PEPTIDEK ÉS El JÁRÁS ÉÉŐAEIJTÁSEKRA
A találmány gyógyhatású anyagokra, nevezetesen olyan biológiailag aktív anyagokra vonatkozik, amelyek immunsz&bályozó tslajdonságokkal rendelkeznek, így gyógyszerhatóanyagként alkalmazhatók a humán és állatgyógyászatban és biokémiai kísérletekben.
Annak fontosságát, hogy áj, veszélymentes Immunszahályozó peptideket fejlesszünk ki, amelyek megakadályozzák az olyan betegségek kifejlődését, mint a rosszindulatú seopiaxmák, szepszis, az immunhiáuyosságok miatt kifejlődő krónikus és lappangó fertőzések, az e területen folytatott nagyszámú kutatás igazolja.
Az új peptidek izolálásának legáltalánosabban, hasznait módszere- abban áll, hogy aktív peptíd-irakciókat Izolálnak szőveiienyészetekhől, majd ezeket tovább írakcionálják és tisztítják és végül azonosítják az egyes anyagokat (lásd pl. az Sö-i 737798 számú, 19.95. március 20-án engedélyezett szovjet feltalálói tanúsítvány leírását). A gyakorlati gyógyászatban a tínrasz-exiraktumokról ismert, hogy az ínnmmfolyamatokat szabályozzák, ilyen például a timosin S, frakció (Goldsteio AA.. Guna A., Latz M.M., Bardy HA, White A.) és a tsamalm (CH-659 586). Az extraktumok polipepiid természetű anyagokat tartalmaznak, ezek természetes nyersanyagokból történő előállítását korlátozza az eljárás bonyolultsága, az aktív anyag kapott alacsony hozama és a polipeptidek fizikai és kémiai jellemzőinek, valamint biológiai tulajdonságainak jelentős különbözősége. Emellett a természetes dmusz-készitményekben jelenlevő kísérő anyagok néha mellékhatásokat okoznak. Ez utóbbi körülmény ösztönözte a kutatókat arra, hogy szintetikus peptideket fejlesszenek ki. Számos ünmunszabáiyozó pepiidet szintetizáltak már, ilyeneket ismertet például a WO-89/G6134, EP-23Ö052, EP-4Ö6931, US-5Ö21551, US-5013723 számú dokumentum, A?, egyes szintetikus peptidek a szükséges tulajdonságok korlátozott kombinációjával rendelkeznek, miközben magas az aktivitásuk, alacsony a toxicitásuk, nem okoznak, mellékhatásokat, így a gyógyászatban alkalmazhatók,

A találmány tárgya továbbá alapvetően áj teelmelógís peptidek szbtetizálására, -amely minimális és egyszerű lépésekből áll, és a fent említett szerkezetű peptidek nagy hozammal történő előállítását teszi lehetővé, ami döntő fontosságú gyógyszerek előállitásánál.
A találmány szerinti áj eljárás lényege abban áll . hogy oldatban glatamíntartainsé pepiideket szintetizálunk oly módon, hogy terc~bual-oxi-kíaboöil-(10 vagy Lj-ghaaminsav belső anhidridjét a megfelelő D~Trp~Y származék alkalmazásával megbontjuk, majd az a- és γ-izomereket kromatográfiásan elválasztjuk. A szintézis folytatásaképpen a. peptíáíáacot a tere-butil~oxi-karbotül~aminosav aktivált éterének alkalmazásával építjük,
A találmány tárgya közelebbről eljárás
X-A-D-Trp-Y (I) általános képietü peptidek elöállitására, a képletben X. A és Y jelentése a. fenti, oly módon, hogy a pepiidet oldatban szintetizáljuk a tere-butoxi-karhouii-(B vagy IJ-glutammsav belső anhidridjének DTíp-Y káltumsója segítségével végzett megbontásával majd a további láncéphést. aktivált éterek és vegyes- adúdrrdek alkalmazásával végezzük, a ierc-buioxi-karhonn-csopori előzetes, hangyasavvai történő -eltávolítása utam és végül az a- és γ-izomereket kromatográSásan elválasztjuk.
Az 1. táblázatban az új peptid-analógok Rs (kforofornumetanol/32 térf%-os eeeísav 60:45:20 térfogatarányá elegye) és (butmokpridiirZví^ecetsav 5:5:4:1 térfogatarányú elegye) értékeit adjuk meg.

14.7 g (0,1 -mól) H-D-Glu-Gfí-t tékíldunk 200 ml desztillált vízben, pH-ját 1 mol/l-es KO.Hval 10,2 értékre állítjuk. és intenzív keverés közben hozzáadjuk 33,0 g (0,3 mol): BOC4O dioxános oldatát. A pH-t egy p.O-mérövel folyamatosam ellenőrizzük, A reakció befejezése után az elegyet átvisszük egy elválasztó tölcsérbe, és az aíkálikus oldatot 3x150 ml etil-acetáttal extrabáljuk. A vizes fázis pH-át 0,2 terfíé-os kénsav lassú adagolásával 3,0 értékre állítjuk, majd a Boe-D-Glu-OH-t a szerves fázisba (3x200 ml) extraháljuk, A szerves fázist 3x200 mi telített nátriius-szulfáí-oláattal semleges pH-ig mossuk, nátrium-szulfáton szárítjuk, és vákuumban bepároljuk, így .16,7 g (68%) olajat kapunk,

16.7 g (0,068 mól) Boc-D-öto-OH-t feloldunk. 200 ml dimetll-formamklban, 0 A5-ra hntlük, és 'keverés közben hozzáadjuk 20,6 g (0,1 mól) NjN’~dielkl:ohexíl-kari?odilmid 100 ml dhnetdformamlddal készült oldatai. Az elegyet 4 °C-on 4 óra hosszat keverjük, majd szobahőmérsékleten óra hosszat állni hagyjuk. A kicsapódott dtclklohexll-karbodüuddet leszűrjük és 2x50 ml dimeiilformamiddal -mossuk. Á szürietet vákuumban felére bepároljuk, és 4 °C~on, erős keverés közben hozzáadunk 24,3 g (0,1 mól) H-D-Trp-OM-t, majd az -oldatot hagyjuk szobahőmérsékletre melegedni, A reakció végbemenetelét TLC útján ellenőrizzük, kloroform/etil-acetáV metanol 6:3:1 férfogatarányú elegyével eluálva, a Ifoe-D-Gla-sav belső anhidridje által képzett folt eltűnését ügyeljük. A maradék dicikfohexii-karhodihnidet lesxütjük, és a dimeül-formamidot vákuumban .(«desztilláljuk. Az olajos maradékhoz hozzáadunk 200 ml etil-acetátot és 200 mi 0,2 tért%~os kénsavoldatót A szerves: fázist elválasztjuk telheti nátríum-szulfot-oldattal semleges pH-ig mossuk, és nátnum-szulfáton szárítjuk. Az eril-aeetátos oldatot vákuumban bepároijuk. Á kapott olajos maradék (összesen 25,4 g; 70%) a fíoc-D-GIn-D-Trp-OIl és a Boc-y-D-Gln~D-Trp-Oí-l elegye.

24,5 g (0,048 mól) fenti elegyet íéloldunk 200 ml hangyasavban és 40 *C-on 1 óra hosszat keverjük. Ezután az oldószert vákuumban ledeszhlláljuk, igv sűrű olajat kapunk. A peprtdeket elválasztjuk, és Sephadex oszlopon ioncserélő kromatográfiával tisztítjuk, 0,01-0,2 mohi pirídingradíensn acetáí-pnffer segítségével. így 5,7 g (35%) H-D-Glu-D-Trp-OH-t és 5,7 g (35%) Η-γ-DGl u-D-Trp-OH-t kapunk.

Az KV-spektrum a 250-300 sm tartományban 28ö±2 nm-nél egy maximumot, míg 28712 nm-nél egy vállat unhat,
A peptsd biológiai aktivitását Balbró egereken vizsgáltuk. Az egerek lépseisjeit 2 ml glbtamint és 5 térf% irsakíiváh. embriószérammal kiegészített R'PMI 1640 íápközegben szuszpendáltuk, majd. 100 plhnéiyedés (20ÖGÖ0 sejt) mennyiségben íaposfenekö lemezekre vittük. A vizsgák készítményt a tenyésztés kezdetekor hozzáadtuk. Mitogéoként koakavalnt A-ΐ alkalmaztunk 2 pg/mélyedés stogkoneentráeiőbsn. A lemezeket 37 öC-on 5 térf% szén-dioxidot tartalmazó légtérben 48 óra hosszat iakubáltuk. A sejtszaporodást ’fí-tánídin bevitelével mértük, amelyet 5 pg/ml mennyiségben adagoltunk 24 órával a tenyésztés befejezése előtt. A mérést szemtillácíós- -számlálóval végeztük, és az eredményt beatés/perc <CFM) értékben adtuk meg. Az eredményeket a 2. táblázatban foglaltuk össze.

A pepiid biztonságosságának ellenőrzésére akut toxicitási vizsgálatot végeztünk az Orosz
Föderáció Farmakológia! Bizottságának módszertan! ajánlása szerint (,JJj gyógyhatású anyagok általános toxikus hatása prekhnika! vizsgálatának kívánalmai”: 1985),
A kapott eredmények szerint a pepiid 1000-szeres iatraperitoneális dózisa nem. okozott akut toxikus hatást, és ion tndtuk elérni az Wjo értéket.
A találmány szerinti biológiailag aktív poptidek lehat széles körben használhatók a humán és állatgyógyászatban.
