Bei den Halogenatomen handelt es sich um Chlor, Fluor, Brom, insbesondere Chlor und Fluor. Bei den oben genannten niedrigmolekularen Alkyl-, Alkenyl- und Hydroxyalkylgruppen handelt es sich um solche mit 1 bis 6 Kohlenstoffatomen, insbesondere 1 bis 4 Kohlenstoffatomen. Die Alkylreste der Dialkylaminogruppe bestehen aus 1 bis 6 Kohlenstoffatomen, insbesondere 1 bis 4 Kohlenstoffatomen. Bei den aliphatischen Acylgruppen handelt es sich um solche mit 2 bis 6 Kohlenstoffatomen ; es kommen die gesättigten Acylgruppen in Frage. Bei den Dicarbonsäuren handelt es sich insbesondere um solche mit 3 bis 6, vorzugsweise 3 bis 5 Kohlenstoffatomen. Beispiele hiefür sind Malonsäure, Bernsteinsäure, Glutarsäure, Adipinsäure. Die Alkylgruppen als solche oder als Bestandteil anderer Gruppen können gerade oder verzweigt sein.
Beispiele für die zuletzt genannten Bedeutungen sind : Methyl, Äthyl, Isopropyl, Butyl, tert. Butyl, Hexyl, Isobutyl, Cyclopropyl, Cyclohexyl, Cyclohexylpropyl, Cyclopropylmethyl, Cyclohexylpentyl, Oxyäthyl, Oxypentyl, Dimethylamino, Diäthylamino, Dibutylamino, Acetyl, Propionyl, Butyryl, Pentanol, Isovaleroyl, Isobutyryl, Cyclobutylmethyl, Allyl, Butenyl- (2), Piperidinoäthyl, Morpholinoäthyl.
Die erfindungsgemäss erhältlichen Verbindungen haben wertvolle pharmakodynamische Eigenschaften.
Beispielsweise besitzen sie psychosedative und insbesondere anxiolytische Eigenschaften. Darüber hinaus ist auch eine antiphlogistische Wirkung vorhanden.
Die Herstellung der Verbindungen der Formel (1) erfolgt erfindungsgemäss dadurch, dass man eine Verbindung der allgemeinen Formel

worin R,R,R,R und Z die oben angegebenen Bedeutungen haben, entsprechend acyliert bzw. alkyliert insbesondere mit einer Verbindung der Formel R4 Y, worin R4 die vorstehend genannte Bedeutung hat und Y entweder, für den Fall R4 gleich niedrigmolekulare Alkylgruppe, ein Halogenatom oder ein Sulfonsäure-oder Monoalkylsulfatrest oder, für den Fall R gleich entsprechende niedere Acylgruppe, ein Halogenatom, eine niedere aliphatische Alkoxygruppe oder Acyloxygruppe ist, in einem Lösungs- oder Suspensionsmittel bei Temperaturen zwischen 0 und 200 0,

Als Alkylierungsmittel kommen beispielsweise in Betracht : Ester der FormelR Hal, ArSO ORR und SO (OR4 )2 wobei Hai ein Halogenatom (insbesondere Chlor, Brom oder Jod) und Ar ein aromatischer Rest wie z. B. ein gegebenenfalls durch einen oder mehrere niedere Alkylreste substituierter Phenyl- oder Naphthylrest ist und R4 die oben angegebenen Bedeutungen hat.
Beispiele sind p-Toluolsulfonsäurealkylester, niedere Dialkylsulfate u. ähnl. Die Alkylierungsreaktion wird, gegebenenfalls unter Zusatz von üblichen säurebindenden Mitteln, wie Alkalicarbonaten, Pyridin oder andern üblichen tertiären Aminen, bei Temperaturen zwischen 0 und 1500c in inerten Lösungsmitteln wie Alkoholen, Dioxan, Dimethylformamid, Dimethylsulfoxyd, aromatischen Kohlenwasserstoffen wie Benzol, Toluol oder in Aceton vorgenommen.
Die Acylierung kann in inerten Lösungs- bzw. Suspensionsmitteln wie Dioxan, Dimethylformamid, Benzol, Toluol bei Temperaturen zwischen 0 bis 2000C erfolgen. Als Acylierungsmittel kommen Ketone sowie Säurehalogenide, Säureanhydride oder Säureester aliphatischer Carbonsäuren mit 2 bis 6 Kohlenstoffatomen bzw. von Kohlensäurehalbesterhalogeniden mit 1 bis 6 Kohlenstoffatomen, gegebenenfalls unter Zusatz eines säurebindenden Mittels wie Kaliumcarbonat oder Natriumäthylat oder eines tertiären Amins, z. B. Triäthylamin in Betracht. Bei den Estern handelt es sich insbesondere um solche mit niederen aliphatischen Alkoholen.
Bei der Alkylierung und Acylierung kann man auch so vorgehen, dass man erst von der umzusetzenden Verbindung (II) eine Alkaliverbindung herstellt, indem man sie in einem inerten Lösungsmittel wie Dioxan, Dimethylformamid, Benzol oder Toluol mit einem Alkalimetall, Alkalihydrid oder Alkaliamid (insbesondere Natrium oder Natriumverbindungen) bei Temperaturen zwischen 0 und 1500C umsetzt und dann das alky- lerende oder acylierende Agens zufügt.
An Stelle der angeführten Alkylierungs- und Acylierungsmittel können auch andere in der Chemie gebräuchliche chemisch äquivalente Mittel verwendet werden (s. z. B. auch : L. F. und Mary Fieser'Reagents for Organic Synthesis", John Wiley and Sons, Inc. New York, Vol. 1 [1967], S. 1303 bis 1304 und Vol. 2, S. 471).
Erhaltene basische Verbindungen der allgemeinen Formel (t) können nach bekannten Methoden in die Salze übergeführt werden. Als Anionen für diese Salze kommen die bekannten und therapeutisch verwendbaren Säurereste in Frage.
Erhalten die Verbindungen der Formel (I) saure Gruppen, so können sie auf die übliche Weise in ihre Alkali-, Ammonium-oder substituierten Ammoniumsalze überführt werden. Als substituierte Ammoniumsalze kommen insbesondere in Betracht : Salze von tertiären Alkylaminen, niederen Aminoalkoholen sowie bis- und tris-(Hydroxyalkyl)-aminen (Alkylreste jeweils mit 1 bis 6 Kohlenstoffatomen) wie Triäthylamin, Amin ä hanol und Di (hydroxyäthyl) amin.
Aus den Salzen der Verbindungen (t) können in üblicher Weise wieder die freien Basen hergestellt werden, beispielsweise durch Behandeln einer Lösung in einem organischen Mittel, wie Alkoholen (Methanol) mit Soda oder Natronlauge.
Die Verbindungen der Formel (I) enthalten mindestens ein asymmetrisches Kohlenstoffatom und fallen

in der Regel als Racemate an ; sie können in an sich bekannter Weise, beispielsweise mit Hilfe einer optisch aktiven Säure, in die optisch aktiven Isomeren gespalten werden. Es ist aber auch möglich, von vornherein eine optisch aktive Ausgangssubstanz einzusetzen, wobei dann als Endprodukt eine entsprechende optisch aktive bzw. diastereomere Form erhalten wird.
Die Verbindungen (1) können auch ganz oder teilweise in einer der möglichen tautomeren Formen vorliegen. Im allgemeinen liegt unter den normalen Arbeits undAufbewahrungsbindungen ein Gleichgewicht vor. Die erfindungsgemäss erhältlichen Verbindungen sind zur Herstellung pharmazeutischer Zusammensetzungen geeignet.
Beispielsweise kommen solche Verbindungen der allgemeinen Formel (1) in Betracht, worin die Symbole

Besonders günstige Wirkung besitzen solche Verbindungen der Formel (1), worin R1 Chlor, R2 und R3 gleich oder verschieden sind und Wasserstoff, Fluor oder Chlor vorzugsweise in o-Stellung bedeuten, Z ein Stickstoffatom ist, R4 eine Acylgruppe mit 2 bis 4 Kohlenstoffatomen ist und R5 Wasserstoff oder eine niedere Alkylgruppe mit 1 bis 4 Kohlenstoffatomen, insbesondere die Methylgruppe darstellt.
Die im erfindungsgemässen Verfahren verwendeten Ausgangsverbindungen können, soweit sie nicht bekannt sind, beispielsweise nach der deutschen Offenlegungsschrift 2259471 erhalten werden.
Beispiel l : 3- (3-Carboxpropionyloxy) -5- (0-chlorphenyl) -6-aza-7-chlor-l, 2- - dihydro-3H-1, 4-benzodiazepinon- (2)

1 gNatriumsalz des 3-Hydroxy-5- (o-chlorphenyl)-6-aza-7-chlor-l, 2-dihydro-3H-l, 4-benzodiazepinons- - (2) (hergestellt aus einer äthanolischen Lösung der 3-Hydroxy-Verbindung mit der äquivalenten Menge Na- triumäthylat-Lösung und Ausfällen des Natriumsalzes mit Äther), 0,7 g Bernsteinsäureanhydrid und 2 ml Dimethylsulfoxyd wurden 20 min auf dem Wasserbad erwärmt, dann abgekühlt und mit 3 ml Wasser versetzt.
Die gewünschte Verbindung kristallisiert beim Reiben aus. Ausbeute 0, 4 g ; Fp. 170 bis 171 oe.
Aus der Mutterlauge konnten noch weitere 0, 4 g Substanz erhalten werden, die aus Äthanol/Wasser umkristallisiert wurde.

21 g (0, 065 Mol) 3-Hydroxy-5- (o-chlorphenyl)-6-aza-7-chlor-l, 2-dihydro-3H-benzodiazepinon- (2) werden in 300 ml Dioxan gelöst und 3 ml Dimethylformamid zugesetzt. Dann werden 4, 1 g 80%iges Natriumhydrid in Weissöl zugefügt und 30 min bei Zimmertemperatur gerührt. Dann wird auf 65 C erwärmt, in 15 min 28, 4 g (0,2 Mol) Methyljodid zugetropft und 1 h bei dieser Temperatur weitergerührt.
Die Reaktionsmischung wird mit 5%iger Essigsäure bis zum Ausfallen des Reaktionsproduktes versetzt, dieses abgesaugt und aus Äthanol umkristallisiert. Ausbeute 10,2 g ; Fp. 247 bis 2490C.
