背景技术
益生菌是通过定殖在人体内,改变宿主某一部位菌群组成的一类对宿主有益的活性微生物。通过调节宿主黏膜与系统免疫功能或通过调节肠道内菌群平衡,促进营养吸收保持肠道健康的作用,从而产生有利于健康作用的单微生物或组成明确的混合微生物。
药品和保健食品领域已知的包装材料包括PE瓶、PET瓶、铝塑泡罩、泡腾管、ADP(Advanced Desiccant Polymers)干燥剂瓶、铝管等。
现有技术中的益生菌原料或其制剂放置一段时间后,活菌数会显著下降。仍需要提供一种具有改善的稳定性的包含益生菌的产品。

本发明的目的是提供一种具有改善的稳定性的包含益生菌的产品。
本发明涉及一种包含益生菌的产品,包括包装(例如瓶)和位于包装中的益生菌,所述包装具有外壳和盖,所述外壳包含高密度聚乙烯材料,所述盖包含分子筛、硅胶或其组合。
所述产品中的益生菌具有改善的稳定性。

在一个方面,本发明涉及一种包含益生菌的产品,包括包装(例如瓶)和位于包装中的益生菌,所述包装具有外壳和盖,所述外壳包含高密度聚乙烯(HDPE)材料,所述盖包含分子筛、硅胶或其组合。所述硅胶即硅酸干凝胶,是经脱水干燥后的硅凝胶,一般人们称为硅胶。硅胶的化学组成为mSiO2·nH2O,其基本结构单元是硅氧四面体。硅氧四面体以不同方式联结,若联结规则则可得SiO2晶体,无规则堆积则成类似于玻璃态的SiO2胶粒,构成硅凝胶的骨架。堆积时的孔隙在为水凝胶时被水填充,干凝胶时为空孔隙。硅凝胶的结构类似于玻璃态SiO2的结构。优选A型硅胶。A型硅胶又称细孔硅胶,外观呈透明或半透明玻璃状,分球形和块状。平均孔径2.0-3.0nm,比表面650-800m2/g。

在一个方面,所述包装的吸湿量可以为0.001至0.1g/ml,优选0.005至0.05g/ml;任选地,水蒸气透过量为0-15mg/24h*L,优选0-5mg/24h*L;任选地,吸湿速率为48h后1%-60%,优选48h后5%-10%。

实施例1.制剂的生产

a.将食品用香精、酸味剂(柠檬酸)、果粉(草莓粉)混合均匀,记为物料A;
b.将益生菌粉与益生元(菊粉)混合均匀,记为物料B;
c.将甜味剂(糖醇)、物料A、物料B加入料斗混合均匀;
d.将赋形剂(硬脂酸镁)加入料斗混合均匀,备用;
e.总混完成后用高速压片机进行压片。
(2)益生菌片剂B

实施例2.稳定性实验
1.选用6种不同的包材,分别对它们的吸湿量、水蒸气透过量和吸湿速率进行考察,结果如下:

HDPE瓶和分子筛盖:如本说明书所述,HDPE为高密度聚乙烯,其密度为0.935g/cm3至0.965g/cm3,分子筛为具有立方晶格的硅铝酸盐化合物,其化学组成通式为:(M)2/nO·Al2O3·xSiO2·pH2O,其中M代表金属离子,如K+、Na+、Ca2+或Ba2+,n代表金属离子价数,x代表SiO2的摩尔数,即硅铝比,p代表水的摩尔数,为A型分子筛,其比表面积:750-800m2/g,孔径为:0.2-0.5nm,分子筛2.5g;例如,使用了3A型分子筛,其孔径约为0.3nm。
HDPE瓶和硅胶盖:硅胶2.5g;
HDPE瓶和硅胶分子筛盖:分子筛与硅胶为6∶4,总量为2.5g;
泡腾管:管身为PP材质,分子筛与硅胶为6∶4;
ADP干燥剂瓶:瓶身为PP材质,干燥剂为ADP树脂;
铝管:管身为铝,干燥剂为硅胶。
测定方法如下:
(1)吸湿量的测定方法(恒温恒湿的条件是25℃,75%RH):
采用恒温恒湿试验箱(T=±2℃、RH=±2%)。
将单个防潮瓶称重,记为M0,将称量好的样品瓶盖打开,平置放入试验条件已达平衡的恒温恒湿试验箱内,每24h后取出玻璃皿称重,直至两次连续称重至恒重,记为M1
吸湿量X1按以下公式计算:
X1=(M1-M0)/V
式中:X1——样品的吸湿量,单位为g/ml;
M0——样品的初始质量,单位为g;
M1-样品吸附平衡后的质量,单位为g;
V——样品体积,单位为ml;
(2)水蒸气透过量的测定方法
采用YBB00092003-2015进行测定,采用恒温恒温箱(T=±0.6℃、RH=±2%、风速为0.5~2.5m/s),试验条件为25℃,RH95%。
取试验瓶适量,用干燥绸布擦净每个试瓶,将瓶盖连续开、关30次后,在试瓶内加入干燥剂无水氯化钙(除去过4目筛的细粉,置110℃干燥1小时):20ml或20ml以上的试瓶,加入干燥剂至距瓶口13mm处;小于20ml的试瓶,加入的干燥剂量为容积的2/3),立即将盖盖紧。另取两个试剂瓶装入与干燥剂相等量的玻璃小球,作对照用。试瓶紧盖后分别精密称定,然后将试瓶置于恒温恒湿箱中,放置72小时,取出,用干燥绸布擦干每个试瓶,室温放置45分钟,分别精密称定。按下式计算水蒸气透过量:

(3)吸湿速率
采用60%-10%-5%三点湿度指示卡和40℃/75%RH的恒温恒湿箱;
首先将湿度指示卡放置已经平衡的恒温恒湿箱箱里30min,确保湿度指示卡的湿度达到75%RH;然后将已经平衡的湿度指示卡放置于干燥瓶内封盖;每隔一定的时间开盖查看湿度指示卡的变化来判断瓶内湿度的变化快慢;
所用包装的来源均为市售。
从结果分析,选用的这几种包材综合吸湿量和水蒸气透过量,可认为这几种包材的防潮效果一样。吸湿速率是吸湿性能的另一重要参数,但吸湿速率检测是短期的,所考察的是包材对益生菌产品在长期储存的影响,故可认为短期吸湿速率对于长期益生菌片剂储存效果造成的影响较小。
2.分别对制得的益生菌片剂选用上述不同的包材进行加速试验,考察不同包材对益生菌的损耗影响。乳酸菌总数的测试采用GB4789.35法进行检测。测试结果如下表所示:

从以上结果可知,通过益生菌片剂不同包材的加速试验,在加速条件(温度为37±2℃、相对湿度为RH75±5%)下,使用“HDPE瓶+分子筛盖”、“HDPE瓶+硅胶盖”或“HDPE瓶+硅胶分子筛盖”包材的活菌总数降低最少,较其他包材更加稳定。由于这几种包材本身的防潮效果相近,使用“HDPE瓶+分子筛盖”、“HDPE瓶+硅胶盖”或“HDPE瓶+硅胶分子筛盖”包材获得的显著较高的稳定性是预料不到的。尤其预料不到的是,根据第6个月的数据,使用“HDPE瓶+分子筛盖”、“HDPE瓶+硅胶盖”或“HDPE瓶+硅胶分子筛盖”包材获得的稳定性更加显著地优于其它包材,说明“HDPE瓶+分子筛盖”、“HDPE瓶+硅胶盖”或“HDPE瓶+硅胶分子筛盖”包材更加适用于益生菌的长期储存。
实施例3
采用HDPE瓶+分子筛盖包装其它益生菌片剂,提供加速3月稳定性数据(加速3月相当于长期18月),发现以下三款产品在加速3月时能保持较高的活菌数。

Claims (10)

1.一种包含益生菌的产品,包括包装(例如瓶)和位于包装中的益生菌,所述包装具有外壳和盖,所述外壳包含高密度聚乙烯材料,所述盖包含分子筛、硅胶或其组合。
2.根据权利要求1所述的包含益生菌的产品,其中所述高密度聚乙烯的密度为0.935g/cm3至0.965g/cm3
3.根据权利要求1所述的包含益生菌的产品,其中所述分子筛选自水合硅铝酸盐或天然沸石,所述水合硅铝酸盐优选为具有立方晶格的硅铝酸盐化合物,其化学组成通式为:(M)2/nO·Al2O3·xSiO2·pH2O,其中M代表金属离子,如K+、Na+、Ca2+或Ba2+,n代表金属离子价数,x代表SiO2的摩尔数,即硅铝比,p代表水的摩尔数;例如,该分子筛为A型分子筛,其比表面积:750-800m2/g,孔径为:0.2-0.5nm,优选3A型分子筛,其孔径约为0.3nm;所述硅胶优选A型硅胶,例如平均孔径2.0-3.0nm,例如比表面650-800m2/g。
4.根据权利要求1所述的包含益生菌的产品,其中所述包装的吸湿量为0.001至0.1g/ml,优选0.005至0.05g/ml;任选地,水蒸气透过量为0-15mg/24h*L,优选0-5mg/24h*L;任选地,吸湿速率为48h后1%-60%,优选48h后5%-10%。
5.根据权利要求1所述的包含益生菌的产品,其中所述益生菌包含于组合物中,所述组合物包含所述益生菌、甜味剂和赋形剂,任选地包含益生元;例如所述组合物为固体制剂形式,例如选自片剂(例如咀嚼片、分散片、含片、用于吞咽的片剂)、丸剂、胶囊剂、颗粒剂或散剂的形式。
6.根据权利要求5所述的包含益生菌的产品,其中所述益生菌选自双歧杆菌属、乳杆菌属、酵母属或其组合;优选地,选自嗜酸乳杆菌、乳双歧杆菌、长双歧杆菌、动物双歧杆菌、瑞士乳杆菌、鼠李糖乳杆菌、罗伊氏乳杆菌、植物乳杆菌、啤酒酵母或其组合。
7.根据权利要求5所述的包含益生菌的产品,其中所述益生元是菊粉。
8.根据权利要求5所述的包含益生菌的产品,其中所述甜味剂是糖醇或糖,例如所述糖醇或糖选自甘露糖醇、山梨醇、异麦芽酮糖醇、乳糖或其组合。
9.根据权利要求5所述的包含益生菌的产品,其中所述赋形剂为硬脂酸镁。
10.根据权利要求5所述的包含益生菌的产品,其中所述组合物还包含果粉(例如草莓粉)、酸味剂(例如柠檬酸)、奶粉、微晶纤维素、矿物质类(例如碳酸钙)、维生素类(例如维生素C)或香精,或其任何组合;例如,所述组合物包含按重量计0.1%-20%,优选1%-15%的益生菌;0%-30%,优选1%-20%的益生元;0.1%-5%,优选0.1%-2%的赋形剂;30%-90%,优选50%-80%的甜味剂;0%-40%,优选5%-30%的果粉;0%-2.0%,优选0.2%-1.0%的酸味剂;和0%-2%,优选0.2%-0.8%的香精。
