Den foreliggende opfindelse angår hidtil ukendte phosphon-syrer og derivater deraf, som er ejendommelige ved, at de har den almene formel RO-P-A-Hot (I), 5 Ar· hvori R og R' hver for sig betyder hydrogen eller C(l-4)-alkyl, A betyder C(l-4)-alkylen, og Het betyder en 2-Rl-pyrrolidinyl- eller 2-Rl-piperidinylgruppe, og R^· betyder carboxy, carbamoyl, N-mono- eller N,N-di-C(1-4)-10 alkylcarbamoyl eller C(l-4)-alkoxycarbonyl.
Opfindelsen angår også salte af forbindelserne med formlen I.
Opfindelsen angår tillige en fremgangsmåde til fremstilling af forbindelserne med formlen I eller salte deraf, 15 farmaceutiske præparater indeholdende disse forbindelser, forbindelserne til anvendelse ved terapeutisk behandling af det menneskelige eller dyriske legeme samt anvendelsen af forbindelserne til fremstilling af farmaceutiske præparater, især til behandling af tilstande og sygdomme hos 20 pattedyr, som reagerer på blokade af den N-methyl-D-aspartatsensitive excitatoriske aminosyrereceptor.

Abstr. 104: 69130a (1986) kendes visse phosphonosubsti- tuerede pyrrolidin- og piperidin-2-carboxylsyrederivater.
25 Fra GB-patentskrift nr. 2.104.079 er det endvidere kendt, at visse 2-amino-ω-phosphono-alkansyrer udviser NMDA-anta-gonistiske virkninger. Det har vist sig, at forbindelserne ifølge opfindelsen er aktive og nyttige i pattedyr som selektive antagonister for den N-methyl-D-aspartat (NMDA)-30 følsomme excitatoriske aminosyrereceptor. Udtrykket "lav" anvendt i det følgende i forbindelse med organiske grupper eller forbindelser betyder, at disse indeholder højst 7, fortrinsvis højst 4 carbonatomer.

C(l-4)-alkoxycarbonyl indeholder 1-4 carbonatomer i alkoxydelen og betyder eksempelvis methoxycarbonyl, propoxycarbonyl, isopropoxycarbonyl eller fordelagtigt 10 ethoxycarbonyl.
N-mono-C(l-4)-alkylcarbamoyl betyder f.eks. N-methyl-carbamoyl, N-propylcarbamoyl eller fordelagtigt N-ethyl-carbamoyl.
N,N-di-C(l-4)-alkylcarbamoyl betyder f.eks. N,N-dimeth-15 ylcarbamoyl, N-ethyl-N-methylcarbamoyl og fordelagtigt N,N-diethylcarbamoyl.
Salte af forbindelserne ifølge opfindelsen er fortrinsvis farmaceutisk acceptable salte, som kan være metal- eller ammoniumsalte af forbindelserne ifølge opfindelsen, som 20 indeholder en fri phosphonsyregruppe eller carboxygruppe, specielt alkalimetal- eller jordalkalimetalsalte, f.eks. natrium-, kalium-, magnesium- eller calciumsalte, eller fordelagtigt krystalliserende ammoniumsalte afledt af ammoniak eller organiske aminer, såsom methylamin, di-25 ethylamin, triethylamin, dicyclohexylamin, triethanol-amin, ethylendiamin, tris-(hydroxymethyl)aminomethan eller benzyltrimethylammoniumhydroxid. På den anden side kan de forbindelser ifølge opfindelsen, som er basiske aminer, danne syreadditionssalte af fortrinsvis farmaceutisk 30 acceptable uorganiske og organiske syrer, såsom stærke mineralsyrer, f.eks. hydrogenhalogenidsyrer, såsom hydrogenchloridsyre eller hydrogenbromidsyre, svovlsyre, phosphorsyre eller salpetersyre, aliphatiske eller aroma- DK 167811 B1 5 tiske carboxylsyrer eller sulfonsyrer, f.eks. eddikesyre, propionsyre, ravsyre, glycolsyre, mælkesyre, æblesyre, vinsyre, gluconsyre, citronsyre, ascorbinsyre, maleinsyre, 5 fumarsyre, pyrudruesyre, pamoesyre, nicotinsyre, methan-sulfonsyre, ethansulfonsyre, hydroxyethansulfonsyre, benzensulfonsyre, p-toluensulfonsyre eller naphthalen-sulfonsyre.
Til isolerings- eller rensningsformål kan der dannes 10 salte, som ikke kan anvendes til farmaceutiske formål. Imidlertid anvendes kun farmaceutisk acceptable salte til terapeutiske formål, hvorfra disse salte foretrækkes.
Forbindelserne ifølge opfindelsen har som anført ovenfor værdifulde farmakologiske egenskaber. Eksempelvis kan de 15 selektivt blokere de N-methyl-D-aspartat-sensitive excita-toriske aminosyrereceptorer hos pattedyr. Forbindelserne ifølge opfindelsen kan således anvendes til behandling af sygdomme, som reagerer på excitatorisk aminosyreblokade i pattedyr, f.eks. sygdomme eller forstyrrelser i nerve-20 systemet, specielt konvulsive tilstande (epilepsi) og angsttilstande.
Disse virkninger kan påvises ved in vitro-forsøg eller in vivo-dyreforsøg, idet man fordelagtigt anvender pattedyr eller vævs- eller enzympræparater fra pattedyr, f.eks.
25 mus, rotter eller aber. Forbindelserne kan indgives enteralt eller parenteralt, fordelagtigt oralt eller transdermalt, eller subkutant, intravenøst eller intra-peritonealt, f.eks. i gelatinekapsler, eller i form af vandige suspensioner eller opløsninger. Den anvendte in 30 vivo-dosis kan ligge i området fra ca. 0,01 til 100 mg/kg, fortrinsvis fra ca. 0,05 til 50 mg/kg, fordelagtigt fra ca. 0,1 til 10 mg/kg. Forbindelserne kan anvendes in vitro i form af f.eks. vandige opløsninger, og dosis kan variere fra koncentrationer på ca. 10-½ til 10-½.

Den inhiberende virkning på de excitatoriske aminosyre-receptorer af NMDA-typen bestemmes in vitro ved måling af inhiberingen af den af NMDA-fremkaldte afgivelse af 5 3H-acethylcholin (3H-ACh) fra corpus striatum-væv fra rottehjerne, jævnfør J. Lehmenn og B. Scatton, Brain Research 252, 77 - 89 (1982) og Nature 297, 422 - 424 (1982).
Antagonister for excitatoriske aminosyrereceptorer af 10 NMDA-typen antagoniserer kompetitivt NMDA-fremkaldt afgivelse af 3H-acetylcholin (3H-ACh) ’ fra corpus striatum-væv i hjernen.
Inhiberingen af afgivelsen af 3H-acetylcholin (3H-ACh) fremkaldt af NMDA fra skiver af corpus striatum fra rotter 15 ved hjælp af en forbindelse ifølge opfindelsen udtrykkes som den procentvise afgivelse af 3H-ACh i respons på stimulering med 50 /m NMDA sammenlignet til kontrolgrupper. Forsøgene er to-sporede (two-tailed) med et minimum på n = 4 i hver gruppe. IC5ø-værdier angiver den 20 koncentration af forsøgsforbindelsen, der kræves for at inhibere den ved hjælp af NMDA-forøgede 3H-ACh-afgivelse ved 50%.
Ved den in vitro NMDA-fremkaldte 3H-ACh-afgivelsestest opnås følgende ICsø-værdier: 25 cis-4-(Phosphonomethyl)piperidin-2-carboxylsyre (eksempel 2): 8 x 10"6M og blanding af cis- og trans-4-(phosphonomethyl)piperidin-2-carboxylsyre (eksempel 3): 10 x 10“^M.
Den inhiberende virkning på de excitatoriske aminosyre-30 receptorer af NMDA-typen påvises in vivo ved inhibering af NMDA-inducerede kramper hos mus. Der opnås følgende DK 167811 B1 7 ED5Q~værdier: cis-4-(Phosphonomethyl)piperidin-2-carboxylsyre (eksempel 2): 2,3 mg/kg i.p. og 5 cis—4-(phosphonomethyl)piperidin-2-carboxylsyreethylester (eksempel 5a): 47 mg/kg i.p.
De omhandlede forbindelsers antikonvulsive virkning fremgår især af, at de virksomt forebygger audiogent inducerede anfald hos DBA/2 mus, jævnfør Chapman et al., 10 Arzneim.-Forsch. 34, 1261, (1984).
Virkningen bestemmes på følgende måde: 45 minutter efter indgift af forsøgsforbindelse eller bærestof anbringes musene individuelt i et lydtæt kammer. Efter en tilpasningsperiode på 30 sekunder udsættes musene for en lyd-15 stimulering på 110 dB i 1 minut eller indtil fremkomst af et tonisk-klonisk anfald. Kontrolanfald består af en indledende fase med vild og uregerlig løben omkring. Forebyggelsen af vild eller uregerlig løben omkring indicerer en antikonvulsiv virkning.
20 Forsøgsforbindelserne indgives enten i form af opløsninger i destilleret vand eller i en 3%’s (vægt/rumfang) kolloid majsstivelsessuspension indeholdende 5%'s (vægt/rumfang) polyethylenglycol 400 og 0,34%’s (vægt/rumfang) "Tween"® 80 ved oral intubation eller intraperitonealt i 25 et rumfang på 10 ml/kg legemsvægt. Ved denne forsøgsmodel opnås følgende EDgg-værdier: cis-4- (Phosphonomethyl )piperidin-2-carboxylsyre-hydro-chlorid (eksempel 2): 1,8 mg/kg i.p., cis-4-(1-methyl-1-phosphonoethyl)-piperidin-2-carboxyl-30 syre (eksempel 4g): 2,0 mg/kg i.p., 8 UK Tb/sΊ I tn cis-4-(3-phosphonopropyl)piperidin-2-carboxylsyre-ethyl-ester (eksempel 5d): 7,3 mg/kg i.p. og (-)-cis-4-phosphonomethylpiperidin-2-carboxylsyre 5 (eksempel 12): 0,8 mg/kg i.p.
Forbindelserne ifølge opfindelsen er virksomme ved afprøvning efter Cook/Davidson-konfliktmodellen (Psychopharmaco-logia lj3, 159 - 168 (1969), hvilket indicerer anxiolytisk virkning.
10 De ovennævnte fordelagtige egenskaber ved forbindelserne ifølge opfindelsen gør disse anvendelige som antagonister for de excitatoriske aminosyrereceptorer af NMDA-typen i pattedyr og til behandling af tilstande, som reagerer derpå, såsom angsttilstande og konvulsive tilstande.
15 Forbindelserne ifølge opfindelsen kan fremstilles under anvendelse af konventionelle fremgangsmåder. Fremgangsmåden ifølge opfindelsen er ejendommelig ved,, at man a) reducerer en forbindelse med formlen RO-P-A*-Het' (VII) 0R' 20 der er identisk med en forbindelse med formlen I bortset fra, at den indeholder en eller flere dobbeltbindinger i den 5- eller 6-leddede heterocycliske ring Het' og/eller i A', som betyder A eller C(2-4)-alkenylen eller C(2-4)-alkanyliden med dobbeltbindingen knyttet til den hetero-25 cycliske ring, eller b) kondenserer en forbindelse med formlen X-A-Het (VIII) 9 DK 167811 Bl hvori Het og A har de ovenfor under formlen I angivne betydninger, og X betyder reaktionsdygtigt esterificeret hydroxy, med en forbindelse, der indfører phosphonsyrede-5 len, og som har formlen H j or" (ix) eller Ρ<κ»·)3 (X) or" hvori R" betyder C(1-4)-alkyl, og R" ’ betyder halogen eller C(l-4)-alkoxy, og hydrolyserer det dannede produkt, når R”' betyder halogen, og, om nødvendigt, omdanner det 10 dannede phosphonsyrederivat til phosphonsyren -eller et andet C(l-4)-alkylesterderivat deraf, eller c) omdanner en substituent forskellig fra R1 i 2-stillin-gen i den heterocycliske ring i en forbindelse, som i øvrigt er identisk med en forbindelse ifølge opfindelsen, 15 til Ri, idet man om nødvendigt gennemfører fremgangsmåderne under midlertidig beskyttelse af ei eller flere interfererende reaktionsdygtige grupper, hvorpå man frigør den dannede forbindelse, og, om nødvendigt, omdanner en dannet forbindelse til en anden forbindelse ifølge opfin-20 delsen og/eller, om ønsket, omdanner en dannet fri forbindelse til et salt deraf eller et dannet' salt til den fri forbindelse eller til et andet salt og/eller adskiller en blanding af isomerer eller racemater i de enkelte isomerer eller racemater og/eller, om ønsket, resolverer 25 et dannet racemat i de optiske antipoder.
Reaktionsdygtigt esterificeret hydroxy i en vilkårlig af de ovenfor beskrevne fremgangsmåder, f.eks. X i en forbindelse med formlen VIII, er esterificeret med en stærk syre, især en hydrogenhalogenidsyre, f.eks. hydrogen-30 chloridsyre, hydrogenbromidsyre eller hydrogeniodidsyre, 10 υκ lb/a i i κ ι eller svovlsyre, eller med en stærk organisk syre, især en stærk organisk sulfonsyre, såsom en aliphatisk eller aromatisk sulfonsyre, f.eks. methansulfonsyre, 4-methyl-5 phenylsulfonsyre eller 4-bromphenylsulfonsyre. Reaktionsdygtigt esterificeret hydroxy betyder især halogen, f.eks. chlor, brom eller iod, eller aliphatisk eller aromatisk substitueret sulfonyloxy, f.eks. methansulfonyloxy, phenylsulfonyloxy eller 4-methylphenylsulfonyloxy 10 (tosyloxy).
I udgangsforbindelser og mellemprodukter derfor, som omdannes til forbindelserne ifølge opfindelsen på den ovenfor beskrevne måde, er tilstedeværende funktionelle grupper, såsom carboxy, amino (herunder ring-NH) og 15 hydroxy, eventuelt beskyttede med konventionelle beskyttelsesgrupper, som almindeligvis anvendes i den præparative organiske kemi. Beskyttede carboxy-, amino- og hydroxy-grupper er sådanne, som under milde betingelser kan omdannes til frie carboxy-, amino- og hydroxy-grupper, 20 uden at molekylet nedbrydes eller at der foregår andre uønskede sidereaktioner.
Formålet med at indføre beskyttelsesgrupper er at beskytte de funktionelle grupper mod uønskede reaktioner med reaktanter og under de anvendte betingelser ved gennemførel-25 sen af en ønsket kemisk omdannelse. Behovet for og valget af beskyttelsesgrupper til en specifik omsætning er kendt for fagfolk og afhænger af naturen af den funktionelle gruppe, som skal beskyttes (carboxygruppe, aminogruppe osv.), strukturen og stabiliteten af det molekyle, hvori 30 gruppen indgår, og reaktionsbetingelserne.

Fremstillingen af forbindelserne ifølge opfindelsen ifølge fremgangsmåde a) ved reduktion af en forbindelse med formlen VII gennemføres eksempelvis under anvendelse af i og for sig kendte fremgangsmåder til reduktion af pyrrol-, 10 pyridin-, indol- og quinolin-ringe. Reduktionen af pyr-idinringen gennemføres eksempelvis fordelagtigt med et organometalreduktionsmiddel eller ved katalytisk hydrogenering, f.eks. i nærværelse af platinoxid og et surt opløsningsmiddel, såsom eddikesyre, til dannelse af de 15 tilsvarende piperidiner ifølge opfindelsen, dvs. med formlen II og salte deraf. Kvaternære pyridiniumfor-bindelser kan reduceres på lignende måde.
Udgangsforbindelserne med formlen VII, hvori Het' indeholder en eller flere dobbeltbindinger, f.eks. pyridin-20 derivaterne, hvori Het betyder en 2-R^-pyridylgruppe, og A betyder C(l-4)-alkylen, fremstilles eksempelvis ved først at kondensere en forbindelse med formlen Het1-A-X (Vila), f.eks. i form af et halogenid, såsom et chlorid eller bromid, med en diester med formlen IX i nærværelse 25 af en stærk base, f.eks. som beskrevet i Chemical Abstracts 61, 10703, eller med et tri-C(l-4)-alkylphos- phit med formlen X til dannelse af den tilsvarende (3-, 4-eller 5-pyridinyl)-A-P(=0)(0-C(1-4)-alkyl)2~forbindelse, hvori A har den ovenfor angivne betydning. 1 2 3 4 5 6
Ved kondensation med et reaktionsdygtigt cyanid, f.eks.
2 et trialkylsilylcyanid, såsom trimethylsilylcyanid, for 3 trinsvis under basiske betingelser, f.eks. i nærværelse 4 af triethylamin, fås de tilsvarende 2-cyanopyridinderi- 5 vater. Cyanogrupperne i 2-cyanopyridinderivaterne kan 6 derpå omdannes på i og for sig kendt måde til de 2-r1- 12 UIV 10/0 I I Dl substituerede pyridinderivater som defineret ovenfor.
Fremstillingen af forbindelserne med formlen I og derivater af forbindelser med formlen VII, hvori A’ betyder 5 C(2-4)-alkenylen eller C(2-4)-alkanyliden med dobbeltbindingen knyttet til den heterocycliske ring ifølge fremgangsmåde a) gennemføres eksempelvis ved katalytisk hydrogenering.
Kondensationen ifølge fremgangsmåde b) af en forbindelse 10 med formlen VIII med en forbindelse med formlen X, f.eks. triethylphosphit, gennemføres eksempelvis ved opvarmning i et indifferent opløsningsmiddel og under de for en Michaelis-Arbuzov-reaktion kendte betingelser, jævnfør Angew. Chem. Int. Ed. 16, 477 (1977) og Chem. Rev. 81, 415 15 (1981). På tilsvarende måde fører kondensation med f.eks.
phosphortrichlorid og efterfølgende hydrolyse til en forbindelse med formlen I.
Kondensationen ifølge fremgangsmåde b) af en forbindelse med formlen VIII med en forbindelse med formlen IX, f.eks.
20 diethylphosphonat (diethylphosphit), gennemføres i et stærkt basisk medium, f.eks. i nærværelse af et alkalimetal, f.eks. natrium, et alkalimetalhydrid, f.eks. natriumhydrid, eller et alkalimetalalkoxid, f.eks. kalium-t-butoxid, i et indifferent opløsningsmiddel, 25 f.eks. toluen eller dimethylformamid.
Udgangsmaterialerne med formlen VIII er enten kendte forbindelser eller kan fremstilles under anvendelse af kendte fremgangsmåder, f.eks. ved reduktion af de tilsvarende pyridinylforbindelser.

Benzyloxycarbonyl eller nitrobenzyloxycarbonyl kan omdannes til carboxy ved katalytisk hydrogenering, nitrobenzyloxycarbonyl tillige ved omsætning med kemiske reduktionsmidler, f.eks. natriumdi thionit eller zink og i en 10 carboxylsyre. Desuden kan tert-butyloxycarbonyl spaltes med trifluoreddikesyre.
Acetyl kan oxidativt spaltes til carboxy, idet der først foretages omdannelse til trihalogenacetyl, f.eks. tribrom-eller triiodacetyl, ved omsætning med f.eks. natriumhypo-15 bromit, hvorpå der foretages spaltning med f.eks. en vandig base, såsom vandig natriumhydroxid.
Formyl, dilavalkoxymethyl eller alkylendioxymethyl (formyl beskyttet i form af en acetal), f.eks. dimethyl-acetalen, oxideres f.eks. med sølvnitrat, pyridiniumdichromat eller 20 ozon til carboxy.
Vinyl kan omdannes til carboxy ved ozono lyse til formyl, som derpå oxideres til carboxy.
Hydrolyse af trialkoxymethyl til carboxy gennemføres fordelagtigt med uorganiske syrer, f.eks. hydrogen-25 halogenidsyre eller svovlsyre. Hydrolyse af etherificeret hydroxymethyl til hydroxymethyl gennemføres fortrinsvis med opløsninger af uorganiske syrer, f.eks. en hydrogen-halogenidsyre. Hydroxymethyl oxideres derpå til carboxy med et oxidationsmiddel, f.eks. pyridiniumdichromat.
30 Halogenmethyl kan også omdannes til de tilsvarende carbox-aldehyder med f.eks. dimethylsulfoxid i nærværelse af tri-ethylamin og sølvtetrafluorborat eller med chromtrioxid og DK 167811 B1 15 pyridin i methylenchlorid.
Omdannelsen af cyano til C(l-4)-alkoxycarbonyl gennemføres fordelagtigt ved først at foretage behandling med en 5 lavalkanol, f.eks. vandig ethanol, i nærværelse af en stærk syre, f.eks. saltsyre, fortrinsvis ved tilbagesvalingstemperatur, hvorpå der hydrolyseres med vand.
Endvidere gennemføres omdannelsen af cyano til carbamoyl fortrinsvis ved behandling med et alkalimetalhydroxid, 10 f.eks. fortyndet natriumhydroxid, og hydrogenperoxid, fortrinsvis ved stuetemperatur.
C(l-4)-alkoxycarbonyl kan amideres med ammoniak, monoeller di-C(l-4)-alkylaminer, f.eks. methylamin eller dimethylamin i et indifferent opløsningsmiddel, f.eks. en 15 lavalkanol, såsom butanol, til usubstitueret, N-mono-eller N,N-di-C(l-4)-alkylcarbamoyl.
Forbindelserne ifølge opfindelsen kan således også omdannes til andre forbindelser ifølge opfindelsen, f.eks. ved omdannelse af funktionelle grupper, på i og for sig kendt 20 måde.
Omdannelse af carboxylsyreestere eller -amider til carboxylsyrer gennemføres eksempelvis fordelagtigt ved hydrolyse med uorganiske syrer, såsom hydrogenhalogenid-syre eller svovlsyre, eller med vandige baser, fortrinsvis 25 alkalimetalhydroxider, såsom lithium- eller natrium hydroxid.
Fri carboxylsyre kan esterificeres med C(l-4)-alkanoler, såsom ethanol, i nærværelse af en stærk syre, f.eks. svovlsyre, eller diazo-C(1-4)-alkaner, f.eks. diazomethan, 30 i et opløsningsmiddel, såsom ethylether, fordelagtigt ved stuetemperatur til dannelse af de tilsvarende C(l-4)-alk-ylestere.

Endvidere kan de fri carboxylsyrer omdannes ved behandling med et reaktionsdygtigt mellemprodukt, f.eks. et acylhal-ogenid, såsom syrechloridet, eller et blandet anhydrid, 5 f.eks. afledt af et C(l-4)-alkylhalogencarbonat, såsom ethylchlorformiat, med ammoniak eller N-mono- eller N,N-di-C(1-4)-alkylaminer i et indifferent opløsningsmiddel, såsom methylenchlorid, fortrinsvis i nærværelse af en basisk katalysator, såsom pyridin, til forbindelser, hvori 10 r! betyder usubstitueret, N-mono- eller N,N-di-C(1-4)-alkylcarbamoyl.
Phosphonsyreestere kan omdannes til de tilsvarende phosphonsyrer ved omsætning med syre, såsom saltsyre eller hydrogenbromidsyre i iseddike, eller med bromtrimethyl-15 silan, jævnfør J. Chem. Soc. Chem. Comm. 1979, 739.
Benzylestere kan omdannes til syrerne ved hydrogenolyse.
Phosphonsyrer omdannes til C(l-4)-alkylester'e, f.eks. ved kondensation med et C(l-4)-alkylhalogenid, fortrinsvis i et basisk vandfrit medium, f.eks. i nærværelse af tri-20 ethylamin.
Forbindelserne, hvori den heterocycliske ring betyder dihydropyrrolyl, omdannes til de tilsvarende pyrrolidin-ylforbindelser, f.eks. ved katalytisk hydrogenering.
De ovenfor omtalte omsætninger gennemføres under anven-25 delse af standardmetoder, i nærværelse eller fraværelse af fortyndingsmidler, fortrinsvis sådanne, som er indifferente over for reaktanterne og som er opløsningsmidler for disse, af katalysatorer, af kondensationsmidler eller andre midler og/eller i en indifferent gasatmosfære, ved 30 lave temperaturer, ved stuetemperatur eller ved højere temperaturer, fortrinsvis ved kogepunktet for de anvendte opløsningsmidler, og ved atmosfæretryk eller ved overtryk.
De foretrukne opløsningsmidler, katalysatorer og reaktionsbetingelser fremgår af de efterfølgende eksempler.

Opfindelsen omfatter endvidere sådanne udførelsesformer for fremgangsmåden ifølge opfindelsen, hvor udgangsmaterialerne dannes under reaktionsbetingelserne eller 5 hvor reaktanterne anvendes i form af deres salte eller optisk rene antipoder.
Der anvendes hovedsagelig sådanne udgangsmaterialer ved omsætningerne, som fører til dannelsen af de ovenfor anførte særligt foretrukne forbindelser.
10 Afhængigt af valget af udgangsmaterialer og fremgangsmåder foreligger forbindelserne ifølge opfindelsen i form af en af de mulige isomerer eller blandinger deraf, f. eks., afhængigt af antallet af asymmetriske carbonatomer, som rene optiske isomerer, såsom antipoder, eller som blan-15 dinger af optiske isomerer, såsom racemater eller som blandinger af diastereoisomerer eller af geometriske isomerer. Disse mulige isomerer eller blandinger deraf er omfattet af den foreliggende opfindelse. Visse specielle isomerer er foretrukket som beskrevet ovenfor.
20 Dannede blandinger af diastereoisomerer eller blandinger af racemater kan adskilles på basis af bestanddelenes fysi-kemiske forskelle på i og for sig kendt måde i de rene isomerer, diastereoisomerer, racemater eller geometriske isomerer, f.eks. ved kromatografi og/eller frak-25 tioneret krystallisation.
Dannede racemater kan adskilles i de optiske antipoder ved anvendelse af kendte fremgangsmåder, f.eks. ved omsætning af et surt slutprodukt med en optisk aktiv base, som danner salte med den racemiske syre, og adskillelse af de 30 på denne måde dannede salte, f.eks. ved fraktioneret krystallisation i de diastereoisomere salte, hvorfra de optisk aktive frie carboxylsyreantipoder eller phosphon-syreantipoder kan frigøres ved syrning. De basiske racemiske produkter kan ligeledes adskilles i de optiske UK lb/ΰΊΊ bl 18 antipoder, f.eks. ved adskillelse af de diastereoisomere salte deraf, med en optisk aktiv syre, hvorefter den optisk aktive basiske forbindelse frigøres ved behandling 5 med en standardbase. Racemiske forbindelser ifølge opfindelsen kan således adskilles i de optiske antipoder, f.eks. ved fraktioneret krystallisation af d- eller l-(tartrat, mandelat, camphersulfonat) eller af d- eller l-(o:-methylbenzylamin, cinchonidin, cinchonin, quinin, 10 quinidin, ephedrin, dehydroabietylamin, brucin eller strychnin) salte. De sure forbindelser ifølge opfindelsen kan også resolveres ved adskillelse af diastereomere ester- eller amidderivater fremstillet ud fra en optisk aktiv alkohol eller amin, hvorpå den resolverede optisk 15 aktive forbindelse gendannes. Fordelagtigt isoleres den mest aktive af de to antipoder.
Til slut fås forbindelserne ifølge opfindelsen enten på fri form eller i form af et salt. En dannet base kan omdannes til et tilsvarende syreadditionssalt, fortrinsvis 20 under anvendelse af en terapeutisk anvendelig syre eller et anionbytterpræparat, eller dannede salte kan omdannes til de tilsvarende frie baser, f.eks. under anvendelse af en stærkere base, såsom et metal- eller ammoniumhydroxid eller et basisk salt, f.eks. et alkalimetalhydroxid eller 25 -carbonat, eller et kationbytterpræparat, eller et alkylenoxid, såsom propylenoxid. En forbindelse ifølge opfindelsen med en fri carboxylgruppe eller phosphonsyre-gruppe kan således omdannes til det tilsvarende metaleller ammoniumsalt. Disse salte eller andre salte, f.eks.
30 prior aterne, kan også anvendes til rensning af de dannede baser, idet baserne omdannes til salte, saltene isoleres, og baserne frigøres fra saltene.
Som følge af det nære slægtskab mellem de fri forbindelser og forbindelserne på saltform skal der ved en fri forbin-35 delse også forstås et tilsvarende salt og omvendt, forudsat at dette er muligt eller hensigtsmæssigt under de DK 167811 B1 19 foreliggende omstændigheder.
Forbindelserne, herunder deres salte, kan også fås i form af hydraterne, eller de kan indeslutte andre opløsnings-5 midler anvendt til krystallisationen.
De farmaceutiske præparater ifølge opfindelsen er egnede til enteral, såsom oral eller rektal, transdermal og parenteral indgift til mennesker og pattedyr til blokering af den N-methyl-D-aspartatsensitive excitatoriske amino-10 syrereceptor og til behandling af sygdomme, som reagerer på blokering af den N-methyl-D-aspartatsensitive excitatoriske aminosyrereceptor, såsom konvulsive tilstande og angsttilstande. De farmaceutiske præparater ifølge opfindelsen er ejendommelige ved, at de indeholder en 15 forbindelse ifølge opfindelsen i blanding med eller i forbindelse med et eller flere farmaceutisk acceptable bærestoffer.
De farmakologisk virksomme forbindelser ifølge opfindelsen er nyttige til fremstilling af farmaceutiske præparater 20 indeholdende en effektiv mængde af forbindelserne sammen med eller i blanding med strækkemidler eller bærestoffer, der er egnede enten til enteral eller parenteral indgift.
Der foretrækkes tabletter og gelatinekapsler, som indeholder den aktive bestanddel sammen med a) fortyndings-25 midler, f.eks. lactose, dextrose, saccharose, mannitol, sorbitol, cellulose og/eller glycin, b) smøremidler, f.eks. siliciumdioxid, talkum, stearinsyre eller dens magnesium- eller calciumsalt og/eller polyethylenglycol, for tabletters vedkommende tillige c) bindemidler, f.eks.
30 magnesiumaluminiumsilicat, stivelsesklister, gelatine, tragacanth, methylcellulose, natriumcarboxymethylcellulose og/eller polyvinylpyrrolidon, om nødvendigt d) desintegreringsmidler, f.eks. stivelse, agar, alginsyre eller dens natriumsalt eller bruseblandinger og/eller e) absorb-35 tionsmidler, farvestoffer, smagsstoffer og sødemidler.

Injektionspræparater er fortrinsvis vandige isotoniske opløsninger eller suspensioner, og suppositorier fremstilles fordelagtigt ud fra fedtemulsioner eller -suspen-5 sioner. Disse præparater kan være steriliserede og/eller indeholde hjælpestoffer, såsom konserveringsmidler, stabiliseringsmidler, fugtemidler eller emulgeringsmidler, opløsningsfremmende stoffer, salte til regulering af det osmotiske tryk og/eller puffere. De kan desuden også 10 indeholde andre terapeutisk værdifulde stoffer. Præparaterne fremstilles under anvendelse af konventionelle blandings-, granulerings- eller overtrækningsmetoder og indeholder fra ca. 0,1 til 75%, fortrinsvis fra ca. 1 til 50% aktivt stof.
15 Egnede præparater til transdermal indgift indeholder en effektiv mængde af en forbindelse med formlen I med bærestof. Fordelagtige bærestoffer omfatter absorberbare farmakologiske acceptable opløsningsmidler, der understøtter overgangen gennem huden hos det behandlede indi-20 vid. Transdermale præparater har karakteristisk form af en bandage, som omfatter et støttelag eller bagsidelag, et reservoir, som indeholder forbindelsen eventuelt med bærestoffer, eventuelt et hastighedskontrollerende spærrelag til afgivelse af forbindelsen til patientens hud med 25 reguleret eller forudbestemt hastighed over et udstrakt tidsrum, og midler til at fæstne præparatet på huden.
Til blokering af de N-methyl-D-aspartatsensitive excita-toriske aminosyrereceptorer i pattedyr og behandling af sygdomstilstande i pattedyr, f.eks. sådanne, der reagerer 30 på blokering af den N-methyl-D-aspartatsensitive excita-toriske aminosyrereceptor, såsom konvulsive tilstande og angsttilstande, anvendes en effektiv mængde af en forbindelse ifølge opfindelsen som farmakologisk aktivt stof, fortrinsvis i form af de ovenfor omtalte farmaceutiske 35 præparater. Den indgivne dosis af den aktive forbindelse afhænger af den pågældende varmblodede dyreart, legems- vægt, alder og individuel tilstand samt af indgifts måden.

Eksempel 1
En blanding af 1,19 g 4-(diethylphosphonomethyl)-2-pyridincarboxamid og 0,75 g platinoxid i 30 ml eddikesyre hydrogeneres ved 300 kPa i 6 timer ved stuetemperatur.
15 Reaktionsblandingen filtreres, og opløsningsmidlet fjernes i vakuum. Remanensen opløses i methylenchlorid, og opløsningen vaskes med et lille rumfang mættet natriumhydrogen-carbonatopløsning. Efter tørring med magnesiumsulfat fås en methylenchloridopløsning af en 2:l-blanding af cis og 20 trans 4-(diethylphophonomethyl)-2-piperidincarboxamid. I løbet af 3 dage ved stuetemperatur isomeriseres denne blanding til den mere stabile cis-isomer. Opløsningsmidlet fjernes i vakuum, hvorved man får cis-4-(diethyl-phosphonomethyl)-2-piperidincarboxamid i form af en olie, 25 som omdannes til hydrochloridet, som smelter ved 155-158°C.
Udgangsmaterialet fremstilles på følgende måde: En blanding af 2,2 g 4-(diethylphosphonomethyl)pyridin [Roczniki Chem. 38, 625 (1964), Chem. Abst. 61, 10703], 30 2,2 g m-chlorperbenzoesyre og 30 ml chloroform omrøres ved stuetemperatur i 16 timer. Opløsningsmidlet fjernes i vakuum, og der tilsættes vand. Det vandige lag vaskes med ether, hvorefter vandet fjernes i vakuum, og man får 4-(diethylphosphonomethyl)pyridin-N-oxid.

En blanding af 1,2 g 4-(diethylphosphonomethyl)pyridin-N-oxid, 1,35 ml triethylamin og 2,6 ml trimethylsilyl-cyanid opvarmes til 90°C i 1 time. De flygtige bestand-5 dele fjernes i vakuum, og remanensen opløses i ethyl-acetat, hvorpå opløsningen vaskes med et lille rumfang vandig natriumhydrogencarbonatopløsning. Efter tørring med magnesiumsulfat fjernes opløsningsmidlet i vakuum, hvorved man får 2-cyano-4-(diethylphosphonomethyl )pyridin i form 10 af en olie.
En blanding af 1,10 g 2-cyano-4-(diethylphosphonomethyl)-pyridin og 4,5 ml koncentreret svovlsyre opvarmes til 90°C i 5 minutter. Reaktionsblandingen hældes på is, og der neutraliseres med 10%'s natriumhydroxidopløsning. Det 15 dannede bundfald isoleres, og man får 4-(diethylphosphono-methyl)-2-pyridincarboxamid, som smelter ved 140-142°C.
Eksempel 2
En opløsning af 3,0 g cis-4-(diethylphosphonomethyl)-2-piperidincarboxamid i 90 ml 20%’s saltsyre opvarmes 20 under tilbagesvaling med omrøring i 16 timer. Efter fjernelse af opløsningsmidlet krystalliseres remanensen fra ethylacetat/ethanol, hvorved man får cis-4-phosphono-methyl-2-piperidincarboxylsyre, som smelter ved 284-285°C (sønderdeling).
25 Eksempel 3
En blanding af 330 mg 4-phosphonomethyl-2-pyridin-carboxylsyre, 150 mg platinoxid, 20 ml eddikesyre og 100 ml vand hydrogeneres ved 300 kPa ved stuetemperatur i 20 timer. Opløsningsmidlet fjernes i vakuum, hvorved man 30 får en 2:l-blanding af cis- og trans-4-phosphonomethyl-2-piperidincarboxylsyre. Remansen opløses i saltsyre, og opløsningsmidlet fjernes under vakuum, hvorved man får en 2:l-blanding af cis- og trans-4-phosphonomethyl-2-piper- DK 167811 B1 23 idin-carboxylsyre-hydrochlorid med smp. 145°C (sønderdeling) .
Udgangsmaterialet fremstilles på følgende måde: En blan-5 ding af 350 mg 2-cyano-4-diethylphosphonomethylpyridin og 10 ml 20%'s saltsyre tilbagesvales i 16 timer. Efter fjernelse af opløsningsmidlet i vakuum tritureres remanensen med 95%'s ethanol, hvorved man får 4-phosphonomethyl-2-pyridincarboxylsyre, som smelter ved 265-268°C (sønder-10 deling).

Ved omsætning af 3-(diethylphosphonomethyl)pyridin (Biochemistry 19, 3400) med m-chlorperbenzoesyre og derpå med trimethylsilylcyanid fås både 3-(diethylphosphono-methyl)-2-cyanopyridin og 5-(diethylphosphonomethyl)-2-cyanopyridin,som er udgangsmaterialerne for forbin-15 delserne fremstillet ovenfor under a) og b).
2. På tilsvarende måde omdannes 3-(2-diethylphosphono-ethyl)pyridin (Biochemistry 19, 3400) til de tilsvarende udgangsmaterialer for forbindelserne fremstillet ovenfor under c) og d), og 4-(2-diethylphosphonoethyl)pyridin 20 (Chem. Abst. 8j), 35665) omdannes til 4-(2-diethyl-phos- phonoethyl-2-cyanopyridin, som er udgangsmaterialet for den ovenfor under e) fremstillede forbindelse.
3. Ved omsætning af 3-(4-pyridinyl)propylchlorid med diethylphosphit (diethylphosphonat) og natriumhydrid i 25 toluen fås 4-(3-diethylphosphonopropyl)pyridin, som omdannes til 4-(3-diethylphosphonopropyl)-2-cyanopyridin under anvendelse af den ovenfor beskrevne fremgangsmåde. Sidstnævnte er udgangsmaterialet for den under f) fremstillede forbindelse.
30 4. En blanding af 1,0 g 4-(diethylphosphonomethyl)pyridin og 0,5 g 50%'s natriumhydrid i 4 ml dimethyl formamid og DK 167811 B1 25 16 ml tetrahydrofuran opvarmes under tilbagesvaling i 30 minutter. Der tilsættes 0,52 ml methyliodid, og opvarmningen med tilbagesvaling fortsættes i 30 minutter.
5 Derefter inddampes reaktionsblandingen til tørhed, og remanensen kromatograferes på silicagel med methanol/ methylenchlorid (1:10) som elueringsmiddel, hvorved man får 4-(l-methyl-l-diethylphosphonoethyl)pyridin, som omdannes til 2-cyano-4-(l-methyl-l-diethylphosphono-10 ethyl )pyridin, der er udgangsmaterialet for den ovenfor under g) fremstillede forbindelse.
5. Ved omsætning af 4-(diethylphosphonomethyl )pyridin på samme måde som beskrevet ovenfor med 1 molækvivalent methyliodid fås 4-(l-diethylphosphonoethyl)pyridin, som er 15 udgangsmaterialet for den ovenfor under h) fremstillede forbindelse.
Eksempel 5 a) En opløsning af 370 mg cis-4-phosphonomethyl-2-piperidincarboxylsyre i 15 ml mættet ethanolisk saltsyre 20 opvarmes med tilbagesvaling i 16 timer. Opløsningsmidlet fjernes i vakuum. En opløsning af remanensen i ethanol omsættes med 0,21 ml propylenoxid, og blandingen inddampes til tørhed, hvorved man får cis-4-phosphonomethyl-2-piperidincarboxylsyre-ethylester, smp. 230-235°C (sønder-25 deling).

Blandingen inddampes til tørhed, remanensen opløses i vand, og opløsningen inddampes til tørhed. En opløsning af det dannede faste stof i ethanol behandles med 0,21 ml propylenoxid, hvorved man får 15 cis-4-phosphonomethyl-2-piperidincarboxamid, smp.

Eksempel 7
En blanding af 357 mg cis-l-benzyloxycarbonyl-4-phosphono-methyl-2-piperidincarboxylsyre, 550 mg diisopropylethyl-20 amin og 600 mg chlormethylpivalat i 2 ml dimethyl formamid opvarmes til 80°C i 2 timer. Reaktionsblandingen inddampes til tørhed ved 70°C/13 Pa. Remanensen opløses i ethyl-acetat, opløsningen vaskes med vand og inddampes til tørhed, hvorved man får cis-l-benzyloxycarbonyl-4-[ di-25 (pivaloyloxymethyl)phosphonomethyl ]-2-piperidincarboxyl-syre-pivaloyloxymethylester (efter kromatografi på silicagel med ether/methylendichlorid som elueringsmid-del). Produktet opløses i 40 ml ethanol, og der hydrogeneres i nærværelse af 10%'s palladium på trækul, hvor-30 ved man får cis-4-[ di (pivaloyloxymethyl )phosphonomethyl ]-2-piperidincarboxylsyre-pivaloyloxymethylester.

Eksempel 8
En blanding af 2,0 g 4-(diethylphosphonomethyliden)-N-t-butoxycarbonylpyrrolidin-2-carboxy1syre-methyl- og ethylester og 800 mg 10%'s palladium på trækul som 10 katalysator i 30 ml ethanol hydrogeneres ved 300 kPa ved stuetemperatur i 18 timer. Reaktionsblandingen filtreres til fjernelse af katalysatoren, hvorpå denne vaskes med methylenchlorid, og de kombinerede filtrater inddampes til tørhed, hvorved man får en blanding af N-t-butoxy-carbo-15 nyl-4-diethylphosphonomethylpyrrolidin-2-carboxylsyre-methyl- og ethylester.
Det ovenfor fremstillede produkt omsættes med 10 ml tri-fluoreddikesyre i 5 minutter, hvorpå trifluoreddikesyren fjernes under vakuum. En opløsning af remanensen i 30 ml 20 6 N saltsyre opvarmes under tilbagesvaling i 6 timer, hvorpå den inddampes til tørhed. Remanensen opløses i 15 ml ethanol, og der tilsættes 1 ml propylenoxid. Det dannede bundfald frafiltreres, hvorved man får 4-phos-phonomethylpyrrolidin-2-carboxylsyre (4-phosphonometh- 25 ylprolin), iH-NMR (D2O): 4,53, 3,82, 2,88, 2,80, 2,0 ppm.
Udgangsmaterialet fremstilles på følgende måde: En blanding af 10,0 g 4-hydroxyprolin, 13 ml 6 N natriumhydroxidopløsning og 19,0 g di-t-butylcarbonat omrøres 30 kraftigt ved stuetemperatur i 24 timer. Der tilsættes 6,5 ml koncentreret saltsyre, og reaktionsblandingen ekstraheres med 30 ml chloroform. Chloroformfasen hældes bort, hvorpå olien isoleres fra det vandige lag, og man får N-t-butoxycarbonyl-4-hydroxyprolin. En opløsning heraf DK 167811 B1 28 1 70 ml methanol omsættes med etherisk diazomethan (fremstillet ud fra 20 g N-methyl-N-ni trosourinstof, 60 ml 40%'s vandig kaliumhydroxidopløsning og 200 ml ether, 5 hvorved man får N-t-butoxycarbonyl-4-hydroxyprolin-methylester.
En blanding af 12,7 g N-t-butoxycarbonyl-4-hydroxy-prolin-methylester og 44,7 g pyridiniumchlorchromat i 150 ml methylenchlorid omrøres ved stuetemperatur i 10 30 timer, hvorpå den fortyndes med 200 ml ether og filtreres gennem " Florisil" under vaskning med 500 ml ether. Opløsningen inddampes til tørhed, og remanensen renses ved flashkromatografi på silicagel under anvendelse af ethylacetat/hexan (4:6) som elueringsmiddel, hvorved 15 man får N-t-butoxycarbonyl-4-oxoprolin-methylester.
En opløsning af 2,28 M n-butyllithium (4,6 ml) sættes ved -78°C til en opløsning af 3,07 g bis(diethylphono)methan i 50 ml tør tetrahydrofuran. Efter 5 minutters forløb tilsættes en opløsning af 2,5 g N-t-butoxycarbonyl-4-20 oxoprolin-methylester. Reaktionsblandingen opvarmes under tilbagesvaling i 20 timer. Blandingens rumfang formindskes til 20 ml, og produktet fordeles mellem methylenchlorid og 2 N saltsyre. Methylenchloridekstrakten tørres og inddampes til tørhed, og remanensen renses ved flashkromato- 25 grafi på silicagel under eluering med methylenchlorid/ methanol (95:5), hvorved man får en blanding af 4-(di-ethylphosphonomethyliden) -N-t-butoxycarbonylpyrrolidin-2-carboxylsyre-methyl- og ethylester.

Eksempel 10
En opløsning af 2,0 g cis-l-benzyloxycarbonyl-4-phos-phonomethyl-2-piperidincarboxylsyre i 10 ml methylen-15 chlorid omsættes med etherisk diazomethan, indtil der opstår en blivende gul farve. Efter fjernelse af opløsningsmidlet opvarmes triesteren ved 50°C i 10 ml tetra-hydrofuran og 10 ml 1 N natriumhydroxidopløsning. Efter syrning ekstraheres produktet med ether, hvorefter det 20 organiske lag tørres og inddampes, hvorved man får cis -1 -benzy loxycarbony 1 -4-dimethy lphosphonomethyl - 2 -piperidincarboxylsyre.
Eksempel 11
En opløsning af 1,0 g cis-l-benzyloxycarbonyl-4-di-25 methylphosphonomethyl-2-piperidincarboxylsyre i 30 ml ethanol hydrogeneres ved 300 kPa over 500 mg 10%'s palladium på kul som katalysator i 30 ml ethanol. Ved filtrering og fjernelse af opløsningsmidlet får man cis -4- dimethylphosphonomethyl - 2 -piperidincarboxy 1 syre i 30 form af et amorft fast stof.

En opløsning af 3,38 g cis-l-benzyloxycarbonyl-4-di-methylphosphonomethyl-2-piperidincarboxylsyre i 35 ml 5 methylenchlorid omsættes med 1,42 g 1,1'-carbony1di-imidazol og 1,3 g ( + )-o!-methylbenzylamin. Efter omrøring i 16 timer vaskes reaktionsblandingen med 1 N saltsyre og mættet natriumhydrogencarbonatopløsning, hvorpå den tørres, og opløsningsmidlet fjernes. Ved adskillelse af de 10 dannede diastereomerer ved højtryksvæskekromatografi (HPLC) under anvendelse af en Ci8~søjle med omvendt fase og acetonitril/vand (2:3) som elueringsmiddel fås de to diastereomerer af cis-l-benzyloxycarbonyl-4-dimethyl-phosphonomethyl-2-piperidin-(N-l-phenylethyl)carboxamid.
15 Ved hydrogenering af den først eluerede diastereomer i ethanol med palladium på trækul som katalysator i 4 timer ved 300 kPa fås det tilsvarende cis-4-dimethylphosphono-methyl-2-piperidin-(N-l-phenylethyl)carboxamid. Ved behandling med 6 N saltsyre under tilbagesvaling i 20 24 timer fås (-)-cis-4-phosphonomethyl-2-piperidin- carboxylsyre, [ a ]§5 = -5,93° (H2O), som er den enantiomer af forbindelsen ifølge eksempel 2 med størst farmakologisk virkning.
Den tilsvarende (+)-cis-4-phosphonomethyl-2-piperidin-25 carboxylsyre, som er den enantiomer, som har mindre aktivitet, fremstilles på tilsvarende måde.

Udgangsmaterialet fremstilles på følgende måde: Til en suspension af 3,44 g benzyl oxycarbonylme thyl triphenyl-phosphoniumbromid i 20 ml tetrahydrofuran sættes 10 ml 5 0,65 M kaliumhexaraethyldisilazidopløsning i toluen efterfulgt af 1,5 g N-t-butoxycarbonyl-4-oxoprolin-methylester i 4 ml tetrahydrofuran. Efter tilbagesvaling i 72 timer afdampes opløsningsmidlet, og remanensen underkastes flashkromatografi med ethylacetat/hexan (2:8) til 10 dannelse af en blanding af cis- og trans-N-t-butoxy-carbonyl-4-(2-oxo-2-benzyloxyethyliden)pyrrolidin-2-carboxylsyre-methylester.
En opløsning af 0,58 g af den ovenfor fremstillede forbindelse i 15 ml methanol hydrogeneres ved 300 kPa i 3 ti-15 mer i nærværelse af 400 mg 10%r s Pd/C. Reaktionsblandingen filtreres gennem "Celite"®, hvorpå opløsningsmidlet af dampes, og remanensen flashkromatograferes med methanol/methylenchlorid (1:9), hvorved man får N-t-butoxycarbonyl-4-( carboxymethyl )pyrrolidin-2-carboxyl-20 syre-methylester.
Til 0,67 g N-t-butoxycarbonyl-4-(carboxymethyl)- pyrrolidin-2-carboxylsyre-methylester i 1 ml THF sættes ved 0°C 3,5 ml boran (1M) i THF. Efter omrøring i 0,5 timer tilsættes 1 ml vand, hvorefter opløsningsmidlet 25 afdampes, og remanensen flashkromatograferes med ethyl-acetat/hexan (75:25), hvorved man får N-t-butoxycarbonyl- 4- (2-hydroxyethyl )pyrrolidin-2-carboxylsyre-methylester.
Til en opløsning af 0,58 g N-t-butoxycarbonyl-4-(2-hydroxyethyl )pyrrolidin-2-carboxylsyre-methylester i 6 ml 30 methylenchlorid sættes ved 0°C 0,577 g triphenylphosphin og derpå 0,388 g N-bromsuccinimid. Efter omrøring i 0,5 timer afdampes opløsningsmidlet, og remanensen flashkromatograferes under anvendelse af ethylacetat/hexan (35:65), hvorved man får N-t-butoxycarbonyl-4-(2-brom-35 ethyl)pyrrolidin-2-carboxylsyre-methylester.

En blanding af 0,60 g N-t-butoxycarbonyl-4-(2-brom-ethyl )pyrrolidin-2-carboxylsyre-methylester og 3 ml triethylphosphit tilbagesvales i 2 timer. Overskud af 5 triethylphosphit afdestilleres under vakuum, og remanensen flashkromatograferes med methylenchlorid/methanol (95:5), hvorved man får N-t-butoxycarbonyl-4-(2-di-ethylphosphonoethyl)pyrrolidin-2-carboxylsyre-methyl-ester.
10 b) På tilsvarende måde fremstilles (+)-cis-4-(2- phos-phonoethyl)pyrrolidin-2-carboxylsyre, [a ]§5 = +23,7°C, idet man går ud fra D-prolinderivatet.
c) På tilsvarende måde fremstilles 3-(2-phosphono-ethyl)pyrrolidin-2-carboxylsyre.
Eksempel 14 a) En omrørt opløsning af 5,3 ml bis(diethylphosphono)-15 methan i 30 ml vandfrit tetrahydrofuran_ (THF) under nitrogen afkøles til -78°C, hvorpå der dråbevis tilsættes 8,5 ml 2,5 M butyllithium. Efter omrøring i 5 minutter tilsættes dråbevis og hurtigt en opløsning af 4,64 g l-ethoxycarbonyl-3-oxo-pyrrolidin-2-carboxylsyre-ethylester 20 [J. Am. Chem. Soc. 86, 5297 (1964)] i 30 ml tør THF.
Opløsningen opvarmes under tilbagesvaling i 18 timer, hvorpå den afkøles til stuetemperatur og koncentreres. Der tilsættes 40 ml 1 N saltsyre, og blandingen ekstraheres med 100 ml methylenchlorid. Den organiske fraktion vaskes 25 med 25 ml vand, tørres over natriumsulfat, filtreres og koncentreres. Rensning ved flashkromatografi under anvendelse af ethylacetat som opløsningsmiddel giver 1-ethoxy-carbonyl-3-(diethylphosphonomethyl)-2,5-dihydropyrrol-2-carboxylsyre-ethylester.

Eksempel 15
En opløsning af 2,75 g l-ethoxycarbonyl-3-(diethylphosphonomethyl ) -2, 5-dihydropyrrol-2-carboxylsyre-ethyl-5 ester i 35 ml ethanol og 1,5 g 10%’s Pd/C hydrogeneres ved 300 kPa, hvorved man får cis- og trans-l-ethoxycarbonyl- 3-(diethylphosphonomethyl)pyrrolidin-2-carboxylsyre-ethylester.
Eksempel 16 10 a) En opløsning af 2,7 g cis- og trans-1-ethoxycarbony1-3-(diethylphosphonomethyl)pyrrolidin-2-carboxylsyre-ethylester opvarmes under tilbagesvaling med 6 N saltsyre i 6 timer, hvorved man får 3-(phosphonomethyl)pyrrolidin-2-carboxyl-syre-hydrochlorid, 15 iH-NMR (D20): 4,78, 4,40, 3,95, 3,83, 3,72, 3,20, 3,01, 2,78, 2,69 ppm.
Claims (18)
1. Phosphonsyrer og derivater deraf, kendetegnet ved, at de har den almene formel RO-P-A-Hot (I),
2. Forbindelser ifølge krav 1, kendetegnet ved, at de har formlen A—P—OR* /x L i I 0R (II) γν H hvor den phosphonobærende kæde er bundet til 3-, 4- eller 15 5-stillingen i piperidinylringen, og R og R* hver for sig betyder hydrogen eller C(1-4)-alkyl, R^ betyder carboxy, carbamoyl, N-mono- eller N,N-di-C(l-4)-alkylcarbamoyl eller C(l-4)-alkoxycarbonyl, og A betyder C(l-4)-alkylen, og farmaceutisk acceptable salte deraf.
3. Forbindelser ifølge krav 2, kendetegnet ved, at de har formlen DK 167811 B1 (^H2)n—I—OR' /\ °R (III) • · I I Y'v H hvori n er et helt tal på 1, 2 eller 3, R og R' hver for sig betyder hydrogen eller C(l-4)-alkyl, R^ betyder carb-5 oxy, C(l-4)-alkoxycarbonyl eller carbamoyl, og farmaceutisk acceptable salte af sådanne forbindelser med en saltdannende funktionel gruppe.
4. Forbindelser ifølge krav 3, kendetegnet ved, at n er et helt tal på 1, 2 eller 3, R og R' betyder hydrogen 10 og r! betyder carboxy eller C(l-4)-alkoxycarbonyl, samt farmaceutisk acceptable salte deraf.

6. Forbindelse ifølge krav 4, kendetegnet ved, at den er cis-4-(3-phosphonopropyl)-2-piperidincarboxylsyre eller et farmaceutisk acceptabelt salt deraf.
7. Forbindelse ifølge krav 4, kendetegnet ved, at den er cis-4-phosphonomethyl-2-piperidincarboxylsyre- 20 ethylester, samt farmaceutisk acceptable salte deraf.
8. Forbindelser ifølge krav 1, kendetegnet ved, at de har den almene formel \ 4—A—I—0R' (vi) Y 'V OR H hvor R og R' hver for sig betyder hydrogen eller C(l-4)-25 alkyl, R1 betyder carboxy, C(l-4)-alkoxycarbonyl, DK 16781 1 B1 carbamoyl eller N-mono- eller N,N-di-C(l-4)-alkylcarba-moyl, og A betyder C(l-4)-alkylen, og salte deraf.
9. Forbindelser ifølge krav 8, kendetegnet ved, at 5 den phosphonobærende gruppe er bundet i 3- eller 4-stil- lingen, R og R' betyder hydrogen, R1 betyder carboxy, C(l-4)-alkoxycarbonyl eller carbamoyl, og A betyder C(l-3)-alkylen, samt farmaceutisk acceptable salte deraf.

13. Farmaceutisk præparat, kendetegnet ved, at det indeholder en forbindelse ifølge et vilkårligt af kravene 20 2, 3, 5-7 og 9 i blanding med eller i forbindelse med et eller flere farmaceutisk egnede bærestoffer.
14. Farmaceutisk præparat, kendetegnet ved, at det indeholder en forbindelse ifølge et hvilket som helst af kravene 1, 4, 8 og 10 i blanding med eller i forbindelse 25 med et eller flere farmaceutisk egnede bærestoffer.
