Claims (1)
- 【特許請求の範囲】 1.細胞のサンプルから核酸を単離する方法において、 (a)該サンプル中の細胞を固体支持体に結合させて、細胞を該サンプルから 分離し; (b)分離した細胞を溶解し;そして (c)該溶解した細胞から放出された核酸を同じ該固体支持体に結合させるこ とを含む、上記の方法。 2.上記の核酸がDNAである、請求の範囲1に記載の方法。 3.上記の段階(a)で、細胞が沈殿剤を使用して支持体上に沈殿される、請求 の範囲1又は2に記載の方法。 4.上記沈殿剤がアルコールと塩とを含む、請求の範囲3に記載の方法。 5.上記の段階(a)で、細胞が、支持体上又は支持体内に設けられた細胞結合 部分により該支持体に結合される、請求の範囲1又は2に記載の方法。 6.上記細胞結合部分が標的細胞を選択的に結合する、請求の範囲5に記載の方 法。 7.上記固体支持体が粒状物である、請求の範囲1ないし6のいずれか1項に記 載の方法。 8.上記支持体が磁性ビーズを含む、請求の範囲7に記載の方法。 9.上記段階(b)において、細胞が界面活性剤および/又はカオトロープを用 いて溶解される、1ないし8のいずれか1項に記載の方法。 10.上記段階(c)において、核酸が支持体に非特異的に結合される、1ない し9のいずれか1項に記載の方法。 11.上記核酸が沈殿剤を用いて支持体上に沈殿される、請求の範囲10に記載 の方法。 12.上記沈殿剤がアルコール、および任意に塩、および/又は界面活性剤を含 む、請求の範囲11に記載の方法。 13.上記の段階(c)で、核酸が、核酸の選択的捕獲を助けるために上記支持 体上に設けられた結合パートナーにより該支持体に結合される、請求の範囲1な いし9のいずれか1項に記載の方法。 14.結合された核酸が上記支持体から溶離される、請求の範囲1ないし13の いずれか1項に記載の方法。 15.請求の範囲1ないし14のいずれか1項に規定された核酸単離方法の、核 酸に基づく標的細胞検出方法に使用する核酸の製造への使用。 16.サンプル中の標的細胞の存在又は非存在を検出する方法であって、下記段 階を含む方法: (a)該サンプル中の細胞を固体支持体に結合させて、細胞を該サンプルから 分離し; (b)分離した細胞を溶解し; (c)該溶解した細胞から放出された核酸を同じ該固体支持体に結合させ;そ して (d)上記の結合された核酸内の上記標的細胞に特有の核酸の存在又は非存在 を検出する。 17.上記検出段階(d)がin situハイブリッド形成、および/又はイ ン ビトロ増幅および/又は核酸配列決定を含む、請求の範囲16に記載の方法 。 18.サンプルから核酸を単離するためのキットであって: (a)固体支持体; (b)任意に、細胞を該固体支持体に結合するための手段; (c)該細胞を溶解する手段; (d)該溶解した細胞から放出された核酸を同じ該固体支持体に結合させる手 段、 を含むキット。
