Claims (1)
- 【特許請求の範囲】 1. (i)(a)グリセロール−3−リン酸デヒドロゲナーゼ活性をコード する遺伝子; (b)グリセロール−3−ホスファターゼ活性をコードする遺伝 子: (c)デヒドラターゼ活性をコードする遺伝子; (d)1,3−プロパンジオールオキシドレダクターゼ活性をコ ードする遺伝子 の少なくとも1つを含んでなる形質転換カセットで適当な宿主生物を形質転換し 、ただし、形質転換カセットが全てより少ない(a)−(d)の遺伝子を含んで なる場合には、適当な宿主生物が内因性遺伝子を含んでなり、それにより得られ る形質転換された宿主生物が少なくとも1つの各々の遺伝子(a)−(d)を含 んでなり; (ii)形質転換された宿主生物を単糖、オリゴ糖、多糖または1炭素基 質よりなる群から選択された少なくとも1つの炭素源の存在下で適当な条件下で 培養し、それにより1,3−プロパンジオールが生成され;そして (iii)1,3−プロパンジオールを回収すること 含んでなる組み換え生物からの1,3−プロパンジオールの生産方法。 2. 形質転換カセットが遺伝子(a)−(d)の全てを含んでなる請求の範 囲1の方法。 3. 適当な宿主生物がバクテリア、酵母及び糸状菌よりなる群から選択され る請求の範囲1の方法。 4. 適当な宿主生物がシトロバクター、エンテロバクター、クロス トリジウム、クレブシエラ、エロバクター、ラクトバシラス、アスペルギルス、 サッカロミセス、シゾサッカロミセス、ザイゴサッカロミセス、ピチア、クルイ ベロミセス、カンジダ、ハンセヌラ、デバリオミセス、ムコール、トルロプシス 、メチロバクター、エシェリキア、サルモネラ、バシラス、ストレプトミセス及 びシュードモナスよりなる属の群から選択される請求の範囲3の方法。 5. 適当な宿主生物がエシェリキア・コリ、クレブシエラ種及びサッカロミ セス種よりなる群から選択される請求の範囲4の方法。 6. 形質転換された宿主生物が遺伝子dhaB1、dhaB2、dhaB3 及びdhaTを含んでなる形質転換カセットで形質転換されたサッカロミセス種 であり、その場合にそれらの遺伝子がサッカロミセス種ゲノム中に安定に組込ま れる請求の範囲1の方法。 7. 形質転換された宿主生物が遺伝子GPD1及びGPP2を含んでなる形 質転換カセットで形質転換されたクレブシエラ種である請求の範囲1の方法。 8. 炭素源がグルコースである請求の範囲1の方法。 9. グリセロール−3−リン酸デヒドロゲナーゼ酵素をコードする遺伝子が 配列番号11、配列番号12及び配列番号13に示されるアミノ酸配列に対応す る遺伝子よりなる群から選択され、それらのアミノ酸配列がグリセロール−3− リン酸デヒドロゲナーゼ酵素の機能を変えないアミノ酸置換、欠失または付加を 包含する請求の範囲1の方法。 10. グリセロール−3−ホスファターゼ酵素をコードする遺伝子が配列番 号33及び配列番号17に示されるアミノ酸配列に対応する遺伝子よりなる群か ら選択され、それらのアミノ酸配列がグリセロール− 3−ホスファターゼ酵素の機能を変えないアミノ酸置換、欠失または付加を包含 する請求の範囲1の方法。 11. グリセロールキナーゼ酵素をコードする遺伝子が配列番号18に示さ れるアミノ酸配列に対応し、そのアミノ酸配列がグリセロールキナーゼ酵素の機 能を変えないアミノ酸置換、欠失または付加を包含する請求の範囲1の方法。 12. デヒドラターゼ酵素をコードする遺伝子がdhaB1、dhaB2及 びdhaB3を含んでなり、それらの遺伝子がそれそれ配列番号34、配列番号 35及び配列番号36に示されるアミノ酸配列に対応し、それらのアミノ酸配列 がデヒドラターゼ酵素の機能を変えないアミノ酸置換、欠失または付加を包含す る請求の範囲1の方法。 13. 1,3−プロパンジオールオキシドレダクターゼ酵素をコードする遺 伝子が配列番号37に示されるアミノ酸配列に対応し、そのアミノ酸配列が1, 3−プロパンジオールオキシドレダクターゼ酵素の機能を変えないアミノ酸置換 、欠失または付加を包含する請求の範囲1の方法。 14. (a)(1)配列番号11に示されるアミノ酸配列に対応するグリセ ロール−3−リン酸デヒドロゲナーゼ酵素をコードする遺伝子; (2)配列番号17に示されるアミノ酸配列に対応するグリセ ロール−3−ホスファターゼ酵素をコードする遺伝子; (3)配列番号34に示されるアミノ酸配列に対応するグリセ ロールデヒドラターゼ酵素のα−サブユニットをコードする遺伝子; (4)配列番号35に示されるアミノ酸配列に対応するグリセ ロールデヒドラターゼ酵素のβ−サブユニットをコードする遺伝子; (5)配列番号36に示されるアミノ酸配列に対応するグリセ ロールデヒドラターゼ酵素のγ−サブユニットをコードする遺伝子;及び (6)配列番号37に示されるアミノ酸配列に対応する1,3 −プロパンジオールオキシドレダクターゼ酵素をコードする遺伝子、 を含んでなる遺伝子の群、(a)(1)−(6)のそれぞれのアミノ酸配列は遺 伝子(1)−(6)の酵素の機能を変えないアミノ酸置換、欠失または付加を包 含する、並びに (b)(a)の遺伝子の群で形質転換された宿主細胞で、それにより 形質転換された宿主細胞が単糖、オリゴ糖及び多糖よりなる群からまたは1炭素 基質から選択された少なくとも1つの基質で1,3−プロパンジオールを生産す る、 を含んでなる形質転換された宿主細胞。
