- 【特許請求の範囲】 1.式(I) [式中、 R1はC1-6アルキルオキシ、C2-6アルケニルオキシ又はC2-6アルキニルオキシ であり; R2は水素又はC1-6アルキルオキシであるか、 あるいは一緒になってR1及びR2は式 −O−CH2−O− (a−1)、 −O−CH2−CH2− (a−2)、 −O−CH2−CH2−O− (a−3)、 −O−CH2−CH2−CH2- (a−4)、 −O−CH2−CH2−CH2−O− (a−5)、 −O−CH2−CH2−CH2−CH2− (a−6)、 の2価の基を形成することができ、 ここで該2価の基において1つもしくは2つの水素原子はC1-6アルキルで置換 されていることができ、 R3は水素又はハロであり; R4は水素又はC1-6アルキルであり; LはC3-6シクロアルキル、C5-6シクロアルカノン、場合によりアリールで置換 されていることができるC2-6アルケニルであるか あるいはLは式 −Alk−R5 (b−1)、 −Alk−X−R6 (b−2)、 −Alk−Y−C(=O)−R8 (b−3)又は −Alk−Y−C(=O)−NR10R11 (b−4) の基であり、 ここで各AlkはC1-12アルカンジイルであり; R5は水素、シアノ、C1-6アルキルスルホニルアミノ、C3-6シクロアルキル、 C5-6シクロアルカノン、アリール、ジ(アリール)メチル又はHet1であり; R6は水素、C1-6アルキル、ヒドロキシC1-6アルキル、C3-6シクロアルキル、 アリール又はHet2であり; XはO、S、SO2又はNR7であり;該R7は水素、C1-6アルキル又はアリール であり; R8は水素、C1-6アルキル、C3-6シクロアルキル、アリール、アリールC1-6ア ルキル、ジ(アリール)メチル、C1-6アルキルオキシ又はヒドロキシであり; YはNR9又は直接結合であり;該R9は水素、C1-6アルキル又はアリールであ り; R10及びR11はそれぞれ独立して水素、C1-6アルキル、C3-6シクロアルキル、 アリール又はアリールC1-6アルキルであるか、あるいはR10及びR11はR10及 びR11を有する窒素原子と組み合わされてピロリジニル又はピペリジニル環を形 成することができ、両者は場合によりC1-6アルキル、アミノ又はモノもしくは ジ(C1-6アルキル)アミノで置換されていることができるか、あるいは該R10 及びR11はR10及びR11を 有する窒素と組み合わされてピペラジニル又は4−モルホリニル基を形成するこ とができ、両者は場合によりC1-6アルキルで置換されていることができ; 各アリールは非置換フェニル又はそれぞれ独立してハロ、ヒドロキシ、C1-6ア ルキル、C1-6アルキルオキシ、アミノスルホニル、C1-6アルキルカルボニル、 ニトロ、トリフルオロメチル、アミノもしくはアミノカルボニルから選ばれる1 、2もしくは3個の置換基で置換されたフェニルであり; Het1及びHet2はそれぞれ独立してフラン;C1-6アルキルもしくはハロで 置換されたフラン;テトラヒドロフラン;C1-6アルキルで置換されたテトラヒ ドロフラン;ジオキソラン;C1-6アルキで置換されたジオキソラン;ジオキサ ン;C1-6アルキルで置換されたジオキサン;テトラヒドロピラン;C1-6アルキ ルで置換されたテトラヒドロピラン;ピロリジニル;それぞれ独立してハロ、ヒ ドロキシ、シアノもしくはC1-6アルキルから選ばれる1つもしくは2つの置換 で置換されたピロリジニル;ピリジニル;それぞれ独立してハロ、ヒドロキシ、 シアノ、C1-6アルキルから選ばれる1つもしくは2つの置換基で置換されたピ リジニル;ピリミジニル;それぞれ独立してハロ、ヒドロキシ、シアノ、C1-6 アルキル、C1-6アルキルオキシ、アミノならびにモノ及びジ(C1-6アルキル) アミノから選ばれる1つもしくは2つの置換基で置換されたピリミジニル;ピリ ダジニル;それぞれ独立してヒドロキシ、C1-6アルキルオキシ、C1-6アルキル 又はハロから選ばれる1つもしくは2つの置換基で置換されたピリダジニル;ピ ラジニル;それぞれ独立してハロ、ヒドロキシ、シアノ、C1-6アルキル、C1-6 アルキルオキシ、ア ミノ、モノ−及びジ(C1-6アルキル)アミノならびにC1-6アルキルオキシカル ボニルから選ばれる1つもしくは2つの置換基で置換されたピラジニルから選ば れ; Het1はまた式 の基であることもでき、 Het1及びHet2はそれぞれ独立してまた式 の基から選ばれることもでき、 R12及びR13はそれぞれ独立して水素又はC1-4アルキルである] の化合物、その立体化学的異性体、そのN−オキシド形態又はその製薬学的に許 容され得る酸もしくは塩基付加塩。 2.R1がメトキシであり、R2が水素であるか;あるいはR1及びR2が一緒にな って式(a−2)、(a−3)又は(a−4)の基を形成し;R3がクロロであ る請求の範囲第1項に記載の化合物。 3.ヒドロキシ又はメトキシが中心ピペリジン部分上のメチレンに関し てトランス位にある請求の範囲第1〜2項のいずれかに記載の化合物。 4.化合物が8−アミノ−7−クロロ−2,3−ジヒドロ−1,4−ベンゾジオ キシン−5−カルボン酸 (±)−トランス−(1−ブチル−3−ヒドロキシ− 4−ピペリジニル)メチル、製薬学的に許容され得るその酸付加塩又はその立体 化学的異性体である請求の範囲第1〜3項のいずれかに記載の化合物。 5.製薬学的に許容され得る担体及び治療的に活性な量の請求の範囲第1〜4項 のいずれかに記載の化合物を含む製薬学的組成物。 6.治療的に活性な量の請求の範囲第1〜4項のいずれかに記載の化合物を製薬 学的に許容され得る担体と緊密に混合することを特徴とする請求の範囲第5項に 記載の製薬学的組成物の調製法。 7.薬剤として用いるための請求の範囲第1〜4項のいずれかに記載の化合物。 8.式(V) [式中、R1、R2、R3及びR4は式(I)の化合物に関して請求の範囲第1項て 定義された通りである] の中間体、その製薬学的に許容され得る酸付加塩又はその立体化学的異性体。 9.a)R4aが水素、C1-6アルキル又は例えばベンジルもしくはt− ブトキシカルボニルなどの保護基であり、PGが保護基であるシス立体配置を有 する式(XI)の中間体を水素雰囲気下で、CuO.Cr2O3、CaOなどの適 した塩基の存在下において撹拌するか; b)シス立体配置を有する式(XII)の中間体を水素雰囲気下で、CuO.C r2O3の存在下で撹拌するか; c)式(XIII)の中間体をボラン又はその誘導体と反応させ、続いて酸化的 仕上げを行い; 得られる生成物を場合により再結晶させるかあるいはカラムクロマトグラフィー を用いて精製することによる; R4aが上記で定義された通りであるトランス−立体配置を有する式(VIII) の中間体の製造法。 10.a)式(II)の中間体を式(III)のカルボン酸誘導体又は その反応性官能基誘導体、例えば酸クロリドと反応させ;b)Wが適した脱離基、例えばハロを示す式(IV)の中間体を式(V)の試薬 を用いてN−アルキル化し; c)L’=OがC1-12アルカンジイル部分の2つのジェミナル水素原子が=Oに より置換されている式L−Hの化合物である式L’=O(VI)の適したケトン 又はアルデヒド中間体を式(V)のピペリジンと反応させ; d)Xがブロモ又はヨードである式(XIII)の中間体を式(II)の中間体 の存在下に、反応に不活性な溶媒、例えばアセトニトリル又はテトラヒドロフラ ン中で、適した触媒、例えばパラジウムカーボン及び第3アミン、例えばトリエ チルアミンの存在下においてならびに室温ないし反応混合物の還流温度の間の範 囲の温度でアルコキシカルボニル化 し; そして必要に応じて、式(I)の化合物を当該技術分野において既知の転換法に 従って互いに転換し;さらに必要に応じて、式(I)の化合物を酸を用いる処理 により治療的に活性な無毒性酸付加塩に転換するかあるいは逆に酸付加塩の形態 をアルカリを用いる処理により遊離の塩基に転換し;そして必要に応じて、その 立体化学的異性体又はN−オキシド形態を製造する ことを特徴とする式(I)の化合物の製造法。
