Abstract
Postupak proizvodnje mikrofolikula kose sisavaca naznačen time da sadrži korake: (a) osiguravanja barem jedne de novo papile, (b) osiguravanja barem jedne druge stanične populacije koja je odabrana iz skupine fibroblasta, keratinocita i/ili melanocita, te (c) kokultivacije de novo papila sa barem jednom drugom staničnom populacijom kod ne-adherentnih uvjeta kultivacije. Patent sadrži još 5 patentnih zahtjeva.
Claims (6)
1. Postupak proizvodnje mikrofolikula kose sisavaca naznačen time da sadrži korake:
(a) osiguravanja barem jedne de novo papile,
(b) osiguravanja barem jedne druge stanične populacije koja je odabrana iz skupine fibroblasta, keratinocita i/ili melanocita, te
(c) kokultivacije de novo papila sa barem jednom drugom staničnom populacijom kod ne-adherentnih uvjeta kultivacije.
2. Postupak prema zahtjevu 1, naznačen time da je barem jednu drugu staničnu populaciju moguće izvesti i/ili je izvedena iz folikula kose sisavaca, koja je odabrana iz skupine fibroblasta ovojnice vezivnog tkiva, keratinocita i/ili melanocita.
3. Postupak prema zahtjevu 1 ili 2, naznačen time da je de novo papila iz koraka (a) prevučena sa izvanstaničnim matričnim proteinima, poželjno kolagenom IV, fibronektinom i/ili lamininom.
4. Postupak prema jednom od zahtjeva 1 do 3, naznačen time da su de novo papile dobivene prema jednom od postupaka prema bilo kojem od zahtjeva 5 do 6.
5. Postupak proizvodnje de novo papila upotrebljivih u postupku prema jednom od zahtjeva 1 do 4, naznačen time da sadrži korake:
(a) osiguravanja barem jedne dermalne papile (DP) iz barem jedne folikule kose sisavaca,
(b) izoliranja fibroblasta dermalne papile kose (DPF) iz DP pomoću mehaničkog učvršćivanja navedene DP na površini posude sa staničnom kulturom, pri čemu je bazična lamina perforirana kako bi se omogućila migracija DPF-ova prema van,
(c) ekspandiranja izoliranih DPF-ova u jednoslojnoj kulturi bez kolagenske prevlake, pri čemu su navedeni DPF-ovi barem jednom pomaknuti,
(d) kondenziranja ekspandiranih DPF-ova u stanične agregate koji iskazuju veličinu i oblik fiziološke DP, pri čemu su navedeni DPF-ovi diferencirani u ne-adhezivnim posudama sa kulturom u staničnoj koncentraciji na površini posude od 1.000 do 100.000 DPF/cm2, te
(e) prevlačenja de novo papila sa izvanstaničnim matričnim proteinima, poželjno kolagenom IV, fibronektinom i/ili lamininom.
6. Postupak prema zahtjevu 5, naznačen time da su ekspandirani DPF-ovi kondenzirani tijekom barem 48 sati.
