Abstract

Procedimiento para la separación de 14OH-10 -deacetil baccatina-III a partir de la parte recuperable de una planta de una especie Taxus por cristalización, que comprende las siguientes etapas: (a) extracción de las partes pulverizadas y opcionalmente secadas de una planta de la especie Taxus, con un alcohol alifático, (b) preparación de un extracto alcohólico parcialmente concentrado que contiene 10DAB, y una cantidad minoritaria de 14OH-10-DAB, (c) tratamiento del extracto con cetonas alifáticas y separación de los insolubles de la solución madre por centrifugación o filtración, (d) tratamiento del residuo obtenido después de la evaporación de solución madre de la etapa (c) en vacío con una mezcla de cetonas alifáticas y agua, y eliminación de los insolubles a través de una capa de celite por filtración o centrifugación, (e) extracción de la solución cetónica acuosa con hidrocarburos aromáticos para eliminar las sustancias menos polares, seguido de una extracción con ésteres alifáticos inmiscibles en agua o solventes clorinados, (f) evaporación de la capa orgánica de (e) hasta sequedad para obtener un residuo semisólido del cual se obtiene 10-DAB crudo por cristalizaciones selectivas, (g) aislamiento y purificación de 10-DAB, (h) evaporación de la solución madre de (g) y a continuación un tratamiento del residuo con un solvente nitrilo y de un hidrocarburo aromático, y (i) aislamiento y purificación de 14 -OH-10DAB por cristalización del solvente del residuo obtenido después del tratamiento con solvente de nitrilo seguido de solventes de hidrocarburos aromáticos.
