Description
Neue Hetaryloxy-ß-carboline, deren Herstellung und Verwendung in Arzneimitteln
Die Erfindung betrifft neue Hetaryloxy-ß-carbolin-Derivate, deren Her¬ stellung und Verwendung in Arzneimitteln.
In EP-A-237 467 und EP-A-305 322 werden mit einem Hetaryloxy-Rest substi¬ tuierte ß-Carboline beschrieben, die das Zentralnervensystem beeinflussen und als Psychopharmaka verwendet werden. Nach diesen Patentanmeldungen war nicht zu erwarten, daß bei der Einführung der erfindungsgemäßen Het- aryl-Substituenten eine Verschiebung des Wirkprofils der Verbindungen eintritt und daß die Verbindungen wegen der fehlenden Muskelrelaκation ein verbessertes Nebenwirkungsprofil zeigen.
Die erfindungsgemäßen Verbindungen haben die Formel I

Der Substituent R kann im A-Ring in Position 5 - 8, bevorzugt in 5,- 6- oder 7-Stellung stehen.
Alkyl beinhaltet jeweils sowohl gerad- als auch verzweigt-kettige Reste wie beispielsweise Methyl, Ethyl, Propyl, Isopropyl, Butyl, Isobutyl, sek. Butyl, tert. Butyl, Pentyl, Isopentyl und Hexyl.
Unter Halogen ist jeweils Fluor, Chlor, Brom und Jod zu verstehen. Cycloalkyl steht jeweils für Cyclopropyl, Cyclobutyl, Cyclopentyl, Cyclo- hexyl, Cycloheptyl.
Der benzokondensierte Hetarylrest mit 1 - 2 Stickstoffatomen hat 8-12 Ringatome; es seien beispielsweise Chinolin, Isochinolin, Chinoxalin, Benzimidazol genannt.
A Bedeutet R einen substituierten stickstoffhaltigen Hetarylrest, so sei beispielsweise Pyridin, Pyrimidin, Pyrazin, Pyridazin, I idazol genannt.
2 Unter einem Bicycloalkylrest R ist Bicycloheptyl und Bicyclooctyl zu
2 verstehen. Als mono- oder bicyclischer Arylrest R seien beispielsweise
Phenyl, Biphenyl, Naphthyl und Indenyl genannt.

Für R ist -C0 -C, .-Alkyl, -CO-R mit R in der Bedeutung eines C„ - 1-o 3-7
Cycloalkyl oder eines gegebenenfalls substituierten Phenylrestes, und der Isoxazol-3-yl-Rest als bevorzugt anzusehen.
Die physiologisch verträglichen Säureadditionssalze leiten sich von den bekannten anorganischen und organischen Säuren ab wie beispielsweise Salzsäure, Bromwasserstoffsäure, Schwefelsäure, Phosphorsäure, Ameisen¬ säure, Essigsäure, Benzoesäure, Maleinsäure, Fumarsäure, Succinsäure, Weinsäure, Zitronensäure, Oxalsäure, Glyoxylsaure sowie von Alkansulfon- säuren wie beispielsweise Methansulfonsäure, Ethansulfonsäure, Benzol- sulfonsäure, p-Toluolsulfonsäure u.a.
Die Verbindungen der Formel I sowie deren Säureadditionssalze sind auf Grund ihrer Affinität zu Benzodiazepin-Rezeptoren als Arzneimittel verwendbar und üben auf die von den Benzodiazepinen bekannten Eigenschaf¬ ten partial-agonistische Wirkung aus, die dadurch gekennzeichnet ist.d aß die Verbindungen zum Beispiel antikonvulsiv und anxiolytisch wirken und nicht ataktisch/muskelrelaxierend sind. Zur Untersuchung der anxioly- tischen Wirkung werden die Verbindungen im 4-Plattentest nach der Methode von Boissier et al Eur. J. Pharmacol. 4, 145-150 (1968) getestet. In der Tabelle ist die minimale niedrigste Dosis (MED) angegeben, die die loko- motorische Aktivität der gestraften Mäuse nach i.p. Behandlung erhöht.

Auf Grund der guten Wirksamkeit im PTZ-Krampftest und im 4-Platten-Test eignen sich die erfindungsgemäßen Verbindungen insbesondere zur Behand¬ lung von Epilepsie und Angstzuständen.
Zur Verwendung der erfindungsgemäßen Verbindungen als Arzneimittel werden diese in die Form eines pharmazeutischen Präparats gebracht, das neben dem Wirkstoff für die enterale oder parenterale Applikation geeignete pharmazeutische, organische oder anorganische inerte Trägermaterialien, wie zum Beispiel Wasser, Gelatine, Gummi arabicum, Milchzucker, Stärke, Magnesiumstearat, Talk, pflanzliche Öle, Polyalkylenglykole usw. enthält.
Die pharmazeutischen Präparate können in fester Form, zum Beispiel als Tabletten, Dragees, Suppositorien, Kapseln oder in flüssiger Form, zum Beispiel als Lösungen, Suspensionen oder Emulsionen vorliegen. Gegebenen¬ falls enthalten sie darüber hinaus Hilfsstoffe wie Konservierungs- , Sta- bilisierungs- , Netzmittel oder Emulgatoren, Salze zur Veränderung des os otischen Drucks oder Puffer.
Für die parenterale Anwendung sind insbesondere In ektionslösungen oder Suspensionen, insbesondere wässrige Lösungen der aktiven Verbindungen in polyhydroxyethoxyliertem Rizinusöl, geeignet.
Als Trägersysteme können auch grenzflächenaktive Hilfsstoffe wie Salze der Gallensäuren oder tierische oder pflanzliche Phospholipide, aber auch Mischungen davon sowie Liposome oder deren Bestandteile verwendet werden.
Für die orale Anwendung sind insbesondere Tabletten, Dragees oder Kapseln mit Talkum und/oder Kohlenwasserstoffträger oder -binder, wie zum Beispiel Lactose, Mais- oder Kartoffelstärke, geeignet. Die Anwendung kann auch in flüssiger Form erfolgen, wie zum Beispiel als Saft, dem gegebenenfalls ein Süßstoff beigefügt wird.
Die erfindungsgemäßen Verbindungen werden in einer Dosiseinheit von 0,05 bis 100 mg aktiver Substanz in einem physiologisch verträglichen Träger eingebracht.
Die erfidungsgemäßen Verbindungen werden im allgemeinen in einer Dosis von 0,1 bis 300 mg/Tag, vorzugsweise 0,1 bis 30 mg/Tag, besonders bevor¬ zugt 1-20 mg/Tag angewendet, beispielsweise als Anxiolytika analog Diaze- pam.
Die Herstellung der erfindungsgemäßen Verbindungen erfolgt nach an sich bekannten Methoden, Beispielsweise gelangt man zu Verbindungen der Formel I dadurch, daß man

Die reaktionsfähige Verbindung R -Y, worin Y beispielsweise Halogen, Tosylat, Mesylat oder Triflat bedeutet, wird in Gegenwart einer Base wie Erdalkali- oder Alkali-alkoholat oder -hydroxid, Alkali- oder Erdalkali- carbonat in polaren Lösungsmitteln wie Dimethylsulfoxid, Dimethylform- amid, Acetonitril oder Alkoholen bei Raumtemperatur oder erhöhter Temperatur umgesetzt, gegebenenfalls in Gegenwart von Phasentransferka- talysatoren.

Die Einführung des Substituenten R in der Bedeutung -CO-R nach Verfah¬ rensvariante b) erfolgt nach den in WO 91/09858 beschriebenen Methoden. Beispielsweise wird ein ß-Carbolin-3-carbonsäurealkylesterdeπvat oder dessen reaktives Säurederivat wie das Carbonsäureimidazolid mit einer
2 . . Lithium-organischen Verbindung R Li in aprotischen polaren Losungsmitteln wie cyclischen oder acyclischen Ethern oder Kohlenwasserstoffen bei
Temperaturen von - 70 °C bis zu Raumtemperatur umgesetzt. Zweckmässiger- weise kann in 9-Stellung des ß-Carbolins eine Schutzgruppe vorhanden sein wie Tosyl, Mesyl oder eine Trialkylsilyl-Gruppe, die bei der Aufarbeitung des Reaktionsgemisches oder anschließend in üblicher Weise je nach Art der Schutzgruppe abgespalten wird.
Die Cycloaddition der Verbindungen der Formeln IV und VI nach Verfahren c) und d) erfolgt nach den in EP-A-305 322 beschriebenen Methoden. Die Addition wird bei Temperaturen von 0 °C - 40 °C in einem aprotischen Lösungsmittel wie aliphatische oder cyclische Ether, halogenierte Kohlen¬ wasserstoffe, Dimethylformamid u.a. durchgeführt. Gegebenenfalls können
in die Reaktion in 9-Stellung geschützte ß-Carbolin-Derivate eingesetzt werden. Die Schutzgruppe wird in üblicher Weise bei der Aufarbeitung des Reaktionsgemisches oder anschließend durch Behandeln mit Basen oder Säu¬ ren je nach Art der Schutzgruppe abgespalten.
Die Herstellung der Nitriloxide erfolgt beispielsweise durch Umsetzung von ß-Carbolin-3-carbaldehyden zu den entsprechenden Oximen, die beispielsweise mit N-Halogen-Succinimid, tert. Butoxychlorit oder Na-hypochlorit in den vorher genannten aprotischen Lösungsmitteln in Hydroxamsäurehalogenide überführt werden können. Mit Basen wie Na- oder K-alkoholaten. Trialkylaminen, Hünig Base, DBU oder Diazabicyclooctan wird aus den Hydroxamsäurehalogeniden Halogenwasserstoff abgespalten und man erhält die Nitriloxide, die ohne Isolierung der Cycloaddition unter¬ worfen werden (R. Annunziata et al. , 3. Chem. Soc. 1987. 529).
Die Einführung der Oxadiazolreste nach den Verfahrensvarianten e) und f) kann nach den in EP-161 574 beschriebenen Methoden erfolgen. Die Herstel¬ lung von 1 ,2,4-0xadiazol-3-yl-carbolinen erfolgt beispielsweise durch
5 Umsetzung von ß-Carbolιn-3-carboxamιdoxιmen mit Säureanhydriden (R CO] bei Raumtemperatur bis zur Siedetemperatur des Gemisches.
Zur Einführung des 1 , ,4-0xadiazol-5-yl-Restes wird die ß-Carbolin-3-car- bonsäure oder deren reaktives Säurederivat wie Halogenid, Imidazolid oder gemischtes Anhydrid oder Carbonsäurealkylester in Gegenwart von Alkoholat mit einem Amidoxim
in aprotischen Lösungsmitteln wie Kohlenwasserstof en wie Toluol, Ethern oder Dimethylformamid bei Raumtemperatur oder erhöhter Temperatur umge¬ setzt.
Wird eine Umesterung gewünscht, so kann man die in EP-A-237 467 beschrie¬ benen Methoden anwenden, indem man mit Alkalialkoholaten oder dem ent¬ sprechenden Alkohol, gegebenenfalls unter Zugabe von Titan-tetra-isoprop- ylat als Katalysator bei erhöhter Temperatur umestert. Die Einführung der
tert. Butylestergruppe erfolgt z.B. durch Umsetzung der Carbonsäure mit tert. Butoxy-bis-dimethyl-aminomethan. Die Hydrolyse der Estergruppe kann in üblicher Weise sauer oder alkalisch erfolgen, beispielsweise mit Na- oder K-hydroxid in protischen Lösungsmitteln oder nach den in EP-A-161 574 beschriebenen Verfahren.
Die Isomerengemische können nach den üblichen Methoden wie beispielsweise Kristallisation, Chromatographie oder Salzbildung in die Diastereomeren bzw. Enantiomeren getrennt werden.
Zur Bildung der physiologisch verträglichen Säureadditonssalze wird eine Verbindung der Formel I beispielsweise in wenig Alkohol gelöst und mit einer konzentrierten Lösung der gewünschten Säure versetzt.
Soweit die Herstellung der Ausgangsverbindungen nicht beschrieben wird, sind diese bekannt oder analog zu bekannten Verbindungen oder hier beschriebenen Verfahren herstellbar.
Beispielsweise ist die Herstellung der 3-Carbonsäureester der Formel II in EP-A-130 140 beschrieben.
Die nachfolgenden Beispiele sollen das erfindungsgemäße Verfahren erläutern.
Beispiel 1
6-(1-Isochinolyloxy)-4-methoxymethyl-B-carbolin-3-carbonsäureisooroDVl- ester
1,3 g Kaliumhydroxidpulver wird zu 25 ml Dimethylsulfoxid bei Raumtempe¬ ratur zugesetzt. Anschließend werden zunächst 3,14 g 6-Hydroxy-ß-carbo- lin-3-carbonsäureisopropylester portionsweise zu dem Ansatz gegeben und dann wird eine Lösung von 2 g 1-Chlorisochinolin in 2 ml Dimethylsulfoxid eingetropft. Nach 3 Stunden Erhitzen auf 90 - 95 °C Badtemperatur werden noch einmal 375 mg 1-Chlorisochinolin zugegeben und 2 Stunden auf 100 °C erwärmt. Man gießt auf Eis, stellt mit Eisessig auf pH 5 ein, saugt den Niederschlag ab und wäscht mit Essigester nach. Man chromatographiert diesen Rückstand über Kieselgel mit Methylenchlorid : Ethanol = 13 : 1 als Elutionsmittel. Nach Einengen der entsprechend zusammengefaßten Fraktionen und Umkristallisation erhält man 2,5 g (55 7. der Theorie) 6- ( 1-Isochinolyloxy)-4-methoxymethy1-ß-carbolin-3-carbonsäureisopropyl- ester vom Schmelzpunkt 10 °C.

Beispiel 2
6-(1-Isochinolyloxy-4-methoxymethyl-3-(5-methoxymethylisoxazol-3-yl)-6- carbpl-yn
330 mg 6-(1-Isochinolyloxy)-4-methoxymethyl-ß-carbolin-3-carbonsäureiso- propylester werden in 10 ml Toluol mit 0,62 ml Triethylamin und 0,2 Chlortrimethylsilan versetzt und 45 Minuten auf 50 - 60 °C Badtemperatur erwärmt. Nach Einengen auf ca. 6 ml wird auf - 78 °C unter Argon abge¬ kühlt und 1,25 ml einer 1,2 molaren Diisobutylaluminiumhydrid in Hexan eingetropft. Nach 30 Minuten bei - 78 °C wird mit 3 ml Ethanol und 0,75 ml 1N-NaOH versetzt. Es wird mit Essigester und sehr wenig Wasser ver¬ setzt. Die organische Phase wird abgetrennt und ohne Trocknung eingeengt. Der Rückstand wird mit Hexan angerührt und ergibt 440 mg 6-(1-Isochinol- yloxy)-4-methoxymethyl-ß-carbolin-3-carbaldehyd. 254 mg davon werden in 2,5 ml Dimethylfuran mit 67,5 mg Hydroxylaminhydrochlorid und 69 mg Kaliumhydroxidpulver versetzt und 16 Stunden bei Raumtemperatur stehen gelassen. Der Ansatz wird auf Eis gegossen und der Niederschlag abgesaugt sowie mit Wasser nachgewaschen. Man erhält 133 mg 6-( 1-Isochinolyloxy-4- methoxymethyl-ß-carbolin-3-carbaldehydoxim vom Schmelzpunkt 215 - 218 °C.
450 mg dieses Oxims werden in 7 ml Dimethylformamid gelöst und mit 218 mg N-Bromsuccinimid 30 Minuten bei Raumtemperatur gerührt. Nach Zugabe von 0,8 ml Triethylamin und 0,16 ml Methylpropargylether wird 3 Stunden bei Raumtemperatur gerührt. Nach Einengen wird in Essigester/Wasser verteilt und die organische Phase getrocknet, filtriert und eingeengt.Der Rück¬ stand wird zweimal über Kieselgel zunächst mit Methylenchlorid : Ethanol = 10 : 1 und dann mit Methylenchlorid : Ethanol = 12 : 1 chro atogra- phiert. Nach Umkristallisation der entsprechend zusammengefaßten und ein¬ geengten Fraktionen erhält man 200 mg 6- ( 1-Isochinolyloκy)-4-methoxy- methyl-3-(5-methoκymethylisoxazol-3-yl)-ß-carbolin vom Schmelzpunkt 107 - 112 °C (Essigester/Hexan).
Beispiel 3
6-(1-Isochinolyloxy)-4-methoxymethyl-3-benzoyl-B-carbolin
476 mg 6-( 1-Isochinolyloxy)-4-methoκymethyl-9-tosyl-ß-carbolin-3-carbon- säureisopropylester werden in 10 ml Tetrahydrofuran unter Argon bei - 60 °C mit 0,79 ml einer 0,9 m Lösung von Phenyllithium in Benzol ver¬ setzt. Nach 1 Stunde bei - 60 °C wird 16 Stunden bei Raumtemperatur gerührt. Der Ansatz wird mit Eisessig angesäuert und eingeengt. Der Rück¬ stand wird in Essigester/Wasser verteilt und die organische Phase nacheinander mit jeweils gesättigter Natriumbicarbonat- und Kochsalz¬ lösung gewaschen, getrocknet, filtriert und eingeengt. Der Rückstand wird über Kieselgel mit Methylenchlorid : Ethanol = 10 : 1 chromatographiert. Die entsprechenden Fraktionen werden zusammengefaßt, eingeengt und umkristallisiert. Man erhält 30 mg 6-( 1-Isochinolyloxy)-4-methoxymethyl- 3-benzoyl-ß-carbolin vom Schmelzpunkt 159 - 160 °C (Essigester/Hexan).
