Abstract
Description
、特にトラコーマクラミジアの感染によって生じる疾患に対して宿主を保護する
ためのワクチン接種法に関するものである。
クラミジアは結膜および生殖管の上皮に感染し、トラコーマや各種性病(STD
)を起こし、それぞれ失明や不妊に至る場合がある。トラコーマクラミジアには
15種類の血清型があり、そのうちのA、BおよびCがトラコーマの病原菌であ
り、血清型D、E、F、G、H、I、JおよびKがクラミジアSTDの最も一般
的な病原菌である。トラコーマクラミジア感染は世界全体で流行している。トラ
コーマは、発展途上国での予防可能な失明の第1位の原因であり、世界中で6億
人がトラコーマを患い、1000万人もの人々がその疾患によって失明している
と推定されている。米国では、トラコーマクラミジアが原因のSTDが年間30
0万例と推計されている。

であって、結膜炎症のピーク期間に相当している(参考文献5)。ヒトより下等
な霊長類モデルでの実験的な眼球感染後の保護免疫は同型であり、眼球攻撃に対
する抵抗は、涙中の血清型特異的抗体の存在と相関している(参考文献1、2、
6)。感染ヒトからの涙は、フクロウザル眼球における同種トラコーマ血清型の
感染性を中和したが、異種のものは中和せず(参考文献7)、抗トラコーマ抗体
による受動的体液免疫感作は保護効果はなかった(参考文献8)。いくつかの系
統の証拠から、クラミジア感染からの保護またはそれのクリアランスにおける細
胞介在応答の重要性が示されている。B−細胞欠乏マウスは感染を解消すること
ができるが、ヌードマウスは持続的に感染状態となる。少なくともいくつかのク
ラミジア特異的T細胞系またはクローンの養子移入によって持続的に感染したヌ
ードマウスを治癒させることができ、その抗クラミジア活性は恐らくは、T細胞
がインターフェロン−γを分泌する能力の関数である(参考文献9〜16)。
チン感作によって疾患を強化してきた(参考文献1、2)。感作は、トラコーマ
クラミジアの全ての血清型に存在する57kDストレス応答蛋白(SRP)(H
SP60)に対して誘発されることが確認されている。57kD SRPに対す
る繰り返し曝露によって過敏反応遅延が生じて、それが原因でクラミジア感染に
共通に関連する慢性炎症が生じる場合がある。そこで、宿主感作を起こさず、強
力かつ持続的な粘膜中和抗体反応および強力な細胞免疫応答を誘発することがで
きる免疫原性物が有用であると考えられる(参考文献17)。

、24)。クラミジア感染性を中和することができる抗体はMOMPを認識する
(参考文献25、26、27、28)。MOMP特異的中和性モノクローナル抗
体が結合するエピトープが、いくつかの血清型についてマッピングされており(
参考文献21、22、29、30、31、32、33)、合成ワクチンもしくは
サブユニットワクチンの開発に向けた重要な目標となっている。結合部位は、血
清型によって決まるVD IまたはII内にある6〜8個のアミノ酸の連続配列
である。MOMPエピトープを有するサブユニット免疫原(例:単離MOMPま
たは合成ペプチド類)は、完全なクラミジアを認識できる抗体を誘発することが
できる(参考文献25)。しかしながら、従来投与されていたサブユニット免疫
原は、粘膜免疫性誘発剤としては弱い。クラミジア抗原を、それの免疫原性を高
め、体液および細胞介在の両方の免疫応答を誘発するような形で、クラミジア抗
原を製剤することが有用であると考えられる。
レステロールおよび不飽和脂肪酸類の混合物から形成される籠型構造である。I
SCOMの構成要素は疎水性相互作用によって一体となっており、その結果、性
質上疎水性である蛋白(例:MOMP)または疎水性残基を露出または付加する
ような処理をされた蛋白を、それが形成する通りにISCOM中に効果的に組み
入れることができる(参考文献34、35、36)。
局所抗体および局所細胞免疫応答が、粘膜感染からの保護の重要な要素である。
従って、クラミジアワクチンが、細胞要素および抗体要素の両方を含む粘膜免疫
応答を誘発することが有用であると考えられる。
(参考文献37)。蛋白もしくはペプチドに基づくサブユニットワクチンとは異
なり、DNA免疫感作は、宿主細胞による異種蛋白の発現を介して保護免疫を提
供することで、ウィルス感染または細胞内病原による感染時に起こるものに比較
的類似した形での免疫系への抗原の提供を可能とするものである(参考文献38
)。この方法はかなりの関心を集めたが、奏功する免疫が一貫して誘発されたの
はウィルス疾患におけるDNA免疫感作によるものであった(参考文献39)。
非ウィルス系病原体の場合には結果はさらに変動しており、それは病原体の性質
、選択される免疫抗原ならびに免疫感作の経路における違いを反映したものであ
ると考えられる(参考文献40)。DNA予防接種のさらなる開発は、基礎とな
る免疫機序の解明と既存のワクチン開発戦略が奏功していない他の感染症へのそ
れの利用拡大によって決まるであろう。
特にネズミチフス菌の使用が報告されている(参考文献41、42)。この種の
投与は、粘膜免疫応答などの特異的免疫応答を誘発する細胞種に対してDNAを
投与するという利点をさらに提供するものである。この種の予防接種はまた、多
くの安全なサルモネラの弱毒株が容易に得られることから安全であって、コスト
効果が高いという利点をも提供する。

ミドDNAを構築し、マウスでの実験ができるようにした。このモデルでの一次
感染によって、再感染に対する強力な保護免疫が誘発される。ヒト免疫感作の場
合、トラコーマクラミジアの血清型Cなどのヒト病原菌株を用いる。

A,USA)のようなMOMP遺伝子または断片に関して、簡便なプラスミドベ
クターを用いることができる。MOMP遺伝子またはMOMP遺伝子断片は、簡
便な方法でベクターに挿入することができる。その遺伝子または遺伝子断片は、
好適なプライマーを用いるPCRによってトラコーマクラミジアゲノムDNAか
ら増幅することができ、PCR産物はベクターにクローニングする。得られるM
OMP遺伝子保有プラスミドは、例えばエレクトロポレーションによって大腸菌
中にトランスフェクションして、そこでの複製を行わせることができる。大きさ
約54kbのMOMP保有プラスミドpcDNA3/MOMPは、図5に示して
ある。プラスミドは、簡便な方法で大腸菌から抽出することができる。
クトロポレーションなどの標準的なプロトコールに従って弱毒サルモネラ菌をト
ランスフォームすることができる(参考文献43)。

イトメガロウィルス主要中間初期エンハンサー領域(CMVプロモーター)の制
御下での転写によって、PCR産物をpcDNA3真核II選択可能発現ベクタ
ー(Invitrogen,San Diego)中にクローニングした。10
0μg/mLのアンピリシンを含むLB肉汁中で成長させた大腸菌DH5αF中
にエレクトロポレーションによって、MOMP遺伝子コードプラスミドを移し入
れた。ウィザード(商標)プラスマキシプレプ(Wizard Plus Ma
xiprep)DNA精製システム(Promega,Madison)によっ
てプラスミドを抽出した。報告の方法に従って、組換えMOMP遺伝子の配列を
PCR直接配列分析によって確認した(参考文献44)。精製プラスミドDNA
を濃度1mg/mLで生理食塩水に溶かした。DNA濃度をDU−62分光光度
計(Beckman,Fullerton,CA)によって260nmで測定し
、プラスミドの大きさを臭化エチジウム染色アガロースゲル中でDNA標準と比
較した。

とに制限されているほとんどのトラコーマクラミジア菌株とは異なり、MoPn
は天然マウス病原菌である。このモデルでの一次感染は再感染に対する強力な保
護免疫を誘発することが、これまでに明らかになっている。さらに感染のクリア
ランスは、CD4 Th1リンパ球応答に関係し、MHCクラスII抗原提供に
よって決まる(参考文献45)。

、筋肉投与MOMP−DNAよりその効果が高いように思われることを明瞭に示
している。MOMP−DNAの経鼻投与を複数の実験で評価して、その再現性を
確認した。表1に示したように、MOMP−DNAの粘膜投与によって保護免疫
応答が誘発されたが、保護指数の大きさはかなり変動して、各種実験で0.5〜
4.1log10保護の範囲であった。そのような変動性の原因は、予防接種され
た動物にMoPnの比較的高い接種原を負荷することで保護指数が低下したこと
から、裸出DNA予防接種の免疫原性が制限されたためである可能性がある。粘
膜表面に投与された裸出DNAはまた、その動向が非常に多様であると考えられ
、一部は細胞外ヌクレアーゼによって分解され、一部は体細胞によって取り込ま
れる。
レクトロポレーションによってpcDNA3/MOMPおよびpcDNA3でト
ランスフェクションした。プラスミドDNAでトランスフェクションされたサル
モネラの弱毒株を、100μg/mアンピリシンを含むルリア肉汁(LB)培地
中、振盪を行わずに37℃で16〜25時間培養した。細菌を遠心によって回収
し、PBS中に再度懸濁させた。各種濃度のサルモネラをPBSで希釈し、同容
量の10%重炭酸ナトリウムを加えて、直ちに免疫感作を行った。6〜8週齢の
雌Balb/cマウス5〜10匹の群に5〜6時間水を摂取させないようにして
から免疫感作に供した。100μL中約105〜1010CFUの細菌を、給餌針
(Ejay International Inc.)によって摂取させた。2
週間の間隔を設けて4回の摂取を行った。

NAトランスフェクションサルモネラを、2週間の間隔で4回経口免疫感作した
Balb/cマウスの群間で、106cfuで投与したMOMP−DNAトラン
スフェクションサルモネラと比較した。106cfuで免疫感作したマウスでは
、4回目の免疫感作時にISCOM(14)に埋め込まれた1μgのMoPn
MOMPを筋肉投与することで、単一蛋白ブーストがあった。ISCOM取得物
は、上記の米国特許出願08/718236号(WO98/10789)に記載
の方法に従って得たものである。最終免疫感作から2週間後に、マウスに対して
、5000 IFUのMoPn EBの経鼻投与による攻撃を行った。感染マウ
スを、感染から10日目に屠殺した。感染後毎日、マウスを屠殺するまで体重測
定を行った(図3、A)。そのマウスについては、実施例3に記載の方法に従っ
て蛋白ブーストを行わずに108cfuのサルモネラ投与を受けたマウスと比較
して、かなり良好な保護があった。感染後10日での肺におけるクラミジアEB
を、定量的組織培養によって分析した(図3、BおよびC)。図3Bにおいてデ
ータは、マウス5〜6匹の肺当たりのlog10IFUの平均±標準誤差を示して
あり、図3Cは個々のマウスで認められた結果を示している。DNAプライミン
グされた蛋白ブーストマウスも、免疫原性の向上を示し、DTH応答が大きくな
り(図4A)、血清IgG2およびIgG1抗体応答が強くなった(図4Bおよび
C)。最終免疫感作から2週間後に、免疫感作マウスから血清を採取した。Mo
Pn特異的IgG2a(B)およびIgG1(C)抗体を、ELISAによって調
べた。
EB(2×105 IFU)25μLをマウスの右後足蹠に注射し、等容量のS
PG緩衝液を対照として左後足蹠に注射した。足蹠腫脹を、ディラ(dila)
−ゲージキャリパスを用いて注射から48時間後および72時間後に測定した。
2つの足蹠間での厚さの差をDTH応答の尺度として用いた。
