Claims (13)
1. Fremgangsmåde til fremstilling af koagulationsfaktor IX med et forbedret glycosyle-ringsmønster i en cellekultur omfattende: dyrkning af pattedyrceller, som indeholder et gen, som koder for en koagulationsfaktor IX i et cellekulturmedium, der omfatter imellem 10 nM og 600 nM mangan under betingelser og over et tidsrum, som er tilstrækkelige til at tillade ekspression af koagulationsfakator IX, hvor glycosyleringsmønsteret for det eksprimerede koagulationsfaktor IX er mere ekstensivt end det glycosyleringsmønster, som observeres under iøvrigt identiske betingelser i iøvrigt identisk medie, som mangler manganet, hvor et mere ekstensivt glycosyleringsmønster er et større antal kovalent bundne sukkerrester i oligosaccharidkæden.
2. Fremgangsmåde ifølge krav 1, hvor cellekulturmediumet omfatter imellem 20 og 200 nM mangan.
3. Fremgangsmåde ifølge krav 2, hvor cellekulturmediumet omfatter omtrent 40 nM mangan.
4. Fremgangsmåde ifølge ethvert af kravene 1 til 3, hvor cellekulturmediumet omfatter glutamin med en begyndelseskoncentration, som er mindre end eller lig med omtrent 8 mM.
5. Fremgangsmåde ifølge krav 4, hvor begyndelsesglutaminkoncentrationen i cellekulturmediumet er mindre end eller lig med omtrent 4 mM.
6. Fremgangsmåde ifølge ethvert af kravene 1 til 5, hvor volumenet af cellekulturren er i det mindste omkring 500 I.
7. Fremgangsmåde ifølge ethvert af kravene 1 til 6, hvor cellekulturen yderligere forsynes med et næringsmedium, efter den indledende celledyrkning er begyndt.
8. Fremgangsmåde ifølge ethvert af kravene 1 til 7, hvor cellekulturen yderligere tilføres supplerende komponenter.
9. Fremgangsmåde ifølge krav 8, hvor de supplerende komponenter vælges fra gruppen bestående af hormoner og/eller andre vækstfaktorer, uorganiske ioner, buffere, vitaminer, nucleosider, nucleotider, sporelementer, aminosyrer, lipider, glucose eller andre energikilder og kombinationer deraf.
10. Fremgangsmåde ifølge krav 9, hvor cellekulturen suppleres med omtrent 2 g glucose pr. I.
11. Fremgangsmåde ifølge ethvert af kravene 1 til 10, hvor cellekulturmediumet er defineret.
12. Fremgangsmåde ifølge krav 1, hvor koagulationsfaktor IX er rekombinant human koagulationsfaktor IX.
13. Fremgangsmåde ifølge krav 1, hvor den eksprimerede koagulationsfaktor IX isoleres og/eller oprenses.
