Die vorliegende Erfindung betrifft orale Darreichungsformen mit kontrollierter Gesamt-Freisetzung eines Wirkstoffes, wobei derselbe Wirkstoff in Form von wenigstens zwei unterschiedlichen Salzen vorliegt, die im festen Aggregatzustand in der Darreichungsform vorhanden sind und eine unterschiedliche in-vitro Freisetzung dieses Wirkstoffes aufweisen.

Description

Orale Darreichunqsformen
Die vorliegende Erfindung betrifft orale Darreichungsformen mit kontrollierter Gesamt-Freisetzung eines Wirkstoffes, wobei derselbe Wirkstoff in Form von wenigstens zwei unterschiedlichen Salzen vorliegt, die im festen Aggregatzustand in der Darreichungsform vorhanden sind und eine unterschiedliche in-vitro Freisetzung dieses Wirkstoffes aufweisen.
Die Verabreichung eines Wirkstoffes in Form von Zubereitungen, aus denen dieser Wirkstoff kontrolliert freigesetzt wird, ist für viele Therapien vorteilhaft. So wird durch eine kontrollierte Freisetzung eines Wirkstoffes mit einer relativ kurzen Halbwertszeit dessen Verfügbarkeit im Organismus verlängert. Außerdem kann dadurch ein gleichmäßiger Blutspiegel eingestellt und damit ggf. unerwünschte Begleiterscheinungen vermindert sowie die Einhaltung von Dosierungsvorschriften verbessert werden.
Die kontrollierte Freisetzung eines Wirkstoffes aus oralen Darreichungsformen kann üblicherweise nur durch relativ aufwendige Formulierungsverfahren, wie beispielsweise durch das Überziehen von Wirkstoff-haltigen oralen Darreichungsformen mit retardierenden Filmüberzügen oder durch Einbettung in eine Retardmatrix erreicht werden. Sofern eine unterschiedliche Freisetzung von Teilmengen eines Wirkstoffes zur Steuerung seines Gesamt- Freisetzungsprofils gewünscht wird, wird ein- und derselbe Wirkstoff oder ein- und dasselbe Wirkstoffsalz zu unterschiedlichen Formulierungen getrennt verarbeitet, die dann z.B. als retardierte und unretardierte Form zu einer Darreichungsform vereinigt werden.
Aufgabe der vorliegenden Erfindung war es daher, orale Darreichungsformen eines Wirkstoffes zur Verfügung zu stellen, aus denen dieser Wirkstoff kontrolliert freigesetzt wird, ohne daß aufwendige, getrennte
Formulierungsschritte zur Einstellung des Gesamt-Freisetzungsprofils des Wirkstoffes aus einer Darreichungsform erforderlich sind. Erfindungsgemäß wird diese Aufgabe durch die Bereitstellung oraler Darreichungsformen mit kontrollierter Gesamt-Freisetzung eines Wirkstoffes gelöst, in denen derselbe Wirkstoff in Form von wenigstens zwei unterschiedlichen Salzen vorliegt, die im festen Aggregatzustand in der Darreichungsform vorhanden sind und eine unterschiedliche in-vitro Freisetzung dieses Wirkstoffes aufweisen.
In einer bevorzugten Ausführungsform der vorliegenden Erfindung weisen alle verschiedenen Salze des Wirkstoffes einer Darreichungsform eine jeweils voneinander unterschiedliche Wasserlöslichkeit auf, die in aller Regel zu einer unterschiedlichen Lösungsgeschwindigkeit des Wirkstoffes führt.
Vorzugsweise unterscheidet sich die Wasserlöslichkeit aller in einer erfindungsgemäßen Darreichungsform zum Einsatz kommenden unterschiedlichen Wirkstoffsalze jeweils um wenigstens den Faktor 2 voneinander.
Das Gesamt-Freisetzungsprofil für den jeweiligen Wirkstoff ist bei den erfindungsgemäßen oralen Darreichungsformen durch die Wahl der Wirkstoffsalze sowie deren Mengenverhältnisse in der gemeinsamen Darreichungsform in gewünschter Form einstellbar. Dies ermöglicht eine Therapie-gerechte Einsteilung des Plasmaspiegels für einen Wirkstoff, z.B. die Einstellung eines möglichst konstanten Plasmaspiegeis über einen längeren Zeitraum, eine gepulste Freisetzung mit zeitverschobenen Plasmaspiegelspitzen des Wirkstoffes oder die Einstellung eines
Plasmaspiegels des Wirkstoffes in Anlehnung an den zirkadianen Rhythmus im Organismus.
Ein Wirkstoff im Sinne der vorliegenden Erfindung ist jeder Stoff, der im menschlichen oder tierischen Organismus einen Einfluß auf biologische, biochemische, chemische, physikalische oder physiologische Prozesse oder Strukturen ausübt und ein bei 25 °C festes Salz durch Umsetzung mit einer Säure oder Base bilden kann. Diese Wirkstoffsalzbildung kann auch durch Umsetzung mit einem weiteren Wirkstoff mit der entsprechenden Säure- oder Basenfunktion erfolgen.
Vorzugsweise wird der salzbildende Wirkstoff aus der Gruppe der salzbildenden pharmazeutischen Wirkstoffe, Vitamine, Nährstoffe,
Mineralstoffe oder Diagnostika, besonders bevorzugt aus der Gruppe der salzbildenden pharmazeutischen Wirkstoffe ausgewählt.
Sofern es sich bei dem Wirkstoff um einen salzbildenden pharmazeutischen Wirkstoff handelt, kann dieser bevorzugt aus der Gruppe der salzbildenden Analgetika, Anthelminthika, Antiarrhythmika, Antiasthmatika, Antidepressiva, Antidiabetika, Antidota, Antiallergika, Antitussiva, Antibiotika, Antiemetika, Antiinfektiva, Antihistaminika, Antihypertonika, Antihypertensiva, Antikoagulantia, Antirheumatika, Antipyretika, Anxiolytika, Abmagerungsmittel, Acidosetherapeutika, Antivertiginosa, Antihämorrhagika, Antifibrinolytika, Hämostatika, Antihypoglykämika, Antihypotonika, Antimykotika, Antiphlogistika, Expektorantia, Antiepileptika, Arteriosklerosemittel, Betrarezeptorenblocker, Calciumkanalblocker, Hemmstoffe des Renin- Angiotensin-Systems, Broncholytika, Cholagoga, Gallenwegstherapeutika, Cholinergika, Corticoide (Interna), Durchblutungsfördernde Mittel,

Besonders bevorzugt wird der salzbildende pharmazeutische Wirkstoff aus der Gruppe der saizbildenden Analgetika, Antiinfektiva oder Neuroleptika ausgewählt.
Als salzbildende Analgetika können bevorzugt salzbildende Opioide, Verbindungen mit opoider Wirksamkeit, oder nichtsteroidale Analgetika zum Einsatz kommen.
Bevorzugt können als salzbildende Opioide oder Verbindungen mit opioider Wirksamkeit Brifentanil, Carfentanil, Fentatienil, Lofentanil, Ocfentanil, Trefentanil, Codein, Dextropropoxyphen, Dihydrocodein, Diphenoxylat, Meptazinol, Nalbuphin, Pethidin (Meperidin), Tilidin, Tramadol, Viminol, Butorphanol, Dextromoramid, Dezocin, Diacetylmorphin (Heroin), Hydrocodon, Hydromorphon, Ketobemidon, Levomethadon, Levomethadyl, Levorphanol, Morphin, Nalorphin, Oxycodon, Pentazocin, Piritramid, Alfentanil, Buprenorphin, Etorphin, Fentanyl, Remifentanil oder Sufentanil vorliegen. Besonders bevorzugt können Tramadol oder Morphin zum Einsatz kommen.
Als salzbildendes Neuroleptikum kann bevorzugt Promethazin zum Einsatz kommen.
Als Wirkstoffsalze können in den erfindungsgemäßen, oralen, pharmazeutischen Darreichungsformen die physiologisch verträglichen Wirkstoffsalze vorliegen. Als Salzpartner des zum Einsatz kommenden Wirkstoffes kann auch ein weiterer Wirkstoff verwendet werden.
Vorzugsweise sind diese Salze ausgewählt aus der Gruppe Chlorid, Bromid, Sulfat, Sulfonat, Phosphat, Tartrat, Teoclat, Embonat, Formiat, Acetat, Propionat, Benzoat, Oxalat, Succinat, Citrat, Diclofenacat, Naproxenat, Salicylat, Glutamat, Fumarat, Acetylsalicylat, Aspartat, Glutarat, Stearat, Butyrat, Malonat, Lactat, Mesylat, Saccharinat, Cyclamat oder Acesulfamat, besonders bevorzugt aus der Gruppe Chlorid, Sulfat, Saccharinat, Teociat, Embonat, Diclofenacat, Naproxenat oder Salicylat.
Vorzugsweise können in den erfindungsgemäßen, oralen Darreichungsformen als Salze ein- und desselben Wirkstoffes Tramadolhydrochlorid,
Tramadolsaccharinat und Tramadoldiciofenacat oder Morphinhydrochlorid, Morphinsaccharinat und Morphinsulfat nebeneinander vorliegen. Besonders bevorzugt können in den erfindungsgemäßen, oralen Darreichungsformen als Salze ein- und desselben Wirkstoffes Tramadolhydrochlorid und Tramadolsaccharinat oder Tramadolhydrochlorid und Tramadoldiciofenacat nebeneinander vorliegen.
Ebenfalls bevorzugt kann als Wirkstoffsalz ein Alkalimetallsalz, Erdalkaiimetallsalz, Ammoniumsalz, Eisensalz oder Aluminiumsalz des Wirkstoffes, besonders bevorzugt ein Alkalimetallsaiz, ganz besonders bevorzugt das Natrium- oder Kaliumsalz des Wirkstoffes vorliegen.
Die kontrollierte Gesamt-Freisetzung des Wirkstoffes aus den erfϊndungsgemäßen, oralen Darreichungsformen kann zusätzlich auch dadurch modifiziert werden, daß wenigstens eines der Wirkstoffsalze, vorzugsweise mehrere bis alle Wirkstoffsalze in retardierter Form in den Darreichungsformen vorliegen. Ebenfalls bevorzugt kann auch die orale Darreichungsform bei der gemeinsamen Formulierung aller Wirkstoffsalze zu der erfindungsgemäßen Darreichungsform retardiert werden.
In einer bevorzugten Ausführungsform der vorliegenden Erfindung erfolgt die Retardierung durch einen retardierenden Überzug und/oder durch Einbettung in eine retardierende Matrix.
Der retardierende Überzug basiert vorzugsweise auf einem wasserunlöslichen ggf. modifizierten natürlichen oder synthetischen Polymeren ggf. in Kombination mit einem üblichen Weichmacher oder auf einem natürlichen, halbsynthetischen oder synthetischen Wachs oder Fett oder Fettalkohol oder einem Gemisch aus wenigstens zwei der vorstehend genannten Komponenten.
Als wasserunlösliche Polymere werden zur Herstellung eines retardierenden Überzuges vorzugsweise Poly(meth)acrylate, besonders bevorzugt Poly(Cι-4)- Alkyl(meth)acrylate, Poly(Cι- )-dialkylamino-(Cι- )-alkyl(meth)acrylate und/oder deren Copolymere, bevorzugt Ethylacrylat/Methylmethacrylat-Copolymere mit einem molaren Verhältnis der Monomeren von 2 : 1 (Eudragit NE30D®), Ethylacrylat/Methylmethacrylat/Trimethylammoniumethylmethacrylat-chlorid- Copolymere mit einem molaren Verhältnis der Monomeren von 1 : 2 : 0,1 (Eudragit RS®),
Ethylacrylat/Methylmethacrylat/Trimethylammoniumethyl-methacrylatchlorid- Copolymere mit einem molaren Verhältnis der Monomeren von 1 : 2 : 0,2 (Eudragit RL®) oder ein Gemisch aus wenigstens zwei dieser vorstehend genannten Polymeren als Überzugsmaterial eingesetzt.
Diese Überzugsmaterialien sind als 30 Gew.%-ige wäßrige Latexdispersionen, d.h. als Eudragit RS30D®, Eudragit NE30D® bzw. Eudragit RL30D® am Markt erhältlich und werden als solche auch als Überzugsmaterial bevorzugt eingesetzt.
Ebenfalls bevorzugt können als wasserunlösliche Polymere zur Herstellung des retardierenden Überzugs der erfindungsgemäßen Darreichungsformen insgesamt oder der einzelnen Wirkstoffsalze Polyvinylacetate ggf. in Kombination mit weiteren Hilfstoffen eingesetzt werden. Diese sind als wäßrige Dispersion enthaltend 27 Gew.-% Polyvinylacetat, 2,5 Gew.-% Povidon und 0,3 Gew.-% Natriumlaurylsulfat (Kollicoat SR 30 D ®) am Markt erhältlich.
Die retardierenden Überzüge können auch auf wasserunlöslichen Cellulosederivaten, vorzugsweise Alkylcellulosen wie z.B. Ethylcellulose, oder Celluloseestern, wie z.B. Celluloseacetat basieren. Die Überzüge aus Ethylcellulose werden bevorzugt aus wäßriger Pseudolatexdispersion aufgebracht. Wäßrige Ethylcellulose-Pseudolatexdispersionen werden als 30 Gew.%-ige Dispersionen (Aquacoat®) oder als 25 Gew.%-ige Dispersionen (Surelease®) am Markt geführt.
Als natürliche, halbsynthetische oder synthetische Wachse, Fette bzw. Fettalkohole kann der retardierende Überzug vorzugsweise auf
Camaubawachs, Bienenwachs, Glycerinmonostearat, Glycerinmonobehenat (Compritol ATO888®), Glycerinditripalmitostearat (Precirol AT05®), mikrokristallines Wachs, Cetylalkohol, Cetylstearylalkohol oder einem Gemisch aus wenigstens zwei dieser Komponenten basieren.
Sofern der retardierende Überzug auf einem wasserunlöslichen, gegebenenfalls modifizierten natürlichen und/oder synthetischen Polymeren basiert, kann die Überzugsdispersion oder Lösung neben dem entsprechenden Polymeren einen üblichen, dem Fachmann bekannten, physiologisch verträglichen Weichmacher aufweisen, um die notwendige
Mindestfilmtemperatur zu senken oder die Filmeigenschaften zu modifizieren.
Geeignete Weichmacher sind beispielsweise lipophile Diester aus einer aliphatischen oder aromatischen Dicarbonsäure mit C-6-C 0 und einem aliphatischen Alkohol mit
Cι-C8) wie z.B. Dibutylphthalat, Diethylphthalat, Dibutylsebacat oder Diethylsebacat, hydrophile oder lipophile Ester der Zitronensäure, wie z.B. Triethylcitrat, Tributylcitrat, Acetyltributylcitrat oder Acetyltriethylcitrat, Polyethylenglycole, Propylenglycol, Ester des Glycerins, wie z.B. Triacetin, Myvacet® (acetylierte Mono- und Diglyceride, C23H44θ5 bis C25H470 ), mittelkettige Triglyceride (Miglyol®), Ölsäure oder Gemische aus wenigstens zwei der genannten Weichmacher. Vorzugsweise wird für wäßrige Dispersionen von Eudragit RS® und gegebenenfalls Eudragit RL® Triethylcitrat als Weichmacher verwendet.
Vorzugsweise enthält der retardierende Überzug den/die Weichmacher in Mengen von 5 bis 50 Gew.-%, besonders bevorzugt 10 bis 40 Gew.-% und ganz besonders bevorzugt 10 bis 30 Gew.-%, bezogen auf die Menge an des/der eingesetzten Polymer(en). In Einzelfällen, beispielsweise für Celluloseacetat können auch höhere Mengen an Weichmachern, vorzugsweise bis zu 110 Gew.-% eingesetzt werden.
Desweiteren kann der retardierende Überzug weitere übliche, dem Fachmann bekannte Hilfsstoffe, wie z.B. Gleitmittel, vorzugsweise Talkum oder Glycerinmonostearat, Farbpigmente, vorzugsweise Eisenoxide oder Titandioxid, Tenside, wie z.B. Tween 80® oder Hilfsstoffe zur Modulation der Filmeigenschaften, wie z.B. wasserlösliche Porenbildner, beispielsweise Lactose, Polyethylenglykol 1000 (PEG 1000) oder Saccharose aufweisen.
Die erfindungsgemäßen oralen Darreichungsformen können auch wenigstens ein Wirkstoffsalz, vorzugsweise mehrere bis alle Wirkstoffsalze in einer retardierenden Matrix, bevorzugt gleichmäßig verteilt, enthalten. Als Matrixmaterialien können bevorzugt physiologisch verträgliche, hydrophile Materialien verwendet werden, die dem Fachmann bekannt sind. Besonders bevorzugt basiert die retardierende Matrix auf Celluloseethern, Celluloseestern und/oder Acrylharzen, ganz besonders bevorzugt auf Ethylcellulose, Hydroxypropylmethylcellulose, Hydroxypropylcellulose, Hydroxymethylcellulose, Poly(meth)acrylsäure und/oder deren Salzen, Amiden und/oder Estern.
Ebenfalls bevorzugt können als Matrixmaterialien physiologisch verträgliche hydrophobe Materialien verwendet werden, die dem Fachmann bekannt sind. Besonders bevorzugt basiert die Matrix auf hydrophoben Polymeren,
Wachsen, Fetten, langkettigen Fettsäuren, Fettalkoholen oder entsprechenden Estern oder Ethern oder deren Gemischen, besonders bevorzugt auf Mono- oder Diglyceriden von Cι2-C30-Fettsäuren und/oder Cι2-C3u-Fettalkoholen und/oder Wachsen oder deren Gemischen.
Es ist auch möglich, Mischungen der vorstehend genannten hydrophilen und hydrophoben Materialien als retardierendes Matrixmaterial einzusetzen. Sofern die erfindungsgemäßen oralen Darreichungsformen Wirkstoffsalze enthalten, deren Säurekomponente eine schwächere Säure ist als die im Magen des menschlichen oder tierischen Organismus vorkommende Salzsäure, sollten diese einen Schutzüberzug aufweisen, der vorzugsweise magensaftresistent ist. Durch diesen Schutzüberzug kann erreicht werden, daß der Wirkstoff aus den in den erfindungsgemäßen, oralen Darreichungsformen vorliegenden Wirkstoffsalzen höchstens verzögert oder überhaupt nicht im Magen freigesetzt wird. Die magensafttresistenten Überzüge bewirken, daß die erfindungsgemäßen oralen Darreichungsformen den Magentrakt unaufgelöst passieren und der Wirkstoff erst im Darmtrakt zur Freisetzung gelangt. Vorzugsweise löst sich der magensaftresistente Überzug bei einem pH-Wert von 5 bis 7,5 auf. Das gewünschte Gesamt- Freisetzungsprofil kann vom Fachmann durch einfache in vitro Vorversuche mit Hilfe der bekannten Meßmethode zur Bestimmung der Wirkstofffreisetzung überprüft und dementsprechend eingestellt werden.
Ein magensaftresistenter Überzug besteht bevorzugt aus
Methacrylsäure/Methylmethacrylat-Copolymeren mit einem molaren Verhältnis der Monomeren von 1 : 1 (Eudragit L®), Methacrylsäure/Methylmethacrylat- Copolymeren mit einem molaren Verhältnis der Monomeren von 1 : 2 (Eudragit
S®), Methacrylsäure/Ethylacrylat-Copolymeren mit einem molaren Verhältnis der Monomeren von 1 : 1 (Eudragit L30-D55®)
Methacrylsäure/Methylacrylat/Methylmethacrylat mit einem molaren Verhältnis der Monomeren von 7 : 3 : 1 (Eudragit FS®), Schellack, Hydroxypropylmethylcelluloseacetatsuccinat, Celluloseacetatphthalat oder aus einer Mischung aus wenigstens zwei dieser Komponenten, ggf. auch in
Kombination mit Poly(meth)acrylaten, vorzugsweise Eudragit NE30D® und/oder Eudragit RL® und/oder Eudragit RS®.
Die Überzugsdispersion oder Lösung aus der der magensaftresistente
Überzug aufgebracht wird, kann neben dem entsprechenden Polymeren einen der vorstehend genannten Weichmacher aufweisen. Desweiteren können auch die oben aufgeführten Retardüberzugsmaterialien als Schutzüberzug gegen die Magensäure in unterschiedlicher, dem Fachmann bekannter Dicke zum Einsatz kommen.
Die Retard- und/oder Schutzüberzüge der erfindungsgemäßen oralen Darreichungsformen können nach den üblichen, für den jeweiligen Überzug geeigneten, dem Fachmann bekannten Verfahren, wie z.B. durch Aufsprühen von Lösungen, Dispersionen oder Suspensionen, durch Schmelzverfahren oder durch Pulverauftragsverfahren aufgebracht werden. Die Lösungen, Dispersionen oder Suspensionen können in Form von wäßrigen oder organischen Lösungen oder Dispersionen eingesetzt werden. Dabei werden wäßrige Dispersionen bevorzugt eingesetzt. Als organische Lösungsmittel können Alkohole, beispielsweise Ethanol oder Isopropanol, Ketone, wie z.B. Aceton, Ester, beispielsweise Ethylacetat, chlorierte Kohlenwasserstoffe, wie z.B. Dichlormethan verwendet werden, wobei Alkohole und Ketone bevorzugt eingesetzt werden. Es ist auch möglich Mischungen aus wenigstens zwei der vorstehend genannten Lösungsmittel einzusetzen.
Diese Verfahren sind aus dem Stande der Technik, z.B. H. Sucker, Georg Thieme Verlag, 1991 , Seiten 347 ff. bekannt. Sie werden hiermit als Referenz eingeführt und gelten somit als Teil der Offenbarung.
In einer bevorzugten Ausführungsform der vorliegenden Erfindung liegen die erfindungsgemäßen oralen Darreichungsformen in Form von Tabletten, Kautabletten, Kaugummis, Dragees oder Pulver, ggf. in Kapseln abgefüllt, besonders bevorzugt in Form von Tabletten vor.
In einer weiteren bevorzugten Ausführungsform der vorliegenden Erfindung liegen die erfindungsgemäßen oralen Darreichungsformen in multipartikulärer Form, vorzugsweise in Form von Mikrotabletten, Mikrokapseln, Granulaten, Wirkstoffkristallen oder Pellets, besonders bevorzugt in Form von
Mikrotabletten, Granulaten oder Pellets, ggf. in Kapseln abgefüllt oder zu Tabletten verpreßt, vor. Sofern die erfindungsgemäßen oralen Darreichungsformen in Form von Granulaten oder Pellets vorliegen, können diese bevorzugt eine Größe im Bereich von 0,1 bis 3 mm, besonders bevorzugt im Bereich von 0,5 bis 2 mm aufweisen.
Liegen die erfindungsgemäßen oralen Darreichungsformen in Form von Mikrotabletten vor, können diese bevorzugt einen Durchmesser im Bereich von 0,5 bis 5 mm, besonders bevorzugt im Bereich von 1 bis 3 mm und ganz besonders bevorzugt im Bereich von 1 bis 2 mm aufweisen.
Liegen die erfindungsgemäßen oralen Darreichungsformen in Form von Wirkstoffkristallen, Mikropartikeln, Mikropellets oder Mikrokapseln vor, so können diese bevorzugt einen Durchmesser im Bereich von 10 μm bis 1 mm, besonders bevorzugt im Bereich von 15 μm bis 0,5 mm und ganz besonders bevorzugt im Bereich von 30 μm bis 200 μm aufweisen.
Die erfindungsgemäßen oralen Darreichungsformen können außerdem je nach Ausführungsform als weitere Bestandteile die üblichen, dem Fachmann bekannten Hilfsstoffe enthalten.
Sofern die erfindungsgemäßen oralen Darreichungsformen in Form von Tabletten oder Mikrotabletten vorliegen, können diese als weitere Hilfsstoffe vorzugsweise mikrokristalline Cellulose, Celluloseether, Lactose, Stärke und Stärkederivate, Zuckeralkohole, Calciumhydrogenphosphat sowie die üblichen, dem Fachmann bekannten Bindemittel, Fließregulationsmittel, Gleitmittel und gegebenenfalls Sprengmittel enthalten.
Liegen die erfindungsgemäßen oralen Darreichungsformen in Form von Pellets, Granulaten oder Mikropellets vor, können diese als weitere Hilfsstoffe bevorzugt mikrokristalline Cellulose, Celluloseether, Lactose, Stärke und Stärkederivate, Zuckeralkohole, Calciumhydrogenphosphat, Fettalkohole, Ester des Glycerins oder Fettsäureester enthalten. Liegen die erfindungsgemäßen oralen Darreichungsformen in Form von Mikrokapseln oder Mikropartikeln vor, so können diese je nach Art des zu ihrer Herstellung eingesetzten Verfahrens die üblichen, dem Fachmann bekannten Hilfsstoffe enthalten.
Zur Herstellung der erfindungsgemäßen, oralen Darreichungsformen werden die Wirkstoffsalze und ggf. weitere Hilfsstoffe vorzugsweise in einem hochtourigen Mischer oder in der Rotorwirbelschicht vorzugsweise homogen gemischt. Anschließend erfolgt die Formulierung nach verschiedenen, dem Fachmann bekannten Methoden und ggf. das Aufbringen der vorzugsweise magensaftresistenten Schutzüberzüge aus den obenstehend genannten Überzugsmaterialien nach den obenstehend angegebenen Methoden.
Sofern die erfindungsgemäßen oralen Darreichungsformen in Form von Tabletten vorliegen, werden die unterschiedlichen festen Wirkstoffsalze vorzugsweise homogen vermischt, mittels Feucht-, Trocken-, oder Schmelzgranulation zu Granulaten verabeitet und zu Tabletten verpreßt oder durch direkte Tablettierung der Wirkstoffsalze, ggf. mit weitereren Hilfsstoffen hergestellt. Desweiteren können die Tabletten bevorzugt durch Verpressen ggf. überzogener Pellets, Wirkstoffkristalle, Mikropartikel oder Mikrokapseln hergestellt werden.
Die erfindungsgemäßen oralen Darreichungsformen in Form von Pellets können vorzugsweise durch Mischen der Wirkstoffsalze, Extrusion und Spheronisation, durch Aufbaupelletisierung oder durch Direktpelletisierung in einem hochtourigen Mischer oder in der Rotorwirbelschicht hergestellt werden. Besonders bevorzugt ist die Herstellung der Pellets durch Extrusion feuchter Massen und anschließender Spheronisation. Die Herstellung von Mikrokapseln erfolgt nach üblichen Mikroverkapselungsverfahren, wie z.B. durch Sprühtrocknung, Sprüherstarrung oder Koazervation. Sofern die erfindungsgemäßen oralen Darreichungsformen in multipartikulärer Form vorliegen, wird der retardierende Überzug vorzugsweise so aufgebracht, daß man die multipartikulären Formen enthaltend die Salze des Wirkstoffes nach ihrer Herstellung mit den jeweiligen Polymeren und ggf. weiteren Hilfsstoffen aus wässrigen und/oder organischen Medien, vorzugsweise aus wässrigen Medien, mit Hilfe des Wirbelschichtverfahrens überzieht und den Überzug vorzugsweise gleichzeitig bei üblichen Temperaturen in der Wirbelschicht trocknet, ohne den Überzug anschließend zu tempern. Vorzugsweise erfolgt die Trocknung des Überzuges für Poly(meth)acrylatüberzüge bei einer Zulufttemperatur im Bereich von 30 bis 50 °C, besonders bevorzugt im Bereich von 35 bis 45 °C.
Für Überzüge auf Cellulosebasis, wie z.B. Ethylcellulose oder Celluloseacetat, erfolgt die Trocknung bevorzugt bei einer Temperatur im Bereich von 50 bis 80 °C, besonders bevorzugt im Bereich von 55 bis 65 °C.
Wachsüberzüge können durch Schmelzüberziehen in der Wirbelschicht aufgebracht werden und bei Temperaturen unterhalb des jeweiligen Schmelzbereiches nach dem Überziehen zur vollständigen Verfestigung abgekühlt werden. Das Aufbringen von Wachsüberzügen kann auch durch Aufsprühen von deren Lösungen in organischen Lösungsmitteln erfolgen.
Die an den Patienten zu verabreichende Menge des Wirkstoffes variiert in Abhängigkeit von der Art des eingesetzten Wirkstoffes, vom Gewicht des Patienten, von der Indikation sowie ggf. dem Schweregrad der Schmerzen bzw. der Erkrankung.
Vorzugsweise wird die zu verabreichende Menge des Wirkstoffes sowie dessen Gesamt-Freisetzung aus den Salzen des Wirkstoffes so eingestellt, daß eine Verabreichung der erfindungsgemäßen oralen Darreichungsformen höchstens zweimal, vorzugsweise nur einmal täglich erfolgen muß. Die erfindungsgemäßen, oralen Darreichungsformen haben den Vorteil, daß der Wirkstoff nach dem angestrebten Gesamt-Freisetzungsprofil, z.B. pulsatil oder mehrphasig über einen bestimmten Zeitraum, kontrolliert freigesetzt werden kann, ohne daß aufwendige, getrennte Formulierungsschritte für den Wirkstoff erforderlich sind.
Dies ermöglicht es, die Zeit und damit auch die Kosten für die Herstellung der erfindungsgemäßen oralen Darreichungsformen zu verringern.
Die Wasserlöslichkeit der Wirkstoffsalze wurde wie folgt bestimmt:
Von dem jeweiligen Wirkstoffsalz wurde bei 25 °C eine so große Menge in ionenfreies Wasser gegeben (z.B. für Tramadolsaccharinat ca. 1 g auf 10 ml ionenfreies Wasser), daß eine bei dieser Temperatur gesättigte Lösung entstand, die auch noch nach 20 Stunden rühren bei 25 °C gesättigt war. Die hierzu erforderliche Menge des jeweiligen Wirkstoffsalzes kann ggf. durch Vorversuche ermittelt werden.
Nach dem Absetzen lassen des nicht gelösten Wirkstoffsalzes wurde der klare Überstand abpipettiert und bei mindestens 3000 Umdrehungen pro Minute für 5 Minuten zentrifugiert.
Ein Teil des so erhaltenen klaren Überstandes wird in ein HPLC- Probenfläschen überführt und die Konzentration des Wirkstoffsalzes gegen einen entsprechenden Standard bestimmt.
Die Freisetzungsprofile der erfindungsgemäßen oralen Darreichungsformen wurde wie folgt bestimmt:
Die erfindungsgemäße Darreichungsform wurde in der Körbchenapparatur des Pharm. Eur. bei einer Temperatur des Freisetzungsmediums von 37 ± 0,5 °C bei einer Umdrehungsgeschwindigkeit von 50 Umdrehungen pro Minute 2 Stunden lang in 600 ml künstlichem Magensaft ohne Enzyme bei pH 1 ,2 geprüft. Anschließend wurde die Darreichungsform weitere 8 Stunden lang in 900 ml künstlichem Darmsaft ohne Enzyme bei pH 7,2 geprüft. Die jeweils zu einem Zeitpunkt freigesetzte Menge an Wirkstoff wurde mittels HPLC bestimmt. Die dargestellten Werte sind die Mittelwerte aus jeweils 3 Proben. Im folgenden wird die Erfindung anhand von Beispielen erläutert. Diese Erläuterungen sind lediglich beispielhaft und schränken den aligemeinen Erfindungsgedanken nicht ein.

Herstellung der Pellets:
50 g Tramadolhydrochlorid, 280 g [1e-(m-Methoxyphenyl)-2e- dimethyiaminomethyl-cyclohexan-1(a)-ol]-[2-(2',6'-dichloraniiino)-phenylacetat] (Tramadoldiciofenacat) und 330 g mikrokristalline Cellulose (Avicel PH 101®, FMC) werden für 10 Minuten in einem Kenwood Chef Mixer homogen gemischt und anschließend mit einer ausreichenden Menge demineralisiertem Wasser granuliert, so daß man ein für die Extrusion und Spheronisation geeignetes Granulat erhält. Das feuchte Granulat wird im NICA E140 Extruder mit einer 1 ,0 x 2,0 mm Extrusionsmatrize extrudiert und das feuchte Extrudat im NICA Spheroniser Typ S450 spheronisiert. Die Pellets werden anschließend 24 Stunden bei 50 °C im Trockenschrank getrocknet. Die Ausbeute an Pellets durch Siebfraktionierung mit einer Partikelgröße im Bereich von 800 bis 1250 μm beträgt > 90 %.
Aufbringen des Überzuges:
500 g dieser Pellets (800 bis 1250 μm) werden in der Wirbelschicht (Hüttlin HKC05) mit einer wäßigen Dispersion der nachstehenden Zusammensetzung bei einer Zulufttemperatur von 40 °C bis zu einer Gewichtszunahme von 7,6 % (bezogen auf das Ausgangsgewicht der Pellets) überzogen. Wäßrige Dispersion für 500 g Pellets:

Die Löslichkeit des [1e-(m-Methoxyphenyl)-2e-dimethylaminomethyl- cyclohexan-1 (a)-ol]-[2-(2',6'-dichloranilino)-phenylacetat] wurde nach obenstehender Methode zu ca. 0,3 mg/ml bestimmt, die des Tramadolhydrochlorids zu > 300 mg/ml.
Jeweils 710 mg der mit einem Überzug versehenen Pellets werden auf einer Zanasi E6 Steckkapselmaschine in Hartgelatine-Kapseln der Größe 0EL abgefüllt.
Das Freisetzungsprofil wurde gemäß der oben angegebenen Methode in der Körbchenapparatur bestimmt und ist in der nachfolgenden Tabelle 1 sowie in Figur 1 (in % der Gesamtdosis, berechnet als Tramadolhydrochlorid) wiedergegeben. Abweichend von den obenstehend beschriebenen Bedingungen wurden die überzogenen Pellets 18 Stunden in künstlichem Darmsaft ohne Enzyme bei pH 7,2 geprüft.

50 g Tramadolhydrochlorid, 280 g 1e-(m-Methoxyphenyl)-2e- dimethylaminomethyl-cyclohexan-1 (a)-ol]-[2-(2',6'-dichloranilino)-phenylacetat, 120 g Lactose Monohydrat, 90 g mikrokristalline Cellulose (Avicel PH 101®, FMC) und 90 g kolloidale mikrokristalline Cellulose (Avicel RC 591®, FMC) werden für 10 Minuten in einem Kenwood Chef Mixer homogen gemischt und anschließend mit einer ausreichenden Menge demineralisiertem Wasser granuliert, so daß man ein für die Extrusion und Spheronisation geeignetes Granulat erhält. Das feuchte Granulat wird im NICA E140 Extruder mit einer 1 ,0 x 2,0 mm Extrusionsmatrize extrudiert und das feuchte Extrudat im NICA Spheroniser Typ S450 spheronisiert. Die Pellets werden anschließend 24 Stunden bei 50 °C im Trockenschrank getrocknet. Die Ausbeute an Pellets durch Siebfraktionierung mit einer Partikelgröße im Bereich von 800 bis 1250 μm beträgt > 90 %.
Aufbringen des Überzuges:
500 g dieser Pellets (800 bis 1250 μm) werden in der Wirbelschicht (Hüttlin HKC05) mit einer wäßigen Dispersion der nachstehenden Zusammensetzung bei einer Zulufttemperatur von 60 °C bis zu einer Gewichtszunahme von von 2,4 Gew.-% (bezogen auf das Ausgangsgewicht der Pellets) überzogen.

Das Freisetzungsprofil wurde gemäß der oben angegebenen Methode in der
Körbchenapparatur bestimmt und ist in der nachfolgenden Tabelle 2 sowie in
Figur 2 (in % der Gesamtdosis, berechnet als Tramadolhydrochlorid) wiedergegeben. Abweichend von den obenstehend beschriebenen Bedingungen wurden die überzogenen Pellets 10 Stunden in künstlichem
Darmsaft ohne Enzyme bei pH 7,2 geprüft. Tabelle 2:
323 mg der überzogenen Pellets enthalten 25 mg Tramadolhydrochlorid und 140 mg
[1e-(m-Methoxyphenyl)-2e-dimethylaminomethyl-cyclohexan-1(a)-ol]-[2-(2',6'- dichloranilino)-phenylacetat] entsprechend einer Wirkstoffmenge von insgesamt 100 mg Tramadolhydrochlorid.
Der aufgebrachte Ethylcelluloseüberzug bewirkt keine Retardierung der Wirkstoffsalze sondern bewirkt lediglich, daß das [2-(2',6'-dichloranilino)- phenylacetat-lon nicht durch die Magensäure aus seinem Salz verdrängt wird. Aus dem sehr leicht wasserlöslichen Tramadolhydrochlorid wird der Wirkstoff Tramadol sehr schnell freigesetzt, aus dem wesentlich schwerer löslichen 1e- (m-Methoxyphenyl)-2e-dimethylaminomethyl-cyclohexan-1 (a)-ol]-[2-(2',6'- dichloranilino)-phenylacetat wird das Tramadol über einen Zeitraum von 10 Stunden retardiert abgegeben.
Beispiel 3:
Herstellung der Pellets:
20 g Tramadolhydrochlorid, 188 g [1e-(m-Methoxyphenyl)-2e- dimethylaminomethyl-cyclohexan-1(a)-ol]-[2-(2',6'-dichloranilino)-phenylacetat], 84 g Lactose Monohydrat und 332g mikrokristalline Cellulose (Avicel PH 101®, FMC) werden für 10 Minuten in einem Kenwood Chef Mixer homogen gemischt und anschließend mit einer ausreichenden Menge demineralisiertem Wasser granuliert, so daß man ein für die Extrusion und Spheronisation geeignetes Granulat erhält. Das feuchte Granulat wird im NICA E140 Extruder mit einer 1 ,0 x 2,0 mm Extrusionsmatrize extrudiert und das feuchte Extrudat im NICA Spheroniser Typ S450 spheronisiert. Die Pellets werden anschließend 24 Stunden bei 50 °C im Trockenschrank getrocknet. Die Ausbeute an Pellets durch Siebfraktionierung mit einer Partikelgröße im Bereich von 800 bis 1000 μm beträgt > 90 %.
Aufbringen des Überzuges:
500 g dieser Pellets (800 bis 1000 μm) werden in der Wirbelschicht (Hüttlin HKC05) mit einer wäßigen Dispersion der nachstehenden Zusammensetzung bei einer Zulufttemperatur von 40 °C bis zu einer Gewichtszunahme von 5,3 % (bezogen auf das Ausgangsgewicht der Pellets) überzogen.

219 mg der mit einem magensaftresistenten Überzug versehen Pellets enthalten 10 mg Tramadolhydrochlorid und 94 mg 1e-(m-Methoxyphenyl)-2e- dimethylaminomethyl-cyclohexan-1 (a)-ol]-[2-(2',6'-dichloranilino)-phenylacetat entsprechend einer Wirkstoffmenge von insgesamt 60 mg Tramadolhydrochlorid.
Der magensaftresistente Überzug bewirkt, daß während der Prüfung in künstlichem Magensaft keine Freisetzung des Wirkstoffes Tramadol erfolgt. Bei der Prüfung in künstlichem Darmsaft wird der Wirkstoff Tramadol aus Tramadolhydrochlorid sehr schnell, aus [1e-(m-Methoxyphenyl)-2e- dimethylaminomethyl-cyclohexan-1 (a)-ol]-[2-(2',6'-dichloranilino)-phenylacetat] über einen Zeitraum von 8 Stunden retardiert freigesetzt. Beispiel 4:
Herstellung der Pellets:
30 g Tramadolhydrochlorid, 254 g Tramadolsaccharinat (Wasserlöslichkeit, ca. 20 mg/ml, bestimmt nach obenstehend angegebener Methode), 284 g mikrokristalline Cellulose (Avicel PH 101®, FMC) werden für 10 Minuten in einem Kenwood Chef Mixer homogen gemischt und anschließend mit einer ausreichenden Menge demineralisiertem Wasser granuliert, so daß man ein für die Extrusion und Spheronisation geeignetes Granulat erhält. Das feuchte Granulat wird im NICA E140 Extruder mit einer 1 ,0 x 2,0 mm Extrusionsmatrize extrudiert und das feuchte Extrudat im NICA Spheroniser Typ S450 spheronisiert. Die Pellets werden anschließend 24 Stunden bei 50 °C im Trockenschrank getrocknet. Die Ausbeute an Pellets durch Siebfraktionierung mit einer Partikelgröße im Bereich von 800 bis 1250 μm beträgt > 90 %.
Aufbringen des Überzuges:
500 g dieser Pellets (800 bis 1250 μm) werden in der Wirbelschicht Hüttlin HKC05 mit einer wäßigen Dispersion der nachstehenden Zusammensetzung bei einer Zulufttemperatur von 40 °C bis zu einer Gewichtszunahme von 15 % (bezogen auf das Ausgangsgewicht der Pellets) überzogen.

Das Freisetzungsprofil wurde gemäß der oben angegebenen Methode in der Körbchenapparatur bestimmt und ist in der nachfolgenden Tabelle 4 sowie in Figur 4 (in % der Gesamtdosis, berechnet als Tramadolhydrochlorid) wiedergegeben:

327 mg der mit einem magensaftresistenten Überzug versehen Pellets enthalten 15 mg Tramadolhydrochlorid und 127 mg Tramadolsaccharinat entsprechend einer Wirkstoffmenge von insgesamt 100 mg Tramadolhydrochlorid.
Beispiel 5:
Herstellung der Pellets:
50 g Tramadolhydrochlorid, 526 g Tramadolsaccharinat und 384g mikrokristalline Cellulose (Avicel PH 101®, FMC) werden für 10 Minuten in einem Kenwood Chef Mixer homogen gemischt und anschließend mit einer ausreichenden Menge demineralisiertem Wasser granuliert, so daß man ein für die Extrusion und Spheronisation geeignetes Granulat erhält. Das feuchte Granulat wird im NICA E140 Extruder mit einer 1 ,0 x 2,0 mm
Extrusionsmatrize extrudiert und das feuchte Extrudat im NICA Spheroniser Typ S450 spheronisiert. Die Pellets werden anschließend 24 Stunden bei 50 °C im Trockenschrank getrocknet. Die Ausbeute an Pellets durch Siebfraktionierung mit einer Partikelgröße im Bereich von 800 bis 1250 μm beträgt > 90 %.
Aufbringen des Überzuges:
500 g dieser Pellets (800 bis 1250 μm) werden in der Wirbelschicht (Hüttlin HKC05) mit einer wäßigen Dispersion der nachstehenden Zusammensetzung bei einer Zulufttemperatur von 40 °C bis zu einer Gewichtszunahme von 14,4 % (bezogen auf das Ausgangsgewicht der Pellets) überzogen.

Das Freisetzungsprofil wurde gemäß der oben angegebenen Methode in der Körbchenapparatur bestimmt und ist in der nachfolgenden Tabelle 5 sowie in Figur 5 (in % der Gesamtdosis, berechnet als Tramadolhydrochlorid) wiedergegeben: Tabelle 5
549 mg der mit einem Überzug versehen Pellets enthalten 25 mg Tramadolhydrochlorid und 263 mg Tramadolsaccharinat entsprechend einer Wirkstoffmenge von insgesamt 200 mg Tramadolhydrochlorid.
Beispiel 6:
Herstellung von Tabletten:
15 g Promethazinhydrochlorid, 39 g Promethazinteoclat, 120 g mikrokristalline Cellulose, 75 g Methylhydroxypropylcellulose (50 mPa-s, Metolose 60 SH), 2,5 g hochdisperses Siliziumdioxid und 2,5 g Magnesiumstearat werden 10 Minuten in einem Freifallmischer (Bohle, LM 40) homogen gemischt. Dieses Gemisch wird auf einer Korsch EK0 Exzenterpresse mit einem Stempelwerkzeug zu runden, geölbten Tabletten mit einem Durchmesser von 9 mm verpreßt. Das Freisetzungsprofil wurde nach der obenstehend angegebenen Methode in der bestimmt. Abweichend von den obenstehend beschriebenen Bedingungen wurden die überzogenen Pellets in der Blattrührerapparatur des Pharm. Eur. geprüft.

Claims

Patentansprüche:
1. Orale Darreichungsformen mit kontrollierter Freisetzung eines Wirkstoffes, dadurch gekennzeichnet, daß derselbe Wirkstoff in Form von wenigstens zwei unterschiedlichen Salzen vorliegt, die im festen Aggregatzustand in der Darreichungsform vorhanden sind und eine unterschiedliche in-vitro Freisetzung dieses Wirkstoffes aufweisen.
2. Orale Darreichungsformen nach Anspruch 1 , dadurch gekennzeichnet, daß die verschiedenen Wirkstoffsalze eine unterschiedliche
Wasserlöslichkeit aufweisen.
3. Orale Darreichungsformen nach Anspruch 2, dadurch gekennzeichnet, daß sich die Wasserlöslichkeit aller unterschiedlichen Wirkstoffsalze jeweils um wenigstens den Faktor 2 voneinander unterscheidet.
4. Orale Darreichungsformen nach einem oder mehreren der Ansprüche 1 bis 3, dadurch gekennzeichnet, daß der Wirkstoff aus der Gruppe der salzbildenden pharmazeutischen Wirkstoffe, Vitamine, Mineralstoffe, Nährstoffe oder Diagnostika, vorzugsweise aus der Gruppe der salzbildenden pharmazeutischen Wirkstoffe ausgewählt worden ist.
5. Orale Darreichungsformen nach Anspruch 4, dadurch gekennzeichnet, daß der salzbildende pharmazeutische Wirkstoff ausgewählt ist aus der Gruppe der salzbildenden Analgetika, Anthelminthika, Antiarrhythmika,
Antiasthmatika, Antidepressiva, Antidiabetika, Antidota, Antiallergika, Antitussiva, Antibiotika, Antiemetika, Antiinfektiva, Antihistaminika, Antihypertonika, Antihypertensiva, Antihypotonica, Antikoagulantien, Antirheumatika, Antipyretika, Anxiolytika, Abmagerungsmittel, Acidosetherapeutika, Antivertiginosa, Antihämorrhagika, Antifibrionolytika,
Hämostatika, Antihypogylkämika, Antihypotonika, Antimykotika, Antiphlogistika, Expektorantia, Arteriosklerosemittel, Betrarezeptorenblocker, Calciumkanalblocker, Hemmstoffe des Renin- Angiotensin-Systems, Broncholytika, Cholagoga, Gallenwegstherapeutika, Cholinergika, Corticoide (Interna), Durchblutungsfördernde Mittel, Entwöhnungsmittel, Geriatrika, Gichtmittel, Grippemittel, Mittel gegen Erkältungskrankheiten, Gynäkologika, Hepatika, Hypnotika, Hypophysenhormone, Hypothalamushormone, regulatorische Peptide oder deren Hemmstoffe, Immunmodulatoren, Kardiaca, Analeptika, Antihypoxämika, Antianämika, Antidementiva (Nootropika), Appetitzügler, Koronarmittel, Laxantia, Chemotherapeutika, Diuretika, Enzyme, Fungistatika, Hormone, Lipidsenker, Neuraltherapeutika, Magen-Darm- Mittel, Migränemittel, Mineralstoffpräparate, Muskelrelaxantia,

Orale Darreichungsformen gemäß Anspruch 5, dadurch gekennzeichnet, daß die salzbildenden Analgetika aus der Gruppe der salzbiidenden Opioide, Verbindungen mit opioider Wirksamkeit oder sterioidfreien Analgetika ausgewählt worden sind.
7. Orale Darreichungsformen gemäß Anspruch 6, dadurch gekennzeichnet, daß die salzbildenden Analgetika ausgewählt sind aus der Gruppe Brifentanil, Carfentanil, Fentatienil, Lofentanil, Ocfentanil, Trefentanil, Codein, Dextropropoxyphen, Dihydrocodein, Diphenoxylat, Meptazinol, Nalbuphin, Pethidin (Meperidin), Tilidin, Tramadol, Viminol, Butorphanol,
Dextromoramid, Dezocin, Diacetylmorphin (Heroin), Hydrocodon, Hydromorphon, Ketobemidon, Levomethadon, Levomethadyl, Levorphanol, Morphin, Nalorphin, Oxycodon, Pentazocin, Piritramid, Alfentanil, Buprenorphin, Etorphin, Fentanyl, Remifentanil oder Sufentanil, vorzugsweise Tramadol oder Morphin.
8. Orale Darreichungsformen gemäß Anspruch 5, dadurch gekennzeichnet, daß das salzbildende Neuroleptikum Promethazin ist.
9. Orale Darreichungsformen gemäß einem oder mehreren der Ansprüche 1 bis 8, dadurch gekennzeichnet, daß die Wirkstoffsalze aus der Gruppe Chlorid, Bromid, Sulfat, Sulfonat, Phosphat, Tartrat, Teoclat, Embonat, Formiat, Acetat, Propionat, Benzoat, Oxalat, Succinat, Citrat, Diclofenacat,
Naproxenat, Salicylat, Acetylsalicylat, Glutamat, Fumarat, Aspartat, Glutarat, Stearat, Butyrat, Malonat, Lactat, Mesylat, Saccharinat, Cyclamat oder Acesuifamat, vorzugsweise aus der Gruppe Chlorid, Sulfat, Saccharinat, Teoclat, Embonat, Diclofenacat, Naproxenat oder Salicylat ausgewählt worden sind.
10. Orale Darreichungsformen gemäß einem oder mehreren der Ansprüche 1 bis 6, dadurch gekennzeichnet, daß als Wirkstoffsalz ein Alkalimetallsalz, Erdalkalimetallsalz, Ammoniumsalz, Eisensalz oder Aluminiumsalz des Wirkstoffes, vorzugsweise ein Alkalimetallsalz, besonders bevorzugt das
Natrium- oder Kaliumsalz des Wirkstoffes eingesetzt worden ist.
11. Orale Darreichungsformen gemäß einem der Ansprüche 1 bis 10, dadurch gekennzeichnet, daß sie in Form von Tabletten, Kautabletten, Kaugummis, Dragees, Pulver ggf. in Kapseln abgefüllt, vorzugsweise in
Form von Tabletten vorliegen.
12. Orale Darreichungsformen gemäß einem der Ansprüche 1 bis 10, dadurch gekennzeichnet, daß sie in multipartikulärer Form, vorzugsweise in Form von Mikrotabletten, Mikrokapseln, Granulaten, Wirkstoffkristallen oder
Pellets, besonders bevorzugt in Form von Mikrotabletten, Granulaten oder Pellets, ggf. in Kapseln abgefüllt oder zu Tabletten verpreßt, vorliegen.
13. Orale Darreichungsformen gemäß Anspruch 12, dadurch gekennzeichnet, daß die Granulate oder Pellets eine Größe im Bereich von 0,1 bis 3 mm, vorzugsweise im Bereich von 0,5 bis 2 mm aufweisen.
14. Orale Darreichungsformen gemäß Anspruch 12, dadurch gekennzeichnet, daß die Mikrotabletten einen Durchmesser von 0,5 bis 5 mm, vorzugsweise 1 bis 3 mm, besonders bevorzugt 1 bis 2 mm aufweisen.
15. Orale Darreichungsformen gemäß Anspruch 12, dadurch gekennzeichnet, daß die Wirkstoffkristalle, Mikropartikel, Mikropellets oder Mikrokapseln einen Durchmesser von 10 μm bis 1 mm, vorzugsweise 15 μm bis 0,5 mm, besonders bevorzugt 30 μm bis 200 μm aufweisen.
16. Orale Darreichungsformen nach einem oder mehreren der Ansprüche 1 bis 15, dadurch gekennzeichnet, daß wenigstens eines der Wirkstoffsalze in retardierter Form vorliegt.
17. Orale Darreichungsformen Anspruch 16, dadurch gekennzeichnet, daß alle Wirkstoffsalze in retardierter Form vorliegen.
18. Orale Darreichungsformen nach einem oder mehreren der Ansprüche 1 bis 17, dadurch gekennzeichnet, daß die jeweilige Darreichungsform retardiert ist.
19. Orale Darreichungsformen gemäß einem der Ansprüche 16 bis 18, dadurch gekennzeichnet, daß die Retardierung durch einen retardierenden Überzug und/oder Einbettung in eine retardierende Matrix erfolgt ist.
20. Orale Darreichungsformen gemäß Anspruch 19, dadurch gekennzeichnet, daß der retardierende Überzug auf einem wasserunlöslichen ggf. modifizierten natürlichen oder synthetischen Polymeren ggf. in Kombination mit einem üblichen Weichmacher oder auf einem natürlichen, halbsynthetischen oder synthetischen Wachs oder Fett oder Fettalkohol oder einem Gemisch aus wenigstens zwei dieser Komponenten basiert.
21. Orale Darreichungsformen gemäß Anspruch 20, dadurch gekennzeichnet, daß als wasserunlösliche Polymere Poly(meth)acrylate, bevorzugt
Poly(Cι-4)-Alkyl(meth)acrylate,
Poly(Cι-4)-dialkylamino-(Cι-4)-alkyl(meth)acrylate und/oder deren
Copolymere, bevorzugt Ethylacrylat/Methylmethacrylat-Copolymere mit einem molaren Verhältnis der Monomeren von 2 : 1 (Eudragit NE30D®), Ethylacrylat/Methylmethacrylat/Trimethylammoniumethylmethacrylat- chlorid-Copolymere mit einem molaren Verhältnis der Monomeren von 1 :
2 : 0,1 (Eudragit RS®),
Ethylacrylat/Methylmethacrylat/Trimethylammoniumethyl- methacrylatchlorid-Copolymere mit einem molaren Verhältnis der Monomeren von 1 : 2 : 0,2 (Eudragit RL®) oder ein Gemisch aus wenigstens zwei dieser vorstehend genannten Polymeren als
Überzugsmaterial vorliegen.
22. Orale Darreichungsformen gemäß Anspruch 20, dadurch gekennzeichnet, daß als wasserunlösliche Polymere Cellulosederivate, vorzugsweise
Alkylcellulose, besonders bevorzugt Ethylcellulose, oder Celluloseester, vorzugsweise Celluloseacetat als Überzugsmaterial vorliegen.
23. Orale Darreichungsformen gemäß Anspruch 21 oder 22, dadurch gekennzeichnet, daß die Polymere aus wäßrigem Medium, vorzugsweise aus wäßriger Latex oder Pseudolatex-Dispersionen aufgetragen worden sind.
24. Orale Darreichungsformen gemäß Anspruch 19, dadurch gekennzeichnet, daß als Überzugs-Polymere ein Gemisch aus Polyvinylacetat und
Polyvinyipyrollidon, vorzugsweise in Form wäßriger Pseudolatex- Dispersionen eingesetzt worden ist.
5. Orale Darreichungsformen gemäß Anspruch 20, dadurch gekennzeichnet, daß als Wachs Camaubawachs, Bienenwachs, Glycerinmonostearat, Glycerinmonobehenat (Compritol AT0888®), Glycerinditripalmitostearat (Precirol ATO5®), mikrokristallines Wachs oder ein Gemisch aus wenigstens zwei dieser Komponenten als Überzugsmaterial vorliegt.
26. Orale Darreichungsformen gemäß einem der Ansprüche 21 bis 23, dadurch gekennzeichnet, daß die Polymere in Kombination mit üblichen Weichmachern eingesetzt worden sind.
27. Orale Darreichungsformen gemäß Anspruch 26, dadurch gekennzeichnet, daß als Weichmacher lipophile Diester aus aliphatischen oder aromatischen Dicarbonsäuren mit Cβ-C4o und aliphatischen Alkoholen mit Ci-Cβ, vorzugsweise Dibutylphthalat, Diethylphthalat, Dibutylsebacat oder Diethylsebacat, ein hydrophile oder lipophile Ester der Zitronensäure, vorzugsweise Triethylcitrat, Tributylcitrat, Acetyltributylcitrat oder Acetyltriethylcitrat, Polyethylenglycole, Propylenglycole, Ester des Glycerins, vorzugsweise Triacetin, acetylierte Mono- und Diglyceride, mittelkettige Triglyceride, Ölsäure oder ein Gemisch aus wenigstens zwei dieser Weichmacher eingesetzt worden sind.
28. Orale Darreichungsformen gemäß Anpruch 26 oder 27, dadurch gekennzeichnet, daß der Weichmacher in Mengen von 5 bis 50 Gew-%., vorzugsweise 10 bis 40 Gew.-%, besonders bevorzugt 10 bis 30 Gew-%, bezogen auf das polymere Überzugsmaterial, vorliegt.
29. Orale Darreichungsformen gemäß einem der Ansprüche 19 bis 28, dadurch gekennzeichnet, daß die Matrix auf einem hydrophilen Matrixmaterial, vorzugsweise hydrophilen Polymeren, besonders bevorzugt auf Celluloseethern, Celluloseestern und/oder Acrylharzen, ganz besonders bevorzugt auf Ethylcellulose, Hydroxypropylmethylcellulose, Hydroxypropylcellulose, Hydroxymethylcellulose, Poly(meth)acrylsäure und/oder deren Salzen, Amiden und/oder Estern basiert.
30. Orale Darreichungsformen gemäß einem der Ansprüche 19 bis 29, dadurch gekennzeichnet, daß die Matrix auf einem hydrophoben
Matrixmaterial, vorzugsweise hydrophoben Polymeren, Wachsen, Fetten, langkettigen Fettsäuren, Fettalkoholen oder entsprechenden Estern oder Ethern oder deren Gemischen, besonders bevorzugt auf Mono- oder Diglyceriden von C-ι2-C3o-Fettsäuren und/oder C-ι2-C3o-Fettalkoholen und/oder Wachsen oder deren Gemischen basiert.
31. Orale Darreichungsformen gemäß einem der Ansprüche 1 bis 31 , dadurch gekennzeichnet, daß sie einen Schutzüberzug, vorzugsweise einen magensaftresistenten Schutzüberzug aufweist.
32. Orale Darreichungsformen gemäß Anspruch 31 , dadurch gekennzeichnet, daß der magensaftresistente Überzug aus
Methacrylsäure/Methylmethacrylat-Copolymeren mit einem molaren Verhältnis der Monomeren von 1 : 1 (Eudragit L®), Methacrylsäure/Methylmethacrylat-Copolymeren mit einem molaren
Verhältnis der Monomeren von 1 : 2 (Eudragit S®), Methacrylsäure/Ethylacrylat-Copolymeren mit einem molaren Verhältnis der Monomeren von 1 : 1 (Eudragit L30-D55®) Methacrylsäure/Methylacrylat/Methylmethacrylat mit einem molaren Verhältnis der Monomeren von 7 : 3 : 1 (Eudragit FS®), Schellack,
Hydroxypropylmethylcelluloseacetatsuccinat, Celluloseacetatphthalat oder aus einer Mischung aus wenigstens zwei dieser Komponenten, ggf. auch in Kombination mit Poly(meth)acrylaten, vorzugsweise Eudragit NE30D® und/oder Eudragit RL® und/oder Eudragit RS® besteht.
33. Orale Darreichungsformen nach einem oder mehreren der Ansprüche 1 bis 32 erhältlich durch Mischen von wenigstens zwei unterschiedlichen Salzen desselben Wirkstoffes, von denen ggf. mindestens ein Salz in retardierter Form voriiegt und die eine unterschiedliche in-vitro Freisetzung dieses Wirkstoffes aufweisen, Formulieren der Mischung und Überziehen mit einem Schutzüberzug, der retardierend und/oder magensaftresistent ist.
