La présente invention concerne un dérivé de la
théophylline et un dérivé de la théobromine, et des procé-
dés pour leur préparation. Ces composés sont nouveaux et
doués de propriétés pharmacologiques très intéressantes.
Ces dérivés de la théophylline et de la théobro- mine répondent à la formule

Dans les définitions de R, X1, X2, Y1 et Y2 de la formule générale [I], l'expression "groupe alkyle inférieur" désigne des groupes alkyle linéaires ou ramifiés en C1 à C6, tels que les groupes méthyle, éthyle, npropyle, isopropyle,
isobutyle, 1-méthylpropyle, tert-butyle, n-pentyle, 1-éthyl-
propyle, isoamyle et n-hexyle. L'expression "groupe alcoxy inférieur" désigne des groupes alcoxy correspondant aux

Parmi les dérivés de la théobromine définis ci-

Le composé [I] de la présente invention peut aisé-
ment être transformé en un sel d'addition avec un acide par
réaction avec un acide minéral ou organique pharmacologique-
ment acceptable. Comme acides minéraux, on peut citer l'a-
cide -chlorhydrique, l'acide bromhydrique, l'acide iodhydri-
que et l'acide sulfurique, et comme acides organiques, l'a-
cide maléique, l'acide fumarique, l'acide succinique, l'a-
cide acétique, l'acide malonique, l'acide citrique et l'a-
cide benzoique.

Les dérivés de la théophylline et de la théobro-
mine de l'invention sont des composés nouveaux non décrits dans la littérature. Ils ont des effets vasodilatateurs
et d'augmentation du débit sanguin très prononcés et amé-
liorent efficacement la circulation dans les artères céré- brales et coronaires et dans les vaisseaux capillaires. En outre, les composés de l'invention ont une action de régulation de la coagulation des plaquettes du sang. Ils sont donc utilisables comme remèdes dans diverses maladies provoquées par des perturbations du flux sanguin dans les
vaisseaux capillaires, par des troubles des vaisseaux san-
guins cérébraux et leurs séquelles, par l'angine de poi-
trine et par l'infarctus du myocarde. En outre, les dérivés de la théophylline et de la théobromine de l'invention ont d'autres effets pharmacologiques très intéressants: une
action sur le système nerveux central, une action anti-
histaminique, une action analgésique, une action anti-
asthmatique et une action hypotensive. En conséquence,
les composés de l'invention peuvent être utilisés effica-
cement comme psychodynamisants, agents anti-histaminiques, agents analgésiques, agents anti-asthmatiques et agents hypotenseurs.
Les composés III de l'invention peuvent se prépa-
rer par divers procédés. On adopte ordinairement un procédé représenté par le schéma réactionnel suivant

sulfonyloxy, et R, X, Z et n sont tels que définis ci-
dessus. En bref, un composé III de l'invention peut être obtenu en faisant réagir un composé de formule générale V
avec un composé de-formule générale VI.
La réaction est effectuée en l'absence de solvant
ou en présence d'un solvant ne participant pas à la réac-
tion, choisi de manière adéquate parmi des alcools infé-
rieurs tels que le méthanol, l'éthanol, le propanol et l'isopropanol, des solvants benzéniques tels que le benzène, le toluène et le xylène et des éthers tels que l'éther
éthylique et le tétrahydrofurane. La réaction peut s'effec-
tuer même à la température ambiante, mais on préfère opérer à chaud, à une température pouvant aller jusqu'au point d'ébullition du solvant. La réaction peut être effectuée de façon plus douce en ajoutant un agent de fixation des acides tels que la triéthylamine, un bicarbonate de métal alcalin, un carbonate de métal alcalin ou la pyridine au
mélange réactionnel.

parer par divers procédés. On adopte ordinairement un pro-

-. 5cH)n - N N -Z IV dans lequel X désigne un atome d'halogène ou un groupe p-toluène-sulfonyloxy, et R, Z et n sont tels que définis cidessus. En bref, un composé IV de l'invention peut être obtenu en faisant réagir un composé de formule générale
VII avec un composé de formule générale VIII.
Cette réaction est effectuée en l'absence de sol-
vant ou en présence d'un solvant ne participant pas à la
réaction, choisi de manière adéquate parmi des alcools infé-
rieurs tels que le méthanol, l'éthanol, le propanol et l'isopropanol, des solvants benzéniques tels que le benzène, le toluène et le xylène et des éthers tels que l'éther
éthylique et le tétrahydrofurane. La réaction peut s'effec-
tuer même à la température ambiante, mais on préfère opérer à chaud, à une température pouvant aller jusqu'au point d'ébullition du solvant. La réaction peut être effectuée CH= -J de façon plus douce en ajoutant un agent de fixation des acides tel que la triéthylamine, un bicarbonate de métal alcalin, un carbonate de métal alcalin ou la pyridine au
mélange réactionnel.
On décrira à présent les remarquables effets phy- siologiques des dérivés de la théophylline de l'invention
en se référant à des composés typiques.
Action d'augmentation du flux sanguin.
1 - Méthodes.
On utilise comme animaux d'essai des chiens bâtards mâles et femelles d'un poids corporel de 8 à 20 kg, et on mesure les débits sanguins dans les artères vertébrales et fémorales. A cet effet, on fixe des sondes d'un débit-mètre électromagnétique (modèle MF-27 de la Société Nippon Koden)
sur une artère vertébrale latérale et sur une artère fémo-
rale latérale d'un chien anesthésié par du diéthyl barbital
(240 mg/kg, injection hypodermique) et du pentobarbital so-
dique (10 mg/kg, injection intraveineuse), et on mesure
simultanément les débits sanguins des deux artères. On admi-
nistre le composé d'essai par piqûre dans une artère à la
dose de 0,1, 1 ou 10 lig par kg de poids corporel.
2 - Résultats.
Les résultats obtenus sont donnés dans le tableau 2. L'intensité de l'augmentation du débit sanguin dont il est question dans le tableau 2 est déterminée en utilisant de la papavérine comme échantillon de référence. L'intensité de l'échantillon ayant une dose efficace minimale de 0,1 i'g/kg est-désignée par A, et celle d'un échantillon ayant une dose efficace minimale de 1,0 vg/kg est désignée par B. Les échantillons ayant des doses minimales efficaces de
lig/kg sont répartis en 2 groupes, l'un ayant une inten-
sité supérieure à celle de la papavérine. L'intensité du premier groupe est désignée par C, et celle du second est désignée par D. L'intensité de l'échantillon ayant une activité nulle est désignée par E. Les résultats concernant
l'augmentation du débit sanguin et la durée d'action, ob-
tenus dans 10 cas en utilisant la papavérine, sont donnés
dans le tableau 1.

Exemple 1.
Synthèse du chlorhydrate de 7-'2-[4-p-chlorobenzhydrylpi-
pérazinyl ( 1)7-éthyl -théophylline. On agite dans du benzène sous reflux, pendant 18,5
heures, 6,3 g de 7-(2-brométhyl)théophylline, 5,7 g de 1-
(p-chlorobenzhydryl)pipérazine et 4,0 g de triéthylamine.
On élimine le chlorhydrate de triéthylamine par filtration
et on extrait le filtrat par de l'acide chlorhydrique dilué.
On alcalinise l'extrait par de l'hydroxyde de sodium dilué, puis on l'extrait par du chloroforme. On lave la couche chloroformique à l'eau et on la sèche sur du carbonate de potassium anhydre.On élimine le solvant par distillation et on transforme les cristaux bruts résiduels en chlorhydrate
de la manière habituelle. On fait recristalliser le chlorhy-
drate dans du méthyl cellosolve et de l'eau, et l'on obtient
4,8 g du chlorhydrate de 7- 2-[4-p-chlorobenzhydrylpipéra-
zinyl-(1l-éthyl} -théophylline désiré (rendement 42,5 %).
Pf = 250-252 C.
Composition élémentaire pour C26H2902N6CH.2HCl:
Calculé: C = 55,16 %, H = 5,53 %, N = 14,85 Trouvé C = 55,19 %, H = 5,38 %, N = 14,87 %.
ExemDle 2.
Synthèse de la 7- 4-/4-o-méthoxyphénylpipérazinyl-(1)]-n-
butyl1 -théophylline.
On agite dans du benzène sous reflux pendant 18 heures 6,9 g de 7-(4bromo-n-butyl)théophylline, 3,8 g d'o-méthoxyphénylpipérazine et 4,0 g de triéthylamine. Les traitements ultérieurs sont effectués de la même manière qu'à l'exemple 1. On fait recristalliser les cristaux bruts
* obtenus dans de l'éthanol, et l'on obtient 3,8 g de la 7-
14-[4-o-méthoxyphénylpipérazinyl- ( l?7-n-butyl} -théophylline
désirée (rendement 37,6 %).
Pf = 117-118 C.
Composition élémentaire pour C22H3003N6:
Calculé: C = 61,94 %, H = 7,10 %, N = 19,71 %.
Trouvé: C = 62,10 %, H = 7,21%, N = 19,86 %.
Exemple 3.
Synthèse de la 7- 5-[4-o,m-diméthylphénylpipérazinyl-(1)! n-heptyl1théophylline. On agite dans du toluène sous reflux pendant 11,5 heures 9, 9 g de 1-(5-bromo-n-heptyl)théophylline, 3,8 g d'o,mdiméthylphénylpipérazine et 4,0 g de triéthylamine, et on effectue les traitements ultérieurs comme à l'exemple 1. On fait recristalliser les cristaux bruts obtenus dans
de l'éthanol, et l'on obtient 4,3 g de la 7-[5-{4-o,m-dimé-
thylphénylpipérazinyl- ( 1)j-n-heptyl -théophylline désirée.
Pf = 115-117oC.
Composition élémentaire pour C24H3402N6:
Calculé: C = 65,71 %, H = 7,38 %, N = 19,16 %.
Trouvé: C = 65,42 t, H = 7,92 %, N = 19,31 %.
Exemple 4.
Synthèse du chlorhydrate de 7-t7-/(3-méthyl-4-m-méthylphé-
nyl)-pipérazinyl- (l)j-n-heptyl) -théophylline.
On agite dans du toluène sous reflux pendant 11 heures 7,8 g de 7-(7bromo-n-heptyl)théophylline, 3,8 g de N-(m-méthylphényl)-2-méthyl-Npipérazine et 4,0 g de triéthylamine. On effectue les traitements ultérieurs comme à l'exemple 1, et l'on obtient 10 g de cristaux bruts. On purifie les cristaux bruts obtenus par chromatographie sur gel de silice et on les transforme en chlorhydrate par les procédés habituels. On obtient 5,3 g du chlorhydrate de
7-{7-Z3-méthyl(4-m-méthylphényl)pipérazinyl-(1)7-n-heptyl -
théophylline désiré.
Pf = 222-265 C.

Synthèse de la 1-t4-f4-o,m-diméthylphénylpipérazinyl(1)]7
-n-butyl3 -théobromine.
On agite dans du toluène sous reflux pendant 13 heures 9,5 g de 1-(4bromo-n-butyl)théobromine, 3,8 g
d'o,m-diméthylphénylpipérazine et 4,0 g de triéthylamine.
On élimine le chlorhydrate de triéthylamine par filtration et on extrait le filtrat par de l'acide chlorhydrique
dilué. On alcalinise l'extrait par de l'hydroxyde de so-
dium dilué, puis on l'extrait par le chloroforme. On lave la couche chloroformique à l'eau et on la sèche sur du carbonate de potassium anhydre. On élimine le solvant par distillation et on fait recristalliser les cristaux bruts résiduels dans le méthyl cellosolve. On obtient 3,7 g de
la 1- {4-[4-o,m-dimiéthylphénylpipérazinyl-( 1)7-n-butyl3 -
théobromine désirée (rendement 43,64 %).
Pf = 134-135oC.
Composition élémentaire pour C23H3202N6:
Calculé: C = 65,05 %, H = 7,61%, N = 19,80 %.
Trouvé: C = 65,11 %, H = 7,72 %, N = 19,46 %.
Exemple 97.
Synthèse du chlorhydrate de 1-j5-[4-benzhydrylpipérazinyl-
(1)7-n-pentyl3 -théobromine.
On agite dans du toluène sous reflux pendant 30 heures 7,9 g de 1-(5bromo-n-pentyl)théobromine, 5,0 g de
N-benzhydrylpipérazine et 4,0 g de triéthylamine. On effec-
tue les traitements ultérieurs de la même manière qu'à l'exemple 96. On transforme les cristaux bruts obtenus
en chlorhydrate par les procédés habituels, et l'on ob-
tient le chlorhydrate de 1-5-f4-benzhydrylpipérazinyl-(1)]
-n-pentyl -théobromine désiré (rendement 40, 9 %).
Pf = 262-264 C (décomp.).
Composition élémentaire pour C29H3602N6.2HCl:
Calculé: C = 60,93 %, H = 6,71 % N = 14,71%.
Trouvé: C = 60,57 %, H = 7,15 %, N = 14,65 %.
Exemple 98.
Synthèse de la 1-[7-[4-o-méthoxyphénylpipérazinyl-(1)7-n-
heptyl} -théobromine.
On agite dans du toluène sous reflux pendant 11,5 heures 7,5 g de 1-(7bromo-n-heptyl)théobromine,
3,8 g de N-o-méthoxyphénylpipérazine et 4,0 g de triéthyl-
amine, et on effectue les traitements ultérieurs de la même manière qu'à l'exemple 96. On purifie les cristaux bruts obtenus par chromatographie sur gel de silice et l'on obtient 4,6 g de la 1-{7-[4-ométhoxyphénylpipérazinyl-(1)i
-n-heptyl3-théobromine désirée (rendement 49,1 %).
Pf = 97-98oC.
Composition élémentaire pour C25H3603N6:
Calculé: C = 64,08 X, H = 7,74 %, N = 17,94 %.
Trouvé: C = 63,90 %, H = 7,67 %, N = 18,01%.

suivant la revendication 1, associé à un véhicule pharma-
cologiquement acceptable.
