:

EMI1.3

3. The method according to claim 1 for the preparation of 1 - [(2-chlorophenyl) diphenylmethyl] imidazole, characterized in that o-chlorophenyldiphenylmethanol is reacted in a solvent with tri- (l -imidazolyl) phosphine.
The present invention relates to a new, improved process for the preparation of l- (triphenylmethyl) imidazoles of the following general formula:
EMI1.4
wherein Rl, R3, which can be the same or different, hydrogen atoms, alkyl radicals and / or phenyl radicals, Xl, X2 and X3, which can be the same or different, alkyl radicals, halogen atoms or nitro, trifluoromethyl, cyano, alkylthio or Alkoxy groups and ni, n2 and n3 are integers, namely the numbers 0, 1 and / or 2.
The 1-tritylimidazole compounds of Formula I are already known as antifungal and antibacterial agents (e.g. U.S. Patents 3,321,366 and 3,705,172).
As can be seen from said patents, the 1-tritylimidazole compounds can be obtained by reacting halides and salts of tritylcarbinol derivatives with silver or sodium salts of imidazole derivatives. However, this method has certain disadvantages. For example, the halides and salts of tritylcarbinol derivatives are not stable in water and are therefore difficult to manufacture. The silver salts of imidazole derivatives, on the other hand, are expensive and unstable when left to stand under the influence of light. Furthermore, the yield of the desired compounds is poor.
The 1-tritylimidazole compounds can also be obtained by reacting tritylcarbinol derivatives with imidazole derivatives. Compared to the aforementioned known method, this method does not require expensive and unstable starting materials, but the reaction must be carried out at very high temperatures for a long period of time in order to obtain the desired compounds even in low yields, because this reaction does not proceed smoothly under ordinary conditions is going. This process is therefore unsatisfactory for the economical production of l-tritylimidazole compounds.
Based on extensive studies, a more convenient and economical process for the preparation of 1-tritylimidazole compounds has now been developed, and it has been found that the reaction of tritylcarbinol derivatives with tri- (l -imidazolyl) phosphine derivatives under
mild conditions, the l-tritylimidazole compounds are obtained in high yields.
It has also been found that o-chlorophenyldiphenylmethanol, which is used as a starting material for the preparation of the extremely effective fungicidal compound known as clotrimazole in the present process, can be obtained in high yield by the fact that the Friedel Crafts reaction of o -Chlorbenzotrichlorid with benzene in the presence of aluminum chloride reaction mixture obtained is treated with water.
The present invention therefore provides a process for the preparation of 1-tritylimidazole compounds
Formula I, which consists in that a tritylcarbi nol derivative of the following general formula:
EMI2.1
wherein Xl, X2, X3, nl, n and n3 have the above meanings, with a tri- (l-imidazolyl) phosphine derivative of the following general formula:
EMI2.2
wherein Rl, R2 and R3 have the above meanings, in a solvent at a temperature in the range between -5 C and 1300C.
According to a preferred embodiment, 1 - [(2-chlorophenyl) diphenylmethyl] imidazole is obtained by treating the reaction mixture, which is obtained by reacting o-chlorobenzotrichloride with benzene in the presence of aluminum chloride, with water, the o -Chlorphenyldiphenylmethan arrives, whereupon the o-chlorophenyldiphenylmethanol is reacted with tri- (l -imidazolyl) phosphine.
In the definition mentioned above and, if appropriate, also in the description below, the alkyl groups Rs, R2, R3, Xl, X2 and X3 can be both straight-chain and branched alkyl groups having 1 to 12 carbon atoms and preferably having 1 to 4 carbon atoms . Examples of preferred alkyl groups are methyl, ethyl, n-propyl, isopropyl, n-butyl, isobutyl, sec-butyl etc.
Halogen atoms are e.g. Fluorine, chlorine, bromine or iodine in question. As alkylthio and alkoxy, those with 1 to 4 carbon atoms will be preferred.
According to the invention, the l-tritylimidazole compounds of the formula I are obtained by reacting a tritylcarbinol derivative of the formula II with a tri- (l-imidazolyl) phosphine derivative of the formula III usually at a temperature in the range from approximately -5 ° C. to 1300 ° C. and preferably in Reacts ranges from about 0 to 1 00 C in a suitable solvent.
When carrying out the reaction, the tri (l-imidazolyl) phosphine derivative of the formula III will generally be used in an amount of more than 1/3 mole per mole of the tritylcarbinol derivative of the formula II, but good results are also obtained if 2/3 to 3 mol of tri (1-imidazolyl) phosphine derivative of the formula III per 1 mol of the tritylcarbinol derivative of the formula II is used.
Aromatic hydrocarbons, such as e.g. Benzene or toluene, halogenated aliphatic hydrocarbons, e.g. Name chloroform or dichloromethane, tetrahydrofuran, methyl isobutyl ketone, dimethylformamide, pyridine, acetonitrile and mixtures of such compounds.
The tritylcarbinol derivatives of the formula II and tri- (l-imidazolyl) -phosphine derivatives of the formula III used as starting materials are known compounds and can be prepared by known methods. [J.
Org. Chem., 7,392 (1942); J. Am. Chem. Soc., 33,531 (1911); Angew. Chem., 73, 143 (l961) j.
The tri (l-imidazolyl) phosphine derivatives of the formula III can also be obtained by using an imidazole derivative of the following general formula:
EMI2.3
wherein Rl, Rund R3 have the above meanings, with phosphorus trichloride in the presence of an acid-binding agent in a suitable solvent.
The tri- (l-imidazolyl) phosphine derivatives of the formula III are sensitive to moisture. It is therefore advisable to carry out the reaction in solution with the tritylcarbinol derivatives of the formula II without isolation.
The invention is illustrated by the following examples without, however, being restricted thereto.

A solution of 4.48 g of imidazole and 6.96 g of triethylamine in 80 ml of acetonitrile is added dropwise with 3.92 g of phosphorus trichloride at room temperature and the mixture is stirred for 1 hour. 5.90 g of the o-chlorophenyldiphenylmethanol obtained as described in Example 1 (B) are then added and the mixture is heated to boiling under reflux for 2 hours. The solvent is then evaporated and the residue is heated to boiling under reflux for 2 hours. The solvent is then evaporated and the residue extracted with chloroform. The extract is treated in the same manner as in Example 3 to obtain 5.72 g of l- (o-chlorophenyldiphenylmethyl) imidazole.
Yield: 83%. Mp 142 to 143 c.
Example 7
Preparation of l- (o-chlorophenyldiphenylmethyl) imidazole
3.43 g of phosphorus trichloride are added dropwise to a solution of 10 g of imidazole in 50 ml of chloroform at 10 ° C. After the mixture has been left to stand at 5 to 10 ° C. for 1 hour, the imidazole hydrochloride is precipitated and then filtered off with suction , whereupon the filtrate is concentrated. 7.08 g of o-chlorophenyldiphenylmethanol are then added and the mixture is heated for 5 minutes to 40 to 60 ° C. Water is then added and the mixture is extracted with chloroform.
The extract is treated in the same manner as in Example 3 to obtain 6.95 g of l- (o-chlorophenyldiphenylmethyl) imidazole. Yield: 84%. 140 to 142 "C.
Example 8
Preparation of 1 -triphenylmethylimidazole
The reaction is carried out in the same manner as in Example 3, but using triphenylmethanol instead of o-chlorophenyldiphenylmethanol. The crystalline residue thus obtained is washed with isopropyl ether, crude 1-triphenylmethylimidazole being obtained in a yield of 98.4%. Recrystallization from acetonitrile gives pure 1-triphenylmethylimidazole in a yield of 83%. M.p. 220-221 "C.
Elemental analysis for C22HIsN2: Calc .: C 85.13%; H 5.85%; N 9.03%.
Found: C 85.10%; H 5.69%; N 8.97%.
Example 9
Preparation of 1 -triphenylmethylimidazole
The reaction is carried out in the same manner as in Example 7, using triphenylmethanol instead of o-chlorophenyldiphenylmethanol. By recrystallizing the crystalline residue thus obtained from a mixture of acetonitrile and chloroform, pure is obtained
1 -Triphenylmethylimidazole in a yield of 90%. The melting point is 220 to 221 C.
Example 10
Preparation of 1 -triphenylmethylimidazole
The reaction is carried out in the same way as in Example 2, but using triphenylmethanol instead of o-chlorophenyldiphenylmethanol. By recrystallizing the crystalline mass thus obtained from acetonitrile, pure 1-triphenylmethylimidazole is obtained in a yield of 81%. The melting point is 219 to 220 "C.
