Den foreliggende opfindelse angår en fremgangsmåde til dannelse af et forbedret hydrofilt overfladelag på et substrat af et polymert materiale, hvilket overfladelag er forbedret med hensyn til bibeholdelse af sin glathed i et længere tidsrum, og som har meget lav 5 friktionskoefficient ved befugtning med en vandbaseret væske, ved hvilken fremgangsmåde man danner et ikke-reaktivt overfladelag af en hydrofil polymer på substratet,
Opfindelsen angår tillige en medicinsk genstand, som er belagt med 10 et sådant forbedret hydrofilt overfladelag.
Et hydrofilt overtræk har langt lavere friktionskoefficient i våd tilstand end i tør tilstand. Dannelse af et hydrofilt overtræk på substrater har således mange anvendelser, men er i særdeleshed 15 særlig ønskværdig ved mange biomedicinske anvendelser. F.eks. er sådanne overtræk ønskelige til biomedicinske formål, såsom sårdræn, katetere, kirurgiske redskaber og andre medicinske .redskaber beregnet til at blive indført i et legemshulrum, fordi instrumentet giver et godt greb for hånden i tør tilstand samtidig med, at det 20 bliver meget glat, når det kommer i kontakt med vandbaserede væsker, hvorfor det let kan indsættes uden at fremkalde ulemper for patienten. Desuden er en artikel med et hydrofilt overfladeoverlag ønskelig for at mindske tendensen til trombose, krystaldannelse, vævstraumer, vævsvedhængning til medicinske instrumenter og 25 fremmed!egemereaktioner.
Ved inden for teknikken kendte metoder har man gjort overflader hydrofile ved sådanne metoder som højenergi bestråling, ved polymerisationsprocesser in situ, ved podning, ved dannelse af et 30 interpolymernetværk eller ved en direkte kemisk binding, f.eks. med isocyanater eller silaner. Sådanne hydrofile polymeroverflader til medicinsk anvendelse er beskrevet i f.eks. GB 1.600.963, US 4.373.009, US 4.459.317, WO 83/03977 og EP patentansøgning nr. 83850090.8. Omfattende undersøgelser tyder imidlertid på, at det 35 hydrofile overtræk kan blive udtørret og derved gøre genstanden utilstrækkelig hydrofil.
Det er den foreliggende opfindelses formål at tilvejebringe et forbedret hydrofilt overfladelag på et allerede hydrofilt 2 DK Ί69552 bl polymeroverfladelag på et substrat.
Dette formål opnås ved en fremgangsmåde til dannelse af et forbedret hydrofilt overfladelag på et substrat af et polymert materiale, 5 hvilket overfladelag er forbedret med hensyn til bibeholdelse af sin glathed i et længere tidsrum, og som har meget lav friktionskoefficient ved befugtning med en vandbaseret væske, ved hvilken fremgangsmåde man danner et ikke-reaktivt overfladelag af en hydrofil polymer på substratet, hvilken fremgangsmåde ifølge 1° opfindelsen er ejendommelig ved, at man påfører det ikke-reaktive overfladelag en opløsning' indeholdende over 2% v/r af en osmol al itetsforøgende forbindelse og eventuelt en polymer, hvor opløsningsmidlet er vand eller en lavere alkohol, og den osmolalitetsforøgende forbindelse er et mono- eller disaccharid, en *5 sukkeralkohol eller et ugiftigt organisk eller uorganisk salt, med undtagelse af et trihalogenid, såsom Kig (KI/^), hvorpå man afdamper opløsningsmidlet fra opløsningen.
Opfindelsen angår også en medicinsk genstand med meget lav 20 friktionskoefficient ved befugtning med en vandbaseret væske, hvilken genstand er ejendommelig ved, at der på genstandens overflade af polymert materiale er et ikke-reaktivt hydrofilt polymerlag, og at der på polymerlaget er et inddampet overtræk fra en opløsning indeholdende over 2% v/r af en osmolalitetsforøgende 25 forbindelse og eventuelt en polymer, hvor opløsningsmidlet er vand eller en lavere alkohol, og den osmolalitetsforøgende forbindelse er et mono- eller disaccharid, en sukkeralkohol eller et ugiftigt organisk eller uorganisk salt, undtagen et trihalogenid, såsom Kig (KI/I2).

Under forsøg med hydrofile polymere overtræk på forskellige substrater viste det sig, at når en genstand overtrukket med et hydrofilt polymert overfladelag dyppes i vand, befugtes det godt, men der er stor risiko for, at den polymere overflade mister sit 55 vand, når den koraner i kontakt med en slimhinde eller lignende. Dette beror på forskellen i det osmotiske potentiale mellem den hydrofile overflade og slimhinden. Slimhinden har højere osmotisk potentiale, dvs. en højere saltkoncentration, end den hydrofile overflade, hvilket resulterer i, at vandet går fra det hydrofile DK 169552 B1 3 overfladelag til slimhinden, hvorved forskellen i saltkoncentrationen opvejes.
Ifølge opfindelsen har det overraskende vist sig muligt samtidig at 5 nedsætte den lave friktion af den hydrofile overflade yderligere og at forøge osmolaliteten af den hydrofile overflade ved, at man påfører et overtræk af en osmolalitetsforøgende forbindelse på det hydrofile polymeroverfladelag. Der kan bruges forskellige typer af osmolalitetsforøgende forbindelser, såsom uorganiske eller organiske salte, mono- eller disaccharider eller sukkeralkoholer. Eksempler på sådanne osmolalitetsforøgende forbindelser er glucose, sorbitol, natriumchlorid, natriumcitrat, natriumbenzoat, calciumchlorid, kaliumchlorid, kaliumjodid og kaliumnitrat. Denne osmolalitetsforøgende forbindelse må være ugiftig. Fremgangsmåden har vist sig at være særdeles nyttig, når der anvendes natriumchorid. Den osmolalitetsforøgende forbindelse blandes eventuelt med en polymer og opløses i vand eller en lavere alkohol, fortrinsvis i vand til en passende viskositet. Denne opløsning påføres på det hydrofile polymeroverfladelag ved dypning, sprøjtning eller lignende, hvorpå 20 opløsningsmidlet afdampes ved lufttørring eller ved tørring ved forhøjet temperatur. Den til satte polymer bør være godt forenelig med polymeren i polymeroverfladelaget og er fortrinsvis den samme polymer. Polymeren er ikke et nødvendigt additiv i opløsningen, men den forøger viskositeten, og i kraft heraf kan modtagelsen af den 25 osmolalitetsforøgende forbindelse reguleres. Desuden har polymeren smøringsegenskaber og virkning med hensyn til styret afgivelse.
Fremgangsmåden ifølge opfindelsen kan bruges til overtrækning af mange forskellige typer velkendte hydrofile overflader, hvor den 50 hydrofile polymer er udvalgt blandt polyvinyl forbindelser, poly-saccharider, polyurethaner, polyacrylater og copolymerer af vinyl-forbindelser og acrylater eller anhydrider. Navnlig egnet er poly-ethylenoxid, polyvinylpyrrolidon, heparin, dextran, xantan, polyvinyl al kohol, hydroxypropylcellulose, methylcellulose, copolymerer 55 af vinylpyrrol idon og hydroxyethylenmethylacrylat eller copolymerer af polymethylvinylether og maleinsyreanhydrid. Den foretrukne hydrofile polymer er polyvinylpyrrolidon. Polymeroverfladen må gøres ikke-reaktiv, f.eks. ved hærdning.

Substratet kan være et hvilket som helst polymert materiale, som er velkendt, og hvortil de hydrofile polymerer hænger ved, f.eks. polyurethaner, latexgummier, andre gummier, polyvinylchlorid, andre vinylpolymerer, polyestere eller polyacrylater.

Den hydrofile overflade indeholder en kendelig mængde delvis frie bevægelige polymerkæder. Det er påvist, at nogle overtræk kan kompleksbinde en væsentlig mængde frit jod, hvilket f.eks. er tilfældet med frit polypyrrol idon og jod, når en hydrofil overflade 10 af polyvinyl pyrrol idon behandles med en Kl/Ig-opløsning. PVP-overtrukne katetere behandlet med en Kl/Ig-opløsning har således en mindre glat overflade end en tilsvarende ubehandlet overflade, og sådanne KI3-behandlede katetere tørrer hurtigere.
15 Eksempel
Et trimeriseret hexamethylendiisocyanat af biuret-type (betegnet Desmodur® L 2291 fra Bayer AG) opløstes i methylenchlorid til en koncentration på 6% v/r (vægt/rumfang, dvs. forholdet mellem fast 20 stof og væske såsom mellem gram og milliliter). Et urinkateter af PVC blev dyppet i denne opløsning i 30. sek. Derefter tørredes kateteret ved 70eC i 60 sek., hvorpå det dyppedes i 5 sek. i en opløsning indeholdende 33 g polyvinyl pyrrol idon (K25; gennemsnitlig molekylvægt 25.000) pr. 100 ml methylenchlorid. Denne opløsning 25 indeholdt også 0,33 g tri ethyl endiamin (DABC0®) pr. 100 ml opløsning. Kateteret blev derefter tørret ved omgivelsernes temperatur i 60 sek., hærdedes derpå i 40 min. ved 70*C, afkøledes til omgivelsestemperatur og skylledes med vand. Kateteret blev dyppet i en opløsning indeholdende 20% v/r natriumchlorid og 5% v/r 30 polyvinyl pyrrolidon (PVP) opløst i vand i 1 time ved 20eC. Derefter lod man kateteret tørre ved 70eC i 8 timer. Kateteret havde en meget glat og vedhængende overflade i befugtet tilstand.
Forsøasafprøvning 35
Et antal katetere fremstillet i overensstemmelse med foranstående eksempel blev dyppet i 10 min i forskellige opløsninger indeholdende osmol al itetsforøgende forbindelser og anbragtes derefter i vand i 30 sek, ved 23°C og en relativ fugtighed på 49% og blev derefter tørret DK 169552 B1 5 ved stuetemperatur. Glatheden blev vurderet 1 min., 2 min. og så videre efter påbegyndt tørring. Værdien for glatheden blev opnoteret i en relativ skala, hvor 8 betyder en meget glat overflade og 0 betyder en tør overflade.

Dette forsøg blev gentaget med et antal tilsvarende PVP-overtrukne katetere, med og uden belægning med en osmol al itetsforøgende forbindelse. Resultaterne fremgår af tabel 1.

Tabel 1 viser, at de katetere, som har et overtræk af en ugiftig, osmol al itetsforøgende forbindelse, bevarer deres glathed i længere tid end de tilsvarende ubehandlede overflader, dvs. de overtrukne katetere tørrer langsommere. De osmol al itetsforøgende forbindelser 30 forhindrer den hydrofile polymeroverflade i at udtørre.
Katetere, som er forsynet med en osmolalitetsforøgende forbindelse, såsom natriumchlorid, i overensstemmelse med opfindelsen, tørrer således langsommere end tilsvarende ubehandlede katetere. Katetere behandlet med natriumchlorid bevarer deres glathed i meget længere tid, hvilket er meget ønskeligt.
Medicinske prøver viser, at katetere, som er påført et overtræk af en osmolalitetsforøgende forbindelse, såsom natriumchlorid, er bedre 5 DK 169552 B1 6 ved indføring og ved fjernelse af kateteret fra urinrøret.

Claims (10)
1. Fremgangsmåde til dannelse af et forbedret hydrofilt overflade-5 lag på et substrat af et polymert materiale, hvilket overfladelag er forbedret med hensyn til bibeholdelse af sin glathed i et længere tidsrum, og som har meget lav friktionskoefficient ved befugtning med en vandbaseret væske, ved hvilken fremgangsmåde man danner et ikke-reaktivt overfladelag af en hydrofil polymer på substratet, 10 kendetegnet ved, at man påfører det ikke-reaktive overfladelag en opløsning indeholdende over 2% v/r af en osmolalitets- forøgende forbindelse og eventuelt en polymer, hvor opløsningsmidlet er vand eller en lavere alkohol, og den osmolalitetsforøgende forbindelse er et mono- eller disaccharid, en sukkeralkohol eller et 15 ugiftigt organisk eller uorganisk salt, med undtagelse af et tri-halogenid, såsom Kig (KI/I^)> hvorpå man afdamper opløsningsmidlet fra opløsningen.

Fremgangsmåde ifølge krav 1, kendetegnet ved, at 20 forbindelsen i opløsningen er udvalgt blandt glucose, sorbitol, natriumchlorid, natriumcitrat, natriumbenzoat, calciumchlorid, kaliumchlorid, kaliumjodid eller kaliumnitrat, fortrinsvis blandt glucose, natriumchlorid, natriumcitrat, natriumbenzoat, calciumchlorid, kaliumchlorid, kaliumjodid og kaliumnitrat. 25

Medicinsk genstand med meget lav friktionskoefficient ved befugtning med en vandbaseret væske, kendetegnet ved, at der på genstandens overflade af polymert materiale er et ikke-reaktivt hydrofilt polymerlag, og at der på polymerlaget er et 10 inddampet overtræk fra en opløsning indeholdende over 2% v/r af en osmol al itetsforøgende forbindelse og eventuelt en polymer, hvor opløsningsmidlet er vand eller en lavere alkohol, og den osmolali-tetsforøgende forbindelse er et mono- eller disaccharid, en sukkeralkohol eller et ugiftigt organisk eller uorganisk salt, undtagen et I5 trihalogenid, såsom KI3 (KI/I2).
9. Medicinsk genstand ifølge krav 8, kendetegnet ved, at det inddampede overtræk består af glucose, sorbitol, natrium-chlorid, natriumcitrat, natriumbenzoat, calciumchlorid, kalium- 20 chlorid, kaliumjodid eller kaliumnitrat, fortrinsvis glucose, natriumchlorid, natriumcitrat, natriumbenzoat, calciumchlorid, kaliumchlorid, kaliumjodid eller kaliumnitrat.
