Die Erfindung betrifft eine Packung umfassend ein Packmittel und Weichkapseln, ein Verfahren zur Stabilisierung von Weichkapseln sowie ein Verfahren zur Herstellung der Packung.

Description

PACKUNG ENTHALTEND WEICHKAPSELN
Die Erfindung betrifft eine Packung umfassend ein Packmittel und Weichkapseln, ein Verfahren zur Stabilisierung von Weichkapseln sowie ein Verfahren zur Herstellung der Packung.
Weichkapseln sind Kapseln die auf Grund von in der Kapselhülle enthaltenen Zusätzen an Weichmachern wie z. B. Glycerin, Sorbit, Maltitol und Polyethylenglykole eine gewisse Elastizität und Geschmeidigkeit aufweisen und zur Verabreichung von verschiedenen Stoffen wie z. B. Arzneistoffe, Nahrungsergänzungsstoffe und/oder Functional Ingredients eingesetzt werden. Weichkapseln können beispielsweise auf Gelatineoder Stärkebasis hergestellt werden. Weichkapseln auf Gelatinebasis sind als Weichgelatinekapseln seit langem bekannt und werden von verschiedenen Anbietern kommerziell angeboten. Weichkapseln auf Stärkebasis sind eine relativ neue Entwicklung, ein Beispiel hierfür sind Weichkapseln, die von der Firma Swiss Caps, Schweiz, unter der Handelsbezeichnung VegaGels® vertrieben werden.
Soweit mit dem Weichkapseln flüssige oder halbfeste Stoffe verabreicht werden sollen können diese direkt verkapselt werden, Feststoffe werden zuvor in flüssigen oder halbfesten Grundlagen, wie z. B. fetten, Ölen, Wachsmischungen oder Polyethylenglykole, eventuell in Kombination mit Emulgatoren gelöst oder suspendiert. Je nach Applikationsweg, wie z. B. oral, rektal oder vaginal, können Weichkapseln verschiedene Formen aufweisen, sie können z. B. rund, oval, oblong oder torpedoförmig sein. Weichkapseln können nach den im Stand der Technik bekannten Verfahren hergestellt werden, wie z. B. nach dem Scherer- Verfahren, dem Accogel-Verfahren oder dem Tropf- oder Blasverfahren.
Die weich machende Wirkung des in den Hüllen von Weichkapseln enthaltenen Weichmacheranteils beruht im wesentlichen auf dessen Hygroskopizität und der damit verbundenen Einlagerung von Wasser die Kapselhülle. Bei Raumtemperatur (22°C / 45 bis 65 % rF (rF = relative Feuchte)) weisen Weichkapseln einen Wassergehalt von ca. 7 bis 8 Gew.- % auf. Wird der Wassergehalt, beispielsweise durch zu trockene Lagerung oder Trocknung, wesentlich unterschritten führt dies zu einer Versprödung der Kapselhülle mit der Folge von erhöhter Brüchigkeit der Kapselwand und der Bildung von Rissen. Wird der Wassergehalt, beispielsweise durch zu feuchte Lagerung, deutlich überschritten kommt es zu einem Aufquellen der Kapselhülle und zur Verklebung der Weichkapseln. Sowohl zu trockene als auch zu feuchte Lagerung führt auch zu unvorhersehbaren Auswirkungen auf die zu verabreichenden Stoffe sowie der ggf. weiterhin enthaltenen Hilfsstoffe, sodass die mit der Einnahme von Weichkapseln beabsichtigte Versorgung mit den hierin enthaltenen Stoffen nicht mehr sichergestellt werden kann. Schwankende Umwelteinflüsse führen zu völlig unkalkulierbaren Einflüssen auf die Stabilität von Weichkapseln, sodass ihre Lagerfähigkeit, d. h. der Zeitraum über welchen die Weichkapseln innerhalb der jeweiligen für sie geltenden Spezifikationen liegen, die zu erfüllen sind, damit sie bestimmungsgemäß eingenommen werden können, nicht mehr mit der erforderlichen Sicherheit vorhergesagt werden kann. Dies ist, insbesondere auch aus Sicherheitsgründen, nicht akzeptabel.
Es war Aufgabe der vorliegenden Erfindung eine einfache und kostengünstige Möglichkeit zu finden, mit der sowohl die mit der Versprödung der Kapselhülle als auch die durch Aufquellen und Verklebung sich ergebenden Qualitätsverluste vermieden werden und mit welcher die Lagerfähigkeit von Weichkapseln bei hoher Trockenheit, hoher Luftfeuchtigkeit, ggf. auch zusammen mit hohen Temperaturen, wie sie z. B. in den Klimazonen III und IV vorherrschen, und auch bei stark schwankender Luftfeuchtigkeit deutlich erhöht wird.
Die Aufgabe konnte überraschenderweise gelöst werden, indem die Weichkapseln in ein Packmittel eingebracht wurde, in dessen Innenwandung/en zumindest teilflächig ein Absorptionsmittel und mindestens ein Kanalbildner eingebettet ist. Gegenstand der Erfindung ist somit eine Packung umfassend ein wiederverschließbares Behältnis, in/an dessen Innenwandung/en zumindest teilflächig mindestens ein Kanalbildner zusammen mit mindestens einem Absorptionsmittel eingebettet ist, sowie mindestens einer Weichkapsel, wobei die Weichkapsel/n in dem Behältnis enthalten ist/sind.
Überraschenderweise führt das Verbringen der Weichkapseln zu einer Erhöhung der Lagerstabilität, obwohl das Behältnis mit einem Absorptionsmittel ein Mittel enthalten ist, welches ein trocknende Wirkung zur Folge hat. Denn Trocknung von Weichkapseln führt über Wasserentzug gerade zur Versprödung und Brüchigkeit der Kapselhülle bis hin zur Rissbildung, also genau zu dem Zustand, der durch die Erfindung vermieden wird.
Nach einer vorteilhaften Ausführungsform enthält die erfindungsgemäße Packung als Weichkapseln Weichgelatinekapseln. Gegenstand der Erfindung ist daher eine Packung die dadurch gekennzeichnet ist, dass die hierin enthaltene/n Weichkapsel/n (eine) Weichgelatinekapsel/n ist/sind.
Nach einer weiteren vorteilhaften Ausführungsform enthält die erfindungsgemäße Packung als Weichkapseln auf Stärkebasis. Gegenstand der Erfindung ist daher eine Packung die dadurch gekennzeichnet ist, dass die hierin enthaltene/n Weichkapsel/n (eine) Weichkapsel/n auf Stärkebasis ist/sind.
Das Behältnis ist wiederverschließbar und kann alle Raumformen, wie z. B. die eines Zylinders oder Quaders, aufweisen, vorausgesetzt dass diese groß genug ist, um die Weichkapseln in sich aufzunehmen.
Wiederverschließbar bedeutet, dass das Behältnis wiederholt, d. h. mindestens einmal, bevorzugt mehrmals, besonders bevorzugt mindestens so oft geöffnet und wieder geschlossen werden kann, wie es der Anzahl der in dem Behältnis enthaltenen Weichkapseln entspricht. Das Behältnis ist nach jedem Öffnungs- und Verschlussvorgang so dicht verschlossen, dass ein Eindringen von Feuchtigkeit und Gasen in das Behältnisinnere wirksam verhindert wird. Öffnen und Verschließen des Behältnisses erfolgt durch einen dem Behältnis angepassten, dicht schließenden Verschluss. Es können alle Arten von Verschlüssen eingesetzt werden, soweit diese auch nach wiederholtem Öffnen und Schließen des Behältnisses sicherstellen, dass Gase und/oder Feuchtigkeit im geschlossenen Zustand nicht oder nur in äußerst geringen Mengen in das Behältnisinnere eindringen können.
Nach einer Ausführungsformen der Erfindung werden als Verschluss Deckel verwendet. Beispiele von Deckeln sind Schraubverschlüsse, Kappen, die über den oberen Rand der Gefäße gestülpt werden, oder in das Innere des Gefäßes eingeführt werden. Bei Raumformen, die Ecken aufweisen, wie z. B. Quadern, können die Ecken der Öffnung und der hierzu passende Deckel auch leicht gerundet sein, um die Dichtigkeit des Behältnisses gegenüber Gasen und Feuchtigkeit zu erhöhen.
Nach einer bevorzugten Ausführungsform hat das Behältnis eine Quaderförmige Raumform, gerundete Ecken und ist gut stapelbar. Eine derartige Ausführungsformen ist in Figur 1 beispielhaft dargestellt. Das Behältnis umfasst Wandungen (2), deren nach innen gerichtete Seiten zumindest teilflächig mindestens einen Kanalbildner zusammen mit mindestens einem Absorptionsmittel enthalten, hat gerundete Ecken (7) und weist als Verschluss einen Deckel (1) auf.
Die im Behältnis enthaltenen Absorptionsmittel und Kanalbildner können gemeinsam entweder direkt in der/den Innenwandung/en des das Behältnis ausbildenden Polymeren enthalten oder als Schicht auf der/den Innenwandung/en des Behältnisses aus Polymeren aufgebracht sein. Ebenso können Absorptionsmittel und Kanalbildner in ein Inlay eingebettet werden, das als Einschub in das Behältnis eingebracht wird, sodass zumindest ein Teil der Innenwandungen des Behältnisses hierdurch ausgekleidet werden.
Unter Innenwandung/en wird die nach innen gerichtete Fläche der Wand/Wände des Behältnisses verstanden, also die Fläche/n des Behältnisses, die im Kontakt zu der/den hierin enthaltenen Weichkapsel/n steht/stehen.
Als Materialien für das Behältnis in Frage kommen Polymere. Polymere, die in Mischung mit Absorptionsmittel und Kanalbildner eingesetzt werden können, sind insbesondere Thermoplaste wie z.B. Polyolefine, wie Polyethylen und/oder Polypropylene, Polyisoprene, Polybutadiene, Polybutene, Polysiloxane, Polyamide, Ethylen-Vinylacetat-Copolymere, Ethylen-Methacrylat-Copolymere, Polystyrole, Polyester, Polyanhydride, Polyacrylatnitrile, Polysulfonate, Polyesteramide, Polyacrylatester, Propylen- Maleinsäureanhydrid, Polyethylen- Maleinsäureanhydrid, Polyethylen- Urethane, Polyethylen-Ethylvinylalkohole, Polyethylen-Nylon und/oder Polyurethane sein. Die an ihrer Innenfläche mit Absorptionsmittel und Kanalbildner ausgerüsteten Wände weisen, bezogen auf das Gesamtgewicht der Mischung aus Polymer, Kanalbildner und Absorptionsstoffen, einen Gehalt an Polymer von 10 - 90 Gew.-% auf.
Als Absorptionsmittel kann grundsätzlich jede Art von Trockenmitteln, d. h. feuchtigkeitsbindende Bindemittel, enthalten sein. Es kommen drei Gruppen von Trockenmitteln in Betracht:
Die erste Gruppe beinhaltet chemische Stoffe, die mit Wasser Hydrate bilden. Beispiele derartige chemische Stoffe sind wasserfreie Salze, die dazu neigen Wasser oder Feuchtigkeit zu absorbieren, und hierbei ein stabiles Hydrat bilden. Es wird die Feuchtigkeit gebunden und deren Freisetzung durch eine chemische Reaktion verhindert.
Die zweite Gruppe der Trockenmittel enthält Stoffe, die reaktiv sind. Die Stoffe reagieren mit Wasser oder Feuchtigkeit, indem sie einen neuen Stoff bilden. Die neu gebildeten Stoffe sind normalerweise bei niedrigen Temperaturen stabil, die nur unter Aufwendung hoher Energie reversibel ist. Diese Art von Trockenmitteln wird hauptsächlich zum Trocknen von Lösungsmitteln und als Wasser absorbierendes Material bei Polymeren, die selber in einem feuchtigkeitsreduzierten Zustand bleiben müssen, verwendet.
Die dritte Gruppe der Trockenmittel bindet die Feuchtigkeit durch physikalische Adsorption. Das Trockenmittel enthält Teilchen mit feinen Kapillaren, in welche die Feuchtigkeit eingezogen wird. Dabei bestimmen die Porengröße der Kapillaren sowie deren Dichte im Trocknungsmittel die Absorptionseigenschaften. Beispiele solcher Trockenmittel sind Molekularsiebe, Kieselgele, bestimmte synthetische Polymeren, wie z. B. solche, die in Babywindeln verwendet werden, und Stärken. Trockenmittel der dritten Gruppe sind bevorzugt im Behältnis enthalten, da sie weitgehend inert und wasserunlöslich sind. Besonders bevorzugt sind dabei Molekularsiebe mit einer Porengröße von 3 bis 15 Angström und/oder Kieselgele mit einer Porengröße von 24 Angström.
Als Kanalbildner in Betracht kommen hydrophile Stoffe wie z. B. Polyglykole, Ethylvinylalkole, Glycerin, Polyvinylalkohole, Polyvinylpyrrolidon, Vinylpyrrolidon, N-Methylpyrrolidon, Polysaccharide, Saccharide und/oder Zuckeralkohole. Als Polyglykole werden Polyethylenglykol und/oder Polypropylenglykol bevorzugt. Als Saccharide können z.B. Glucose, Mannose, Galactose und/oder Fructose verwendet werden. Als Zuckeralkohole in Frage kommen z.B. Mannitol, Sorbitol, Hexitol, Dulcitol, Xylitol, Ribitol und/oder Erythrol. Unter Polysacchariden zu verstehen sind z.B. Dextrine und/oder hydrolisierte Stärke.
In den mit Absorptionsmitteln und Kanalbildnern ausgerüsteten Innenwandungen können die Kanalbildner, bezogen auf das Gesamtgewicht der Mischung aus Polymer, Kanalbildner und Absorptionsmittel, einen Anteil von 10 - 40 Gew.-% aufweisen.
Absorptionsmittel und Kanalbildner sind in der/den Innenwandung/en des Behältnisses teilflächig oder ganzflächig eingebettet. Teilflächig bedeutet, dass zumindest ein Teil der gesamten die Innenwandung/en ausbildenden Fläche des Behältnisses Absorptionsmittel und Kanalbildner enthält. Ganzflächig bedeutet, dass die gesamten, die Innenwandungen ausbildende Fläche des Behältnisses Absorptionsmittel und Kanalbildner enthält. Nach einer vorteilhaften Ausführungsform sind, bezogen auf die gesamte Innenfläche des Behältnisses, Absorptionsmittel und Kanalbildner in mindestens 10 %, bevorzugt in mindestens 50 %, besonders bevorzugt in mindestens 90 % der Innenwandungen enthalten.
Behältnisse aus Polymeren, die Absorptionsmittel und Kanalbildner enthalten und die als Behältnis für die erfindungsgemäße Packung geeignet sind, sind im Stand der Technik bekannt und werden z. B. beschrieben in WO 97/32663 A1 , EP 1000873 A2 und WO 03/086900 A1 , EP 1421991 A1. Behältnisse, die in der erfindungsgemäßen Packung eingesetzt werden können, sind kommerziell erhältlich und werden beispielsweise von der Firma Capitol Specialty Plastics Inc., 2039 McMillan Street Auburn, Alabama, USA, unter dem Warenzeichen Activ-Vial oder von der Firma Süd Chemie, Ostenrieder Str. 15, 85368 Moosburg, Deutschland, unter dem Markennamen 2 AP Multipolymer angeboten.
Die erfindungsgemäße Packung wird hergestellt, indem die Weichkapsel/n in ein wiederverschließbares Behältnis abgefüllt wird, in/an dessen Innenwandung/en zumindest teilflächig mindestens ein Kanalbildner zusammen mit mindestens einem Absorptionsmittel eingebettet ist, und das Behältnis anschließend verschlossen wird. Gegenstand der vorliegenden Erfindung ist daher auch ein Verfahren zur Herstellung einer Packung, das dadurch gekennzeichnet ist, dass die Weichkapsel/n in ein wiederverschließbares Behältnis verbracht wird/werden, in/an dessen Innenwandung/en zumindest teilflächig mindestens ein Kanalbildner zusammen mit mindestens einem Absorptionsmittel eingebettet ist, und das Behältnis anschließend verschlossen wird.
Die erfindungsgemäße Packung führt zu einer Erhöhung der Lagerstabilität der hierin enthaltenen Weichkapseln. Gegenstand der Erfindung ist daher auch ein Verfahren zur Erhöhung der Lagerstabilität von Weichkapseln, das dadurch gekennzeichnet ist, dass diese in ein wiederverschließbares Behältnis verbracht werden, in/an dessen Innenwandung/en zumindest teilflächig mindestens ein Kanalbildner zusammen mit mindestens einem Absorptionsmittel eingebettet ist, und das Behältnis anschließend verschlossen wird.
Die stabilisierende Wirkung der erfindungsgemäßen Packung beruht auf dem Einfluss des Behältnisses auf die in Weichkapsel/n, die hierdurch lagerstabil zur Verfügung gestellt werden kann. Die Erzielung der erfindungsgemäßen Wirkung erfordert somit, dass die Weichkapsel/n in dem Behältnis enthalten ist/sind, die Weichkapsel/n und Behältnis also zusammen als Packung vorliegen.
Neben der stabilisierenden Wirkung auf die Weichkapselhülle führt in die erfindungsgemäße Packung auch zu einer Stabilisierung der in diesen enthaltenen Wirk- und/oder Hilfsstoffe, also der gesamten Formulierung. Dies gilt insbesondere dann wenn die enthaltenen Wirk- und/oder Hilfsstoffe empfindlich gegenüber Feuchtigkeit sind. Beispiele für feuchtigkeitsempfindliche Wirkstoffe, die somit bevorzugt in den Weichkapseln in der Packung enthalten sind, sind viele pharmazeutische Wirkstoffe wie Hormone oder Proteine, Vitamine, Zellen, wie z.B. probiotische Kulturen.
Die Beispiele, ohne hierauf beschränkt zu sein, erläutern die Erfindung.
Beispiel 1
Untersuchung der Stabilität von Weichkapseln in der erfindungsgemäßen Packung im Vergleich zu Weichkapseln in üblichen Packmitteln
Multivitamin-Weichkapseln (Wirkstoffzusammensetzung sowie Kapselhülle wie unten beschrieben) werden in eine tiefgezogene Muldenfolie aus PVC 250 μm + 40 g/m2 PVDC vereinzelt und mit Aluminiumfolie (20 μm) versiegelt bzw. in ein wiederverschließbares Behältnis, in/an dessen Innenwandung/en zumindest teilflächig mindestens ein Kanalbildner zusammen mit mindestens einem Absorptionsmittel eingebettet ist, verbracht (Typ: M-3006-16 Activ-Vial), in dessen Innenwandungen ein Molekularsieb mit einer Kapazität von 1 ,92 g eingebettet ist.
Die die Multivitamin-Weichkapseln enthaltenden Blister und Behältnisse werden bei 400C und 75% relativer Feuchte (rF) eingelagert, nach vorbestimmten Zeiten ausgelagert, hinsichtlich ihres Aussehens visuell beurteilt sowie im Hinblick auf die enthaltenen Wirkstoffmengen analytisch untersucht. Die Ergebnisse sind in Tabellen 1 und 2 zusammengestellt.

Beispiel 2
Untersuchung der Stabilität von Weichkapseln in der erfindungsgemäßen Packung
Multivitamin-Weichkapseln (Wirkstoffzusammensetzung sowie Kapselhülle wie unten beschrieben) werden in ein wiederverschließbares Behältnis, in/an dessen Innenwandung/en zumindest teilflächig mindestens ein Kanalbildner zusammen mit mindestens einem Absorptionsmittel eingebettet ist, verbracht (Typ: M-3006-16 Activ-Vial), in dessen Innenwandungen ein Molekularsieb mit einer Kapazität von 1 ,92 g eingebettet ist.
Die die Multivitamin-Weichkapseln werden bei 300C und 65% relativer Feuchte (rF) eingelagert, nach vorbestimmten Zeiten ausgelagert, hinsichtlich ihres Aussehens visuell beurteilt sowie im Hinblick auf die enthaltenen Wirkstoffmengen analytisch untersucht. Die Ergebnisse sind in Tabelle 3 zusammengestellt.

Claims

Patentansprüche
1. Packung umfassend ein wiederverschließbares Behältnis, in/an dessen Innenwandung/en zumindest teilflächig mindestens ein Kanalbildner zusammen mit mindestens einem Absorptionsmittel eingebettet ist, sowie mindestens eine Weichkapsel, wobei die Weichkapsel/n in dem Behältnis enthalten ist/sind.
2. Packung nach Anspruch 1 , dadurch gekennzeichnet, dass die Weichkapsel/n (eine) Weichgelatinekapsel/n ist/sind.
3. Packung nach Anspruch 1 , dadurch gekennzeichnet, dass die Weichkapsel/n (eine) Weichkapsel/n auf Stärkebasis ist/sind
4. Packung nach einem oder mehreren der Ansprüche 1 bis 3, dadurch gekennzeichnet, dass das Behältnis eine Quader-förmige Grundform aufweist.
5. Packung nach einem oder mehreren der Ansprüche 1 bis 4, dadurch gekennzeichnet, dass das Behältnis als Absorptionsmittel ein Trockenmittel enthält, das Feuchtigkeit durch physikalische Adsorption bindet.
6. Packung nach Anspruch 5, dadurch gekennzeichnet, dass das Trockenmittel ein Molekularsieb oder Kieselgel ist.
7. Verfahren zur Herstellung einer Packung nach einem oder mehreren der Ansprüche 1 bis 6, dadurch gekennzeichnet, dass die Weichkapseln/n in ein wiederverschließbares Behältnis verbracht wird/werden, in/an dessen Innenwandung/en zumindest teilflächig mindestens ein Kanalbildner zusammen mit mindestens einem Absorptionsmittel eingebettet ist, und das Behältnis anschließend verschlossen wird.
8. Verfahren zur Erhöhung der Lagerstabilität von Weichkapseln, dadurch gekennzeichnet, dass diese in ein wiederverschließbares Behältnis verbracht werden, in/an dessen Innenwandung/en zumindest teilflächig mindestens ein Kanalbildner zusammen mit mindestens einem Absorptionsmittel eingebettet ist, und das Behältnis anschließend verschlossen wird.
