Abstract

A találmány eljárás baktériumi gazdasejtben exprimált és oldhatatlanzárványként (inclusion bodies) a sejtben tárolt rekombináns proteinekoldására és tisztítására. A találmány szerinti eljárásra jellemző,hogy (i) az elválasztott oldhatatlan proteinaggregátumokat szervesdenaturáló reagensben feloldják, (ii) az (i) szerint kapott oldatotlegalább egy kromatográfiás lépésben tisztítják lényegében szervetlen,puffermentes, bázikus, sótartalmú eluens alkalmazásóval, (iii) azutolsó tisztító lépésben kapott, az oldott, renaturált és biológiailagaktív proteint tartalmazó bázikus oldatot semlegesítik. Az eluensbenlévő sók milyenségének és mennyiségének alkalmas megválasztásával azoldott tisztított protein mindjárt fiziológiai sóoldatban van jelen. Ó
