Description
- Die Erfindung bezieht sich auf eine Wasch- bzw. Spülvorrichtung für Titerplatten od. dgl. mit einer Mehrzahl von Kavitäten zur Aufnahme eines zu untersuchenden bzw. zu bestimmenden Mediums, beispielsweise Antigene, mit Zuführeinrichtungen mit Düsen zum Einbringen einer Spülflüssigkeit in jede der Kavitäten und mit Absaug-und/oder Ausblaseinrichtungen, um Flüssigkeit aus den Kavitäten zu entleeren und mit Führungseinrichtungen, die die Titerplatte zur Waschstellle bewegen und dort lagegenau halten. Eine derartige Vorrichtung gehört zum Stand der Technik.
- In der medizinischen Labortechnik wird beispielsweise zur Messung von Antigenen und/oder Antikörpern das Elisa Verfahren angewendet (Enzym linked immunosorbent assay), bzw. die verschiedenen Verfahren dieses Systems. Es werden beispielsweise zur Messung von Antigenen in einem ersten Verfahrensschritt Antikörper an der Titerplatte absorbiert, anschliessend wird die Titerplatte gewaschen, dann erfolgt in einem weiteren Verfahrensschritt die Beimengung einer Restlösung, welche das Antigen enthält, darauf erfolgt wiederum eine Waschung, worauf enzymmarkierte spezifische Antikörper auf die Titerplatte gegeben werden, dann erfolgt wieder ein Waschen der Titerplatte, worauf die Beimengung des Enzymsubstrates erfolgt.
- Die eigentliche Messung erfolgt in einem Photometer.
- Aus dem zuvor beschriebenen Beispiel lässt sich erkennen, dass das Waschen der Titerplatten ein wesentlicher Bestandteil des Verfahrens ist, der einerseits relativ zeitaufwendig ist und andererseits noch den Nachteil mit sich bringt, dass sehr oft mit hochgiftiger, d.h. in höchstem Masse krankheitserregender, lebender Materie umgegangen werden muss, was die händische Handhabung der Titerplatte nicht wünschenswert macht.
- Das wesentliche Problem, das beim Waschen der Titerplatten entsteht, ist jedoch das folgende:
- Die derzeit im Gebrauch befindlichen Titerplatten weisen 96 Kavitäten auf, von denen jede praktisch einer Küvette entspricht. Während des Verfahrens bzw. während der Feststellung bzw. Bestimmung der Antigene oder der Antikörper befinden sich Proben in jeder Kavität (beispielsweise Proben von verschiedenen Patienten). Es muss nun absolut verhindert werden, dass während der Handhabung der Platte, und dies ist insbesondere beim Waschen schwierig zu verhindern, Teile der Proben von einer Kavität in die andere gelangen.
- Die FR-A-2358206 zeigt ein Verfahren und eine Maschine zum Waschen von Mikrotestplatten, wobei ein Absaugkopf und ein Wascherverteiler vorgesehen ist, unter die jede Vertiefungsreihe der Titerplatten nacheinander eingestellt wird. Weiters ist ein Schlitten vorgesehen, der die Titerplatte transportiert.
- Die FR-A-2277630 beschreibt eine Vorrichtung zum Absaugen einer Serumflüssigkeit auf einer Mikrotiterplatte und zur anschliessenden Waschung der Mikrotiterplatte. Hierbei sind kombinierte Ansaug- und Ausströmdüsen vorgesehen, die einen inneren Düsengang aufweisen, der zum Ansaugen der Serumflüssigkeit dient und einen äusseren Düsenganq, der bei der Titerplatte in einen Ringspalt mündet, durch den die Waschflüssigkeit ausströmt. Es sind keine Vorkehrungen getroffen, um während des Waschvorganges die Kavitäten der Titerplatte in bezug aufeinander abzudichten.
- Eine Vorrichtung und ein Verfahren zur automatischen Analyse von Blut oder anderen Flüssigkeitsproben ist in der US-A-3533744 beschrieben.
- Aufgabe der Erfindung ist es daher, eine Wasch- bzw. Spülvorrichtung der eingangs erwähnten Art zu schaffen, mit der die Titerplatten funktionssicher gewaschen werden und bei der verhindert wird, dass Probenteile von einer Kavität zur anderen gelangen bzw. wobei überhaupt jede Verschmutzung der Proben vermieden wird.
- Dies wird erfindungsgemäss erreicht durch eine mit Düsen versehene Waschplatte, an die die zu spülende Titerplatte mittels einer Führungseinrichtung abdichtend bringbar und an der die Titerplatte während des Wasch- bzw. Spülvorganges mit Duck gehalten ist, wobei jeder Kavität der Titerplatte eine Düse und ein die Düse umgebender Ringspalt, der die Kavität mit einem Abflusskanal verbindet, zugeordnet sind und jede einzelne Kavität gegenüber den anderen Kavitäten abgedichtet ist.
- Es soll hier noch einmal erwähnt werden, dass das eingangs beschriebene Verfahren nur ein Beispiel des Anwendungsgebietes des Automaten beschreiben soll. Ähnliche Verfahren bzw. ähnliche Bestimmungen werden auch in der Landwirtschaft und in der Biologie und Biochemie angewandt. Des weiteren soll auch der Begriff «Titerplatte» nicht einschränkend verstanden werden. Das wesentliche ist, dass eine Mehrzahl von nebeneinander befindlichen Kavitäten gleichzeitig gewaschen werden soll. Es könnte sich hier durchaus um eine Reihe handeln, oder es könnten auch einzelne Küvetten in einen Trägerrahmen befestigt und so dem Wasch- bzw. dem Spülautomaten zugeführt werden.
- In der Grundversion wird für den erfindungsgemässen Spülautomaten als Wasch- bzw. Spülflüssigkeit Leitungswasser verwendet. Als Trocknungsmedium dient Druckluft.
- Es kann jedoch beim erfindungsgemässen Automaten vorgesehen sein, dass andere Spülflüssigkeiten, beispielsweise destilliertes Wasser oder eine Pufferlösung, verwendet werden. Ebenso kann statt Druckluft ein anderes gasförmiges Medium, beispielsweise Sauerstoff verwendet werden.
- Vorzugsweise ist vorgesehen, dass das gasförmige Medium pulsierend durch die Düsen geführt wird, wodurch der Reinigungs- bzw. Trocknungseffekt wesentlich verbessert wird.
- Ein Ausführungsbeispiel der Erfindung sieht vor, dass sich die Waschplatte in einer abgedichteten Waschkammer befindet. In der Waschkammer kann ein Fühler angeordnet sein, der die Abdichtung zwischen Waschplatte und den zu reinigenden Titerplatten prüft und eventuell nach erkannter Undichtheit einen vorprogrammierten Selbstreinigungsvorgang für die Waschplatte auslöst, damit es zu keiner Verschmutzung der nachfolgenden Titerplatten kommt.
- Eine derartige Selbstreinigung kann jedoch auch profilaktisch nach Durchlauf einer vorbestimmten Anzahl von zu reinigenden Titerplatten erfolgen. Dies kann durch der Waschplatte an ihrer Düsenseite gegenüberliegende Reinigungsdüsen erfolgen. Es kann jedoch auch eine Vorrichtung zum Abheben der Waschplatte von der Titerplatte (oder umgekehrt) vorgesehen sein, wobei nach erfolgtem Flüssigkeitsdurchlass durch die Düsen in der Waschplatte ein weiterer Flüssigkeitsdurchlass durch die Düsen bei abgehobener Waschplatte erfolgt. Dadurch, dass sich die Waschplatte und die Titerplatte in geringem Abstand voneinander befinden, weden bei entsprechendem Druck des Spülmediums die Oberflächen der Waschplatte und der Titerplatte gereinigt.
- Vorteilhaft weist der erfindungsgemässe Wasch- bzw. Spülautomat Führungseinrichtungen auf, die die Titerplatten bodenfrei halten, d.h. die Titerplatte wird entlang ihrer Seiten bzw. entlang ihrer Seitenunterkanten geführt, sodass es an der Plattenunterseite zu keiner Verunreinigung kommt. Dies ist für die nachfolgende Messung der Proben von Bedeutung.
- Ein weiteres Ausführungsbeispiel der Erfindung sieht eine Kippvorrichtung für die Titerplatte, vorzugsweise im Waschbereich, vor. Dabei kann die Titerplatte gemeinsam mit der Waschplatte kippbar sein. Der Vorteil dieser Ausführung ist, dass die Spülflüssigkeit nach unten abrinnen kann und unter Umständen kein eigener Trocknungsvorgang mittels Druckluft notwendig ist.
- Nachstehend wird die Erfindung anhand der Figuren der beiliegenden Zeichnungen eingehend beschrieben, doch soll die Erfindung nicht auf die gezeigten Ausführungsbeispiele eingeschränkt sein, da verschiedene Ausführungsvarianten denkbar und möglich sind, ohne dass dadurch der Erfindungsgedanke verlassen wird.
- Die Fig. 1 zeigt schematisch ein Schaubild einer zu reinigenden Titerplatte, die Fig.2 zeigt ebenfalls schematisch den erfindungsgemässen Wasch- bzw. Spülautomat, die Fig.3 zeigt eine Draufsicht auf die erfindungsgemässe Waschplatte, die Fig.3a zeigt einen Schnitt nach der Linie I-I der Fig. 3 mit angelegter Titerplatte, die Fig. 4 bis 6 zeigen Ausschnitte durch die Waschplatte mit verschiedenen Düsenformen, die Fig. 7 zeigt schematisch einen Schnitt durch eine Waschplatte, die Fig.8 zeigt einen Querschnitt durch ein weiteres Ausführungsbeispiel einer Waschplatte nach der Linie I-I der Fig. 3, und die Fig. zeigt einen gleichen Schnitt bei einem weiteren Ausführungsbeispiel einer Waschplatte mit angelenkter Titerplatte.
- In der Fig. 1 ist eine Titerplatte 1 mit Kavitäten 2 gezeigt. Bei den heute gebräuchlichen Titerplatten 1 sind im allgemeinen acht Reihen mit jeweils zwölf Kavitäten 2 vorgesehen, was eine Gesamtsumme von sechsundneunzig ergibt.
- Der erfindungsgemässe Wasch- bzw. Spülautomat weist im Ausführungsbeispiel eine Abstapelvorrichtung 3, eine Waschstelle 6 und eine Aufstapelvorrichtung 4 auf.
- Die zu waschenden bzw. zu spülenden Titerplatten 1 werden beispielsweise in einem Stapel von zehn Stück in die Abstapelvorrichtung 3 eingelegt, einzeln von einer Fördereinrichtung, die Zahmriemen 7 aufweist, in der Richtung des Pfeiles V zur Waschstelle unterhalb der Waschplatte 5 gebracht und kommen nach erfolgter Spülung in die Aufstapelvorrichtung 4.
- Unterhalb der Waschplatte 5 kommen die Titerplatten 1 auf einer Tragplatte 8 zu liegen, die eine Zentriereinrichtung 9, beispielsweise Zentrierbolzen, für die genaue Positionierung der Titerplatte 1 aufweist.
- Befindet sich die Titerplatte 1 in ihrer gewünschten Position, wird die Tragplatte 8 mittels der Hebevorrichtung 10, die beispielsweise ein Pneumatikzylinder ist, angehoben und die Titerplatte 1 an die Waschplatte 5 gepresst.
- Wie insbesondere aus den Fig. 3 und 3a ersichtlich, weist die Waschplatte 5 eine Vielzahl von Düsen 11 auf , deren Anzahl und Position genau der Anzahl und Position der Kavitäten 2 in der Titerplatte 1 entspricht. Im Ausführungsbeispiel weist also die Waschplatte 5 sechsundneunzig Düsen 11 auf. Bei an die Waschplatte 5 angepresster Titerplatte 1 ist, wie aus der Fig. 3a ersichtlich, jeder Kavität 2 eine Düse 11 zugeordnet.
- Um die Düsen 11 befinden sich Ringspalte 12, die den Durchfluss des Spülmediums bzw. auch den Durchtritt der Trocknungs- bzw. Druckluft in die Abflusskanäle 13 gestatten. Die Abflusskanäle 13 münden in den Auslass A der Waschplatte 5.
- Ist nun die Titerplatte 1 angehoben und an die Waschplatte 5 gepresst, wird in einem ersten Arbeitsgang Spülmedium, bespielsweise Wasser, durch einen oder mehrere Einlässe E in die Waschplatte 5 gepresst und gelangt durch die Düsen 11 unter Druck von bis zu 4 Bar in die Kavitäten 2 der Titerplatte 1. Es kommt zur Wirbelbildung und das Spülmedium gelangt jeweils durch den Ringspalt 12 in den Abflusskanal 13.
- Um diese Entleerung zu verbessern bzw. um einen gewissen Absaugeffekt zu erreichen, sind die Düsenköpfe 11' der Düsen 11 so ausgebildet, dass sie von vorne nach hinten abnehmenden Querschnitt aufweisen.
- In den Fig.4 bis 6 sind verschiedene Ausführungsbeispiele der Düsenköpfe 11' gezeigt.
- Nach erfolgter Spülung wird die Flüssigkeitsmedium-Zufuhr abgeschaltet und es wird durch dieselben Einlässe E und durch dieselben Düsen 11 Druckluft in die Kavitäten 2 eingepresst.
- Die Feuchtigkeit und die Temperatur der Druckluft ist einstell- bzw. regelbar. Durch die Druckluft wird der Rest der Spülflüssigkeit durch die Ringspalte 12 in die Abflusskanäle 13 gepresst und dem Auslass A zugeführt.
- Nun kann die Titerplatte wieder mittels der Tragplatte 8 abgesenkt werden und mittels der Zahnriemen 7 der Aufstapelvorrichtung 4 zugeführt werden.
- Es ist jedoch auch möglich, dass die Titerplatte 1 nur leicht von der Waschplatte 5 abgesenkt wird, sodass nur ein kleiner Spalt zwischen der Deckfläche 1' der Titerplatte 1 und der Dichtplatte 14 der Waschplatte 5 entsteht. Wird nun noch einmal Spülmedium durch die Düsen 11 gepresst, kommt es auch zu einer Reinigung der Dichtplatte 14 der Titerplatte 1 und zur Reinigung der Deckfläche der Waschplatte 5. Eine derartige Nachspülung erfolgt zweckmässigerweise vor dem Trocknungszyklus, d.h. vor dem oben beschriebenen Zuführen der Druckluft.
- Die Titerplatte 1 kann nun mittels der Tragplatte 8 wieder an die Waschplatte 5 angepresst werden, worauf nun die Zufuhr der Druckluft erfolgt und die Titerplatte 1 getrocknet wird. Im Ausführungsbeispiel ist an der Waschplatte 5 eine Dichtplatte 14 aus Kunststoffmaterial vorgesehen.
- Bei dem Ausführungsbeispiel der Waschplatte 5 nach der Fig.9 ist die Waschplatte 5 in einen feststehenden Oberteil 5a, der die Düsen 11 trägt, und einen beweglichen Unterteil 5b geteilt. Im Oberteil 5a befinden sich auch die Vorkammer 16 oder die Zuführleitungen 15. Der bewegliche Unterteil 5b hat zu den Düsen 11 konzentrische Bohrungen, die mit den Düsen 11 den Ringspalt 12 bilden. Bei an den Oberteil angelegtem Unterteil ragen die Düsen 11, wie in der Figur der Zeichnung gezeigt, durch die Bohrungen des Unterteiles 5b. Im Unterteil 5b befinden sich weiters die Abflusskanäle 13, die Dichtplatte 14 zwischen der Titerplatte 1 und der Waschplatte 5, und zwischen den beiden Teilen 5a und 5b befindet sich eine weitere Dichtplatte 14'.
- Die Düsen 11 sind gegenüber den voran beschriebenen Ausführungsbeispielen verlängert und bei Beginn des Waschvorganges bzw. unmittelbar davor ist der Unterteil 5b bis zum unteren Ende der Düsen 11 abgesenkt. Beim Hochdrücken dichtet zunächst die Titerplatte 1 mit dem Unterteil 5b der Waschplatte 5 ab. Dann wird der Unterteil 5b zusammen mit der Titerplatte 1 an den Oberteil 5a herangeführt. Dabei dringen die Düsen 11 in die Kavitäten 2 der Titerplatte 1 ein und verdrängen die zu entleerende Flüssigkeit teilweise in Richtung der Abflusskanäle 13, bis der Unterteil 5b dicht am Oberteil 5a anliegt. Damit ist das System geschlossen und der Spülvorgang kann beginnen.
- Die Vorteile dieses Ausführungsbeispieles sind folgende:
- Die konzentrischen Bohrungen für jeden Ringspalt 12 sind volumenmässig so ausgelegt, dass die durch das Eintauchen der Düse 11 verdrängte Flüssigkeit in den konzentrischen Bohrungen (Ringspalt 12) aufgenommen wird und dadurch Kontamination vermieden wird. Durch die vorzugsweise zylindrisch ausgelegte Düse 11 und deren Eintauchen in die Kavität 2 wird das zu spülende bzw. zu trocknende Volumen der Kavität 2 verringert. Die Düsen 11 tauchen vorzugsweise zu zwei Dritteln in die Kavitäten 2 ein. Die dadurch entstehende höhere Strömungsgeschwindigkeit erhöht die Reinigungs- bzw. Trocknungseffizienz.
- Um insbesondere beim Spülmedium einen gleichmässigen Druck an den Düsen zu erzielen, kann die Waschplatte 5 entweder, wie in der Fig. 7 gezeigt, mit H-förmigen Zuführkanälen 15 versehen sein, wobei der Einlass E jeweils in der Mitte des Verbindungssteges angeordnet ist, oder es kann, wie in der Fig.8 gezeigt, die Waschplatte 5 eine oder mehrere Vorkammern 16 mit einer schrägen Wand 16' aufweisen, sodass der Kammerquerschnitt vom Einlass E für das Spülmedium gesehen, von Düsenreihe zu Düsenreihe abnimmt.
- Bei dem erfindungsgemässen Wasch- und Spülautomaten befinden sich die Luft-, Wasser- und Stromanschlüsse vorteilhaft an der Rückseite des Gerätes, weiters sind eingebaute Druckreduzierventile und Manometer zur Druckkontrolle für Spülmedium und Druckluft vorgesehen. Der Antrieb des Zahnriemens 7 erfolgt vorteilhaft über einen Gleichstrommotor. Die Waschplatte 5 ist vorteilhaft mittels eines Schnellverschlusses austauschbar. Die Steuerung des Gerätes erfolgt über einen Mikroprozessor. Die Zeiten für Wasch-, Verweil- und Trocknungsvorgang sind frei wählbar, ebenso sind der Ablauf und die Wiederholzyklen frei wählbar.
- Kontrollsysteme sind vorgesehen für Kontrolle auf leeren Abstapler, Prüfung auf vorhandenen Platz im Aufstapler, Prüfung auf richtige Druckverhältnisse im Gerät und Prüfung auf Undichtheit im System. Die Meldungen erfolgen durch akustische Signale und/oder entsprechende LED-Anzeige.
- Die Waschung einer Titerplatte nach der anderen erfolgt automatisch. Nach dem Startbefehl wird das eingegebene Spül- und Trockenprogramm so lange wiederholt, bis sich entweder keine Titerplatte 1 in der Abstapelvorrichtung 3 mehr befindet, oder das Programm durch eine Stopptaste beendet wird.
