Abstract

【課題手段】 配列番号:1のアミノ酸配列を有する60アミノ酸のホメオボックスドメインを含むポリペプチドであって、かつヒトの成長に対する制御活性を有するポリペプチドをコードする単離されたヒト核酸分子、該核酸分子又はそれによりコードされるタンパク質を含有してなる医薬組成物。

Description
a)配列番号:1のアミノ酸配列を有する60アミノ酸のホメオボックスドメインを含有するポリペプチドであって、かつヒト成長に対する制御活性を有するポリペプチドをコードする単離されたヒト核酸分子。
b)本質的に図2、図3または図4に示された核酸配列、とりわけ配列番号:10、配列番号:12または配列番号:15に示された核酸配列を含む単離されたDNA分子。
c)b)のDNA分子にハイブリダイズしうるDNA分子。
d)60〜70℃の温度で、かつ標準緩衝液の存在下に、2.のDNA分子とのハイブリダイゼーションが可能な前記c)のDNA分子。
e)配列番号:10、配列番号:12または配列番号:15のヌクレオチド配列と70%以上の相同性を有し、かつヒト成長に対する制御活性を有するポリペプチドをコードするヌクレオチド配列を含有するDNA分子。
f)配列番号11、13もしくは16のアミノ酸配列を有するヒト成長タンパク質またはその機能的断片。
g)f)のヒト成長タンパク質またはその抗原性改変体での動物の免疫により得られた抗体。
h)ヒト成長タンパク質またはその機能的断片を含有する、ヒト成長遺伝子の遺伝子変異により引き起こされた疾患を治療するための医薬組成物。
i)物質による宿主細胞の処置に応答するDNA分子の発現レベルにおける増大を測定するために、a)〜e)に記載されたDNA分子のいずれかにハイブリダイズするメッセンジャーRNAを検出する工程を含む、前記h)の疾患の治療に有用な物質のスクリーニング方法。
j)DNA分子を哺乳類細胞において発現させることが可能である、前記a)〜e)記載の核酸分子のいずれかを含む発現ベクターまたはプラスミド。
k)体の組織または体液の生物学的試料中における、低身長の原因となる1若しくは複数の遺伝子の決定方法。
研究した全6人の患者はデ・ノボでの性染色体異常を有した。
CCは、核型45,X/46,X psu dic (X)(Xqter→Xp22.3::Xp22.3 → Xqter)の女子である。6歳半の年齢の最後の検査で、彼女の身長は114cmであった〔25−50%百分位数〕。彼女の母親の身長は155cmであり、父親は解析の求めに応じなかった。詳細は、ヘンケ(Henke)ら、1991を参照のこと。
SHOXaの変異について、特発性の低身長を患った250個体を試験した。該患者は、下記特徴で選抜した:暦年齢の身長は、国民身長平均の3番目の百分位数より下であり、マイナス2の標準偏差(SDS)であった;特に原因となる疾患は知られなかった:妊娠年齢として正常な体重(身長)、正常な体型、慢性器官疾患無し、正常な食事量、精神医学的疾患無し、骨形成不全無し、甲状腺または成長ホルモン欠陥無し。
症例1および2は、ドイツ人の血を分けていない家族の低身長の子供である。男子(症例1)は帝王切開により妊娠第38週目に生まれた。誕生時の体重は、2660gであり、誕生時の身長は47cmであった。彼は、普通より劣る成長を除いて正常に発育した。6.4歳の年齢での検査で、彼は、比例して小さく(106.8cm,−2.6SDS)、肥満(22.7kg)であったが、他の点では正常であった。彼の骨の年齢は遅延しておらず(6歳)、X線解析により骨形成不全が除外された。IGF−IおよびIGFBP−3レベルならびに血清中の甲状腺パラメーターはGHまたは甲状腺ホルモン欠陥がありそうもなかった。女子(症例2)は、帝王切開により出産予定日に生まれた。誕生時の体重は2920gであり、誕生時の身長は47cmであった。彼女の発育の時点は正常であったが、12か月の年齢までに成長不良が明らかとなった(身長:67cm、−3.0SDS)。4歳のとき、彼女は89.6cmの身長であった(−3.6SDS)。形態不全の特徴すなわち体型不全は明らかではなかった。彼女は、肥満ではなかった(13kg)。彼女の骨年齢は3.5歳であり、骨形成不全は排除された。ホルモンパラメーターは正常であった。該女子と該男子の両方はターナー症候群の女性の50百分位数の成長曲線で成長した。母親は、ファミリーのなかで最も小さく、軽い四肢体型不全を有する(142.3cm、−3.8SDS)。彼女の2人の姉妹の1人(150cm、−2.5SDS)および母方の祖母(153cm、−2.0SDS)は、いかなる体型不全をももたずに全員低かった。一方の姉妹は、正常な身長を有する(167cm、+0.4SDS)。父親の身長は166cm(−1.8SDS)であり、母方の祖父の身長は165cm(−1.9SDS)であった。他の患者は生まれが日系人であり、同一の変異を示した。
A.イン・サイチュハイブリダイゼーション
a)蛍光イン・サイチュハイブリダイゼーション(FISH)
Xp/Yp偽常染色体領域(PAR1)に存在するコスミドを用いる蛍光イン・サイチュハイブリダイゼーション(FISH)を行った。コスミド64/75cos(LLNLc110H032)、E22cos(2e2)、F1/14cos(110A7)、M1/70cos(110E3)、P99F2cos(43C11)、P99cos(LLNLc110P2410)、B6cosb(1CRFc104H0425)、F20cos(34F5)、F21cos(ICRFc104G0411)、F3cos2(9E3)、F3cos1(11E6)、P117cos(29B11)、P6cos1(ICRFc104P0117)、P6cos2(LLNLc110E0625)およびE4cos(15G7)を用いたFISH研究を公開された方法〔リヒター(Lichter)およびクレマー(Cremer),1992〕に従って行った。簡単には、それぞれのコスミドクローン1マイクログラムをビオチンで標識し、反復性DNA配列からのシグナルを抑制する条件下にヒト分裂中期の染色体とハイブリダイズさせる。ハイブリダイゼーションシグナルの検出はFITC共役アビジンを介した。FITCのイメージを冷却電荷共役装置カメラシステム(cooled charge coupled device camera system)〔Photometrics,Tucson,AZ)を用いて撮影された。
コスミドはローレンス・リバーモア・ナショナル・ラボラトリー(Lawrence Livermore National Laboratory)X−およびY−染色体ライブラリーならびにインペリアル・キャンサー・リサーチ・フンド・ロンドン(Imperial Cancer Research Fund London)〔現マックス・プランク・インスティチュート・フォア・モレキュラー・ジェネティックス・ベルリン(Max Planck Institute for Molecular Genetics Berlin)X染色体ライブラリーから得られた。DXYS15に遠位のコスミド、すなわち、E4cos、P6cos2、P6cos1、P117cosおよびF3cos1を用いて、2コピーのE4cos、P6cos2、P6cos1が、まだ存在し、1コピーのP117cosおよびF3cos1が存在する。AKとSSの両方の患者のブレークポイントは、コスミドP6cos1上にお互いから10kbの最大物理距離で位置づけした。AKとSSの異常なX染色体が、約630kbのDNAを欠失したことを結論づけた。
c)サザンブロットハイブリダイゼーション
ビオチン化コスミドDNA(挿入断片サイズ32−45kb)またはコスミド断片(10−16kb)を以前に記載された(リヒター(Lichter)およびクレマー(Cremer),1992)ような条件下に患者の刺激されたリンパ球由来の分裂中期染色体とハイブリダイズさせた。ハイブリダイズしたプローブは、アビジン共役FITCを介して検出された。
全てのPCRは、100pg〜200ng鋳型、20pmolの各プライマー、200μM dNTP(Pharmacia)、1.5mM MgCl2 、75mM Tris/HCl pH9、20mM (NH4 )2 SO4 、0.01%(w/v) Tween20および2UのGoldstar DNAポリメラーゼ(Eurogentec)を含む50μl容量中で行った。サーマルサイクルはThermocycler GeneE(Techne)で行った。
コスミド整列群由来の各々4〜5クローンからなる4つのコスミドプールをエキソン増幅実験に用いた。各コスミドプール中のコスミドはSau3Aで部分的に切断した。4〜10kbのサイズ範囲のゲル精製画分をBamHI切断pSPL3Bベクター〔バーン(Burn)ら、1995〕にクローン化し、以前に記載されたように〔チャーチ(Church)ら,1994〕、エキソン増幅実験に用いた。
2つのコスミドLLOYNCO3’M’15D10およびLLOYNCO3’M’34F5の音波処理断片を別々にM13mp18ベクターにサブクローニングした。各コスミドライブラリーから、少なくとも1000プラークを取り、M13DNAを調製し、ダイ・ターミネーター、ThermoSequenase(Amersham)およびユニバーサルM13−プライマー(MWG−BioTech)を用いてシークエンスした。ゲルをABI−377シークエンサーで流し、データーを集め、GAP4プログラム(Staden)で編集した。

〔(+)〕ブレークポイント領域;ブレークポイントがFISHにより示 されたコスミド内に生じることを示す。
最小170kb重要領域内の転写ユニットを探索するために、6種類のコスミド( 110E3、F2cos、43C11、P2410、15D10、34F5) のエキソントラッピングとcDNA選択を行った。エキソントラッピングにより、3種類の陽性クローン( ET93、ET45、およびG108) を単離したが、これらのものはいずれもコスミド34F5に逆マッピングされた。cDNA選択プロトコルと25種類の過剰cDNAライブラリーを用いた過去の研究は不成功に終わったことから、この部分に存在する遺伝子は、発現頻度が非常に低いことが示唆された。
全3つのエキソンクローンET93、ET45およびG108は同一の遺伝子の一部であると仮定し、これらをまとめてプローブとして用い、胎児組織(肺、肝臓、脳1および2)ならびに成人組織(卵巣、胎盤1および2、繊維芽細胞、骨格筋、骨髄、脳、脳幹、視床下部、下垂体)の12種類から14種類のcDNAライブラリーをスクリーニングした。約1,400万個のプレート培養クローンからはただ一つの単一クローンも検出されなかった。全長転写物を単離するために、3’および5’RACEを行った。3’RACEについては、G108が発現することが示されている組織の胎盤、骨格筋、および骨髄繊維芽細胞由来のRNA上でエキソンG108由来プライマーを使用した。これら3 種類の組織すべてから1173bpと652bpの2 種類の3 ’RACEクローンが得られ、2 種類の3 ’エキソンaとbが存在することが示唆された。該2 種類のエキソン型をSHOXaおよびSHOXbと命名した。
RT−PCRとcDNAライブラリーの構築
心臓、膵臓、胎盤、骨格筋、胎児腎臓および肝臓のヒトポリA+ RNAをClontech社から購入した。製造業者により記載されたように、TRIZOL試薬(Gibco-BRL 社)を用いて、骨髄繊維芽細胞株から全RNAを単離した。オリゴ(dT)- アダプタープライマー(GGCCACGCGTCGACTAGTAC[ dT] 20Nを用い、スーパースクリプト第一鎖cDNA合成キット(Gibco-BRL 社)により、100ngのポリA+ RNAまたは10μgの全RNAから第一鎖cDNA合成を行った。第一鎖cDNA合成終了後、反応混合物を10倍に希釈した。この希釈液5 μlずつをさらにPCR実験に供した。
5’および3’RACE由来クローンの配列を分析することにより、SHOXaおよびSHOXb(1349および1870bp)のコンセンサス配列を決定した。1870bp(SHOXa)および1349bp(SHOXb)の単純オープンリーディングフレームが同定され、292アミノ酸(SHOXa)および225アミノ酸(SHOXb)の2つのタンパク質が見つかった。これらaとbの転写物の両方は共通の5 ’末端を持っていたが、異なる最後の3’エキソンを有し、知見は、択一的スプライシングシグナルの利用可能性を示唆する。2つのcDNAとコスミドLL0YNCO3”M”15D10およびLL0YNC3”M”34F5由来のシークエンスしたゲノムDNAとの完全なアラインメントを行い、エキソン−イントロン構造を確定することができた(図4 )。この遺伝子は、58bp(エキソンIII)から1146bp(エキソンVa)のサイズにわたる6 つのエキソンから構成される。エキソンIはCpG島と開始コドンと5 ’領域を含む。終止コドンならびに3’非コード領域は、択一的スプライシングエキソンVaとVbのそれぞれの中に局在される。
単一のエキソンをハイブリダイゼーションプローブとして用いるノーザンブロット分析で、各エキソンごとに異なる発現プロファィルが明らかとなり、異なるサイズと強度のバンドは、GとCの多い他の遺伝子配列とのクロス−ハイブリダイゼーション産物であることをしめすことが示唆される。SHOXaとbの両遺伝子のより詳細な発現プロファィルを達成するために、異なる組織に由来するRNAにつき、RT−PCR実験を行った。骨格筋、胎盤、膵臓、心臓、および骨髄繊維芽細胞ではSHOXaの発現が見られたのに対し、SHOXbの発現は、胎児の腎臓、骨格筋、および骨髄繊維芽細胞に限定されており、骨髄繊維芽細胞で一層高い発現が認められた。
a)5’および3’RACE
SHOXaおよびb転写物の5 ’末端をクローン化するために、上記で構築した「マラソンcDNAライブラリー」を用いて、5’RACEを行った。下記オリゴヌクレオチドプライマー:SHOX B rev、GAAAGGCATCCGTAAGGCTCCC(697- 718位、逆鎖[r])およびアダプタープライマーAP1 を用いた。下記タッチダウンパラメーター:94℃2分間、94℃30秒間、70℃30秒間、72℃2分間を5サイクル、94℃30秒間、66℃30秒間、72℃分間を5サイクル、94℃30秒間、62℃30秒間、72℃2分間を25サイクルによりPCRを行った。PCR産物の1/ 100量と下記ネスティッドオリゴヌクレオチドプライマー:SHOX A rev、GACGCCTTTATGCATCTGATTCTC(617- 640位r)およびアダプタープライマーAP2を用いて、第2ラウンド目の増幅を行った。アニーリング温度60℃で35サイクルのPCRを行った。
既報の方法〔オリタら(Orita et al.)、1989〕に従い、患者のゲノム増幅DNAのSSCP分析を行った。1〜5μlのPCR産物を、95%ホルムアミドと10mMのEDTA pH8を含む変性液5μlと混合し、95℃で10分間変性させた。試料をただちに氷上で冷やし、2%グリセリンと1xTBEを含む10%ポリアクリルアミドゲル(アクリルアミド:ビスアクリルアミド=37.5:1および29:1;Multislotgel、TGGEベース、Qiagen社)に負荷した。ゲルを15℃、500Vで3〜5時間泳動させ、TGGEハンドブック(Qiagen社、1993)の記載に従い銀染色を行った。
Amersham社から入手したpMOSBlueT- ベクターキットを用いて、PCR産物をpMOSBlueにクローン化した。単一のコロニーの一夜培養物を、100μlのH2 O中で10分間煮沸することで、溶解した。溶解物をクローン化PCR産物に特異的なプライマーを用いるPCRの鋳型として使用した。PCR産物のSSCPにより、異なる対立遺伝子を含むクローンの同定ができた。CY5標識ベクタープライマーUniおよびT7を用い、メーカー指定のサイクルシークエンシング(ThermoSequenaseキット(Amersham社))により、ALF高速自動シークエンサー(Pharmacia 社)で、クローンの配列を決定した。

受胎後5日目から受胎後18.5日目のマウス胚ならびに胎児の動物および新生動物のイン・サイチュハイブリダイゼーションを行い、発現パターンを確定した。発現は、発生段階の肢芽、鼻と口蓋の形成に関与する上顎骨前頭突起の中胚葉、眼瞼、大動脈、発生段階の雌性生殖腺、発生段階の脊髄(分化中の運動神経に限定)、および脳で見られた。この発現パターンおよびヒトホモログであるSHOTのマッピング位置からみて、SHOTは、低身長を含むコルネリア・ド・ランゲ症候群の原因遺伝子の恐らく候補であろうことを示す。
ネズミOG12遺伝子とヒトSHOX遺伝子との最も高い相同性を有するSHOTと呼ばれる新規遺伝子(第3染色体上のSHOX相同染色体)をヒトから単離した。ヒトSHOT遺伝子とネズミOG12遺伝子は高度に相同的であり、タンパク質レベルで99%同じである。まだ証明はなされていないが、SHOTとSHOXの間には顕著な相同性があるため(ホメオドメイン内のみの同一性)、SHOTも恐らく低身長やヒトの成長に関与する遺伝子であろう。
本発明のDNA配列を、PCR、LCR、およびその他の公知技術に用いて、低身長患者が低身長遺伝子に小型欠失または点突然変異を有しているかどうかを調べた。
上記と同じ状況が、日系人の低身長患者でも見られた。

- 下記の群:
a)(i)配列番号:1のアミノ酸配列を有する60アミノ酸のホメオボックスドメインを含むポリペプチドであって、かつヒトの成長を制御する生物学的活性を有するポリペプチドをコードするヌクレオチド配列、または(ii)1以上のアミノ酸残基が欠失、付加もしくは置換されていることを除いて配列番号:1のアミノ酸配列を有する60アミノ酸のホメオボックスドメインを含むが、ヒトの成長を制御する同等の生物学的活性を有するポリペプチドをコードするヌクレオチド配列を含有してなる単離されたDNA分子;

- 低身長遺伝子の変異に関連する疾患の治療用の医薬組成物を調製するための請求項1〜24いずれか記載のDNA配列の使用。
- 減少した(deminished)ヒトの成長の原因となる遺伝子欠陥を有する個体の同定用のキットを調製するための請求項1〜24いずれか記載のDNA配列の使用。
- ヒトの低身長の原因となる遺伝子の同定のための請求項33記載のDNA配列の使用。
- 検査対象の生物学的試料分子が配列番号:2〜配列番号:7記載のいずれかのDNA配列に完全にまたは部分的に相補的な2つのヌクレオチドプローブの存在下に増幅され、つづいて適切な検出系で決定される、RNAまたはDNA分子に基づく低身長の決定方法。
- 低身長の原因となる遺伝子欠陥を有する人の同定のための請求項35記載の方法の使用。
- 請求項1〜24いずれか記載のDNA配列を含む低身長の原因となる遺伝子で形質転換されたトランスジェニック動物。
- 請求項1〜24いずれか記載のDNA配列で形質転換された細胞。
- 請求項38記載の細胞を含有するヒト低身長の治療に有用な医薬剤を同定またはスクリーニングするための試験系。
- 請求項39記載の試験系を提供する工程、および該細胞と該医薬剤の候補物とを接触させたのち、該細胞の表現型における変化または該細胞の発現産物における変化を決定する工程を含む、低身長遺伝子中の変異に関連する疾患の治療に有用な医薬剤の候補物を同定またはスクリーニングする方法。
- コードされたポリペプチドの発現をもたらしうる請求項1〜8いずれか記載のDNA分子を含有してなる発現ベクター。
- ヒトの宿主細胞における発現をもたらす発現プロモーターの下流に請求項1〜8いずれか記載のDNA分子を組み込んだ発現プラスミドをヒト細胞に導入する工程を含む、SHOXまたはSHOT遺伝子における少なくとも1種の変異に関連したヒトの成長疾患の遺伝子治療によるイン・ビボ治療方法。
- ターナー症候群または低身長の治療のための請求項42記載の方法。
- 転写因子A、BもしくはCまたはそれらの抗原性断片を用いて哺乳類を免疫し、かつかかる哺乳類から抗体を単離することにより得られた抗体。
