Claims (15)
1. Fremgangsmåde til fremstilling af en herbicidresistent plante, hvilken fremgangsmåde omfatter: (a) at indføre i en plantecelle en genreparation-oligonukleobase (GRON) med en målrettet mutation i et acetohydroxysyresyntase (AHAS)-gen til fremstilling af en plantecelle med: (i) et AHAS-gen, der udtrykker et AHAS-protein omfattende en substitution af asparaginsyre med glutaminsyre ved aminosyrepositionen svarende til position D376 af SEQ ID NO: 1 og (ii) et AHAS-gen, der udtrykker et AHAS-protein omfattende en substitution af serin med asparagin ved aminosyrepositionen svarende til position 653 af SEQ ID NO: 1; (b) at identificere en plantecelle med i det væsentlige normal vækst og katalytisk aktivitet i forhold til en tilsvarende vildtypeplantecelle i nærvær af et AHAS-hæmmende herbicid; og (c) at regenerere en ikke-transgen herbicidresistent plante med et muteret AHAS-gen fra plantecellen.
2. Fremgangsmåde ifølge krav 1, hvor genreparation-oligonukleobasen er en enkeltstrenget oligodeoxynukleotid-mutationsvektor.
3. Fremgangsmåde ifølge krav 1 eller 2, hvor AHAS-proteinet er modstandsdygtigt over for hæmning gennem et AHAS-hæmmende herbicid, især hvor det AHAS-hæmmende herbicid udvælges fra gruppen bestående af herbicider af: imidazolinon, sulfonylurea, triazolopyrimidin, pyrimidinylthiobenzoat, sul-fonylamino-carbonyltriazolinon og blandinger deraf.
4. Fremgangsmåde ifølge et hvilket som helst af de foregående krav, hvor AHAS-genet koder for et protein omfattende 70 % identitet eller derover med en eller flere af aminosyresekvenserne fra figur 2.
5. Fremgangsmåde ifølge et hvilket som helst af de foregående krav, hvor planten er udvalgt blandt plantearter fra gruppen bestående af canola, solsikke, tobak, sukkerroe, bomuld, majs, hvede, byg, ris, hirse, tomat, mango, fersken, æble, pære, jordbær, banan, melon, kartoffel, gulerod, salat, løg, soja arter, sukkerrør, ærter, hestebønner, poppel, vindruer, citrus, lucerne, rug, havre, græstørv og fodergræsser, hør, raps, agurk, ipomoea, balsamin, peber, aubergine, morgenfrue, lotus, kål, bellis, nellike, tulipan, iris, lilje og nøddepro-ducerende planter.
6. Fremgangsmåde ifølge et hvilket som helst af de foregående krav, hvor planten er en Brassica napus-plante.
7. Plante omfattende et acetohydroxysyresyntase (AHAS)-gen, hvor genet koder for et AHAS-protein omfattende en substitution af asparaginsyre med glutaminsyre ved aminosyrepositionen svarende til position D376 af SEQ ID NO: 1, og en substitution af serin med asparagin ved aminosyrepositionen svarende til position 653 af SEQ ID NO: 1, og hvor planten ikke udelukkende opnås ved hjælp af en i det væsentlige biologisk proces.
8. Plante ifølge krav 7, hvor planten er ikke-transgen, og hvor planten ikke udelukkende opnås ved hjælp af en i det væsentlige biologisk proces.
9. Plante ifølge krav 7 eller 8, hvor AHAS-proteinet er modstandsdygtigt over for hæmning gennem et AHAS-hæmmende herbicid.
10. Plante ifølge et hvilket som helst af kravene 7 til 9, hvor AHAS-genet koder for et protein omfattende 70 % identitet eller derover med en eller flere af ami-nosyresekvenserne fra figur 2.
11. Plante ifølge et hvilket som helst af kravene 7 til 10, hvor planten er udvalgt blandt plantearter fra gruppen bestående af canola, solsikke, tobak, sukkerroe, bomuld, majs, hvede, byg, ris, hirse, tomat, mango, fersken, æble, pære, jordbær, banan, melon, kartoffel, gulerod, salat, løg, soja-arter, sukkerrør, ærter, hestebønner, poppel, vindruer, citrus, lucerne, rug, havre, græstørv og fodergræsser, hør, raps, agurk, ipomoea, balsamin, peber, aubergine, morgenfrue, lotus, kål, bellis, nellike, tulipan, iris, lilje og nøddeproducerende planter.
12. Plante ifølge et hvilket som helst af kravene 7 til 11, hvor planten er en Brassica napus-plante.
13. Frø produceret af planten ifølge et hvilket som helst af kravene 7 til 12, hvor frøet omfatter et acetohydroxysyresyntase (AHAS)-gen, der koder for et AHAS-protein omfattende en substitution af asparaginsyre med glutaminsyre ved aminosyrepositionen svarende til position D376 af SEQ ID NO: 1, og en substitution af serin med asparagin ved aminosyrepositionen svarende til position 653 af SEQ ID NO: 1, og hvor frøet ikke udelukkende opnås ved hjælp af en i det væsentlige biologisk proces.
14. Isoleret nukleinsyre, der koder for et acetohydroxysyresyntase (AHAS)-protein omfattende en substitution af asparaginsyre med glutaminsyre ved aminosyrepositionen svarende til position D376 af SEQ ID NO: 1, og en substitution af serin med asparagin ved aminosyrepositionen svarende til position 653 af SEQ ID NO: 1.
15. Ekspressionsvektor omfattende en nukleinsyre, der koder for et acetohydroxysyresyntase (AHAS)-protein omfattende en substitution af asparaginsyre med glutaminsyre ved aminosyrepositionen svarende til position D376 af SEQ ID NO: 1, og en substitution af serin med asparagin ved aminosyrepositionen svarende til position 653 af SEQ ID NO: 1.
