Med., 20 314, 745 (1986) og Mensink og Katan, Lancet, side 122 (17. januar 1987) redegør for, at en kost med et højt indhold af oliesyre og et lavt indhold af mættede fedtsyrer kan nedsætte indholdet af lavdensitetslipoprotein-cholesterol (LDL-cholesterol), mens mere højdensitetslipoprotein-cholesterol (HDL-cholesterol) bevares i større udstrækning end ved kost med højt ind-25 hold af linolsyre. Dette kan være at foretrække set ud fra et synspunkt om nedsættelse af risikoen for hjertesygdomme.
Holman, Prog. Chem. Fats & Other Lipids, 9, 279 (1971) anfører på side 382, at α-linolensyre kan fungere som en essentiel fedtsyre, idet den fremmer 30 vækst næsten lige så godt som linolsyre. Neuringer et al., J. Clin. Invest. 73, 272 (1984) angiver, at α-linolensyre i kosten forbedrer synskarpheden hos unge voksende aber ved at virke som præcursor for docosahexaensyre, som er en bestanddel af fotoreceptormembraner. Et nyere arbejde af Renaud et DK 174768 B1 2 al., Am. J. Clin. Nutr, 43, 136 (1986), forbinder et forøget indhold af a-linolensyre i kosten hos mennesker med nedsat blodpladesammenklumpning (hvilket tyder på eventuel nedsat risiko for blodprop). Berry og Hirsch, Am. J.
Clin. Nutr., 44, 336 (1986), forbinder forøget α-linolensyreindhold i humant 5 fedtvæv (og dermed forøget kostindhold af α-linolensyre) med nedsat blodtryk.
US patentskrift nr. 4.277.412, Logan, udstedt den 7. juli 1981, redegør for en fremgangsmåde til adskillelse af glyceridolier efter umættethedsgraden ved 10 anvendelse af et faststofadsorbens. Ved fremgangsmåden bringes en organisk opløsningsmiddelopløsning af olien i kontakt med et ionbytningsadsor-bens, som selektivt adsorberer de umættede forbindelser, og derefter desor-beres de umættede forbindelser fra udbytningsadsorbenset, og opløsningsmidlet fordampes. I patentskriftets eksempel 3 behandles solsikkeolie 15 ifølge fremgangsmåden. Den fremkomne umættethedsberigede oliefraktion indeholder (analyseret som Me-estre) 0,54% mættede fedtsyrer, 21,13% oliesyre og 78,33% linolsyre. Olien angives at være egnet som en salat- eller madlavningsolie.
20 US patentskrift nr. 4.297.292, Logan et al., udstedt den 27. oktober 1981, redegør for en fremgangsmåde til adskillelse af triglyceridolier efter graden af umættethed på en kationbytningsharpiks. Tabel VI og VII i patentskriftet fremlægger adskillelsesresultater for en blanding af tristearin, triolein og trili-nolein. I eksempel IV i tabel VI fremstilles en olie bestående af 88% triolein 25 og 12% trilinolein. I eksempel D i tabel VII fremstilles en olie bestående af 3% tristearin, 91% triolein og 6% trilinolein.
Den foreliggende opfindelse angår triglyceridmadlavnings-ysalatolie-sammensætninger med et afbalanceret fedtsyreesterindhold til opnåelse af 30 sundhedsmæssige fordele. Indholdet af fedtsyreestre i oliens triglycerid omfatter fra 60% til 92% oliesyre, fra 5% til 25% linolsyre, fra 0% til 15% a-lino-lensyre og under 3%, fortrinsvis under 1% mættede fedtsyrer.

Den foreliggende opfindelse angår madlavnings-/sa1atoliesammensætninger, hvori fedtsyresammensætningen af triglyceriderne er afbalanceret med henblik på opnåelse af sundhedsmæssige fordele. Fordele tilvejebragt med diæter, som indeholder olierne ifølge opfindelsen, indbefatter sænkning af se-5 rumcholesterolkoncentrationen og muligvis formindsket risiko for blodprop og kræft, stor fordøjelighed samt tilvejebringelse af essentielle fedtsyrer i kosten.
Triglyceriderne i olien ifølge den foreliggende opfindelse har følgende fedtsyresammensætning.

Mindst 20% af glyceriderne er glycerider med blandede fedtsyrer.
Alle procentdele og forhold heri er "på vægtbasis", med mindre andet er anført. Fedtsyresammensætningen heri er udtrykt i procentdele på vægtbasis 20 og refererer til andelene af fedtsyrer, som er forestret til triglyceridernes glycerinrygrad.
Oliesyreindholdet er fortrinsvis på fra 60% til 80%, og a-linolensyreindholdet er fortrinsvis på fra 3% til 15%, mere fortrinsvis 8% til 12%. Det mættede 25 fedtsyreindhold er fortrinsvis under 2,5%, mere fortrinsvis under 1% og mest fortrinsvis under 0,5%.
Som nævnt ovenfor er mindst 20% af oliens glycerider glycerider af blandede fedtsyrer. Et glycerid med blandede fedtsyrer er ét, hvori mindst to af de fedt-30 syrer, som er forestret til triglyceridets glycerinrygrad, er forskellige. Typisk vil olier ifølge den foreliggende opfindelse indeholde mere end 30% glycerider med blandede fedtsyrer.

Olierne kan udvindes alene fra plantekilder, hvorved indførelse af cholesterol i kosten undgås.
Ved at have et lavt eller fortrinsvis så godt som intet indhold af triglycerider, 5 som indeholder mættede fedtsyrer, nedsætter de heri omhandlede sammensætninger indførelse i kosten af forbindelser, som vides at forårsage forhøjede koncentrationer af serumcholesterol i kroppen, hvilket igen forøger risikoen for coronar-hjertesygdomme.
10 Det store forhold mellem oliesyre og linolsyre muliggør tilvejebringelse af en praktisk talt fuldstændig umættet olie og nedsætter samtidig muligvis risikoen for fremkaldelse af kræft, hvilken er blevet forbundet med kost med højt linol-syreindhold.
15 Tilstedeværelsen af en væsentlig mængde α-linolensyre i de foretrukne sammensætninger tilvejebringer de fordele, som essentielle fedtsyrer giver, til trods for det nedsatte indhold af linolsyre i olien. Det er endvidere blevet vist, at α-linolensyre fremmer vækst og forbedrer udviklingen af synskarpheden.
Den formindsker muligvis også risikoen for blodprop og nedsætter blodtryk-20 ket.
Olier ifølge den foreliggende opfindelse bør i det væsentlige ikke indeholde forbindelser, såsom organiske opløsningsmidler (f.eks. carbonhydrider såsom hexan, lavmolekylvægtige estre, såsom ethylacetat, ethere, såsom 25 diethylether og alkoholer, såsom ethanol), der på ugunstig måde ville påvirke deres egnethed til brug som madlavnings-/salatolie.
Olierne ifølge den foreliggende opfindelse kan fremstilles ved en traditionel syntese ved forestring af glycerin med en blanding af syrechloriderne af de 30 oprensede olie-, linol- og α-linolensyrer i de forhold, som ønskes i den færdige olie. Alternativt kan olierne ifølge den foreliggende opfindelse fås ved fjernelse af mættede forbindelser fra en naturlig planteoliekilde, som indeholder glycerider af de ønskede syrer i de ønskede forhold. I begge tilfælde vil olier- 5 DK 174768 B1 ne indeholde en væsentlig andel af glycerider med blandede fedtsyrer. Et eksempel på en sådan olie er canolaolie (rapsolie med lavt indhold af eruca-syre), hvori triglyceriderne typisk har følgende fedtsyresammensætning: 5 Oliesyre 62,0%

Hvis alle mættede forbindelser fraskilles denne olie ved en fremgangsmåde som beskrevet i US patentskrift nr. 4.277.412, Logan, udstedt den 7. juli 1981, der medtages heri ved denne henvisning, er resultatet en olie ifølge nærværende opfindelse, som omtrentlig har følgende fedtsyresammensæt- 15 ning:

En anden olie, der kan anvendes som kilde til olier ifølge nærværende opfindelse, er en solsikkeolie med et højt oliesyreindhold såsom den, der forhandles under navnet Tri Sun-80" af S.V.O. Enterprises of Columbus, Ohio.
25 Triglyceriderne i denne olie har en fedtsyresammensætning på ca. 8% mættede fedtsyrer, 80% oliesyre, 9% linolsyre og 3% andet. Hvis alle de mættede fedtsyrer fraskilles denne olie ved en fremgangsmåde som beskrevet i ovennævnte US patentskrift nr. 4.277.412, er resultatet en olie ifølge nærværende opfindelse med følgende fedtsyresammensætning: oliesyre 87%, linolsyre 30 9,8%, andre 3,2%. En anden oliekilde, for hvilken mættede forbindelser kan fraskilles under frembringelse af en olie ifølge nærværende opfindelse, er farvetidselolie (safflowerolie) med højt indhold af oliesyre, som markedsføres af Spectrum Marketing, Inc. of Petaluma, California 94952. Denne olie har DK 174768 B1 6 følgende omtrentlige sammensætning: mættede fedtsyrer 8,7%, oliesyre 71,9%, linolsyre 19,0%, andre 0,3%.
En anden potentiel metode til fremstilling af olier ifølge nærværende opfin-5 delse er gensplejsning af oliefrøplanter med henblik på en sådan ændring af fedtsyresammensætningen, at den svarer til de heri anførte.
Olier ifølge den foreliggende opfindelse er egnede til emballering, transportering og anvendelse på samme måde som enhver traditionel salat-10 /madlavningsolie. Tilsætningsstoffer, som forefindes i traditionelle salat-/madlavningsolier, såsom antioxidanter, emulgatorer og smagsstoffer, kan også anvendes i de normale koncentrationer i olierne ifølge den foreliggende opfindelse.
15 Med henblik på opnåelse af fordele i form af nedsat kalorieindhold kan olierne ifølge den foreliggende opfindelse anvendes i sammensætninger, hvori de er kombineret med ikke-fordøjelige fedtstoffer og olier, såsom C3-C22-fedtsyrepolyestre af carbohydrater og carbohydratalkoholer, hvori carbo-hydratet eller carbohydratalkoholen har fra 4 til 8 hydroxylgrupper, og hvori 20 mindst 4 hydroxylgrupper er forestret, se US patentskrift nr. 3.600.186, Mattson et al., udstedt den 17. august 1971, nr. 4.005.196, Jandacek, udstedt den 25. januar 1977, og nr. 4.034.083, Mattson, udstedt den 5. juli 1977, som alle medtages heri ved denne henvisning.
25 Selv om olierne ifølge den foreliggende opfindelse primært er egnede som madlavnings-/salatolier til human ernæring, er de også anvendelige som et sundhedsmæssigt afstemt fedtstof i kosten til andre dyr end mennesket.
Opfindelsen vil blive illustreret af følgende eksempler.

Eksempel 1 Følgende blanding af "oprensede" fedtsyrer blev fremstillet: 7 DK 174768 B1
Oliesyre 42,1 g (70%)
Linolensyre 12,0 g (20%) a-Linolensyre 6.0 a (10%) 5 60,1 g (100%)
Denne blanding blev omsat med 131 g oxalylchlorid i toluen til omdannelse af syrerne til de tilsvarende syrechlorider. Reaktionen blev udført i en 500 ml trehalset kolbe med tør nitrogentilgang, magnetisk omrørerpind, termometer 10 og reflux-kondensator og med gasafgang gennem en Drierit-kolonne til en toliters kolbe med natronkalk spredt ud over bunden. Blandingen blev opvarmet til 60°C under langsom udluftning med nitrogen. Infrarød-analyse efter en reaktionstid på fire timer viste ingen rester af carboxylsyre. Udbyttet var 66,5 g (teoretisk udbytte = 64,1 g). Overskuddet var sandsynligvis en rest 15 toluen. Gaskromatografanalyse er en lille prøve af syrechloriderne (omdannet til methylestre) viste 31% linol- + a-linolensyre, 67% oliesyre og 1% af en top, som sandsynligvis var eleostearat.
Syrechloridblandingen (66,5 g) blev derefter omsat med 6,0 g glycerin i 170 g 20 pyridin til dannelse af triglycerid. Glycerinen blev opløst i pyridin i en 500 ml rundbundet tre-halset kolbe under en nitrogenstrøm. Syrechloridet blev dryppet ned i kolben med en sådan hastighed, at temperaturen blev holdt på ca.
40°C. Efter tilendebringelse af syrechloridtilsætningen blev omrøringen fortsat natten over.
25 200 ml 1 N HCI blev tilsat reaktionsblandingen, som overførtes til en skilletragt. Denne blanding blev derefter ekstraheret med 1:1 diethyletherhexan. Adskillelse af det øverste ravgult farvede lag fra den vandige bobbleemulsion foregik langsomt. Fortyndet HCI og mættet vandig NaCI blev tilsat til adskil-30 lelse af lagene. Ekstraktion med vandig HCI og NaCI blev gentaget to gange.
Det organiske lag blev tørret over Na2S04, filtreret og opløsningsmidlet af-dampet. Udbyttet var 65 g af en mørk ravgul olie med en vis pyridinlugt.
DK 174768 B1 8 65 g af den mørke ravgule olie blev oprenset i en Waters Prep500 kromato-graf under anvendelse af et 85:15 hexan:diethylether-elueringsmiddel. En fraktion på 3,5 g (efter afdampning af opløsningsmidlet) af renfraktioneret olie og en fraktion på 50,4 g (efter fordampning af opløsningsmidlet) af næsten 5 renfraktioneret olie blev isoleret fra denne kromatografpassage.
Prøven på 50,4 g af næsten ren oliefraktion blev genkromatograferet på nyfremstillede kolonner i det samme apparat under anvendelse af et 94:6 he-xan:ethylacetat-elueringsmiddel, som var blevet gennemboblet med argon.
10 Forud for indførelse af prøven blev kolonnerne gennemskyllet med en 80:20 hexan.diethylether-blanding og derefter med en 94:6 hexan:ethylacetat-blanding. 1 g BHT-Antioxidant blev også sendt igennem systemet ca. 500 ml foran triglyceridol i en. En delfraktion på 41,5 g (efter afdampning af opløsningsmidlet) af yderligere oprenset svagt farvet olie blev isoleret fra denne 15 anden passage gennem kromatografen og forenet med den første rene frak tion på 3,5 g fra den første passage gennem kromatografen under tilvejebringelse af 45 g ren svagt gullig klar olie. En lille mængde af olien blev omdannet til de tilsvarende methylestre og analyseret ved gaskromatografi, hvilket gav følgende resultat: 20

Eksempel II
Dette eksempel illustrerer fremstilling af en olie ifølge den foreliggende opfindelse ved adskillelse af mættede fedtsyrer fra de umættede i en planteolie 5 ved en faststof/væske-adsorptionsproces som beskrevet i US patentskrift nr. 4.227.412, Logan, udstedt den 7. juli 1981.
Apparatur 10 Der blev anvendt et prøveanlægsafprøvningsapparatur (undertiden omtalt som en demonstrationsenhed). Apparaturet drives ligesom en enhedsoperation med et kontinuerligt, simuleret, bevægende leje. Apparaturet omfatter 24 søjler, som er serieforbundne i en sløjfe, således at procesvæsken kan strømme i én retning. Hver søjle har en længde på 61,0 cm og en indre 15 diameter på 2,29 cm og er fyldt med ca. 237 ml af et adsorbens (på basis af våd pakning). Hver søjle er udstyret med to ventiler (top og bund) med fire indstillinger, hvilke er forbundet til fire indløbs- og fire udløbsledninger. Når en ventil er lukket, strømmer der kun væske henimod den for ventilen ned-strøms søjle. Ved at vælge mellem de otte åbningsindstillinger (fire i toppen 20 og fire i bunden) kan føden bringes til indføring i systemet (f.eks. indstilling 1), opløsningsmiddel kan bringes til indføring i systemet (f.eks. indstilling 2), en raffineret strøm kan udtages fra systemet (f.eks. indstilling 3), og en ekstraktstrøm kan udtages fra systemet (f.eks. indstilling 4), eller en opløsningsmiddelstrøm kan udtages fra systemet (f.eks. indstilling 5). Tilba-25 gestrømningskontrolindstillinger er anbragt i hver af bundventilerne. Disse anvendes til at afskære zoner af systemet fra tilbagestrømning; dvs. adskille højtryksindløbet (opløsningsmiddel) fra lavtryksudløbet. Driften foregår som følger: Til ethvert tidspunkt udgør apparaturet et enkelt trin. Det drives med fire arbejdszoner (adsorption, oprensning, desorption og puffer). Én tilba-30 gestrømningskontrolventil er hele tiden i en lukket position for at udelukke tilbagestrømning mellem opløsningsmiddelindløbet og lavtryksudløbet. Der anvendes ikke recirkulation. De 24 søjler er fordelt mellem adsorptions-, oprensnings-, desorptions- og pufferzonerne med et valgt antal af søjler i serie DK 174768 B1 10 til at udgøre hver zone. Føde indføres i den første søjle i adsorptionszonen, opløses i opløsningsmidlet og bringes i kontakt med adsorbenset. Medens væske strømmer nedstrøms gennem adsorptionszonen, adsorberes triglyceridkomponenter med højere iodtal selektivt efterladende et raffinat, 5 der er beriget med triglycerid med lavere iodtal. I oprensningszonen afdrives ikke-adsorberede komponenter fra adsorpbenset og tvinges således med strømmen henimod fødepunktet. Ekstraktet udtages ved indløbet til oprensningszonen og er beriget med adsorberede komponenter. Opløsningsmidlet tilsættes ved indløbet til desorptionszonen og forårsager 10 desorption af adsorberede komponenter fra adsorbenset med henblik på nedstrømsudtagning i ekstraktpunktet. I pufferzonen adsorberes triglycerid, og opløsningsmidlet desorberes. En strøm, der heri benævnes opløsningsmiddeludløbsstrømmen, og som væsentligst består af opløsningsmiddel, udtages ved udløbet fra pufferzonen. Med fastlagte 15 intervaller rykker et styringsorgan strømningsmønstret (ind og ud af søjlerne) én søjle frem (med andre ord indstiller styringsorganet ventilerne således, at punkterne for raffinatets udstrømning, fødeindstrømning, ekstraktudstrømning, opløsningsmiddelindstrømning og opløsningsmiddeludstrømning hver rykker et trin frem, dvs. til det næste væske-20 tilgangspunkt i væskestrømningsretningen) med henblik på at "rykke fremad" for at holde trit med væskestrømningen. En cyklus består af det antal trin, der svarer til antallet af søjler. "Trintiden" vælges således, at den tillader, at de ikke-adsorberede komponenter føres hurtigere frem end fødepunktet og når raffinatpunktet. Det adsorberede triglycerid bevæger sig langsommere end 25 fødepunktet og falder tilbage til ekstraktpunktet.

Dette eksempel illustrerer adskillelse af canolaolie i en ekstraktfraktion inde-30 holdende en væsentlig nedsat procentdel af triglycerid med mættede fedtsyrekæder og en raffinatfraktion. Processen udføres under anvendelse af en kontinuerlig, simuleret, bevæget Jejeproces i demonstrationsenheden som beskrevet ovenfor.
11 DK 174768 B1 Fødesammensætningen er renset, bleget og desodoriseret canolaolie, som er forbehandlet til fjernelse af resterende urenheder (f.eks. frie fedtsyrer, mo-noglycerider, diglycerider, spor af vand) ved opløsning i hexan og passage 5 gennem en Florisil-pakket søjle. Fedtsyresammensætningen i oliens triglyce-rider er omtrentlig 5,5% mættede fedtsyrer, 62% oliesyre, 22% linolsyre, 10% α-linolensyre og 0,5% andre. Fødesammensætningen er i det væsentlige fri for urenheder.
10 Adsorbenset er Decalso Y, som er modificeret således, at det indeholder 1,0 mmol sølv (Ag+1 )/100 m2 adsorbensoverfladeareal (på basis af 100% natriumsubstitution). De kationsubstituenter i adsorbenset, som ikke er sølv-substituenter, er natriumsubstituenter. Adsorbenset foreligger i form af partikler, som (på bulkvandfri og opløsningsmiddelfri basis) i det væsentlige 15 fuldstændig består af udbytningsadsorbens, der har en størrelse på fra ca. 40 mesh til ca. 20 mesh, og som har et vandindhold på under 4 vægtprocent. Adsorbenset er kendetegnet ved et forhold mellem siliciumatomer og aluminiumatomer på 3:1 og et overfladeareal, bestemt på basis af 100% natriumsubstitution, på 233 m2 pr. g. Adsorbenset fremstilles ved anbringelse af De- 20 calso Y-partikler (sigtet gennem 20 mesh og på 40 mesh) i en vandig sølvnitratopløsning (105% af det støkiometriske forhold) i tre timer, vask med vand og indstilling af vandindholdet ved ovntørring ved 130°C.
Opløsningsmidlet består af 90% ethylacetat og 10% hexan på volumenbasis.

Styringsorganet og ventilerne i demonstrationsenheden indstilles således, at adsorptionszonen omfatter seks søjler, oprensningszonen omfatter otte søjler, desorptionszonen omfatter otte søjler, og pufferzonen omfatter to søjler.

Adskillelse sker på grundlag af iodtallet, dvs. med henblik på at opnå fraktioner med højere iodtal og lavere jodtal.

Triglyceridfraktionen i ekstraktet indeholder på vægtbasis (på fedtsyrebasis) mindre end 1 % mættede fedtsyrer, omtrentlig 65% oliesyre, omtrentlig 23% linolsyre og omtrentlig 11 % α-linolensyre. Efter fjernelse af opløsningsmidlet er triglyceridfraktionen fra ekstraktet egnet som en salat- eller madlavnings-15 olie ifølge den foreliggende opfindelse.
En solsikkeolie med højt indhold af oliesyre, som sælges under navnet ’Tri Sun 80" af S.V.O. Enterprises of Columbus, Ohio, underkastes ovenstående adskillelsesproces. Fedtsyresammensætningen i oliens triglycerider før be-20 handling var 8% mættede fedtsyrer, 80% oliesyre, 9% linolsyre og 3% andre.
Efter gennemførelse af adskillelsesprocessen er fedtsyresammensætningen af oliens triglycerider omtrentlig: 0,6% mættede fedtsyrer, 86,4% oliesyre, 9,7% linolsyre og 3,2% andre. Efter fjernelse af opløsningsmiddel er olien egnet som en madlavnings- eller salatolie.

Dette eksempel illustrerer anvendelsen af HPLC (engelsk: "high performance liquid chromatography") til fjernelse af mættede fedtsyrer fra canolaolie. Den 30 som føde anvendte canolaolie, havde en fedtsyresammensætning på 0,3% C14, 5,0% C16, 2,0% C18, 56,3% C18:1, 20,8% C18:2 og 8,1% C18:3. Fremgangsmåden blev udført ved at adskille de forskellige canolatriglyceri-der og selektivt opsamle triglycerider med umættede fedtsyrekæder. Et 13 DK 174768 B1 HPLC-apparatur bestående af to pumper og to Beckman Ultrasphere ODS-kolonner (ODS: octadecylsilansiliciumoxidgel) med en 5 mikrometer pakning blev anvendt. Kolonnerne havde en indre diameter på 4,6 mm og var 25 cm lange.

Canolaolie blev tilsat kromatografen. Ved forsøgets begyndelse bestod den mobile fase af 35% methylenchlorid og 65% acetonitril. Under forsøget øgedes methylenchloridkoncentrationen lineært til 55% i løbet af 40 minutter. De ønskede triglycerider blev opsamlet i en Erlenmeyer-kolbe. Opløsningsmidlet 10 blev fjernet med en nitrogenstrøm i et dampbad.
Den fremkomne, med umættede fedtsyrer berigede lipidfraktion blev omdannet til de tilsvarende methylestre, og analyse med gaskromatograf gav følgende resultat: 15

Procentdelen af blandede triglycerider i lipidfraktionen før omdannelse til 25 methylestre beregnes til at være ca. 73%.
Claims (9)
1. Madlavnings-/salatoliesammensætning, som i det væsentlige består af triglycerider, kendetegnet ved, at fedtsyresammensætningen 5 af triglyceriderne på vægtbasis omfatter: fra 60% til 92% oliesyre, fra 5% til 25% linolsyre, 10 fra 0% til ca. 15% α-linolensyre og mindre end 3% mættede fedtsyrer, 15 og hvori mindst 20% af triglyceriderne er triglycerider med blandede fedtsyrer.
