Abstract

Die Erfindung betrifft ein Verfahren zur Herstellung von Fettsäureamiden durch Umsetzen einer Fettsäure und eines Amins, insbesondere Serotonin neben Tryptamin, Tyramin und deren Derivaten, in Gegenwart eines Kopplungsreagenzes, welches in der Lage ist, die Carbonsäure zu aktivieren und eines bevorzugt apolaren, aprotischen Lösungsmittels. Die Kopplungsreagenzien sind insbesondere ausgewählt aus Phosphoniumsalzen, Uronium- und/oder Guanidiniumsalzen. Überraschenderweise müssen Hydroxylgruppen und andere reaktive Gruppen im Amin nicht geschützt werden.

Description

VERFAHREN ZUR HERSTELLUNG VON 3-INDOLYL-FETTSÄUREAMIDEN MIT GESÄTTIGTEN, UNGESÄTTIGTEN ODER HYDROXY- FETTSÄUREN
Die Erfindung betrifft ein Verfahren zur Herstellung von Fettsäureamiden aus gesättigten, ungesättigten oder Hydroxy-Fettsäuren mit Aminen, insbesondere Serotonin neben Tryptamin, Tyramin und deren Derivaten. Die Erfindung betrifft außerdem die Verwendung von durch das erfindungsgemäße Verfahren hergestellten Fettsäureamiden zur Herstellung von Standardlösungen für die Analytik und als pharmazeutische Wirkstoffe.
Amide von Fettsäuren (z. B. mit Serotonin, Tryptamin oder Tryptaminderivaten als Amin) treten als sekundäre Pflanzenstoffe in zahlreichen Nutz- und Kulturpflanzen auf. Dort können sie sowohl erwünschte Wirkungen als auch unerwünschte Nebenwirkungen für den Menschen haben. Eine besondere Rolle spielen sie in der Analytik zur Qualitätsüberprüfung von Lebensmitteln. So werden beispielsweise im deutschen Einzelhandel Kaffees angeboten, die durch eine besondere Behandlung mit Wasserdampf vor der Röstung als magenschonender und reizärmer als unbehandelte Kaffees gelten. Der Marktanteil solcher mild behandelter Kaffees lag im Jahr 2006 bei 18 % (Deutscher Kaffeeverband e.V., Jahresbericht 2006). Als Parameter zur Überprüfung einer erfolgten Dämpfung des Rohkaffees kann der Gehalt an Carbonsäure-5-Hydroxytryptamiden (C-5HT) herangezogen werden, denn durch die Dämpfung des Rohkaffees werden etwa 30 % dieser Verbindungen aus dem Kaffee entfernt (Wurziger J. (1973), 6th International Colloquium on the Chemistry of Coffee, 332-42, ASIC, Paris, Hinkel C. u. Speer K (2006), 21 st International Colloquium on the Chemistry of Coffee, 133-42, ASIC, Paris).
Auch bei der Qualitätsprüfung verschiedener Kakaoprodukte spielt die Quantifizierung von Fettsäuretryptamiden eine Rolle. So stellt der Kakaoschalenanteil in Kakaobutter einen wichtigen Qualitätsparameter für die Schokoladenproduktion dar und wird analytisch über den Gehalt der Fettsäuretryptamide Lignocerinsäuretryptamid und Behensäuretryptamid bestimmt. Auch bei der Herstellung von Kakaobutter kann über den Gehalt dieser beiden Verbindungen ermittelt werden, ob es sich um ein qualitativ höherwertiges Produkt aus geschälten Kakaobohnen oder eine minderwertigere sogenannte Expellerbutter aus ganzen, ungeschälten Kakaobohnen handelt. Die Bestimmung erfolgt dabei über herkömmliche Verfahren wie HPLC oder GC.
Für all diese Verfahren benötigt man zum Abgleich Standardlösungen der zu bestimmenden Substanzen, die daher für eine Quantifizierung zunächst aufwändig aus dem jeweiligen Rohmaterial isoliert oder chemisch synthetisiert werden müssen.
Fettsäureamide von Serotonin, das auch als 5-Hydroxytryptamin bezeichnet wird, spielen auch pharmakologisch eine Rolle. Von N-arachidonoyl-Serotonin wurde gezeigt, dass es in vitro und im Mausmodell stark analgetische Eigenschaften besitzt (Maione et al. British Journal of Pharmacology (2007) 150, 766-781 ).
Bisherige Syntheseansätze zur Herstellung von Fettsäureamiden gehen dabei von Fettsäurechloriden aus und setzen diese im Sinne einer Aminolyse mit Serotonin zum Säureamid um. Dabei wird die Hydroxylgruppe am Indolgrundkörper als potentieller zweiter Reaktionsort durch eine Benzylgruppe geschützt, die nach erfolgter Reaktion wieder entfernt werden muss (Hubert P. et al. (1977), Z. Anal. Chem. 285, 242-50). Alternativ wird das Fettsäurechlorid tropfenweise zu einer Aminlösung zugegeben (Lang R. et al. (2005), Eur. Food Res. Technol. 220, 638-43) oder mit dieser direkt umgesetzt (Münch M et al. (1999), Z. Lebensm. Unters. Forsch. A 208, 39-46).
Aufgrund der Abspaltung der Benzylschutzgruppe durch Niederdruckhydrierung können keine C-5HT mit ungesättigten Fettsäuren synthetisiert werden, da durch diesen Schritt der Doppelbindungscharakter der Fettsäure verloren geht. Bei Verzicht auf die Schutzgruppe besteht jedoch die Gefahr von Nebenreaktionen durch die mangelnde Selektivität des verwendeten Fettsäurechlorids. Die Ausbeuten der angewandten Verfahren lagen mit 57- 75 % nur in einem mittleren Bereich.
Die der Erfindung zugrunde liegende Aufgabe ist daher die Bereitstellung eines einfachen, schnellen und kostengünstigen Verfahrens zur Herstellung von Fettsäureamiden aus Aminen, insbesondere Serotonin, Tyramin, Tryptamin oder aus davon abgeleiteten Derivaten, und einer beliebigen freien Fettsäure, darunter sowohl gesättigte als auch ungesättigte oder Hydroxy-Fettsäuren.
Erfindungsgemäß wird die Aufgabe gelöst durch ein Verfahren zur Herstellung von Fettsäureamiden durch Umsetzen einer Fettsäure und eines Amins in Gegenwart eines Kopplungsreagenzes, welches in der Lage ist, die Carbonsäure zu aktivieren. Bevorzugte Fettsäuren und Kopplungsreagenzien werden weiter unten genannt. Letztere sind insbesondere ausgewählt aus Phosphoniumsalzen, Uronium- und/oder Guanidiniumsalzen.
Das Amin ist vorzugsweise ein primäres oder sekundäres acyclisches oder cyclisches Amin. Bevorzugt sind bioaktive Amine ausgewählt aus primären und sekundären Aminen, die einen aromatischen Rest, bevorzugt einen Phenyl- oder Indolrest enthalten. Besonders bevorzugte bioaktive Amine sind ausgewählt aus Amphetamin, Capsaicin, Tyramin, Tryptamin und deren Derivaten. Amphetamin-Derivate sind insbesondere Metamphetamin und Pervertin. Tyramin-Derivate sind insbesondere Dopamin und Dopa. Tryptamin-Derivate sind insbesondere Serotonin, Methylserotonin, Methyltryptamin und Methoxytryptamin. Die Amine enthalten bevorzugt keine Carboxylgruppen, ausgenommen davon ist Dopa.
Besonders bevorzugt eignet sich das erfindungsgemäße Verfahren zur Herstellung von Tryptamin-Derivaten der allgemeinen Formel:
Dazu wird im erfindungsgemäßen Verfahren ein Amin der allgemeinen Formel
mit n = 0 bis 5, bevorzugt n = 1 oder 2, und R1, R2, R3, R4 R5 unabhängig voneinander ausgewählt aus H (1H oder auch 2H), Halogen, OH, substituierten oder unsubstituierten Alkyl, Alkenyl, Alkinyl und Alkoxy,
mit einer Fettsäure der allgemeinen Formel,

Bevorzugt ist mindestens einer der Reste R1, R2, R3, R4 R5, besonders bevorzugt R2, ein Halogen (bevorzugt Br, Cl oder F), NH2, oder OH. Überraschenderweise müssen bedingt durch das erfindungsgemäße Verfahren diese reaktiven Gruppen nicht mit Schutzgruppen versehen werden. Die Halogene, OH und NH2 liegen bevorzugt vielmehr als freie, ungeschützte Gruppen vor.
Bevorzugte Verbindungen der allgemeinen Formel I sind Tryptamin oder eines seiner Derivate, bevorzugt ausgewählt aus Serotonin, Methylserotonin, Methyltryptamin oder Methoxytryptamin.
Alkyl bezeichnet in der vorliegenden Erfindungsbeschreibung einen unsubstituierten oder substituierten, geradkettigen oder verzweigten, vorzugsweise nicht aromatischen Kohlenwasserstoffrest. Alkenyl steht für einen ungesättigten Alkyl-Rest nach vorgenannter Definition. Alkoxy steht für einen über eine Sauerstoffbrücke verknüpften Alkyl-Rest nach vorgenannter Definition. Halogen ist bevorzugt ausgewählt aus F, Cl, Br und I.
Die Kohlenwasserstoffreste in R1, R2, R3, R4 und R5 haben bevorzugt 1 bis 10 Kohlenstoffatome. Der Kohlenwasserstoffrest in R6 hat bevorzugt von 4 bis 40, vorzugsweise 10 bis 32, besonders bevorzugt 12 bis 24 Kohlenstoffatome.
Fettsäuren im Sinne der Erfindung sind aliphatische, gesättigte oder ungesättigte Monocarbonsäuren, bevorzugt mit 4 bis 40 Kohlenstoffatomen, vorzugsweise über 10, besonders bevorzugt mit 12 bis 24 Kohlenstoffatomen. Die Fettsäuren haben eine nahezu ausschließlich unverzweigte Kohlenstoff-Kette (maximal bis zu 3 Verzweigungen), bevorzugt sind sie vollständig unverzweigt. Der Begriff Fettsäuren schließt jedoch methylverzweigte Fettsäuren, Hydroxysäuren, methoxy- und/oder acetoxysubstituierte Fettsäuren, Cyclopropancarbonsäuren, acetylenische Carbonsäuren, Furanfettsäuren und Urofuranfettsäuren mit ein. Die Fettsäuren tragen somit gegebenenfalls Substituenten, wie z. B. bis zu drei Hydroxylgruppen. Die Fettsäuren können eine gerade (natürlich vorkommende Fettsäuren) oder ungerade (synthetische Fettsäuren) Kohlenstoffanzahl aufweisen. Die Fettsäuren enthalten bevorzugt keine Aminogruppen.
Fettsäuren im Sinne der Erfindung sind auch die sich von der Arachidonsäure ableitenden Eicosanoide, die als Lipidhormone verschiedenste Aufgaben im tierischen und menschlichen Körper haben. Sie umfassen unter anderem Prostaglandine, die Leukotriene und Hydroxy- und Hydroperoxyfettsäuren.
Das für die Reaktion verwende Lösungsmittel ist bevorzugt ein apolares aprotisches Lösungsmittel, bevorzugt halogenierte Kohlenwasserstoffe, wie z. B. Dichlormethan, Chloroform, Tetrachlorkohlenstoff, Hexafluorbenzol, Alkane (wie z. B. Pentan, Hexan, Heptan, Cyclopentan, Cyclohexan), Alkene, Alkine, Aromaten ohne Substituenten oder mit ausschließlich aliphatischen und aromatischen Substituenten (z. B. Benzol, Toluol), Silane oder Borane mit ausschließlich aliphatischen und aromatischen Substituenten (wie z. B. Tetramethylsilan), Kohlenstoffdisulfid und bei hohen Drücken auch Kohlenstoffdioxid. Das verwendete Lösungsmittel oder Lösungsmittelgemisch ist möglichst wasserfrei, d. h. es enthält bevorzugt unter 1 %, besonders bevorzugt unter 0,1 % Wasser. Das Lösungsmittel enthält keine Amidgruppen. Ein aprotisches Lösungsmittel verfügt über keine funktionellen Gruppen, aus denen durch Laugen wie NaOH oder KOH Wasserstoffatome im Molekül als Protonen abgespalten werden können. Die Dielektrizitätskonstante des Lösungsmittels bei 25 0C beträgt bevorzugt unter 12, bevorzugt unter 10. Das Dipolmoment des Lösungsmittels beträgt bevorzugt unter 6 10"30 Cm, bevorzugt unter 5,5 10"30 Cm.
Vorteilhaft lässt sich das erfindungsgemäße Verfahren auch auf ungesättigte Fettsäuren anwenden. Diese enthalten bevorzugt 1 bis 10, vorzugsweise 1 bis 6 Doppelbindungen und gegebenenfalls auch bis zu drei Dreifachbindungen.

Auch die Hydroxygruppen der Hydroxyfettsäuren müssen bedingt durch das erfindungsgemäße Verfahren überraschenderweise nicht mit Schutzgruppen versehen werden. Tabelle 5: ungewöhnliche Fettsäuren
Weitere Fettsäuren, die mit dem erfindungsgemäßen Verfahren zu Amiden umgesetzt werden können, sind Prostaglandine, bevorzugt Prostaglandin G2, Prostaglandin D2, Prostaglandin E2, Prostaglandin F, Thromboxan A2 und Prostacyclin I2, und Leukotriene, bevorzugt Leukotrien A4, Leukotrien B4, Leukotrien C4, Leukotrien D4, und Leukotrien E4. Die Prostaglandine und Leukotriene können auch Amingruppen enthalten. Für die Reaktion wird das Amin - vorzugsweise nach der allgemeinen Formel (I) - bevorzugt zunächst in einem organischen Lösungsmittel gelöst. Das Lösungsmittel ist bevorzugt ein apolares aprotisches Lösungsmittel (bevorzugt wie oben genannt) oder ein Gemisch aus einem apolaren aprotischen Lösungsmittel (wie z. B. Dichlormethan) und einem polaren protischen Lösungsmittel (wie z. B. Ethanol). Das verwendete Lösungsmittel oder Lösungsmittelgemisch ist möglichst wasserfrei, d. h. es enthält bevorzugt unter 1 %, besonders bevorzugt unter 0,1 % Wasser. Das Lösungsmittel enthält bevorzugt keine Amidgruppen.
Parallel oder anschließend erfolgt die Zugabe eines Kopplungsreagenzes, welches in der Lage ist, die Carbonsäure zu aktivieren. Derartige Kopplungsreagenzien sind aus der Peptidsynthese bekannt und gehören unterschiedlichen Klassen an, wie z. B.:
• Phosphoniumsalze (z. B. BOP),
• Uronium- und/oder Guanidiniumsalze (z. B. HBTU, HATU),
• Immonium-Reagenzien,
• Carbodiimid-Reagenzien (z. B. DCC),
• Imidazolium-Reagenzien,
• Organophosphorige Reagenzien,
• Saure halogenierende Reagenzien,
• Chloroformate.
Bevorzugt werden als Kopplungsreagenzien Phosphoniumsalze, Uronium und/oder Guanidiniumsalze verwendet.
Die Kopplungsreagenzien haben bevorzugt folgende allgemeine Struktur:

A steht dabei für zwei oder drei Aminreste, die an ein Zentralatom ausgewählt aus P und C gebunden sind. Die Aminreste sind bevorzugt vollständig mit aliphatischen und/oder cyclischen Alkylresten substituiert. Ist das Zentralatom ein Kohlenstoff, sind daran bevorzugt 2 Aminstickstoffe gebunden. Ist das Zentralatom ein Phosphor, sind daran bevorzugt 3 Am in Stickstoffe gebunden. Die aliphatischen Alkylreste enthalten bevorzugt 1 bis 4 C-Atome. Die cyclischen Alkylreste enthalten bevorzugt 4 bis 6 C-Atome. Der Stickstoff des Amins ist bevorzugt Teil des Ringsystems. W steht für ein Halogen, Azid (N3) oder einen substituierten oder unsubstituierten Alkyl- oder Alkylenrest, das bzw. der direkt oder über einen Sauerstoff oder einen Schwefel an das P- oder C-Atom des Restes A gebunden ist. Bevorzugt ist ein substituierter oder unsubstituierter cyclischer Alkylrest, der ein aromatisches, heteroaromatisches oder nichtaromatisches Ringsystem mit bevorzugt 1 bis 3 Ringen und bevorzugt 5 bis 12 C-Atomen und bis zu 6 N-Atome enthält.
Y" ist ein geeignetes Anion, bevorzugt ausgewählt aus einwertigen anorganischen Anionen. Y" ist vorzugsweise ausgewählt aus Halogen, wie Cl", F" und Br", sowie BF4 ", PF6 ", und CIO4 ".

NOP (6-Nitrobenzotriazol-1 -yloxytris(dimethylamino)phosphoniumhexafluoro-phosphat): Der Ansatz aus Aminkomponente und Kopplungsreagenz wird bevorzugt mit einer im gleichen organischen Lösungsmittel gelösten Fettsäure der allgemeinen Formel (III) versetzt. Die Fettsäure wird bevorzugt in einem Molverhältnis im Bereich von 1 : 0,5 bis 1 : 2, besonders bevorzugt von 1 : 0,8 bis 1 : 1 ,2 zu dem eingesetzten Amin eingesetzt. Die Edukte werden bevorzugt in einem Konzentrationsbereich von 5 bis 500 mmol/l, besonders in Konzentrationen zwischen 20 bis 100 mmol/l eingesetzt.
Der pH-Wert sollte während der ganzen Reaktion in einem Bereich von pH 7 bis 11 , bevorzugt in einem Bereich pH 8 bis pH 10, gehalten werden. Der pH-Wert wird dazu durch Zugabe einer Base, bevorzugt einem basischen Lösungsmittel, besonders bevorzugt einem tertiären Amin oder einem cyclischen Amin als Base eingestellt. Bevorzugte tertiäre Amine sind Trimethylamin, Triethylamin oder Tripropylamin, entsprechend gemischte Amine, wie z. B. Methyldiethylamin, oder ein anderes Trialkylamin.
Die Reaktion wird bevorzugt bei einer Temperatur zwischen 4°C und 42°C, besonders bevorzugt zwischen 16°C und 25°C durchgeführt. Die Reaktion wird bevorzugt solange durchgeführt, bis keine Abnahme der Fettsäurekonzentration mehr zu beobachten ist.
Das erhaltene Amid, bevorzugt gemäß der allgemeinen Formel
wird nach Abtrennung von Verunreinigungen vorzugsweise durch Waschen des Ansatzes mit wässrigen Lösungen verschiedener Salze wie beispielsweise KHSO4, NaHCC>3 und NaCI aus dem Reaktionsgemisch beispielsweise durch Kristallisation gewonnen. Sofern notwendig, kann eine Reinigung vorzugsweise durch Säulenchromatographie, Festphasenextraktion oder Umkristallisation erfolgen.
Durch die vorteilhafte Anwendung von Kopplungsreagenzien wie Phosphonium-, Uronium- oder Guanidiniumsalzen, wie beispielsweise BOP, NOP, TBTU, HBTU oder HATU werden in einer einfachen Ein-Stufen-Synthese Tryptamin und Tryptaminderivate wie z.B. Serotonin, Methylserotonin, Methyltryptamin oder Methoxytryptamin mit freien Fettsäuren zu den entsprechenden Fettsäureamiden umgesetzt.
Vorteilhaft ermöglichen die milden Reaktionsbedingungen des beschriebenen Verfahrens die Umsetzung von sowohl gesättigten als auch ungesättigten sowie Hydroxy-Fettsäuren zur Synthese von Fettsäureamiden. Vorteilhaft ergeben sich dabei Ausbeuten von über 90%.
Gegenstand der Erfindung ist auch die Verwendung der mit dem erfindungsgemäßen Verfahren hergestellten Verbindungen zur Herstellung einer Standardlösung für die Analytik, in der Lebensmittelchemie und in der Pharmazie.
In der Lebensmittelchemie werden die Verbindungen bevorzugt als Standards in der Qualitätskontrolle von Kaffee und Kakao eingesetzt.
Bei der Verwendung in der Pharmazie erleichtert die Kopplung des Amins an die Fettsäuren die Passage durch Epithelien bzw. Zellmembranen, wie z. B. das Darmepithel, und erhöht damit bei oraler Gabe die Aufnahme im Darm. Darüber hinaus wird durch die Kopplung des Amins an die Fettsäuren die Passage durch die Blut-Hirn-Schranke möglich. Auch bei einer transdermalen, rektalen, vaginalen oder nasalen Anwendung erleichtert die Kopplung des Amins an die Fettsäuren die Passage durch die Mucosa bzw. Dermis. Darüber hinaus kann durch die Kopplung des Amids an die Fettsäuren ein Depoteffekt bewirkt werden, insbesondere bei subkutaner Verabreichung.
Ausführungsbeispiele:
Das erfindungsgemäße Verfahren wird näher erläutert durch die folgenden Ausführungsbeispiele, ohne auf diese beschränkt zu sein:
Ausführungsbeispiel 1 :
Zu einer Lösung von 0,4 mmol der Aminkomponente Serotonin in einer Mischung aus 15 ml Dichlormethan und der gleichen Menge Ethanol wird unter Rühren eine Lösung des Kopplungsreagenzes 0-(1 H-Benzotriazol-1-yl)-N,N,N',N'-tetramethyl-uronium Hexafluoro- phosphat (HBTU, 0,44 mmol in 40 ml Dichlormethan) gegeben. Anschließend wird eine zuvor bereitete Lösung von 0,44 mmol Palmitinsäure in Dichlormethan unter stetem Rühren zugefügt und mit 200 μl Triethylamin als Base versetzt. Der pH-Wert wird während der ganzen Reaktion kontrolliert und in einem Bereich von pH 8 bis pH 10 gehalten, bei Bedarf wird dieser durch zusätzliche Zugabe der Base eingestellt. Der Reaktionsverlauf kann mittels DC-Kontrolle des Fettsäuregehaltes überwacht werden. Wird keine Abnahme der Fettsäurekonzentration mehr verzeichnet, ist die Reaktion abgeschlossen. Der gesamte Ansatz wird unter Nachspülen des Reaktionsgefäßes mit 40 ml Ethylacetat in einen Scheidetrichter übergeführt und dreimal mit je 30 ml einer 5 %igen Kaliumhydrogensulfat- Lösung ausgeschüttelt. Anschließend wird nach dreimaliger Extraktion mit je 30 ml einer 5%igen Natriumhydrogencarbonat-Lösung viermal mit je 30 ml einer gesättigten Kochsalzlösung gewaschen. Die organische Phase wird über Nacht mit Na2SC>4 getrocknet, das Lösungsmittel im Vakuum entfernt und aus Aceton umkristallisiert.
Das Produkt 16-5HT wurde anschließend durch eindimensionale Kernspinresonanz (1 H- NMR), Flüssigchromatographie mit Massenspektrometrie-Kopplung (LC-MS) und hochauflösende Massenspektrometrie (HRMS) charakterisiert.

Claims

Patentansprüche:
1. Verfahren zur Herstellung von Verbindungen der allgemeinen Formel
mit n = 0 bis 5, R-i, R2, R3, R4, R5 ausgewählt aus H, Halogen, OH, NH2, Alkyl, Alkenyl,
Alkinyl und Alkoxy und R6 ausgewählt aus substituierten oder unsubstituierten Alkyl, Alkenyl, Alkinyl, dadurch gekennzeichnet, dass ein Amin der allgemeinen Formel
mit einer Fettsäure der allgemeinen Formel

OH mit R6 ausgewählt aus substituierten oder unsubstituierten Alkyl, Alkenyl, Alkinyl unter Verwendung eines Kopplungsreagenzes, ausgewählt aus Phosphoniumsalzen, Uroniumsalzen und/oder Guanidiniumsalzen oder einem Kopplungsreagenz der allgemeinen Struktur
(A-W)+ Y" (IV) umgesetzt wird, wobei
A für zwei oder drei Aminreste steht, die an ein Zentralatom ausgewählt aus P und C gebunden sind,
W ausgewählt ist aus Halogen, Azid und substituierten oder unsubstituierten Alkyl- oder Alkylenresten, wobei W direkt oder über einen Sauerstoff oder einen Schwefel an das P oder C-Atom des Restes A gebunden ist und Y" ein geeignetes Anion ist.
2. Verfahren nach Anspruch 1 , dadurch gekennzeichnet, dass mindestens eine der Gruppen R1, R2, R3, R4, R5 ausgewählt ist aus Halogen, OH und NH2.
3. Verfahren nach Anspruch 1 oder 2, dadurch gekennzeichnet, dass das Amin der allge meinen Formel I Serotonin, Methylserotonin Tryptamin, Methyltryptamin oder Methoxytryptamin ist.
4. Verfahren nach einem der Ansprüche 1 bis 3, dadurch gekennzeichnet, dass als Kopplungsreagenz ein Phosphoniumsalz mit der allgemeinen Formel

verwendet wird, wobei R7, R8, Rg, Rio, Rn und Ri2 unabhängig voneinander ausgewählt sind aus aliphatischen und/oder cyclischen Alkylresten oder Alkoxyresten, die jeweils substituiert oder unsubstituiert sind, wobei gegebenenfalls zwei oder mehrere der Reste R7 bis R12 jeweils zusammen einen zweiwertigen Alkylrest darstellen, der zusammen mit dem N einen Ring bildet.
5. Verfahren nach Anspruch 4, dadurch gekennzeichnet, dass als Kopplungsreagenz ein Phosphoniumsalz der allgemeinen Formel ((CHs)2N)3P+-W Y" oder
verwendet wird.
6. Verfahren nach einem der Ansprüche 1 bis 5, dadurch gekennzeichnet, dass W ausgewählt ist aus Halogen (bevorzugt Br, Cl), N3 und
mit T1 und T2 bevorzugt unabhängig voneinander ausgewählt aus H, NO2 und CF3,
sowie
7. Verfahren nach einem der Ansprüche 1 bis 3, dadurch gekennzeichnet, dass als Kopplungsreagenz ein Guanidiniumsalz und/oder Uroniumsalz ausgewählt aus der allgemeinen Formel

16 verwendet wird, wobei Ri3, Ri4, Ri5 und Ri6 unabhängig voneinander ausgewählt sind aus aliphatischen und/oder cyclischen Alkylresten oder Alkoxyresten, die jeweils substituiert oder unsubstituiert sind, wobei gegebenenfalls zwei oder mehrere der Reste R13 bis Ri6 jeweils zusammen einen zweiwertigen Alkylrest darstellen, der zusammen mit dem N einen Ring bildet.
8. Verfahren nach Anspruch 7, dadurch gekennzeichnet, dass als Kopplungsreagenz ein Guanidiniumsalz und/oder Uroniumsalz ausgewählt aus den folgenden allgemeinen Formeln
verwendet wird.
9. Verfahren nach einem der Ansprüche 1 bis 8, insbesondere Anspruch 7 oder 8, dadurch gekennzeichnet, dass W ausgewählt ist aus Halogen (bevorzugt F, Cl oder Br) oder den folgenden Formeln a) bis I),
10. Verfahren nach einem der Ansprüche 1 bis 9, dadurch gekennzeichnet, dass das Anion Y" ausgewählt ist aus PF6 ", BF4 ", Cl", F", Br" und CIO4 ".

Verfahren nach einem der Ansprüche 1 bis 10, dadurch gekennzeichnet, dass das Kopplungsreagenz ausgewählt ist aus N-[(1 H-benzotriazol-1-yl)-(dimethylamino)- methylen]-N-methylmethanaminiumhexafluorophosphat-N-Oxid (HBTU), 1 H-Benzo- triazol-1-yl-1 ,1 ,3,3-tetramethyluroniumtetrafluoroborat (TBTU), (1 H-Benzotriazol-1- yloxy)-tris(dimethylamino)phosphonium-hexafluorophosphat (BOP), 6-Nitrobenzo- triazol-1 -yloxytris(dimethylamino)phosphoniumhexafluoro-phosphat (NOP) und N-[(Dimethylamino)-1 H-1 ,2,3-triazolo[4,5-b]pyridin-1 -ylmethylen]-N-methylmethan- aminiumhexafluorophosphat-N-Oxid (HATU).
12. Verfahren nach einem der Ansprüche 1 bis 1 1 , dadurch gekennzeichnet, dass es bei einem pH-Wert von pH 7 bis pH 11 , bevorzugt pH 8 bis pH 10 durchgeführt wird.
13. Verfahren nach einem der Ansprüche 12, dadurch gekennzeichnet, dass der pH-Wert mit einer Base, bevorzugt einem tertiären oder cyclischen Amin eingestellt wird.
14. Verwendung einer durch das Verfahren gemäß einem der Ansprüche 1 bis 13 hergestellten Verbindung in der Analytik, der Lebensmittelchemie in der Pharmazie oder als als Arzneimittel.
