Abstract
Verbindungen mit einem acylierten Stickstoffatom können hergestellt werden, indem man das N-protonierte Addukt der Verbindungen mit einem Chlorid-, Bromidoder Jodid-Anion oder die freie Stickstoff-Base mit Acylhalogenid, vorzugsweise Acylchlorid in Anwesenheit einer Säure umsetzt, die das freigesetzte HCI, HCI bzw. HI austreibt.
Description
Herstellung von N-Acylverhincungsn Beschreibung
Die Erfindung bezieht sich auf ein Verfahren zur Herstellung von N-acylierten Verbindungen.
Die Acylierung von Aminen mit Carbonsäurechloriden ist eine bekannte Reaktion, siehe Methoden der organischen Chemie (Houben-Weyl), 4. Auflage (1958), Band XI/2, Seiten 10 und 1 1 sowie 30 bis 34. Bei der Reaktion wird Chlorwasserstoff abgespalten, der mit einem zweiten Aminmolekül unter Bildung des Aminhydrochlorids gebunden wird. Dadurch wird die Hälfte des Amins für die Bindung der freigesetzten Salzsäure verbraucht. Will man das Amin vollständig in das Amid überführen, so muß der freiwerdende Chlorwasserstoff auf andere Weise gebunden werden, beispielsweise durch Zugabe von feingepulvertem Alkalicarbonat. Alternativ kann man als säurebindendes Mittel Pyridin zusetzen. Auf Seite 34 des zitierten Lehrbuches findet sich die Bemerkung, daß Acetylchlorid zwar ein energisch wirkendes Acetylierungsmittel für Amine sei, daß es jedoch weniger oft als Essigsäureanhydrid angewendet werden, da dieses bequemer zu handhaben sei und fast immer zum Ziel führe. Aufgabe der vorliegenden Erfindung ist es, ein vereinfachtes Verfahren zur Herstellung von Verbindungen unter Acylierung von Stickstoffatomen anzugeben. Diese Aufgabe wird durch das erfindungsgemäße Verfahren gelöst.
Das erfindungsgemäße Verfahren umfaßt die Herstellung von Verbindungen mit einem acylsubstituierten Stickstoff atom, Ausgangsverbindungen sind die entsprechenden Stickstoffverbindungen in Form der N-protonierten Addukte mit einem Chlorid-, Bromid- oder Jodid-Anion und Acylhalogenid, wobei es sich bei dem Acylhalogenid vorzugsweise um Acylchlorid handelt; dabei wird die Acylierung in Anwesenheit derjenigen Carbonsäure durchgeführt, die der verwendeten Acylgruppe entspricht. Das Stickstoffatom muß neben dem Proton mindestens ein weiteres Wasserstoffatom aufweisen; alternativ geht man von der freien Stickstoffbase aus, dabei muß aber mindestens 1 Wasserstoffatom an das Stickstoffatom gebunden sein. Der Begriff "Acyl" steht für die Gruppe RC(O), worin R steht für: lineares oder verzweigtes Alkyl mit 1 bis
6 Kohlenstoffatomen; für Phenyl; für lineares oder verzweigtes Alkyl mit 1 bis 6 Kohlenstoffatomen steht, welches durch ein oder mehr Halogenatome substituiert ist; oder für Phenyl, das durch ein oder mehr Halogenatome substituiert ist. Bevorzugt steht Acyl für Halogenacyl, insbesondere für Trifluoracetyl, Difluoracetyl, Chlordifluoracetyl, C2F5C(0), C2HF C(0) oder C2CIF4C(0). Ganz besonders bevorzugt führt man eine Trifluoracylierung durch, und die entsprechende Säure ist die Trifluoressigsäure.
Im Prinzip kann man beliebige Stickstoffverbindungen wie Amine oder deren Hydrohalogenidaddukte oder Carbonsäureamide acylieren. Bevorzugte Stickstoffverbindungen, die mit dem erfindungsgemäßen Verfahren acyliert werden, sind Aminosäuren oder deren Derivate, beispielsweise die Ester oder Peptide. Ganz besonders bevorzugt verwendet man das erfindungsgemäße Verfahren zur Herstellung von N-acy- lierten Aminosäureestern oder Peptiden, insbesondere mit der Trifluoracetylgruppe. Auch hier können die Hydrohalogenidaddukte, vorzugsweise die Hydrochloride, oder die Verbindungen mit freiem N-Atom eingesetzt werden.
Mittels des erfindungsgemäßen Verfahrens können Acylamide hergestellt werden, die beispielsweise als solche oder in der chemischen Synthese brauchbar sind. Trifluoracetamid, welches ein Handelsprodukt ist, kann beispielsweise in der chemischen Synthese verwendet werden.
Die Einführung einer Acyl- oder Halogenacylgruppe ist auch unter dem Gesichtspunkt der Eigenschaft als Schutzgruppe für das Stickstoffatom von Interesse, insbesondere in der chemischen Synthese. Insbesondere die Trifluoracetylgruppe ist eine bekannte Schutzgruppe, siehe z. B. US-Patent 5,541 ,206, Spalte 6.
Ganz besonders bevorzugt wendet man das erfindungsgemäße Verfahren zur Acylierung, insbesondere zur Trifluoracylierung, von Aminosäuren und ihren Derivaten, besonders den Estern, sowie von Peptiden an.
Sehr gut geeignet ist die Anwendung zwecks Herstellung von N-Trifluoracetyl- L-phenylalaninalkylester, N-Trifluoracetyl-D-phenylalaninalkylester und N-Acyl-Lysin- alkylestem (ω-Aminogruppe), insbesondere den Methyl- und Ethyiestem. Acylieren, insbesondere mit der Trifluoracylgruppe, lassen sich auch Ester, besonders Alkylester, insbesondere Methyl- und Ethylester des Glycins, Alanins, Valins, Leucins, Isoleucins, Phenylalanins, Tyrosins, Prolins, Hydroprolins, Serins, Threonins, Cysteins, Cystins,
Methionins, Tryptophans, der Asparaginsäure, der Glutaminsäure, des Arginins und des Histidins.
Die Phenylalanin-Verbindungen sind brauchbare Zwischenprodukte beispielsweise bei der Herstellung der in dem US- Patent 5,541 ,206 oder 4,816,484 genannten Verbindungen.
Die Isolierung der erfindungsgemäß erhaltenen acylierten Verbindungen kann nach üblichen Methoden bewirkt werden. Eine sehr gut geeignete Methode insbesondere zur Verfestigung der Verbindungen wird in der nicht vorveröffentlichten internationalen Patentanmeldung mit dem Anmeldeaktenzeichen PCT/EP 03/02453 beschrieben. Ein Reaktionsgemisch, das Verunreinigungen enthält sowie eine organische Verbindung, die einen Schmelzpunkt >30 °C besitzt, wird erhitzt, um die Verunreinigungen abzudampfen. Die verbleibende organische Verbindung wird dann auf ein gekühltes Laufband aufgegeben. Eine derartige Methode kann auch für die bei dem vorliegenden Verfahren erhaltenen Verbindungen angewendet werden.
Die Erfindung weist den Vorteil auf, daß kein Hydrochlorid anfällt und beseitigt werden muß.
Die Erfindung wird anhand der folgenden Beispiele weiter erläutert.

Beispiel 1 :
Herstellung von L-Phenylalaninmethylester-Hydrochlorid aus Phenylalanin
C6H5CH2CH(NH2)Cθ2H + MeOH*HCI → C6H5CH2CH(NH2)C02CH3*HCI L-Phenylalanin L-Phenylalaninmethylester*HCI
Ansatz:
61,6 mol (1973,66g) MeOH (32,04g/mol) 9,8 mol (352,8g) HCI (36,0g/mol) 3.05 mol (503,83g) L-Phenylalanin (165, 19g/mol) 1,0 mol (103,0g) Diisopropylether (102,18g/mol)
Durchführung:
In einem 4l-Mehrhalskolben wurde das Phenylalanin im Solvens vorgelegt und mit einer zuvor hergestellten MeOH*HCI- Lösung versetzt und zur Lösung gebracht. Der Ansatz wurde zur besseren Lösung auf 50 °C erwärmt und ca. 5 min. am Rückfluß gekocht (ca. 55 °C). Anschließend wurde Vakuum angelegt, um das überschüssige MeOH und die HCI abzudestillieren. Es wurde so lange destilliert, bis sich der Ansatz eintrübte, viskoser wurde und weiße Flocken bildete. Nach Stehenlassen wurde der ausgefallene Phenylalanin-methylester x HCI abgenutscht. Die Reinheit betrug 99,4 % (GC) mit Smp. 160 °C.
Die Wiederholung des Versuchs mit D- oder D/L-Phenylalanin als Edukt ergab vergleichbare Versuchsergebnisse.
Beispiel 2:
Trifluoracelierung von L-Phenylalaninmethylester-Hydrochlorid
C6H5CH2CH(NH2)C02CH3*HCI in TFA + CF3COCI → C8H5CH2CH(NH-COCF3)C02CH3
L-Phenylalaninmethylester*HCI N-Trifluoracetyl-L-phenylalanin- methylester
Ansatz:
2,0 mol (435,2 g) L-Phenylalaninmethylester-Hydrochlorid (215,68 g/mol) aus Beispiel 1
7,8 mol (889,36 g) Trifluoressigsäure (114,02 g/mol)
2.6 mol (350,5 g) Trifluoressigsäureacetylchlorid (132,47g/mol)
Durchführung:
In einem 4l-Mehrhalskolben wurde der L-Phenylalaninmethylester (lag als HCI-Addukt vor) vorgelegt und in TFA gelöst. Während des Lösungsvorgangs wurde der Ansatz schon langsam auf 90 °C temperiert. Dabei wurde schon HCI frei; zu erkennen an der heftigen Gasentwicklung am Blasenzähler. Nach vollständiger Lösung des Esters und
erreichter Temperatur wurde dann TFAC eingeleitet, wobei immer noch eine Menge HCI freigesetzt wurde.
Es wurde so lange eingeleitet, bis sich aller Ester zu N-Trifluoracetyl-L-phenylalanin- methylester umgesetzt hatte. Das Lösungsmittel TFA wurde im Vakuum abgezogen, zurück blieb nach Abkühlen N-Trifluoracetyl-L-phenylalaninmethylester mit Schmp. 52 °C und einer Ausbeute von 95 % d. Th. In einer Versuchsvariante wurde die aufgeschmolzene Masse auf ein gekühltes Band geleitet. Dies führte zu leicht schüttbaren Pellets als gut handhabbares Produkt.
Beispiel 3:
Trifluoracetylierung von D-Phenylalaninmethylester*HCL und D/L-Phenylalaninmethyl- ester*HCI aus Beispiel 1
Beispiel 2 wurde unter Verwendung von D-Phenylaianinmethylester*HCl und D/L- Phenylalaninmethylester*HCI aus Beispiel 1 wiederholt. Die, Versuchsergebnisse entsprachen denjenigen von Beispiel 2.
Claims
1. Verfahren zur Herstellung von Verbindungen mit acetyliertem Stickstoffatom, wobei Acyl für RC(O) steht und R die Bedeutung hat: lineares oder verzweigtes Alkyl mit 1 bis 6 Kohlenstoffatomen; Phenyl; durch ein oder mehr Halogenatome substituiertes lineares oder verzweigtes Alkyl mit 1 bis 6 Kohlenstoffatomen, oder durch 1 oder mehr Halogenatome substituiertes Phenyl, aus Acylhalogeniden und den entsprechenden Stickstoffverbindungen, die N-protoniert sind und am zu acylierenden Stickstoffatom mindestens ein weiteres Wasserstoffatom aufweisen und als Chlorid-, Bromid- oder Jodid-Anion vorliegen oder in Form der freien Base, in Anwesenheit einer Säure, deren Säurestärke ausreicht, gebildeten Halogenwasserstoff aus dem Reaktionsgemisch auszutreiben.
2. Verfahren nach Anspruch 1 , dadurch gekennzeichnet, daß man Verbindungen mit einer Halogenacyl-Stickstoffgruppe herstellt, wobei Halogenacyltrifluoracetyl, Diflu- oracetyl, Chlordifluoracetyl, C2F5C(0), C2HF4C(0) oder C2CIF4C(0) bedeutet.
3. Verfahren nach Anspruch 2, dadurch gekennzeichnet, daß man in Anwesenheit derjenigen Halogencarbonsäure umsetzt, die der Halogenacylgruppe entspricht.
4. Verfahren nach Anspruch 1 , dadurch gekennzeichnet, daß man Aminosäuren, deren Derivate, vorzugsweise Aminosäureester, oder Peptide acyliert.
5. Verfahren nach Anspruch 1 , dadurch gekennzeichnet, daß man Trifluorace- tylverbindungen herstellt in Anwesenheit von Trifluoressigsäure.
6. Verfahren nach Anspruch 1 , dadurch gekennzeichnet, daß man N-Trifluor- acetylverbindungen von L-Phenylalaninalkylestern, D-Phenylalaninalkylestem oder Lysinalkylestern herstellt.
7. Verfahren nach Anspruch 6, dadurch gekennzeichnet, daß man die Methyloder Ethylester herstellt.
