にして反応を行った。反応時間、得られた(1R)−1−(2−クロロフェニル)−2−ニトロエタノールの収率、光学純度を表3に示す。
溶媒としてジクロロメタンの代わりにアセトンを使用し、反応温度を-70℃とした以外は、実施例11と同様にして反応を行った。その結果、反応は65時間で完結し、(1R)−1−(2−クロロフェニル)−2−ニトロエタノールが定量的に得られた。光学純度は91%eeであった。
各種光学活性β−ニトロアルコールの合成:
2−ナフトアルデヒドの代わりに、表4に示すアルデヒド化合物を使用し、該アルデヒドに対する触媒量を5モル%に、またジイソプロピルエチルアミンの添加量を2.5倍モルとし、表4に示す反応温度とした以外は実施例1と同様にして反応を行った。反応時間、得られた光学活性β−ニトロアルコール{R1−C*H(OH)−CH2NO2}の収率、光学純度を表4に示す。
2−ナフトアルデヒドの代わりにケイ皮アルデヒドを使用し、反応温度を-65℃とした以外は、実施例1と同様にして反応を行った。その結果、反応は90時間で完結し、対応する光学活性β−ニトロアルコールが収率72%で得られた。光学純度は79%eeであった。
前記光学活性金属錯体触媒(c−4)の代わりに、表5に示す(S,S)の絶対立体配置を有する前記光学活性コバルト(II)錯体触媒または前記光学活性コバルト(III)錯体触媒を使用した以外は実施例2と同様にして反応を行った。各実施例における反応温度、反応時間と共に、得られた(1R)−1−(2−ナフチル)−2−ニトロエタノールの収率、光学純度を表5に示す。
光学活性金属錯体触媒として、表6に示す(S,S)の絶対立体配置を有する前記光学活性コバルト(II)錯体触媒を使用し、反応温度を-65℃とした以外は実施例11と同様にして反応を行った。反応時間、得られた(1R)−1−(2−クロロフェニル)−2−ニトロエタノールの収率、光学純度を表6に示す。
前記光学活性金属錯体触媒(c−4)の代わりに、表7に示す(S,S)の絶対立体配置を有する前記光学活性コバルト(III)錯体触媒を使用し、塩基として用いるジイソプロピルエチルアミンの添加量をアルデヒドに対して3倍モルとし、反応温度を-70℃とした以外は実施例11と同様に反応を行った。各実施例における反応時間と共に、(1R)−1−(2−クロロフェニル)−2−ニトロエタノールの収率、光学純度を表7に示す。
前記光学活性金属錯体触媒(c−4)の代わりに、(S,S)の絶対立体配置を有する前記光学活性コバルト(II)錯体触媒(c−7)(2 mol%)を使用し、反応温度を-78℃とした以外は実施例11と同様にして反応を行った。95時間後に反応を停止させたところ、(1R)−1−(2−クロロフェニル)−2−ニトロエタノールが定量的に得られた。光学純度は95%eeであった。
2−クロロベンズアルデヒドの代わりに、表8に示すアルデヒド化合物を使用した以外は実施例35と同様にして反応を行った。反応時間、得られた光学活性β−ニトロアルコールの収率、光学純度を表8に示す。
前記光学活性金属錯体触媒(c−4)の代わりに、(S,S)の絶対立体配置を有する前記光学活性コバルト(II)錯体触媒(c−8)(2 mol%)を使用し、反応温度を-78℃とした以外は実施例1と同様にして反応を行った。47時間後に反応を停止させたところ、(1R)−1−(2−ナフチル)−2−ニトロエタノールが収率12%で得られた。光学純度は51%eeであった。
2−ナフトアルデヒドの代わりに、表9に示すアルデヒド化合物を使用した以外は実施例38と同様にして反応を行った。反応時間、得られた光学活性β−ニトロアルコールの収率、光学純度を表9に示す。
Claims (6)
- アルデヒド類とニトロアルカン類とを不斉触媒の存在下にニトロアルドール反応により光学活性β―ニトロアルコール類を製造する方法において、
一般式(c):
で表される光学活性金属錯体触媒、および塩基の存在下で反応を行うことを特徴とする光学活性β―ニトロアルコールの製造方法。 - 前記一般式(c)における炭化水素基X*が、一般式(x−1)、(x−2)、もしくは(x−3):
のいずれかで表される炭化水素基である請求項1または2記載の製造方法。 - 前記一般式(c)における金属イオンMが、コバルト(II)またはコバルト(III)のいずれかである請求項1〜3のいずれかに記載の製造方法。
- 前記塩基が、1級アミン、2級アミンおよび3級アミンからなる群より選ばれる化合物であることを特徴とする請求項1〜4のいずれかに記載の製造方法。
