Claims (5)
1. Modificeret Corynebacterium sp.-mikroorganisme, der er i stand til at producere L-lysin ved anvendelse af xylose, hvor mikroorganismen omfatter Erwinia carotovora-afledt xyloseisomerase (XylA) og xylulokinase (XylB), hvor XylA omfatter aminosyresekvensen SEQ ID NO: 1 eller en aminosyresekvens med 95% eller mere identitet med aminosyresekvensen ifølge SEQ ID NO: 1, og hvor XylB omfatter aminosyresekvensen SEQ ID NO: 2 eller en aminosyresekvens med 90% eller mere identitet med aminosyresekvensen ifølge SEQ ID NO: 2.
2. Mikroorganisme ifølge krav 1, hvor XylA kodes af en polynukleotidsekvens med SEQ ID NO: 3, og XylB kodes afen polynukleotidsekvens med SEQ ID NO: 4.
3. Mikroorganisme ifølge krav 1, hvor mikroorganismen er Corynebacterium glutami-cum.
4. Fremgangsmåde til fremstilling afen mikroorganisme ifølge krav 1, hvor indføringen af XylA- og XylB-aktiviteter udføres ved at indsætte et polynukleotid, der omfatter nu-kleotidsekvenser, der koder for XylA og XylB, i et kromosom, indføre et vektorsystem, der omfatter polynukleotidet, i mikroorganismen opstrøms for de nukleotidsekvenser, der koder for XylA og XylB, indføre XylA og XylB med en modificeret promotor eller indføre de modificerede nukleotidsekvenser, som koder for XylA og XylB.
5. Fremgangsmåde til fremstilling af L-lysin, som omfatter følgende trin: (i) dyrkning af den modificerede Corynebacterium sp.-mikroorganisme, der er i stand til at fremstille L-lysin ved anvendelse af xylose, ifølge et hvilket som helst af kravene 1 til 3 i et dyrkningsmedium, der indeholder xylose som carbonkilde, for at opnå en dyrkningsvæske; og (ii) udvinding af L-lysin fra dyrkningsvæsken.
