Description
.4.2.30)の阻害剤としてのフタラジン誘導体の使用、PARP−相同酵素阻害
剤としての使用に関し、かつ、特に、これらのフタラジン誘導体はさらにPAR
P−相同酵素の選択的阻害を示す。
リ(ADP−リボース)シンターゼ(PARS)は、細胞核中で見出された調節
酵素である(K.Ikai et al., J. Histochem. Cytochem. 1983, 31, 1261-1264)
。PARPはDNA切断の修復中において含まれるものであると推定される(M.
S. Satoh et al., Nature 1992, 356, 356-358)。DNAストランド中の損傷ま
たは切断は、酵素PARPの活性化を意味し、これが活性化される場合には、N
ADからのADP−リボースの移動を触媒する(Shaw, adv. Radiat. Biol., 19
84, 11, 1-69)。この間において、ニコチンアミドはNADから放出される。ニ
コチンアミドは、エネルギー担体ATPを消費しながら他の酵素によってNAD
に再度変換される。したがって、PARPの過剰な活性化は、ATPの非生理学

70nmol/分/mg総タンパク質)をBIOMOLから購入した(オーダー
番号 SE−165)。ヒトPARP2をバキュロウイルス系(Bac-to-Bac Sys
tem, BRL LifeScience)中で組み換えによって発現させた。この目的のために、
相当するcDNAをpFASTBAC−1ベクター中にクローン化した。E.C
oli中への組み換えによる、組み換えバキュロウイルスDNAの製造の後に、
昆虫細胞(Sf9またはhigh five)を相当する組み換えバキュロウイ
ルスDNAsでトランスフェクトさせた。相当するタンパク質の発現は、ウエス
タンブロット分析によって証明した。ウイルス株を、当業者に公知の方法で増幅
させた。多量の組み換えタンパク質を、MOI(インフェクションの多様性;細
胞に対するウイルスの割合)5〜10で、ウイルスを含有する昆虫細胞培養物5
00ml(2×106細胞/ml)のインフェクションによって感染させ、かつ
3〜4日に亘ってインキュベートした。その後に、昆虫細胞を遠心分離によって

測定した(EAR340AT “Easy Reader”, SAT-Lab instruments, Austria)(45
0nm対620nm)。測定の原理は、図6中に図示されている。
