Abstract
Die Erfindung betrifft Uracilderivate sowie deren Anwendung als therapeutischer Wirkstoff. Die Uracilderivate werden dabei insbesondere zusammen mit einem Zytostatikum zur Unterdrückung oder Reduzierung der Resistenzenbildung bei der Zytostatikabehandlung eingesetzt.
Description
Uracilderivate und deren Verwendung
Die Erfindung betrifft Uracilderivate sowie deren Anwendung als therapeutischer Wirkstoff. Die Uracilde- rivate werden dabei insbesondere zusammen mit einem Zytostatikum zur Unterdrückung oder Reduzierung der Resistenzenbildung bei der Zytostatikabehandlung eingesetzt.
Standardtherapie für Krebserkrankungen ist die Chemotherapie. Zytostatika beeinflussen die Zellteilung und sind dementsprechend besonders toxisch für schnell wachsende Tumorzellen. Zytostatika induzieren Apoptose, d.h. sie führen zum Zelltod der Tumorzel- len. Leider ist die resistenzfreie Behandlungsperiode mit den gegenwärtig auf dem Markt befindlichen Zytostatika meist nicht lang genug, um den Tumor gänzlich zu vernichten. Zur Verbesserung dieser Situation wurden „Chemosensitizer" entwickelt, die bestehender Re- sistenz entgegenwirken.
Wird die Resistenz durch die Amplifikation (Vervielfachung) und Überexpression des „multi-drug resistan- ce"-Gens (MDR-I) hervorgerufen, kann diese durch In- aktivierung von dessen Genprodukt (P-Glykoprotein) vermindert werden (Takara K, Sakaeda T, Okumura K. An update on overcoming MDRl-mediated multidrug resis- tance in Cancer chemotherapy . Curr. Pharm. Des. 2006; 12 (3) :273-86) .
Schwere Nebenwirkungen standen bisher einem Einsatz von P-Glykoprotein-Hemmern im Wege. Substanzen der dritten Generation sind wahrscheinlich wegen ihrer toxischen Wirkung nur für eine KurzZeitbehandlung einsetzbar und auch nur bei den wenigen Tumoren, deren Resistenz ausschließlich auf der Wirkung des „multi-drug resistance"-Gens beruht. Darüber hinaus wurden Inhibitoren der Rezeptoren für Tyrosinkinase oder der Überexpression einzelner Onkogene entwi- ekelt. Behandelt werden können allerdings immer nur einige wenige geeignete Tumoren (Desoize B., Jardil- lier J., Multicellular resistance: a paradigm for clinical resistance? Crit Rev Oncol Hematol . 2000; 36:193-207) .
Aus der EP 0 806 956 sind 5-substituierte Nukleoside zur Hemmung der Resistenzbildung bei der Zytostatika- Behandlung bekannt. Bei den hier ausgeführten Verbindungen handelt es sich um (E) -5' - (2-Bromvinyl) -2 ' - desoxyuridin (BVDU) und (E) -5 ' - (2-Bromvinyl) uracil (BVU) .
Diese verhindern die Resistenzbildung und bekämpfen nicht schon existierende Resistenzen. Im Gegensatz zu den seit Jahrzehnten bekannten und meist erfolglosen Versuchen, schon existierende Chemoresistenzen zu um-
gehen oder zu mindern, gibt es für diesen technologischen Ansatz weltweit keine Konkurrenz (Fahrig, R. , Heinrich, J. C, Nickel, B., Wilfert, F., Leisser, C, Krupitza, G., Praha, C, Sonntag, D., Fiedler, B., Scherthan, H., and Ernst, H. Inhibition of induced chemoresistance by co-treatraent with (E) -5- (2-bro- movinyl) -2 ' -deoxyuridine (RPlOl). Cancer Res . 63 (2003) 5745 -5753) . Das erste Arzneimittel BVDU zeigte in zwei klinischen Studien mit Pankreas (Bauch- Speicheldrüsen) -Krebspatienten eine statistisch signifikante Wirkung. Die Wirkung der Kobehandlung von Zytostatika mit BVDU war wirkungsvoller als jede andere bisher beschriebene Chemotherapie (Fahrig, R. , Quietzsch, D., Heinrich, J. -C, Heinemann, V., Boeck, S., Schmid, R. M. , Praha, C, Liebert, A., Sonntag,
D., Krupitza, G., und Haenel, M.; RPlOl improves the efficacy of chemotherapy in pancreas Carcinoma cell lines and pancreatic Cancer patients . Anti-Cancer Drugs 17, 1045-1056, 2006) .
Daher war es Aufgabe der vorliegenden Erfindung, Substanzen bereitzustellen, die eine gegenüber den aus dem Stand der Technik bekannten Verbindungen höhere Wirksamkeit im Hinblick auf die Unterdrückung oder Reduzierung der Resistenzenbildung bei der Zytostati- ka-Behandlung aufweisen.
Diese Aufgabe wird durch die Uracilderivate mit den Merkmalen des Anspruchs 1 gelöst. In Anspruch 6 wer- den die Uracilderivate zur Anwendung als therapeutischer Wirkstoff bereitgestellt. In Anspruch 8 wird die Verwendung derartiger Uracilderivate zur Unterdrückung oder Reduzierung der Resistenzenbildung bei der Zytostatikabehandlung bereitgestellt. Die weite- ren abhängigen Ansprüche zeigen vorteilhafte Weiterbildungen auf.
Erfindungsgemäß werden Uracilderivate der allgemeinen Formel I bereitgestellt:

Sofern einer oder mehrere der Reste Ri, R2, R3 oder X ein Chiralitätszentrum aufweisen, sind im Rahmen der
vorliegenden Erfindung sowohl die reinen Enantiomere als auch deren Racemate umfasst.
Vorzugsweise sind für die Reste Ri und/oder R4 unabhängig voneinander die unsubstituierten oder substi- tuierten aromatischen Reste ausgewählt aus der Gruppe bestehend aus Methyl, Phenyl, Mono-, Di-, Tri-, Tetra- oder Pentahalogenphenyl , insbesondere 4-Fluor- phenyl, 2, 6-Difluorphenyl, 3 , 4-Difluorphenyl oder Perfluorphenyl, Mono-, Di- oder Tri-Ci-Ci8-Alkoxy- phenyl, insbesondere 4-Methoxyphenyl oder 3,4,5- Triraethoxyphenyl, Mono-, Di- oder Trinitrophenyl, insbesondere 4-Nitrophenyl, oder Vinyl .
Vorzugsweise ist R2 ausgewählt aus der Gruppe bestehend aus : a) Methyl, Ethyl, Propyl, 1, 2-Dichlor-2-hydroxy- ethyl, 1, 2-Dibrom-2-hydroxyethyl, l,2-Diiod-2- hydroxyethyl , b) (E) -2-Chlorvinyl, (E) -2-Bromvinyl, (E) -2-Iod- vinyl, 2 , 2-Dibromvinyl, (E) -2-Cyanovinyl, 2,2-Di- cyanovinyl, (E) -2-Nitrovinyl, 2 , 2-Dinitrovinyl ,

Es ist weiter bevorzugt, dass Ri4 unabhängig vonein- ander ausgewählt ist aus der Gruppe bestehend aus Pi- peridino, Piperazino, Morpholino, Piperidinomethyl, Piperazinomethyl und Morpholinomethyl .
Vorzugsweise sind die erfindungsgemäßen Verbindungen ausgewählt aus der Gruppe bestehend aus :
Erfindungsgemäß werden erstmals auch die zuvor beschriebenen Uracilderivate zur Anwendung als therapeutische Wirkstoffe bereitgestellt.
Diese werden bevorzugt zusammen mit mindestens einem Zytostatikum, in einer gemeinsamen Formulierung oder in getrennten Formulierungen eingesetzt.
Erfindungsgemäß wird auch die Verwendung der zuvor beschriebenen Uracilderivate sowie mindestens eines Zytostatikum zur Unterdrückung oder Reduzierung der Resistenzenbildung bei der Zytostatikabehandlung bereitgestellt.
Anhand der nachfolgenden Beispiele und Figuren soll der erfindungsgemäße Gegenstand im Detail beschrieben werden, ohne diesen auf die hier beschriebenen spe- ziellen Ausführungsformen einschränken zu wollen.
Die Figuren 1 bis 3 zeigen den Einfluss erfindungsgemäßer Verbindungen (gemäß den Beispielen 1 bis 24, wobei eine Zuordnung der einzelnen Verbindungen von A bis Y erfolgte) in Kombination mit Mitomycin C (MMC) im Vergleich zur alleinigen Verabreichung von MMC bzw. zur Verabreichung von MMC mit BVDU auf die ZeIl- zahl von AH13r-Zellen im zeitlichen Verlauf. AH13r- Zellen wurden dabei steigenden Dosen des Zytostatikums MMC ausgesetzt. Aus allen Figuren ist zu erken- nen, dass die Wirkung von MMC zusammen mit den erfin-
dungsgemäßen Verbindungen im Vergleich zu MMC und BVDU deutlich stärker ist.
Beispiele
1) Allgemeine Angaben
Die Bestimmung der Schmelzpunkte erfolgte am Mikro- heiztischmikroskop BOETIUS (VEB NAGEMA) . Die Werte sind korrigiert angegeben.
Die IR-Spektren wurden an einem FT-IR-Spektrometer PC 16 der Firma Perkin Eimer mit Kaliumbromid- Presslingen aufgenommen.

Zur Aufnahme der H- und C-NMR-Spektren wurde ein

Varian Mercury 300 Spektrometer ( H: 300MHz; C: 75MHz; F: 282 MHz), ein Bruker X400 ( H: 400MHz;

600MHz; C: 150MHz) verwendet. Als innerer Standard für die chemische Verschiebung δ, angegeben in ppm,
19 diente das jeweils verwendete Lösungsmittel. Bei F- NMR-Spektren wurde auf Trifluoressigsäure referen- ziert. Falls nicht anders angegeben, kam DMSO-d6 als Lösungsmittel zum Einsatz.
Die EI-Massespektren wurden mit einem Varian MAT CH6- Spektrometer (Fa. Thermo Electron) bzw. mit einem VG ZAB-HSQ-Spektrometer (Fa. Waters) aufgenommen. Zur Messung der ESI-LR-Massespektren diente ein Esquire 3000+ von Bruker Daltonics, die ESI-HR-Messungen er-
TM folgten am 7Tesla Apex II FT-ICR-Massenspektrometer der Firma Bruker Daltonics (kontinuierliche Fließinjektion mittels Spritzenpumpe, Flussrate 120μl/h, Spray-/Trockengas : Stickstoff). Bei den Peaks der
Massespektren werden das Masse/Ladungsverhältnis und die Zuordnung in eckigen Klammern angegeben. Bei 5-
Bromethinyl- und 5- (2, 2-Dibrom-vinyl) uracilderivaten sind zur eindeutigen Zuordnung die monoisotopischen Massepeaks angegeben.
Für säulenchromatographische Trennungen wurde Kiesel- gel 60 der Korngröße 0,063-0, 2mm der Firma Merck verwendet. Soweit nicht anders angegeben, wurde eine Säule mit den Maßen 3cm x 45cm mit 100g Kieselgelfüllung benutzt. Zur Trennung der Verbindungen K und J kam Kieselgel 60 der Korngröße 0, 040-0, 063mm der Fir- ma Merck zum Einsatz. Die analytische Dünnschichtchromatographie wurde an Kieselgel-DC-Folien der Firma Merck (Kieselgel 60 F254) durchgeführt. Die Detektion erfolgte durch Betrachten im UV-Licht (DESAGA HP-UVIS 254nm / 366nm) .
Folgende Fließmittelsysteme wurden für Dünnschicht- und Säulenchromatographie eingesetzt:
I Toluol / Aceton / Methanol 7 : 2 : 1
Dichlormethan / Methanol / etha- nol . Ammoniak
III Ethylacetat / n-Hexan 6 : 4
IV Ethylacetat / n-Hexan 7 : 3
V Ethylacetat / Methanol 9 : 1
Nachfolgende Substanzen wurden nach bestehenden Literaturvorschriften synthetisiert :

Um eine bessere Vergleichbarkeit der NMR-Daten zu gewähren, wurden die Atome der Verbindungen teilweise
abweichend zur bestehenden IUPAC- Nomenklatur durch- nummeriert .

Zu dieser Suspension gibt man 0,45ml (3,75mmol) Benzylbromid und rührt 12h bei Raumtemperatur (Reaktionskontrolle mit DC, Fließmittel He- xan/Ethylacetat = 5/1) . Der Ansatz wird mit 30ml E- ther versetzt und anschließend mehrmals im Scheide- trichter mit Wasser gewaschen. Die organische Phase wird über Na2SO4 getrocknet und abrotiert .

Die Reaktionsmischung wird 8h zum Rückfluss erhitzt, das Lösungsmittel im Vakuum abgezogen und der Ansatz mit 20ml kaltem Ethylacetat versetzt. Der auftretende Niederschlag wird abgesaugt .

5- (2 , 2-Dibromvinyl) -2 ' -desoxyuridin (α-und ß-Anomere, K und J)
590mg (2mmol) 5- (2 , 2-Dibromvinyl) uracil (3), 1,2ml BSA, sowie einige Tropfen Chlortrimethylsilan werden in 50ml abs . Dichlorethan suspendiert und bei Raumtemperatur gerührt bis sich eine klare Lösung gebil- det hat. Anschließend werden dem Ansatz 775mg
(1 , 8mmol) 2-Desoxy-3 , 5-di-O- (p-chlorbenzoyl) -D- ribofuranosylchlorid (4), gelöst in 20ml abs. Dichlorethan, und katalytische Mengen Zinn- (IV) -chlorid zugegeben. Die Umsetzung erfolgt bei Raumtemperatur un- ter Argonatmosphäre innerhalb von 12h. Nach Beendigung der Reaktion wird das Lösungsmittel im Vakuum entfernt und der Rückstand säulenchromatographisch aufgereinigt (Flussmittel III) . Es werden das α- und das ß-Anomer des 5- (2, 2-Dibromvinyl) -1- [2' -desoxy- 3 ', 5 ' -di-O- (p-chlorbenzoyl) ] uridins in einer Gesamtausbeute von 1,03g (74% d. Th.) erhalten. Die zuerst eluierte Substanz ist das ß-Anomer (590mg, 42% d. Th.) .
Zur Entfernung der Schutzgruppen werden 170mg (0,25mmol) ß-Anomer des 5- (2 , 2-Dibromvinyl) -1- [2 '- desoxy-3' , 5' -di-O- (p-chlorbenzoyl) ] uridins in 10ml 0 , IM Natriummethylatlösung suspendiert und auf 00C abgekühlt. Der Reaktionsverlauf wird mittels DC verfolgt (Flussmittel I) und nach ca. zweistündigem Rüh- ren stark saurer Ionenaustauscher für wasserfreies Medium zur Neutralisation der Lösung zugesetzt. Der
abgetrennte Ionenaustauscher wird mehrmals mit Methanol gewaschen und die vereinigten Lösungen eingeengt. Nach Aufnahme des öligen Rückstandes in Ether fällt ein weißer Niederschlag aus. Dieser wird abfiltriert und mit Ether gespült. K wird, ausgehend vom α-Anomer des 5- (2 , 2-Dibrom-vinyl) -1 [2 ' -desoxy-3 ' , 5 ' -di-O- (p- chlor-benzoyl) ] uridins, in analoger Weise dargestellt.

0,31 g (1,0 mmol) 5' -Aminothalidomid-HCl und 0,31g (1,1 mmol) Ölsäure werden in 8 ml trockenem Dichlormethan suspendiert und der Kolben mit einem Septum verschlossen. Nach Zugabe von 170 μl
Triethylamin wird die Suspension im NaCl-Eisbad auf - 15°C abgekühlt. Zum Ansatz gibt man über eine Spritze tropfenweise eine Lösung aus 10mg DMAP und 0,21 g (l,0mmol) DCC in 4 ml trockenem Dichlormethan. Nach Erwärmen auf RT läßt man den Ansatz über Nacht rühren. Man kühlt den Ansatz im Eisbad, saugt den Niederschlag ab und verwirft diesen (Dicyclohexyl- harnstoff) . Zum Filtrat gibt man 10 ml Dichlormethan und schüttelt die Lösung einmal mit mit einer Mischung aus 10 ml Wasser und zwei Tropfen konzentrierter Salzsäure und einmal mit Wasser aus. Man trocknet die organische Phase über Natriumsulfat und entfernt das Lösungsmittel vollständig am Rotations- Verdampfer. Das anfallende Produkt enthält noch geringe Mengen an Dicyclohexylharnstoff . Die weitere Aufreinigung kann durch kurzes Erhitzen zum Rückfluß
in Dichlormethan/Petrolether und heißes Abtritten des Niederschlages erfolgen.

416 mg (2 mmol) getrocknetes 5-TMSU (5-Triτnethylsi- lanylethinyluracil) wird in ca. 50 ml abs . MeCN suspendiert, 1,2 ml (5 mmol) BSA und einige Tropfen
Chlortrimethylsilan über ein Septum zugegeben und bis zur vollständigen Klärung der Suspension bei RT unter Argon gerührt. Zur erhaltenen Lösung gibt man anschließend 585 mg (2,4 mmol) 1- (2-Bromethyl) - adamantan sowie etwas I2. Der Ansatz wird nun unter
Ausschluss von Feuchtigkeit für 4 Tage im Ölbad unter Rückfluss gehalten und noch weitere 3 Tage bei RT. Der Reaktionsverlauf wird dünnschichtchromatogra- phisch verfolgt. (EE/ Hexan 1:2).
Das Lösungsmittel wird im Vakuum abgezogen, der Rückstand mit CHC13/H2O aufgenommen und mit H2O gewaschen, die org. Phase abgetrennt und über Na2SO4 getrocknet. Das Lösungsmittel wird erneut abgezogen und der ölige Rückstand mit etwas CHCl3 aufgenommen und chroma- tographiert . Die erhaltenen Fraktionen werden eingeengt, der ölige Rückstand mit MeOH versetzt und die entstehenden Kristalle abgetrennt.

Zu einer Mischung von 185 mg (0,48 mmol) l-Benzyl-5- (2, 2-dibromvinyl) uracil, 10 mg (0,053 mmol) CuI, 28 mg (0,024 mmol) [(C6Hs)3P]2PdCl2 und 110 μl (0,76 mmol) TMSA in 25 ml abs . EE unter Argon gibt man nach 10 min 176 μl (0,96 mmol) N-Ethyldiisopropylamin bei
RT. Die gelbe Suspension klärt sich nach 10 min auf und färbt sich langsam dunkel. Man rührt 24 h bei RT (DC-Kontrolle) , quencht den Ansatz mit 2 ml Wasser, nimmt in 100 ml EE auf und wäscht mit Wasser. Die or- ganische Phase wird über Na2SO4 getrocknet, das Lösungsmittel im Vakuum abgezogen, das Rohprodukt mit etwas CHCl3 aufgenommen und anschließend chromatographisch gereinigt.
Säulenchromatographie: Säule 400 mm, 30 mm AD, (70g Flash-Gel, 5 % deaktiviert)

120 mg (0,41 mmol) 1- (2-Iodethyl) -adamantan werden in 25 ml DMF gelöst. Zur klaren Lösung gibt man 400 mg
K2CO3 und lässt 24 h bei RT rühren. Das Lösungsmittel wird bei 600C im Vakuum abgezogen und der gelbe Rückstand in einem H2O/CH2C12 (1 : 1) -Gemisch aufgenommen. Man schüttelt noch zweimal mit je 30 ml CH2Cl2 aus und trocknet die org. Phase über Na2SO4. Das Lösungsmittel wird erneut abgezogen und der ölige Rückstand mit etwas CHCl3 aufgenommen und chromatographiert . Die Fraktionen werden eingeengt, der ölige Rückstand mit MeOH versetzt und die entstehenden Kristalle ab- getrennt.

Zur erhaltenen Lösung gibt man anschließend 2,72 g (11 mmol) 1-Bromdiphenylmethan. Der Ansatz wird nun für 24 h im Ölbad unter Ausschluss von Feuchtigkeit bei 840C gerührt. Die dunkelbraune Lösung wird eingeengt und im Eisbad stehengelassen. Das sich abscheidende braune Rohprodukt wird abgetrennt, mit wenig kaltem MeCN ge- waschen und aus ca. 50 ml MeCN umkristallisiert.
Ausbeute: 1,51 g (40,3 % d. Th.)
Beispiel 23
3 -Benzyl - (E) - 5 - (2 -bromvinyl) - 2 ' -deoxyurindin, (J -Anomer (Y)
666 mg (2 mmol) BVDU und 410 mg (2,4 mmol) Benzylbro- mid werden in 25 ml DMF gelöst. Zur klaren Lösung gibt man 1 g K2CO3 und lässt 20 h bei RT rühren. Das Lösungsmittel wird bei 500C abgezogen und der feste Rückstand in einem H2O/CH2Cl2-Gemisch gelöst. Man schüttelt mit je zweimal Wasser/NaClsat. aus und trocknet die org. Phase über Na2SO4. Eine Reinigung erfolgt über eine abschließende Säulenchromatographie :
Kurze Säule (200 mm) mit Fritte, 2,4 cm AD (Flash- Gel, 25g) LM: 100 % CHCl3 110 ml, dann CHCl3: MeOH / 30:1 DC-Kontrolle: EE: Hexan / 2:1
Ausbeute: 631 mg (74 % d. Th.)

1. Uracilderivate der allgemeinen Formel I
mit
Ri ausgewählt aus der Gruppe bestehend aus lineares oder verzweigtes C2-Ci8-Alkenyl, lineares oder verzweigtes C2-Ci8-Alkinyl , unsubstituierte oder substituierte aromatische Reste mit 6-22 Kohlenstoffatomen und/oder unsubstituierte oder substituierte heteroaromatische Reste mit 5-22 Kohlenstoffatomen, C5-C8-Cycloalkyl, C5-C8- Cycloalkenyl, Adamantyl, 0-CH2-CH2-OH,
CH2-O-CH2-PO(OH)2 und der Formeln II bis IV,
II III
IV X eine Einfachbindung oder ausgewählt aus der Gruppe bestehend aus (CHR4)n mit n = 1 bis 3, CO, CNR4, CNOH, SO und SO2, wobei R4 ausgewählt ist aus der Gruppe bestehend aus H, lineares o- der verzweigtes C1-Ci8-AlKyI, und Reste wie für Rl definiert,
R2 ausgewählt aus der Gruppe bestehend aus
a) gesättigten Resten, ausgewählt aus der Gruppe bestehend aus lineares oder verzweigtes C3-Ci8-AIkVl oder Resten der allgemeinen For-
wobei R5 bis R9 jeweils unabhängig voneinander ausgewählt sind aus der Gruppe bestehend aus H, lineares oder verzweigtes Ci-Ci8-Alkyl, lineares oder verzweigtes C2-C18-Alkenyl, Ii- neares oder verzweigtes C2-Ci8-Alkinyl, F, Cl,
Br, I, NO2, CN und OH,
b) ungesättigten Resten der allgemeinen Formel
wobei Rio, Rn und R12 unabhängig voneinander ausgewählt sind aus der Gruppe bestehend aus H, F, Br, Cl I, CN, NO2, Si (CH3) 3, COOR13 oder CON(Ri3J2 mit RX3 = H oder lineares oder verzweigtes Ci-Ci8-Alkyl und die Reste Ri0 und/oder Rn sowohl in (E) - als auch in (Z) -Konformation angeordnet sein können,
c) ungesättigten Resten der allgemeinen Formel
wobei Ri4 ausgewählt ist aus der Gruppe bestehend aus H, Halogenen (F, Cl, Br und/oder I), Si(CH3J3, primärem, sekundärem oder tertiärem Amin oder primärem, sekundärem oder tertiärem Aminomethyl ; oder
d) ein Rest ausgewählt aus der Gruppe von Ver- bindungen der Formeln VIII bis X
VIII IX
H2N. .NH2 N
e) CHO, COOR13, CH2ORi3, CON(R13J2 oder 1,2,3- Triazol-4-yl; und
R3 ausgewählt ist aus der Gruppe bestehend aus H, lineares oder verzweigtes C1-C18-AIkYl, lineares oder verzweigtes C2-C18-Alkenyl, C5- C8-Cycloalkyl, C5-C8-Cycloalkenyl, unsubstitu- iertes oder substituiertes Benzyl oder Ben- zoyl, X-Adamantyl, wobei X die zuvor genannte Bedeutung besitzt, und/oder einem Rest der Formeln II, III, IV oder XI:
mit der Maßgabe , dass folgende Verbindungen ausgeschlossen sind : Ri = H , R2 = Phenyl , R3 = Ethyl und R4 = H ;
Ri = H , R2 = 2 -Hydroxy- 3 -methylphenyl , R3= Ethyl , R4 = H ;
Ri = H , R2 = 4 -Methoxyphenyl , R3 = 1 - Fluor- vinyl und R4 = H .
2. Uracilderivate nach Anspruch 1, dadurch gekennzeichnet, dass R1 und/oder R4 unabhängig voneinander ausgewählt sind aus der Gruppe bestehend aus Methyl, Phenyl, Mono-, Di-, Tri-, Tetra- oder Pentahalogenphenyl, insbesondere 4-Fluorphenyl, 2 , 6-Difluorphenyl, 3,4-Di- fluorphenyl oder Perfluorphenyl, Mono-, Di- oder Tri-Ci-Ciβ-alkoxyphenyl, insbesondere 4-Methoxy- phenyl oder 3 , 4 , 5-Trimethoxyphenyl, Mono-, Di- oder Trinitrophenyl, insbesondere 4-Nitrophenyl oder Vinyl .
3. Uracilderivate nach einem der vorhergehenden Ansprüche, dadurch gekennzeichnet, dass R2 ausgewählt ist aus der Gruppe bestehend aus a) Methyl, Ethyl, Propyl, 1, 2-Dichlor-2-hydroxy- ethyl, 1, 2-Dibrom-2-hydroxyethyl, 1,2-Diiod- 2 -hydroxyethy1 , b) (E) -2-Chlorvinyl, (E) -2-Bromvinyl, (E) -2-Iod- vinyl, 2, 2-Dibromvinyl, (E) -2 -Cyanoviny1 ,
2, 2-Dicyanovinyl, (E) -2-Nitrovinyl , 2, 2 -Di- nitrovinyl, (E) -2-Carboxy-vinyl, (E) -2-Cyano- 2-carboxyvinyl, (E) -2-Carboxy-Ci-C8- alkylester-vinyl, (E) -2-Cyano-2-carboxy-Ci- C8-alkylester-vinyl, (E) -2-Carbonsäureamid- vinyl, (E) -2-Cyano-2-carbonsäureamid-vinyl, (E) - 2 -Carbonsäure-Ci-C8-alkylamid-viny1 , (E) - 2 -Cyano- 2 - carbonsäure-Cx-C8-alkylamid- vinyl oder die jeweiligen (Z) -Isomere oder c) Ethinyl, Bromethinyl, Trimethylsilylethinyl .
4. Uracilderivate nach einem der vorhergehenden Ansprüche , dadurch gekennzeichnet, dass Ri4 unabhängig voneinander ausgewählt ist aus der Gruppe bestehend aus Piperidino, Piperazino, Morpholino, Piperi- dinomethyl, Piperazinomethyl und Morpholino- methyl .
5. Uracilderivate nach einem der vorhergehenden Ansprüche, dadurch gekennzeichnet, dass das Uracilderivat einer Verbindung der folgenden Formeln entspricht:
Uracilderivat nach einem der vorhergehenden Ansprüche zur Anwendung als therapeutischer Wirkstoff.
Uracilderivat nach dem vorhergehenden Anspruch, dadurch gekennzeichnet, dass es zusammen mit mindestens einem Zytostatikum, in einer gemeinsamen Formulierung oder in getrennten Formulierungen angewendet wird.
8. Verwendung eines Uracilderivats nach einem der Ansprüche 1 bis 5 sowie mindestens einem Zytostatikum zur Unterdrückung oder Reduzierung der Resistenzenbildung bei der Zytostatikabehand- lung.
9. Verwendung nach Anspruch 8 , dadurch gekennzeichnet, dass das mindestens eine Nukleosid und das mindestens eine Zytostatikum in einer einzigen Formulierung verwendet werden.
10. Verwendung nach Anspruch 8, dadurch gekennzeichnet, dass das mindestens eine Nukleosid und das mindestens eine Zytostatikum in getrennten Formulierungen verwendet werden.
