Claims (1)
- 【特許請求の範囲】 1. 配列番号:1のヌクレオチド1から14507、ヌクレオチド598 0から14507、ヌクレオチド8671から14507、ヌクレオチド867 1から10515、ヌクレオチド9277から10515、ヌクレオチド867 2から12273、ヌクレオチド10100から14923を含むDNA、発現 増大活性を持つ前述のDNAのフラグメント、前述のDNAと相補性を持つDN A、前述のDNAとヌクレオチド配列で少なくとも約80%同一であり発現増大 活性を持つDNA、および発現増大活性を持つ前述のDNAの組み合わせ、から なる群より選択された、単離された発現増大配列エレメント(EASE)。 2. 本質的に、配列番号:1のヌクレオチド1から14507、ヌクレオ チド5980から14507、ヌクレオチド8671から14507、ヌクレオ チド8671から10515、ヌクレオチド9277から10515、ヌクレオ チド8672から12273、およびヌクレオチド10100から14923か らなるDNAからなる群より選択された、請求項1記載のEASE。 3. 配列番号:1のヌクレオチド14290から14507を含むDNA と連結される、請求項2記載のEASE。 4. 請求項1記載のEASEを含む発現ベクター。 5. 請求項2記載のEASEを含む発現ベクター。 6. 請求項3記載のEASEを含む発現ベクター。 7. 第一のエキソンが興味あるタンパタ質をコードし、第二のエキソンが 増幅可能な優性選択可能マーカーをコードする、バイシストロニックプラスミド である、請求項6記載の発現ベクター。 8. 増幅可能優性選択可能マーカーがジヒドロホレートレダクターゼ(D HFR)である、請求項7記載の発現ベクター。 9. 2つのエキソン間にさらにIRES配列を含む、請求項8記載の発現 ベクター。 10. 請求項7記載の発現ベクターで形質転換されたCHO細胞 11. 請求項8記載の発現ベクターで形質転換されたCHO細胞 12. 請求項9記載の発現ベクターで形質転換されたCHO細胞 13. タンパク質の発現を促進する条件下で請求項10記載の形質転換され た宿主細胞を培養し、タンパク質を回収することを含む、組換えタンパク質を得 る方法。 14. タンパク質の発現を促進する条件下で請求項11記載の形質転換され た宿主細胞を培養し、タンパク質を回収することを含む、組換えタンパク質を得 る方法。 15. タンパク質の発現を促進する条件下で請求項12記載の形質転換され た宿主細胞を培養し、タンパタ質を回収することを含む、組換えタンパク質を得 る方法。 16. 請求項7、8または9のいずれか1項に記載の発現ベクターで形質転 換されたCHO細胞の安定なプール。 17. タンパク質の発現を促進する条件下で請求項16記載のCHO細胞の 安定なプールを培養し、タンパク質を回収することを含む、組換えタンパク質を 得る方法。
