Abstract
Die Erfindung betrifft Cyclodextrin-Clathrate von 5-Cyano-prostacyclin-analoga der allgemeinen Formel (I), worin R1 den Rest OR3, wobei R3 Wasserstoff, Alkyl, Cycloalkyl, Aryl oder einen hetereocyclischen Rest bedeuten kann, oder den Rest NHR4 mit R4 in der Bedeutung eines Säurerestes darstellt, B eine geradkettige oder verzweigtkettige Alkylengruppe mit 2-10 C-Atomen, A eine -CH2-CH2-, cis-CH=CH-, trans-CH=CH- oder -C^C-Gruppe, W eine freie oder funktionell abgewandelte Hydroxymethylengruppe oder eine freie oder funktionell abgewandelte Gruppe (a), wobei die OH-Gruppe α- oder β-ständig sein kann, D und E gemeinsam eine direkte Bindung oder D eine geradkettige oder verzweigte Alkylengruppe mit 1-5 C-Atomen, E ein Sauerstoff- oder Schwefelatom oder eine direkte Bindung, R2 eine Alkyl-, Cycloalkyl-, oder eine gegebenenfalls substituierte Aryl oder eine hetereocyclische Gruppe, R5 eine freie oder funktionell abgewandelte Hydroxygruppe und, falls R3 Wasserstoff darstellt, deren Salze mit physiologisch verträglichen Basen bedeuten. Description
Cyclodextrinclathrate von 5-Cyano-prostacyclinderivaten und ihre Verwendung als Arzneimittel
Beschreibung
Gegenstand der vorliegenden Erfindung sind Cyclodextrinclathrate von 5-Cyano- prostacyclinanaloga und diese enthaltende Mittel.
5-Cyano-prostacyclinaπaloga sind pharmakologisch und medizinisch wertvolle Wirkstoffe, deren Herstellung und Anwendung beispielsweise in DE-OS 2753244 be¬ schrieben werden Diese Substanzen besitzen gegenüber dem natürlichen Prostacyc- lin bei ähnlichem Wirkungsspektrum eine wesentlich verbesserte Spezifität und vor allem wesentlich längere Wirkung.
Die in der erwähnten Offenlegungsschrift beschriebenen 5-Cyano-prostacyclinana- loga liegen oft nicht kristallin vor, wodurch ihrer pharmazeutischen Anwendung Grenzen gesetzt sind. Hinzu kommt noch eine begrenzte Wasserlöslichkeit und Lösungsgeschwindigkeit.
Es wurde nun gefunden, daß Einschlußverbindungen dieser 5-Cyano-prostacyclin- analoga mit Cyclodextrinen die genannten Nachteile nicht besitzen, d.h. ihre Wasserlöslichkeit wird verbessert, die Lösungsgeschwindigkeit wird gesteigert, und die Einschlußverbindungen liegen in kristalliner Form vor. Außerdem wird ihre Stabilität beispielsweise gegenüber Wärme, Licht und Sauerstoff erhöht und ihre galenische Zubereitung (Herstellen von Lösungen oder Tabletten) er¬ leichtert.

Als Alkylgruppe R sind gerade oder verzweigte Alkylgruppen mit 1-10 C-Atomen zu betrachten, wie beispielsweise Methyl, Äthyl, Propyl, Butyl, Isobutyl, tert.-Butyl, Pentyl, Neopentyl. Heptyl, Hexyl, Decyl.
Die Alkylgruppen R können gegebenenfalls 1 bis mehrfach substituiert sein durch Halogeπatome, Alkoxygruppen, gegebenenfalls substituierte Arylgruppen, Dialkylamine und Trialkyla monium. Bevorzugt sind solche Alkylgruppen, die einfach substituiert sind.
Als Substituenten seien bespielsweise genannt Fluor-, Chlor- oder Bromatome, Phenyl, Dimethylamino, Diäthylamino, Methoxy, Äthoxy.
Als bevorzugte Alkylgruppen R sind solche mit 1-4 C-Atomen, wie z. B. Methyl, Äthyl, Propyl, Dimethylaminopropyl, Isobutyl, Butyl zu nennen.
Als Arylgruppen R kommen sowohl substituierte wie auch unsubstituierte Aryl- gruppen in Betracht, wie beispielweise Phenyl, 1-Naphthyl und 2-Naphthyl, die jeweils substituiert sein können durch 1-3 Halogenatome, eine Phenylgruppe, 1-3 Alkylgruppen mit jeweils 1-4 C-Atomen, eine Chlormethyl-, Fluormethyl-, Trifluormethyl-, Carboxyl-, Hydroxy- oder Alkoxygruppe mit 1-4 C-Atomen. Bevor¬ zugt sind die Substituenten in 3- und 4-Stellung am Phenylring, zum Beispiel durch Fluor, Chlor, Alkoxy oder Trifluormethyl oder in 4-Stelluπg durch Hy¬ droxy.
Die Cycloalkylgruppe R kann im Ring 4-10, vorzugsweise 5 und 6 Kohlenstoff¬ atome enthalten. Die Ringe können durch Alkylgruppen mit 1-4 Kohlenstoffatomen substituiert sein. Beispielsweise seien genannt Cyclopentyl-, Cyclohexyl, Me- thylcyclohexyl und Adamantyl.
Als heterocyclische Gruppen kommen 5- und 6-gliedrige Heterocyclen in Frage, die wenigstens 1-Heteroatom, vorzugsweise Stickstoff, Sauerstoff oder Schwefel enthalten. Beispielsweise seien genannt 2-Furyl, 2-Thienyl, 2-Pyridyl, 3-Pyri- dyl, 4-Pyridyl u.a.
Als Säurerest R, kommen physiologisch verträgliche Säurereste in Frage. Bevor- zugte Säuren sind organische Carbonsäuren und Sulforisäureπ mit 1-15 Kohlen- stoffatomen, die der aliphatischen, cyclo-aliphatischen, aromatischen, aroma- tisch-aliphatischen und heterocyclischen Reihe angehören. Diese Säuren können gesättigt, ungesättigt und/oder mehrbasisch und/oder in üblicher Weise substi¬ tuiert sein. Als Beispiele für die Substituenten seien Alkyl-, Hydroxy-, Alk¬ oxy-, Oxo- oder Aminogruppen oder Halogenatome erwähnt.
Beispielsweise seien folgende Carbonsäuren genannt: Ameisensäure, Essigsäure, Propionsäure, Buttersäure, Isobuttersäure, Valeriansäure, Isovaleriansäure,
Capronsäure, Önanthsäure, Caprylsäure, Pelargonsäure, Caprinsäure, Undecyl- säure, Saurinsäure, Tridecylsäure, Myristinsäure, Pentadecylsäure, Trimethyl- essigsäure, Cyclopentylessigsäure, Cyclohexylessigsäure, Cyclopropancarbonsäu- re, Cyclohexancarbonsäure, Phenylessigsäure, Phenoxyessigsäure, Methoxyessig- säure, Äthoxyessigsäure, Mono-, Di- und Trichloressigsäure, Amiπoessigsäure, Diäthyla inoessigsäure, Piperidinoessigsäure, Morpholinoessigsäure, Milchsäure, Bernsteinsäure, Adipinsäure, Beπzoesäure, mit Halogen-, Trifluormethyl-, Hy¬ droxy-, Alkoxy- oder Carboxy-Gruppen substituierte Benzoesäuren, Nikotinsäuren, Isonikotinsäure, Furan-2-carbonsäure, Cyclopentylpropionsäure. Als besonders bevorzugte Acylreste werden solche mit bis zu 10 Kohlenstoffatomen betrachtet.
Als Sulfonsäuren kommen beispielsweise Methansul onsäure, Äthansulfonsäure, Isopropansulfonsäure, ß-Chloräthansulfonsäure, Butansulfonsäure, Cyclopentan- sulfonsäure, Cyclohexansulfonsäure, Benzolsulfonsäure, p-Toluolsulfonsäure, p-Chlorbenzolsulfonsäure, N.N-Dimethylaminosulfonsäure, N,N-Diäthylaminosulfon¬ säure, N,N-Bis-(ß-chloräthyl)-aminosulfonsäure, Pyrrolidino-, Piperidino-, Pi- perazino-, N-Methylpiperazino- und Morpholinosulfonsäure in Frage.
Die Hydroxygruppen R_ und in W können funktionell abgewandelt sein, beispiels-
5 weise durch Verätherung oder Veresterung, wobei die freien oder abgewandelten Hydroygruppen in W α- oder ß-ständig sein können, wobei freie Hydroxygruppen bevorzugt sind.
Als Äther-und Acylreste kommen die dem Fachmann bekannten Reste in Betracht. Bevorzugt sind leicht abspaltbare Ätherreste wie beispielsweise der Tetrahydro- pyranyl-, Tetrahydrofuranyl-, α-Äthoκyäthyl-, Trimethylsilyl-, Dimethyl-tert.- butyl-silyl- und Tri-p-benzyl-silylrest. Als Acylreste kommen die gleichen wie für R genannt in Frage; namentlich genannt seien beispielsweise Acetyl, Pro- pionyl, Butyryl, Beπzoyl.
Als Alkylgruppe R kommen gerad- und verzweigtkettige, gesättigte und unge¬ sättigte Alkylreste, vorzugsweise gesättigte, mit 1-10, insbesondere 1-6 C- Atomen, in Frage, die gegebenenfalls durch gegebenenfalls substituierte Aryl substituiert sein können. Beispielsweise genannt seien Methyl-, Äthyl-, Pro-, pyl-, Butyl-, Isobutyl-, tert.-Butyl-, Pentyl-, Hexyl-, Heptyl-, Octyl-, Bute- nyl-, Isobutenyl-, Propenyl-, Pentenyl-, Beπzyl- und p-Chlorbenzyl.
Die Cycloalkylgruppe kann im Ring 4-10, vorzugsweise 5 und 6 Kohlenstoff¬ atome enthalten. Die Ringe können durch Alkylgruppen mit 1-4 Kohlenstoffato en substituiert sein. Beispielsweise seien genannt Cyclopentyl-, Cyclohexyl, Methyl-cyclohexyl und Adamantyl.
Als substituierte bzw. unsubstituierte Arylgruppen R kommen beispielsweise in Betracht: Phenyl, 1-Naphthyl und 2-Naphthyl, die jeweils substituiert sein können durch 1-3 Halogenatome, eine Phenylgruppe, 1-3 Alkylgruppen mit jeweils 1-4 C-Atomen, eine Chlormethyl- , Fluormethyl-, Trifluormethyl-, Carboxy-, Al- koxy- oder Hydroxygruppe. Bevorzugt ist die Substitution in 3- und 4-Stellung am Phenylring zum Beispiel durch Fluor, Chlor, Alkoxy oder Trifluormethyl oder in 4-Stellung durch Hydroxy.
Als heterocyclische Gruppen R kommen 5- und 6-gliedrige Heterocyclen in Frage, die wenigstens 1-Heteroatom, vorzugsweise Stickstoff, Sauerstoff oder Schwefel enthalten. Beispielsweise seien genannt 2-Furyl, 2-Thienyl, 2-Pyridyl, 3-Pyri- dyl-, 4-Pyridyl u.a.
Als Alkylengruppe B kommen geradkettige oder verzweigtkettige, gesättigte und ungesättigte Alkylenreste, vorzugsweise gesättigte mit 2-10, insbesondere 2-5 C-Atomen, in Frage. Beispielsweise seien genannte äthylen, 1 , 2-propylen, äthyl- äthylen, trimethylen, tetramethylen, pentamethylen u.a.
Als Alkylengruppe D kommen geradkettige mit 1-5 oder verzweigtkettige ge¬ sättigte Alkylenreste mit 2-5 C-Atomen in Frage. Als Beispiele kommen die für B genannten Reste in Betracht.
Zur Herstellung der erfindungsgemäßen Clathrate werden die Verbindungen der allgemeinen Formel I in einem phar akologisch unbedenklichen Lösungsmittel, z.B. einem Alkohol, vorzugsweise Ethanol, einem Keton, z.B. Aceton oder einem Ether, z.B. Diethylether, aufgelöst und mit wäßrigen Lösungen von α-, ß- oder f-Cyclodextrin, vorzugsweise ß-Cyclodextrin, bei 20-80°C vermischt, oder es werden die Säuren in Form der wäßrigen Lösungen ihrer Salze, z.B. der Natrium¬ oder Kaliumsalze mit dem Cyclodextrin versetzt und nach Lösung mit der äquiva¬ lenten Menge einer Säure, z.B. Salzsäure oder Schwefelsäure, versetzt.
Hierbei oder nach dem Abkühlen kristallisieren die entsprechenden Clathrate aus. Man kann aber auch ölige bzw. kristalline 5-Cyano-prostacycline der For¬ mel I durch längeres Rühren bei Raumtemperatur mit einer wäßrigen Lösung von Cyclodextrinen in entsprechende Kristallisate überführen. Die Clathrate können durch Absaugen und Trocknen als feste, frei fließende Kristalle isoliert wer¬ den.
Durch Wahl der geeigneten Cyclodextrin- und Wassermengen lassen sich die Cla¬ thrate in stöchiometrischer Zusammensetzung mit einem reproduzierbaren Wirk¬ stoffgehalt erhalten.
Die entsprechend dieser Erfindung hergestellten Clathrate sind wertvolle Pharmaka.
Die Clathrate können in ihrer wasserfreien, hygroskopischen Form oder in einer wasserhaltigen, wenig hygroskopischen Form Verwendung finden.
Die neuen Clathrate der 5-Cyano-prostacycline nach Formel I liegen im stöchio- methrischen Verhältnis von Carbacyclin : Cyclodextrin vor oder besitzen einen höheren Cyclodextrin-Anteil.
Beispielsweise bildet 5-Cyano-16-methyl-prostacyclin mit ß-Cyclodextrin einen Komplex der Zusammensetzung 1:2,5 (12,12 l des Prostacyclins) oder beliebige niedrigere Werte.
Die entsprechend dieser Erfindung hergestellten Clathrate sind wertvolle Pharmaka.
Die Clathrate dieser Erfindung wirken blutdrucksenkend und bronchodilatorisch. Sie sind weiterhin geeignet zur Hemmung der Thrombozyten-Aggregation. Sie wir¬ ken cytoprotektiv am Magen, Darm, Herz, an der Leber, Niere und am Pankreas. Folglich stellen die neuen Cyclodextrin-Clathrate der Formel I wertvolle phar¬ mazeutische Wirkstoffe dar. Darüber hinaus weisen sie bei ähnlichem Wirkungs¬ spektrum, verglichen mit entsprechenden Prostaglandinen, eine höhere Spezifit t und vor allem eine wesentlich längere Wirksamkeit auf.
Im Vergleich zu PGI zeichnen sie sich durch größere Stabilität aus. Die hohe Gewebsspezifität der neuen Prostaglandiπe zeigt sich bei der Untersuchung an glattmuskulären Organen, wie zum Beispiel am Meerschweinchenileum oder an der isolierten Kaninchentrachea, wo eine wesentlich geringere Stimulation zu beob¬ achten ist als bei der Applikation natürlicher Prostaglandiπe vom E-, A-oder F-Typ.
Die neuen 5-Cyano-prostacyclin-Clathrate besitzen die für Prostacycline ty¬ pischen Eigenschaften, wie zum Beispiel Senkung der peripheren arteriellen und koronaren vaskulären Widerstandes, Inhibierung der Thrombozytenaggregation und Auflösung von Plättchenthromben, myocardiale Zytoprotektion und damit Senkung des systemischen Blutdruckes ohne zugleich Schlagvolumen und koronare Durch¬ blutung zu senken; Behandlung von Schlaganfall, Prophylaxe und Therapie korona- rer Herzerkrankuπgen, koronarer Thrombose, des Herzinfarktes, peripherer Arte¬ rienerkrankungen, Arteriosklerose und Thrombose, Prophylaxe und Therapie ischa- e ischer Attacken des ZNS-Systems, Therapie des Schocks, Inhibierung der Bronchokonstriktion, Inhibierung der Magensäuresekretion, Zytoprotektion der Magen- und Darmschleimhaut, Zytoprotektionen in der Leber, Niere, Herz und im Pankreas, auch bei Organtransplantationen, antiallergische Eigenschaften Sen¬ kung des pulmonalen vaskulären Widerstandes und des pulmonalen Blutdruckes, Förderung der Nierendurchblutung, Anwendung anstelle von Heparin oder als Adju- vans bei der Dialyse der Hämofiltration, Konservierung von Blutplasmakonserven, besonders von Blutplättchenkonserven, Inhibierung von Geburtswehen. Behandlung von Schwangerschaftstoxikose. Erhöhung der zerebralen Durchblutung etc. Außer¬ dem besitzen die neuen Carbacyclinclathrate antiproliferative und antidiarrhoe- gene Eigenschaften. Die Carbacycline dieser Erfindung können auch in Kombina¬ tion, zum Beispiel mit ß-Blockern oder Diuretika, verwendet werden.
Die 5-Cyano-prostacyclin-clathrate dieser Erfindung können auch in Kombination, zum Beispiel mit ß-Blockern, Diuretika, Phosphondiesterasehemmern, Calciumanta- gonisten, nichtsteroidalen Entzündungshemmer, Leukotriensynthesehemmer, Leuko- trienantagonisten, Thromboxansynthesehemmern oder Thromboxanantagonisten ver¬ wendet werden.
Die erfindungsgemäßen Clathrate können in flüssigen oder festen galenischen Formulierungen verwendet werden, wobei die Formulierungen enteral, parenteral, vaginal oder rektal verabreicht werden können oder sie können auch in chirur¬ gisches Nahtmaterial und in Kunststoffe eingebaut werden.
Zur Herstellung von Tabletten wird das Prostaglandin-Cyclodextrin-Clathrat mit Trägersubstanzen und Hilfsstoffen wie Laktoxe. Maisstärke. Polyvinylpyrrolidon und Magnesiumstearat vermischt.
Zur Herstellung von Lösungen für die enterale und parenterale Anwendung werden die wäßrigen Cyclodextrin-Clathrat-Lösungen zusammen mit Laktose lyophilisiert. Anschließend können die Lyophilisate mit physiologischer Kochsalzlösung auf die gewünschte Konzentration gebracht werden.
Die Erfindung umfaßt daher pharmazeutische Präparate und Formulierungen, die als Wirkstoff ein Cyclodextrin-Clathrat eines Carbacyclinanalogen enthalten.
Die Erfindung wird durch folgende Beispiele erläutert:
Beispiel 1
Man rührt 490 mg 5-Cyano-16-methyl-prostacyclin (Nileprost) mit 14,16 g ß-Cyc- lodextrin in 100 ml Wasser 5 Tage bei 24°C. Der Feststoff wird abgesaugt und 8 Stunden bei 0,1 Torr und 24°C getrocknet. Man erhält 10,7 g frei fließende Kristalle des ß-Cyclodextrin-Clathrats des 5-Cyano-16-methyl-prostacyclin.
Der Gehalt an Prostacyclinanolgon im Clathrat wurde durch Nachdruckflüssig¬ keitschromatographie bestimmt und betrug 4 Z.
Beispiel 2
Man löst 1,038 g 5-Cyano-16-methyl-prostacyclin in 34,5 ml 0,1 n-Natriumhyd- roxid-Lösung und verdünnt mit 319 ml Wasser. Dann gibt man 12,49 g ß-Cyclodex- trin hinzu und erwärmt auf 35°C bis zur Lösung. Man kühlt auf 30°C ab und ver¬ setzt dabei langsam mit 34,5 ml 0,1 n-Salzsäure. Man rührt 18 Stunden bei 22°C und 7 Stunden unter Eiskühlung, saugt ab und wäscht den Feststoff mit Wasser chloridfrei. Man trocknet im Vakuum, bis das Kristallisat bei Umgebungstempera¬ tur und -luftfeuchtigkeit wieder Wasser aufnimmt und läßt das Gewicht konstant werden.
Man erhält 8,5 g des ß-Cyclodextrin-Komplexes von 5-Cyano-16-methyl-prostcycliπ mit der stöchiometrischen Zusammensetzung 1:2,5 als Hydrat in Form eines frei fließenden Pulvers, der Wirkstoffgehalt im Komplex beträgt nach Titration 12,2 1 (berechnet auf wasserfreie Substanz), der Wassergehalt 7,1 l .
Beispiel 3
Man löst 943 mg ß-Cyclodextrin bei 22°C in 65 ml Wasser und tropft langsam eine Lösung von 78,3 mg 5-Cyano-16-methyl-prostacyclin in 0,4 ml Ethanol dazu. Man rührt 18 Stunden bei 22°C und 5 Stunden im Eisbad. Man saugt das Kristallisat ab und wäscht mit eiskaltem Wasser, Aceton und wieder mit Wasser. Man trocknet im Vakuum über Phosphorpentoxid und erhält 535 ml eines freifließenden, hygros¬ kopischen Kristallisats , das einen durch Titration ermittelten Wirkstoffgehalt von 12,2 l aufweist.

1 ) Cyclodextrin-Clathrate von 5-Cyano-prostacyclin-analoga der allgemeinen Formel I
worin
R den Rest 0R_, wobei R Wasserstoff, Alkyl, Cycloalkyl, Aryl oder einen hetereocyclischen Rest bedeuten kann, oder den Rest NHR, mit R, in der
Bedeutung eines Säurerestes darstellt.
B eine geradkettige oder verzweigtkettige Alkylengruppe mit 2-10 C-Atomen,
eine -CH -CH2~, cis-CH=CH-, trans-CH=CH- oder -C≡C-Gruppe,
W eine freie oder funktionell abgewandelte Hydroxymethylengruppe oder eine
CH, l 3 freie oder funktionell abgewandelte -C-Gruppe, wwobei die OH-Gruppe α- oder ß-ständig sein kann, OH
-
D und E gemeinsam eine direkte Bindung oder
D eine geradkettige oder verzweigte Alkylengruppe mit 1-5 C-Atomen,
E ein Sauerstoff- oder Schwefelatom oder eine direkte Bindung.
R„ eine Alkyl-, Cycloalkyl-, oder eine gegebenenfalls substituierte Aryl oder eine hetereocyclische Gruppe,
R5 eine freie oder funktionell abgewandelte Hydroxygruppe und, falls R_ Wasser¬ stoff darstellt, deren Salze mit physiologisch verträglichen Basen bedeuten.
2) Arzneimittel, enthaltend eine Verbindung aus Anspruch 1 und übliche Hilfs- und Trägerstoffe.
