Claims (10)
- 配列番号1のアミノ酸配列においてアミノ酸配列の136番目のアミノ酸であるグルタミン(Glutamine)がグルタミン以外の他のアミノ酸に置換された、アセトヒドロキシ酸シンターゼ(acetohydroxy acid synthase、AHAS)活性を有するタンパク質変異体。
- 前記第136番目のアミノ酸が、アスパラギン(Asparagine)、アルギニン(Arginine)、フェニルアラニン(Phenylalanine)、セリン(Serine)、チロシン(Tyrosine)、システイン(Cystenine)、プロリン(Proline)、ヒスチジン(Histidine)、ロイシン(Leucine)、イソロイシン(Isoleucine)、トレオニン(Threonine)、リジン(Lysine)、バリン(Valine)、アラニン(Alanine)、アスパラギン酸(Aspartic acid)、グルタミン酸(Glutamic acid)、グリシン(Glycine)またはトリプトファン(Tryptophan)に置換された、請求項1に記載のアセトヒドロキシ酸シンターゼ(acetohydroxy acid synthase、AHAS)活性を有するタンパク質変異体。
- 前記タンパク質変異体が、配列番号12、配列番号47、配列番号48、配列番号49、配列番号50、配列番号51、配列番号52、配列番号53、配列番号54、配列番号55、配列番号56、配列番号57、配列番号58、配列番号59、配列番号60、配列番号61、配列番号62または配列番号63のアミノ酸配列を含む、請求項1に記載のタンパク質変異体。
- 請求項1~3のいずれか一項に記載のタンパク質変異体をコードするポリヌクレオチド。
- 請求項4に記載のポリヌクレオチドを含むベクター。
- 請求項1~3のいずれか一項に記載のタンパク質変異体を含む微生物。
- 前記微生物が、L-イソロイシンを生産する、請求項6に記載の微生物。
- 前記微生物が、コリネバクテリウム(Corynebacterium sp.)属またはエシェリキア属(Escherichia sp.)である、請求項6に記載の微生物。
- 請求項6に記載の微生物を培地で培養する段階を含む、L-イソロイシンの生産方法。
- 前記培養された微生物または培地からL-イソロイシンを回収する段階をさらに含む、請求項9に記載のL-イソロイシンの生産方法。
