Claims (1)
- 【特許請求の範囲】 1.以下の1または複数の特徴を有する: 40℃にて4時間置かれた後に85%を超える活性を保持するか、または60℃で4時 間置かれた後に30%を越える活性を保持する、 ラクタムのラセミ体100gと緩衝液300mlの初期濃度のものを加水分解し、少な くとも90%の(+)ラクタムを加水分解するまで加水分解を続け、一方で(-)ラクタ ムの加水分解は5%未満である、 二環式ラクタムである2-アザビシクロ[2,2,1]ヘプト-5-エン-3-オンのエナンチ オマーを加水分解し得る酵素。 2.同初期濃度を加水分解し、少なくとも98%の(+)ラクタムが加水分解される まで加水分解を続ける一方、(-)ラクタムの加水分解が2%未満であることに特徴 付けられる、請求項1記載の酵素。 3.コマモナス・アシディボランス(Comamonas acidivorans)から得られる、二 環式ラクタムである2-アザビシクロ[2,2,1]ヘプト-5-エン-3-オンのエナンチオ マーを加水分解し得る酵素。 4.コマモナス・アシディボランス(Comamonas acidivorans)NCIMB 40827から 得られる請求項3記載の酵素。 5.配列表2に記載のアミノ酸配列もしくは、そのフラグメントを有し、二環式 ラクタムである2-アザビシクロ[2,2,1]ヘプト-5-エン-3-オンのエナンチオマー を加水分解し得る酵素。 6.前記各項いずれかに記載の酵素を固定化した、固定化酵素。 7.請求項5記載の酵素をコード化する、単離されたヌクレオチド分子。 8.配列番号1記載の配列を有する、請求項7記載の酵素。 9.請求項5記載の酵素を発現し得る、微生物。 10.コマモナス・アシデイボランス(Comamonas acidivorans)NCIMB 40827の 本質的な性質を有している、請求項9記載の微生物。 11.請求項9または10記載の微生物を培養することを含む、請求項1から5 いずれかに記載の酵素を製造する方法。 12.2-アザビシクロ[2,2,1]ヘプト-5-エン-3-オンのエナンチオマーの混合物 と、請求項1から5いずれかに記載の酵素もしくは請求項9または10記載の微 生物を接触させる行程を含む、二環式ラクタムである2-アザビシクロ[2,2,1]ヘ プト-5-エン-3-オンのエナンチオマーの選択的加水分解方法。 13.加水分解により生成された(+)アミノ酸から残存する(-)ラクタムを分離す る行程をさらに含む、請求項12記載の方法。
