Abstract

Forma de dosificación farmacéutica administrable por vía oral para tratar a un paciente con un riesgo cardiovascular elevado, que comprende una combinación de: (a) una dosificación diaria terapéuticamente eficaz de un agente reductor del colesterol, como primer agente activo; (b) una dosificación diaria terapéuticamente eficaz de un inhibidor del sistema renina-angiotensina, como segundo agente activo; y (c) una dosificación diaria terapéuticamente eficaz de aspirina, como tercer agente activo; en la que por lo menos uno de los agentes activos está presente en una unidad de dosificación que separa físicamente por lo menos a un agente activo de los otros agentes activos, y en la que las unidades de dosificación diaria eficaces de cada uno de los agentes activos de la combinación presentan diferentes perfiles de liberación.

Claims (31)

1. Forma de dosificación farmacéutica administrable por vía oral para tratar a un paciente con un riesgo cardiovascular elevado, que comprende una combinación de:
(a)
una dosificación diaria terapéuticamente eficaz de un agente reductor del colesterol, como primer agente activo;
(b)
una dosificación diaria terapéuticamente eficaz de un inhibidor del sistema renina-angiotensina, como segundo agente activo; y
(c)
una dosificación diaria terapéuticamente eficaz de aspirina, como tercer agente activo;
en la que por lo menos uno de los agentes activos está presente en una unidad de dosificación que separa físicamente por lo menos a un agente activo de los otros agentes activos, y
en la que las unidades de dosificación diaria eficaces de cada uno de los agentes activos de la combinación presentan diferentes perfiles de liberación.
2. Forma de dosificación según la reivindicación 1, en la que por lo menos dos de los agentes activos están presentes en las unidades de dosificación que separan físicamente dichos por lo menos dos agentes activos de los otros agentes activos y entre sí.
3. Forma de dosificación según la reivindicación 1, en la que el agente reductor de colesterol está presente en una primera unidad de dosificación, el inhibidor del sistema renina-angiotensina está presente en una segunda unidad de dosificación, y la aspirina está presente en una tercera unidad de dosificación.
4. Forma de dosificación según la reivindicación 3, en la que la forma de dosificación es una cápsula, y cada una de las unidades de dosificación comprende una pluralidad de perlas o gránulos contenidos en la misma.
5. Forma de dosificación según la reivindicación 3, en la que la forma de dosificación es una cápsula, y cada una de las unidades de dosificación comprende un comprimido contenido en la misma.
6. Forma de dosificación según la reivindicación 3, en la que la forma de dosificación es un comprimido, y cada una de las unidades de dosificación comprende un segmento aislado físicamente del mismo.
7. Forma de dosificación según la reivindicación 6, en la que el comprimido es un comprimido en capas, y cada una de las unidades de dosificación comprende, una capa del mismo.
8. Forma de dosificación según la reivindicación 3, en la que la forma de dosificación es un comprimido, y cada unidad de dosificación comprende una pluralidad de perlas o gránulos contenidos en una mezcla en el mismo.
9. Forma de dosificación según la reivindicación 3, en la que la forma de dosificación está constituida por un núcleo interno recubierto con por lo menos dos capas concéntricas.
10. Forma de dosificación según la reivindicación 9, que contiene dos capas concéntricas.
11. Forma de dosificación según la reivindicación 10, en la que el núcleo interno está constituido por una de las unidades de dosificación y cada capa concéntrica contiene una de las unidades de dosificación.
12. Forma de dosificación según la reivindicación 9, en la que el núcleo interno está constituido por un material inerte.
13. Forma de dosificación según la reivindicación 12, que contiene tres capas concéntricas.
14. Forma de dosificación según la reivindicación 13, en la que cada capa concéntrica contiene una de las unidades de dosificación.
15. Forma de dosificación según cualquiera de las reivindicaciones 2 a 14, en la que por lo menos una de las unidades de dosificación es una unidad de dosificación de liberación inmediata.
16. Forma de dosificación según cualquiera de las reivindicaciones 2 a 15, en la que por lo menos una de las unidades de dosificación es una unidad de dosificación de liberación retardada.
17. Forma de dosificación según cualquiera de las reivindicaciones 2 a 15, en la que por lo menos una de las unidades de dosificación es una unidad de dosificación de liberación sostenida.

18. Forma de dosificación según cualquiera de las reivindicaciones 1 a 17, que comprende además por lo menos una de entre la vitamina B_{6}, la vitamina B_{12} y el ácido fólico.
19. Forma de dosificación según cualquiera de las reivindicaciones 1 a 18, en la que el agente reductor de colesterol se selecciona de entre el grupo constituido por inhibidores de la HMG CoA reductasa, secuestrantes del ácido biliar, probucol, agentes del ácido fíbrico y combinaciones de los mismos.
20. Forma de dosificación según la reivindicación 19, en la que el agente reductor de colesterol es un inhibidor de la HMG CoA reductasa.
21. Forma de dosificación según la reivindicación 20, en la que el inhibidor de la HMG CoA reductasa se selecciona de entre el grupo constituido por atorvastatina, cerivistatina, fluindostatina, fluvastatina, lovastatina, mevastatina, pravastatina, simvastatina y velostatina.
22. Forma de dosificación según la reivindicación 21, en la que el inhibidor de la HMG CoA reductasa se selecciona de entre el grupo constituido por lovastatina y pravastatina.
23. Forma de dosificación según la reivindicación 19, en la que el inhibidor del sistema renina-angiotensina se selecciona de entre el grupo constituido por inhibidores de la enzima convertidora de la angiotensina (ACE) y antagonistas de la angiotensina II.
24. Forma de dosificación según la reivindicación 23, en la que el inhibidor del sistema renina-angiotensina es un inhibidor de ACE.
25. Forma de dosificación según la reivindicación 24, en la que el inhibidor de ACE se selecciona de entre el grupo constituido por captoprilo, cilazaprilo, delaprilo, enalaprilo, fentiaprilo, fosinoprilo, indolaprilo, lisinoprilo, perindoprilo, pivoprilo, quinaprilo, ramiprilo, espiraprilo, trandolaprilo y zofenoprilo.
26. Forma de dosificación según la reivindicación 25, en la que el inhibidor de ACE es el enalaprilo.
27. Forma de dosificación según la reivindicación 23, en la que el inhibidor del sistema renina-angiotensina es un antagonista de la angiotensina II.
28. Forma de dosificación según la reivindicación 20, en la que la primera unidad de dosificación contiene aproximadamente 10 a aproximadamente 120 mg de un inhibidor de la HMG CoA reductasa seleccionado de entre el grupo constituido por atorvastatina, fluvastatina, lovastatina, pravastatina y simvastatina, la segunda unidad de dosificación contiene aproximadamente 1 a aproximadamente 60 mg de un inhibidor del sistema renina-angiotensina seleccionado de entre el grupo constituido por captoprilo, enalaprilo, fosinoprilo, lisinoprilo, quinaprilo, ramiprilo, trandolaprilo y zofenoprilo, y la tercera unidad de dosificación contiene aproximadamente 20 a aproximadamente 600 mg de aspirina.
29. Forma de dosificación según la reivindicación 28, que comprende además por lo menos uno de entre:
(a)
aproximadamente 25 mg a aproximadamente 75 mg de vitamina B_{6},
(b)
aproximadamente 0,25 mg a aproximadamente 2 mg de vitamina B_{12}, y
(c)
aproximadamente 1 mg a aproximadamente 8 mg de ácido fólico.
30. Kit acondicionado para un paciente con un riesgo cardiovascular elevado para su utilización en la autoadministración de formas de dosificación oral indicadas para éste, comprendiendo el kit un recipiente acondicionado que aloja una pluralidad de formas de dosificación oral según la reivindicación 28 ó 29 y las instrucciones para llevar a cabo la administración del fármaco con el mismo.
31. Kit acondicionado según la reivindicación 30, en el que el inhibidor de la HMG CoA reductasa es la lovastatina y el inhibidor del sistema renina-angiotensina es el enalaprilo.
