Claims (15)
1. Enzimski postupak za pripremu celobioze i fruktoze iz saharoze, naznačen time da postupak sadrži korake:
(a) dobivanje prvog pripravka edukta koji sadrži saharozu fosforilazu, saharozu i fosfat;
(b) reagiranje barem dijela saharoze sadržane u prvom pripravku edukta sa najmanje dijelom fosfata sadržanog u prvom pripravku edukta s katalizom pomoću saharoze fosforilaze, pri čemu je dobiven prvi pripravak proizvoda koji sadrži fruktozu i glukozu 1-fosfat;
(c) dobivanje drugog pripravka edukta koji sadrži celobiozu fosforilazu, glukozu i najmanje dio glukoze 1-fosfat dobivene u koraku (b); i
(d) reakciju glukoze s barem dijelom glukoze 1-fosfata sadržanog u drugom pripravku edukta s katalizom pomoću celobioze fosforilaze, gdje je dobiven drugi pripravak proizvoda koji sadrži celobiozu i fosfat;
pri čemu se koraci (b) i (d) izvode u zajedničkom reaktoru; a gdje saharoza fosforilaza i celobioza fosforilaza nisu imobilizirane na čvrstom nosaču, niti su zatvorene u matricu.
2. Postupak prema zahtjevu 1, naznačen time da se saharoza fosforilaza, i/ili celobioza fosforilaza recikliraju.
3. Postupak prema zahtjevu 1 ili 2, naznačen time da se provodi u jednom membranskom reaktoru.
4. Postupak prema zahtjevu 3, naznačen time da saharoza fosforilaza, i/ili celobioza fosforilaza ne mogu proći membranu membranskog reaktora.
5. Postupak prema bili kojem od prethodnih zahtjeva, naznačen time da se koraci (b) i (d) izvode barem djelomično istovremeno.
6. Postupak prema bilo kojem od prethodnih zahtjeva, naznačen time da saharoza fosforilaza je saharoza fosforilaza divljeg tipa bakterije koja pripada rodu Actinobacteria, ili saharoza fosforilaza koja se razlikuje od navedenog divljeg tipa, ali koja sadrži sekvencu aminokiseline koja ima najmanje 80% identiteta u odnosu na saharozu fosforilazu divljeg tipa bakterije koja pripada rodu Actinobacteria.
7. Postupak prema bilo kojem od prethodnih zahtjeva, naznačen time da celobioza fosforilaza je celobioza fosforilaza divljeg tipa bakterije koja pripada rodu Actinobacteria, ili celobioza fosforilaza koja se razlikuje od navedenog divljeg tipa, ali koja sadrži sekvencu aminokiseline koja ima najmanje 80% identiteta u odnosu na celobiozu fosforilazu divljeg tipa bakterije koja pripada rodu Actinobacteria.
8. Postupak prema bilo kojem od prethodnih zahtjeva, naznačen time da je koncentracija saharoze u prvom pripravku edukta danog u koraku (a) najmanje 0.150 mol/l; po mogućnosti najmanje 0.200 mol/l; poželjnije najmanje 0.500 mol/l.
9. Postupak prema bilo kojem od prethodnih zahtjeva, naznačen time da je koncentracija celobioze u drugom pripravku proizvoda dobivenog u koraku (d) najmanje 0.075 mol/l, po mogućnosti najmanje 0.150 mol/l; poželjnije najmanje 0.200 mol/l.
10. Postupak prema bilo kojem od prethodnih zahtjeva, naznačen time da korak (b) dodatno sadrži pomoćni korak (b') koji odvaja barem dio rezultirajuće glukoze 1-fosfat tijekom reakcije saharoze u glukozu 1-fosfat.
11. Postupak prema zahtjevu 10, naznačen time da se u koraku (b') glukoza 1-fosfat odvaja elektrodijalizom i/ili kristalizacijom.
12. Postupak prema bilo kojem od prethodnih zahtjeva, naznačen time da sadrži dodatni korak: (g) odvajanje fosfata dobivenog u koraku (d) i opcionalno vraćanje u korak (a).
13. Postupak prema bilo kojem od zahtjeva 12, naznačen time da je fosfat izdvojen pomoću elektrodijalize.
14. Postupak prema zahtjevu 12 ili 13, naznačen time da je najmanje 50% fosfata koji je izdvojen u koraku (g) vraćeno u korak (a) kao barem dio fosfata koji je sadržan u prvom pripravku edukta.
15. Postupak prema bilo kojem od zahtjeva 12 d o14, naznačen time da se korak (g) izvodi vremenski nakon koraka (d).
