Claims (8)
1. In vitro ili ex vivo postupak proizvodnje T stanica koji sadrži:
(a) aktiviranje populacije T stanica i stimulaciju populacije T stanice da proliferiziraju, naznačen time da su koraci aktivacije i stimulacije izvedeni uz prisutnost PI3K inhibitora;
(b) transdukcije T stanica s virusnim vektorom koji sadrži konstruirani T stanični receptor (TCR) ili kimerni antigen receptor (CAR); i
(c) kultiviranje transduciranih T stanica da proliferiraju; naznačeno time da koraci aktivacije i stimulacije izvedeni uz prisutnost PI3K inhibitora rezultiraju u održavanju proliferacije i smanjenju diferencijacije transduciranih T stanica u usporedbi s proliferacijom i diferencijacijom transduciranih T stanica koje su aktivirane i stimulirane uz odsutnost PI3K inhibitora.
2. Postupak prema patentnom zahtjevu 1, naznačen time da
(a) izvor T stanica su mononuklearne stanice periferne krvi;
(b) aktiviranje T stanica uključuje kontakt T stanica s anti-CD3 antitijelom ili njegovim CD3-vezujućim fragmentom;
(c) stimulacija T stanica sadrži dovođenje u kontakt T stanica s anti-CD28 antitijelom ili njegovim CD28-vezujućim fragmentom, B7-1 ili njegovim CD28- vezujućim fragmentom, ili B7-2 ili njegovim CD28-vezujućim fragmentom;
(d) T stanice su transducirane s virusnim vektorom prije proliferacije T stanica; ili
(e) T stanice su transducirane s virusnim vektorom nakon proliferacije T stanica.
3. Postupak prema bilo kojem od patentnih zahtjeva 1 do 2, naznačen time da:
(a) virusni vektor je retrovirusni vektor;
(b) virusni vektor je lentivirusni vektor; ili
(c) T stanice sadrže CAR.
4. Postupak prema patentnom zahtjevu 3, naznačen time da CAR sadrži
a) ekstracelularnu domenu koja i veže antigen izabran iz skupine sastavljene od: BCMA, alfa folat receptor, 5T4, αvβ6 integrin, B7-H3, B7-H6, CAIX, CD19, CD20, CD22, CD30, CD33, CD44, CD44v6, CD44v7/8, CD70, CD79a, CD79b, CD123, CD138, CD171, CEA, CSPG4, EGFR, EGFR obitelj uključujući ErbB2 (HER2), EGFRvIII, EGP2, EGP40, EPCAM, EphA2, EpCAM, FAP, fetalni AchR, FRα, GD2, GD3, Glipikan-3 (GPC3), HLA-A1+MAGE1, HLA-A2+MAGE1, HLA-A3+MAGE1, HLAA1+ NY-ESO-1, HLA-A2+NY-ESO-1, HLAA3+ NY-ESO-1, IL-11Rα, IL-13Rα2, Lambda, Lewis-Y, Kapa, Mezotelin, Mucl, Muc16, NCAM, NKG2D Ligande, NY-ESO-1, PRAME, PSCA, PSMA, ROR1, SSX, Survivin, TAG72, TEMs, i VEGFR2;
b) transmembransku domenu izveden iz polipeptida odabranog iz grupe koja se sastoji od: CD8α; CD4, CD28, CD45, PD-1, i CD152;
c) jedan ili više intracelularnih kostimulatornih signalnih domena izabrana iz skupine sastavljene od: CD28, CD54 (ICAM), CD134 (OX40), CD137 (41BB), CD152 (CTLA4), CD273 (PDL2), CD274 (PD-L1), i CD278 (ICOS); i
d) CD3ζ signalna domena.
5. Postupak prema ptentnom zahtjevu 4, naznačen time da:
(a) ekstracelularni domen sadrži an antitijelo ili antigen vezujući fragment koji veže antigen;
(b) transmembranska domena je izvedena od CD8α ili CD28;
(c) jedan ili više kostimulatornih signalnih domena je izabranih iz skupine sastavljene od: CD28, CD134, i CD137;
(d) CAR dodatno sadrži polipeptid zglobnog regiona;
(e) CAR dodatno sadrži polipeptid zglobnog područja koji sadrži zglobno područje IgG1 ili CD8α;
(f) CAR dodatno sadrži signalni peptid;
(g) CAR dodatno sadrži signalni peptid koji sadrži signalni polipeptid IgG1 teškog lanca ili CD8α signalni polipeptid.
6. Postupak prema bilo kojem od patentnih zahtjeva 1 do 5, naznačen time da PI3K inhibitor je:
(a) pan-PI3K inhibitor izabran iz skupine sastavljene od: BEZ235, LY294002, i GDC- 0941;
(b) selektivni PI3K inhibitor izabran iz skupine sastavljene od: BYL719, GSK2636771, TGX-221, AS25242, CAL-101, i IPI-145; ili
(c) ZSTK474.
7. Postupak prema bilo kojem od patentnih zahtjeva 1 do 6, naznačen time da populacija T stanica aktivirana i stimulirana uz prisutnost PI3K inhibitora ima povećani broj T stanica koje eksprimiraju jedan ili više markera izabran iz skupine sastavljene od: CD62L, CCR7, CD28, CD27, CD122, i CD127 u usporedbi s populacijom T stanica aktiviranih i stimuliranih u odsutnosti PI3K inhibitora.
8. Postupak prema bilo kojem od patentnih zahtjeva 7, naznačeno time da populacija T stanica aktiviranih i stimuliranih uz prisutnost PI3K inhibitora ne eksprimira CD57 ili KLRG1 ili eksprimira manje CD57 ili KLRG1 u usporedbi s populacijom T stanica aktiviranih i stimuliranih u odsutnosti PI3K inhibitora.
