Abstract
Das neue Verfahren zur Iodierung von substituierten Azolen, insbesondere zur Iodierung von substituierten 1H-Tetrazolen und substituierten 1H-Triazolen, liefert die gewünschten Verbindungen in hoher Reinheit und mit guter Ausbeute.
Description
Verfahren zur Herstellung von Jodierten Azolen
Die vorliegende Erfindung betrifft ein neues Verfahren zur Iodierung von substituierten Azolen, insbesondere zur Iodierung von substituierten 1H-Tetrazolen und substituierten 1 H-Triazolen.
Azole, insbesondere 2-oder 5-substituierte 1H-Triazole und 5-substituierte 1H-Tetrazole, werden unter anderem als pharmazeutisch aktive Substanzen in der Medizin eingesetzt oder werden beispielsweise zum Schutz von Pflanzen oder technischen Materialien als Biozide beschrieben. Zudem können halogenierte 1H-Triazole oder 1H-Tetrazole als Zwischenprodukte bei der Synthese abgeleiteter Derivate eingesetzt werden.
Für die Synthese von 5-substituierten 1 H-Triazolen und -tetrazolen geht man üblicherweise von den entsprechenden 5-H-substituierten Verbindungen aus. Diese werden üblicherweise durch Lithiierung bei sehr tiefer Temperatur und Behandlung mit einem Elektrophil in die entsprechenden 5 -substituierten Derivate umgewandelt. Folgendes Beispiel dient zur Erläuterung des nächstliegenden Standes der Technik.
So wird von Yoshitaka Satoh und Nicholas Marcopulos (vgl. Tetrahedron Letters (1995), 36(1 1), 1759-62) eine Methode zur Anwendung der Lithiierung von 1-Benzyl- und 1-para- Methoxybenzyltetrazolen an der 5-Position beschrieben. Reaktion mit n-Butyllithium gefolgt von Behandlung mit Elektrophilen ergab 5-funktionalisierte 1 -benzylische Tetrazole. Zusammenfassend kann gesagt werden, dass die Lithiierung, als nächstliegender Stand der Technik die Methode der Wahl zur Derivatisierung der 5-Position, zum Beispiel durch Halogene, darstellt, wobei die tiefe Temperatur, die Verwendung von luftempfindlichen und wenig preiswerten Metallierungsreagenzien wie n- Butyllithium und besonders auch die völlige Instabilität der metallierten Zwischenstufe schon bei Temperaturen über -78°C, sehr von Nachteil sind.
Die Aufgabe der vorliegenden Erfindung bestand darin, ein verbessertes Verfahren zur Herstellung von iodierten Azolen bereit zu stellen.
Überraschenderweise wurde nun ein neues Verfahren zur Herstellung von iodierten Azolen gefunden, wodurch die im Stand der Technik beschriebene Lithiierung sowie die Durchführung der Reaktion bei tiefer Temperatur umgangen werden kann.
Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Herstellung von ein oder zweifach iodierten Azolen der allgemeinen Formel (I), oder deren Salzen und Säureadditionsverbindungen,

R3 für jeweils gegebenenfalls substituiertes Alkyl, Alkenyl, Alkinyl, Cycloalkyl, Phenyl oder Phenethyl steht, und
R4 für jeweils gegebenenfalls substituiertes Alkyl, Alkenyl, Alkinyl, Phenyl Phenethyl oder I steht,
durch Umsetzung von Verbindungen der Formel (II) oder deren Salzen und Säureadditionsverbindungen,
worin
A, B und R1 die für Formel (I) angegebene Bedeutung haben,
mit mindestens einem Oxidationsmittel und elementarem Iod und/oder mindestens einer Iodve- rbindung, gegebenenfalls in Gegenwart eines Oxidationskatalysators und gegebenenfalls in Gegenwart von einem Lösungsmittel oder Lösungsmittelgemisch bei einer Temperatur zwischen 00C und 2000C.
Bevorzugt dient das erfindungsgemäße Verfahren zur Herstellung von Verbindungen der allgemeinen Formel (I), in welcher

mit der Maßgabe, dass mindestens einer von A und B für N steht.
Das erfϊndungsgemäße Verfahren dient auch zur Herstellung von Salzen und Säureadditionsverbindungen der Verbindungen der Formel (I), wie z.B. Hydrohalogeniden, Hydrophosphonaten oder Hydrosulfaten wobei als Ausgangsmaterialien dann die entsprechenden Salze und Säureadditionsverbindungen der Formel (II) eingesetzt werden..
Zur Durchführung des erfindungsgemäßen Verfahrens werden Verbindungen der Formel (II) eingesetzt. Die Substituenten R1, A und B in Formel (II) entsprechen in ihrer allgemeinen, bevorzugten, besonders bevorzugten und ganz besonders bevorzugten Bedeutung den entsprechenden, oben aufgeführten Bedeutungen der Substituenten R1, A und B in Formel (I). Die Verbindungen der Formel (II) sind dem Fachmann seit langem aus der Literatur bekannt.
Als Oxidationsmittel für die Durchführung des erfindungsgemäßenVerfahrens eignen sich beispielsweise Wasserstoffperoxid, Kaliumperoxomonosulfat, Persäuren wie z.B. Peressigsäure
oder m-Chloφerbenzoesäure weiterhin Sauerstoff, angeregter Sauerstoff, Hypochlorid, Perchlorate, Perborate, Percarbonate, Luft oder ähnliche aktiven Sauerstoff enthaltende Reagenzien oder Mischungen davon.
Bevorzugt wird Wasserstoffperoxid als wässrige Lösung eingesetzt. Die Oxidationsmittel werden im allgemeinen in Mengen von 0,5 bis 100 Äquivalenten bezogen auf das Azol (II) eingesetzt. Bevorzugt werden 1 bis 50 Äquivalente bezogen auf das Azol (II) eingesetzt.
Das erfindungsgemäße Verfahren wird im allgemeinen bei Temperaturen zwischen 00C und 2000C, bevorzugt zwischen 250C und 1300C und besonders bevorzugt zwischen 600C und 1000C durchgeführt.
Als Lösungsmittel kommen sowohl Wasser alleine als auch die gebräuchlichen organischen Lösungsmittel in Frage, welche durch die Oxidationsmittel nicht angegriffen werden wie beispielsweise Petrolether, n-Octan, n-Pentan, n-Hexan, Cyclohexan, n-Pentan, Toluol, Benzol, THF, Diethylether, MethyW-butylether, Diglyme, Methanol, Ethanol, Isopropanol, n-Butanol, tert- Butanol, 2-Butanol, iso-Butanol, n-Hexanol, CH2Cl2, CHCI3. Vorteilhafterweise können auch Mischungen von zwei oder mehr Lösungsmitteln eingesetzt werden. Vorzugsweise sind die Lösungsmittel untereinander mischbar. Bevorzugt werden Wasser und Alkohole verwendet. Besonders bevorzugt werden Gemische aus Wasser und Methanol oder Ethanol oder Propanol oder Butanol oder Pentanol oder Hexanol verwendet. Die unterschiedlichen stellungsisomeren Derivate der Alkohole eignen sich dabei alle, können jedoch deutliche Unterschiede bei den Ausbeuten liefern. Das Mischungsverhältnis Wasser/Alkohol kann in weiten Grenzen variieren, vorzugsweise liegt das Verhältnis zwischen 10:1 und 1 :10.
Als Iod-Quelle eignet sich besonders elementares Iod. Ebenfalls geeignet sind Ioddepotstoffe wie Iod-Stärke-Verbindungen. Andere Iodverbindungen wie NaI, KI, Natriumperiodat, Iodsauerstoffsäuren oder hypervalente Iodverbindungen können ebenfalls eingesetzt werden. Ebenso können Mischungen der vorgenannten Iodverbindunge untereinander oder mit Iod eingesetzt werden. Iod bzw. die Iodverbindung wird in Mengen zwischen 0,1 bis 10,0 Äquivalenten, vorzugsweise zwischen 0,4 bis 2 Äquivalenten bezogen auf das Azol eingesetzt.
Oxidationskatalysatoren wie Metalloxide, bevorzugt Co-, Fe- oder Ni- Komplexe, wie beispielsweise Co-meso-Tetraphenylpoφhin können zur Verbesserung der Ausbeuten ebenfalls eingesetzt werden. Der Katalysator wird dabei in Mengen zwischen 0,001 und 3,0 Mol-% bezogen auf das Azol eingesetzt.
Man kann die Reaktion bei unterschiedlichen Rührgeschwindigkeiten durchführen um für eine gute Durchmischung der Reaktanden zu sorgen.
Zur Durchführung des erfindungsgemäßen Verfahrens geht man allgemein so vor, dass man die Lösung oder Suspension des Eduktes mit dem Oxidationsmittel und ggf. dem Katalysator in einem geeigneten Lösungsmittelsystem vorlegt und elementares Iod und/oder Iodverbindung, in gelöster oder fester Form, in geeigneter Geschwindigkeit unter Rühren zudosiert. Umgekehrt kann auch zunächst das Edukt mit Iod und/oder Iodverbindung im Lösungsmittelsystem vorgelegt werden und dann das Oxidationsmittel und ggf. der Katalysator in Substanz, oder gelöst in Wasser oder einem gebräuchlichen Lösungsmittel, geeignet zudosiert werden. Während oder nach dem Zudosieren wird die Mischung auf eine geeignete Temperatur erhitzt. Bevorzugt wird die Reaktion innerhalb 1 bis 100 Stunden durchgeführt.
Die optimalen Bedingungen hängen vom Substrat und dessen Reaktivität und Löslichkeit ab und müssen jeweils bestimmt werden.
Zur Aufarbeitung kann beispielsweise das Endprodukt durch Extraktion und gegebenenfalls anschließende Reinigungsschritte wie z.B. durch Chromatographie isoliert werden.
Das erfindungsgemäße Verfahren dient zur Iodierung von Azolverbindungen der Formel (I). In analoger Weise lassen sich auch die entsprechenden bromierten Azolverbindungen herstellen, wenn anstelle von elementarem Iod und/oder Iodverbindungen elementares Brom und/oder Bromverbindungen wie z.B NaBr, KBr oder Bromwasserstoff eingesetzt werden.
Das erfindungsgemäße Vorgehen hat gegenüber bisher angewendeten Verfahren eine Reihe von Vorteilen:
Es kann in sehr preiswerten Lösungsmitteln gearbeitet werden.
Die Reaktion benötigt keine Kühlung.
Alkyl-substituierte Verbindungen können in guten Ausbeuten hergestellt werden.
Iod wird nur stöchiometrisch benötigt, d.h. beide Iodatome des Iodmoleküls werden in das Produkt eingebaut.
Die eingesetzten Reagenzien sind preiswert verfügbar.
Die Reaktion ist leicht auf einen großen Maßstab übertragbar.
Das Endprodukt entsteht in hoher Ausbeute und Reinheit.
Folgende Beispiele sollen zur Veranschaulichung des erfindungsgemäßen Verfahrens dienen ohne dieses jedoch einzuschränken:
Beispjele:
Beispiel 1
Synthese von l-(2-Ethylhexyl)-5-iodo-lH-tetrazol
Es wurden 0.500 g l-(2-Ethylhexyl)-lH-tetrazol (2.74 mmol) in 10 ml 35%igem H2O2 und 10 ml tert-Butanol vorgelegt und unter kräftigem Rühren 0.348 g fein pulverisiertes Iod (1.37 mmol) zugegeben. Danach wurde 60 h bei 80°C gerührt. Der Reaktionsansatz wurde eingeengt, in etwas
Wasser aufgenommen, mit Ethylacetat extrahiert, und die organische Phase mit wässrigem
Natriumthiosulfat ausgeschüttelt. Die organische Phase wurde über Na2SO4 getrocknet und dann eingeengt. Nach Reinigung über Silicagel (Toluol/Ethylacetat) erhielt man 0.623 g l-(2- Ethylhexyl)-5-iodo-lH-tetrazol als farbloses Öl (74% Ausbeute). 1H-NMR (400 MHz, CDCl3) δ
[ppm] 0.90 (t, 3H), 0.92 (t, 3H), 1.30 (m, 8H), 2.02 (m, IH), 4.26 (d, 2H).
Beispiel 2
Synthese von l-(2-Ethylhexyl)-5-iodo-lH-tetrazol
Es wurden 0.500 g l-(2-Ethylhexyl)-lH-tetrazol (2.74 mmol) in 10 ml 35%igem H2O2 und 5.0 ml tert-Butano\ vorgelegt und unter kräftigem Rühren 0.696 g fein pulverisiertes Iod (2.74 mmol) zugegeben. Danach wurde 20 h bei 800C gerührt. Der Reaktionsansatz wurde eingeengt, in etwas
Wasser aufgenommen, mit Ethylacetat extrahiert, und die organische Phase mit wässrigem
Natriumthiosulfat ausgeschüttelt. Die organische Phase wurde über Na2SO4 getrocknet und dann eingeengt. Nach Reinigung über Silicagel (Toluol/Ethylacetat) erhielt man 0.716 g l-(2- Ethylhexyl)-5-iodo-lH-tetrazol als farbloses Öl (85% Ausbeute). 1H-NMR (400 MHz, CDCl3) δ
[ppm] 0.90 (t, 3H), 0.92 (t, 3H), 1.30 (m, 8H), 2.02 (m, IH), 4.26 (d, 2H).
Beispiel 3
Synthese von l-(2-Ethylhexyl)-5-iodo-lH-tetrazol
Die Reaktion wurde wie unter Beispiel 1 angegeben durchgeführt, nur dass zusammen mit dem Tetrazol noch 0.1 mol % Co-meso-Tetraphenylporphin als Katalysator bezogen auf das Tetrazol vorgelegt wurden. Die Ausbeute betrug 78 %.
Beispiel 4
Synthese von 5-Iodo-l-octyl-lH-tetrazol
Es wurden 0.250 g 1-Octyl-lH-tetrazol (1.37 mmol) in 5.0 ml 35%igem H2O2 und 5.0 ml Methanol vorgelegt und unter kräftigem Rühren 0.348 g fein pulverisiertes Iod (1.37 mmol) zugegeben. Danach wurde 20 h bei Rückfluß gerührt. Der Reaktionsansatz wurde eingeengt, in etwas Wasser aufgenommen, mit Ethylacetat extrahiert, und die organische Phase mit wässrigem
Natriumthiosulfat ausgeschüttelt. Die organische Phase wurde über Na2SO4 getrocknet und dann eingeengt. Nach Reinigung über Silicagel (Toluol/Ethylacetat) erhielt man 0.253 g 5-Iodo-l-octyl- lH-tetrazol als leicht gelbes Öl (60% Ausbeute). 1H-NMR (400 MHz, CDCl3) δ [ppm] 0.90 (t, 3H), 1.30 (m, 1 OH), 1.93 (m, 2H), 4.37 (t, 2H).
Beispiel 5
Synthese von 5-Iodo-l-octyl-lH-tetrazol
Es wurden 0.500 g 1-Octyl-lH-tetrazol (2.74 mmol) in 10 ml 35%igem H2O2 vorgelegt und unter kräftigem Rühren 0.696 g fein pulverisiertes Iod (2.74 mmol) zugegeben. Danach wurde 20 h bei 800C gerührt. Der Reaktionsansatz wurde mit Ethylacetat extrahiert, und die organische Phase mit wässrigem Natriumthiosulfat ausgeschüttelt. Die organische Phase wurde über Na2SO4 getrocknet und dann eingeengt. Nach Reinigung über Silicagel (Toluol/Ethylacetat) erhielt man 0.389 g 5- Iodo-1-octyl-lH-tetrazol als farbloses Öl (46% Ausbeute). 1H-NMR (400 MHz, CDCl3) δ [ppm] 0.90 (t, 3H), 1.30 (m, 10H), 1.93 (m, 2H), 4.37 (t, 2H).
Beispiel 6
Synthese von l-(2-Chlorobenzyl)-5-iodo-lH-l,2,4-triazol
Es wurden 0.250 g l-(2-Chlorobenzyl)-lΗ-l,2,4-triazol (1.29 mmol) in 5.0 ml 35%igem H2O2 und 5.0 ml Methanol vorgelegt und unter kräftigem Rühren 0.164 g fein pulverisiertes Iod (0.65 mmol) zugegeben. Danach wurde 20 h bei 65°C gerührt. Der Reaktionsansatz wurde eingeengt, in etwas Wasser aufgenommen, mit Ethylacetat extrahiert, und die organische Phase mit wässrigem Natriumthiosulfat ausgeschüttelt. Die organische Phase wurde über Na2SO4 getrocknet und dann eingeengt. Nach Reinigung über Silicagel (Toluol/Ethylacetat) erhielt man 0.100 g l-(2- Chlorobenzyl)-5-iodo-lH-l,2,4-triazol als weißen Feststoff (24% Ausbeute). Smp. 98°C. 1H-NMR (400 MHz, CDCl3) δ [ppm] 5.50 (s, 2H), 6.86 (m, IH), 7.27 (m, 2H), 7.43 (m, IH), 8.00 (s, IH).
Beispiel 7
Synthese von 3-Iodo-4-(3-methoxy-4-methylphenyl)-4H-l,2,4-triazol [entspricht 5-Iodo-l-(3- methoxy-4-methylphenyl)- 1 H- 1 ,3 ,4-triazol]
Es wurden 0.250 g 4-(3-Methoxy-4-methylphenyl)-4H-l,2,4-triazol (1.32 mmol) in 5.0 ml 35%igem H2O2 und 5.0 ml tert-Butanol vorgelegt und unter kräftigem Rühren 0.164 g fein pulverisiertes Iod (0.65 mmol) zugegeben. Danach wurde 80 h bei 800C gerührt. Der Reaktionsansatz wurde eingeengt, in etwas Wasser aufgenommen, mit Ethylacetat extrahiert, und die organische Phase mit wässrigem Natriumthiosulfat ausgeschüttelt. Die organische Phase wurde über Na2SO4 getrocknet und dann eingeengt. Nach Reinigung über Silicagel (Toluol/Ethylacetat) erhielt man 0.240 g 3-Iodo-4-(3-methoxy-4-methylphenyl)-4H-l,2,4-triazol als weißen Feststoff (58% Ausbeute). RF-Wert: 0.57 (EtOAc/Et3N = 4/1). Smp.: 123°C. 1H-NMR (400 MHz, CDCl3) δ [ppm] 2.29 (s, 3H), 3.87 (s, 3H), 6.73 (s, IH), 6.82 (d, IH), 7.27 (d, IH), 8.31 (s, IH).
Beispiel 8
Synthese von 3,5-Diiodo-4-(3-methoxy-4-methylphenyl)-4H-l,2,4-triazol [entspricht 2,5-Diiodo-l- (3-methoxy-4-methylphenyl)-lH-l ,3,4-triazol]
Es wurden 0.250 g 4-(3-Methoxy-4-methylphenyl)-4Η-l,2,4-triazol (1.32 mmol) in 5.0 ml 35%igem H2O2 und 5.0 ml tert-Butanol vorgelegt und unter kräftigem Rühren 0.164 g fein pulverisiertes Iod (0.65 mmol) zugegeben. Danach wurde 80 h bei 800C gerührt. Der Reaktionsansatz wurde eingeengt, in etwas Wasser aufgenommen, mit Ethylacetat extrahiert, und die organische Phase mit wässrigem Natriumthiosulfat ausgeschüttelt. Die organische Phase wurde über Na2SO4 getrocknet und dann eingeengt. Nach Reinigung über Silicagel (Toluol/Ethylacetat) erhielt man 0.120 g 3,5-Diiodo-4-(3-methoxy-4-methylphenyl)-4H-l,2,4-triazol als hellen Feststoff (21% Ausbeute). RF-Wert: 0.77 (EtOAc/Et3N = 4/1). Smp.: 176°C. 1H-NMR (400 MHz, CDCl3) δ [ppm] 2.30 (s, 3H), 3.87 (s, 3H), 6.61 (s, IH), 6.74 (d, IH), 7.30 (d, IH).
Claims
1. Verfahren zur Iodierung von substituierten Azolen der allgemeinen Formel (I), oder deren Salzen und Säureadditionsverbindungen,

B für N, CH, CR4 oder CI steht,
mit der Maßgabe, dass mindestens einer von A und B für N steht,
R1 für Wasserstoff oder für jeweils gegebenenfalls substituiertes Alkyl, Alkenyl, Alkinyl, Cycloalkyl oder Phenyl steht,
R2 für I steht,
R3 für jeweils gegebenenfalls substituiertes Alkyl, Alkenyl, Alkinyl, Cycloalkyl,
Phenyl oder Phenethyl steht, und
R4 für jeweils gegebenenfalls substituiertes Alkyl, Alkenyl, Alkinyl, Cycloalkyl, Phenyl oder Phenethyl steht,
durch Umsetzung von Verbindungen der Formel (II), oder deren Salzen und Säureadditionsverbindungen,
worin
A, B und R1 die oben für Formel (I) angegebene Bedeutung haben,
mit mindestens einem Oxidationsmittel und elementarem Iod und/oder mindestens einer Iodverbindung in Gegenwart eines Lösungsmittels oder Lösungs- mittelgemischs bei einer Temperatur zwischen 00C und 2000C.
2. Verfahren gemäß Anspruch 1, dadurch gekennzeichnet, dass als Oxidationsmittel Wasserstoffperoxid, Kaliumperoxomonosulfat, Persäuren, Sauerstoff, angeregter Sauerstoff, Hypochlorid, Perchlorate, Perborate, Percarbonate, Luft oder aktiven
Sauerstoff enthaltende Reagenzien oder deren Mischungen eingesetzt werden.
3. Verfahren gemäß wenigstens einem der Ansprüche 1 und 2, dadurch gekennzeichnet, dass das Oxidationsmittel in einer Menge von 1 bis 100 Äquivalenten bezogen auf das Azol der Formel (II) eingesetzt wird.
4. Verfahren gemäß wenigstens einem der Ansprüche 1 bis 3, dadurch gekennzeichnet, dass elementares Iod und/oder Iodverbindungen in einer Menge von 0,1 bis 10,0 Äquivalenten pro Äquivalent an Azol der Formel (II) eingesetzt werden.
5. Verfahren gemäß wenigstens einem der Ansprüche 1 bis 4, dadurch gekennzeichnet, dass als Lösungsmittel Wasser oder Alkohole oder Gemische daraus verwendet werden.
6. Verfahren gemäß wenigstens einem der Ansprüche 1 bis 5, dadurch gekennzeichnet, dass als Oxidationskatalysator mindestens eine Verbindung aus der Reihe der Fe-, Co- oder Ni- Komplexe in einer Menge von 0,001 bis 3,0 Mol-% bezogen auf. das Azol der Formel (II) eingesetzt wird.
