Abstract
Medicinal preparations which are efficaious against bacterial toxins are prepared from liver or spleen by subjecting the tissue to autolysis without addition of acid and then extracting the autolysate with water, or a water-soluble organic solvent, with the addition of a little acid and at a low temperature, and treating the solution with an agent to eliminate ballast substances, particularly proteins. The clear liquid obtained may be dried, and the residue extracted with concentrated methyl alcohol to yield a solution which is dried. Pure or aqueous methyl alcohol may be used as the extracting agent for the autolysate, and iron dialysate or lead acetate may be used to eliminate ballast substances. The Specification as open to inspection under Sect. 91 refers also to extraction of the liver or spleen of normal animals for a prolonged time at low temperature at a pH between 4 and 6 without previous autolysis, and to simple extraction at a feebly acid reaction of the organs of immunized animals; it states also that organic solvents without addition of acid may be used for the extraction and that the extracts so obtained are poor in ballast substances such as albumin and electrolytes. An example relating to the extraction of the spleen of an immunized animal with N/100 hydrochloric acid, followed by purification by means of iron dialysate is included. This subject-matter does not appear in the Specification as accepted.
