Ikke-ioniske analoge af desoxyribonucleinsyrer (DNA) har betydning til undersøgelse af DNA-DNA- og DNA-protein-vekselvirkninger. Især har phosphonsyrees-tere af desoxyribonucleosider interesse som følge af 5 deres kemiske stabilitet, på grund af deres celletilgængelighed samt deres forøgede resistens mod nucleaser.
Hidtil har der været beskrevet fire forskellige synteseveje til fremstilling af nucleotidernes methylphosphonat-analoge: 10 1. Ogilvie m.fl. (M.J. Nemer og K.K. Ogilvie, Tetra hedron Lett. 21, side 4149 (1980)), fremstillede et fuldstændigt beskyttet uridyl-3',5'-uridin-methylphospho-nat ved Michaelis-Arbuzovsche omlejring af det tilsvarende phosphitmellemtrin. Denne reaktion (behandling med me-15 thyliodid i 20 timer ved 50°C) turde som følge af de her herskende drastiske betingelser ikke være alment anvendelig, da man f.eks. kan forvente en methylering af purin-baserne.

Denne metode har phosphotriestermetodens kendte fordele og ulemper, idet man specielt som ulempe kan nævne phosphor (V)-forbindelsens ringe reaktivitet.

Grossbruchhaus og H. Koster i Tetrahedron Lett. 24, 887 (1983), går også ud fra methyldichlorphosphan og syntetiserer nucleotidmethylphosphonaterne på 10 polymer bærer. En karakterisering af de opnåede produkter mangler endnu.
Medens man på den ene side hos methylphosphonsyredi-chloridet er udsat for, at det andet halogens reaktivitet for det meste er for ringe og derfor nødvendiggør en yder-15 ligere aktivering, er tilfældet hos phosphinsyredichloriderne tværtimod, at aktiviteten er for høj. Således forekommer der vanskeligheder ved håndteringen (ekstremt vandfrit medium) , og oven i købet dannes der uundgåeligt symmetrisk phosphorsyr1ingester.

Omsætningen af phosphonyleringsreagenset med den almene formel (II) med et egnet beskyttet nucleosid med den almene formel (III) sker i et moderat polært opløsningsmiddel, fortrinsvis chloroform, under udelukkel-5 se af fugtighed. Som hjælpebase for denne reaktion kan anvendes tertiære aminer, fortrinsvis ethyldiisopro-pylamin (Hiinig-base) . Oparbejdningen sker ved vandig ekstraktion og fældning af produkterne med den almene formel (IV) ved hjælp af et upolært opløsningsmiddel såsom petroleumsether eller pentan. De således udvundne phosphorsyrlingesteramider med den almene formel (IV) fremkommer som farveløse pulvere og kan karakteriseres ved spektroskopiske data såsom H-NMR, P-NMR eller UV- eller elementæranalyse. Yderligere kan de også ved direkte oxidation overføres til phosphonsyreesterami-derne med den almene formel

Forbindelserne med den almene formel (IV) er i tør 30 tilstand og opbevaret ved mindst -20°C stabile i mindst 1 måned, således som påvist ved P-NMR. Denne store bestandighed hos phosphorsyrlingesteramiderne er forbløffende og understreger denne metodes værdi. Deres universelle anvendelighed ved opbygningen af nucleosidernes phosphon-35 syrediestere viser sig ved omsætningen med egnede 3'-beskyttede nucleosider:

Herved opløses de 5'-beskyttede nucleosidphospho-nitter med den almene formel (IV) i et moderat polært opløsningsmiddel, fortrinsvis acetonitril, chloroform eller tetrahydrofuran, og blandes med nucleosidet med den 5 almene formel (V) (beskyttet i 3'-stillingen). Egnede beskyttelsesgrupper G på forbindelserne med den almene formel (V) er acylgrupper såsom benzoyl, acetyl, pivaloyl eller laevulonyl eller silylgrupper såsom tert.butyldimeth-ylsilyl. Reaktionen katalyseres med en syre, fortrinsvis 10 en azol eller et aminhydrochlorid. Særlig egnet er benzo-triazol. Det er bemærkelsesværdigt, at HPLC-analyse af det dannede produkt ikke viser tilstedeværelse af de symmetriske 5',5'-isomere og kun spor af de 3',3'-isomere phos-phonater.
15 Det labile mellemprodukt, phosphorsyrlingtriesteren med den almene formel (VI) oxideres direkte til phosphonat med den almene formel (I), hvor Z betyder oxygen. Ud over de hertil gængse oxidationsmidler, såsom dinitrogentetroxid eller iod, har peroxider, især vandfrit tert.butylhydroper-20 oxid, vist sig velegnede hertil. Reaktionen gennemføres fortrinsvis i et moderat polært opløsningsmiddel som fortrinsvis acetonitril eller chloroform. Især må man tage hensyn til den kendte (jvf. F.W. Hoffmann, R.G. Roth, T.C. Simmons, i J.
Amer. Chem. Soc. 80, 5937-40 (1958)) syrekatalyserede om-25 estering af diacylalkylphosphonitterne.
Karakteriseringen af forbindelserne (som til 31 dels allerede er kendte) sker ved hjælp af P-NMR og ^H-NMR samt ved chromatografiske sammenligninger med autentiske materialer.
30 Fremstillingen af forbindelserne med den almene formel (I), hvori Z betyder svovl eller selen, sker ved direkte omsætning af forbindelserne med den almene formel (VI) med elementært svovl eller selen. Indrøring i et polært opløsningsmiddel såsom tetrahydrofuran sammen 35 med den støkiometriske mængde svovl eller selen fører i gode udbytter til de tilsvarende thio- eller selenphos-

phonater med den almene formel (I). Karakteriseringen 31 sker foruden ved elementæranalyse også via P-NMR og 1H-NMR.
På grund af tilstedeværelsen af et asymmetri-5 centrum på nucleosiddelen og tilvejebringelsen af et nyt ved phosphoratomet foreligger phosphonaterne med den almene formel (I) som diastereomere blandinger (se tabel 6, isomerene 1 og 2). En variation af parametrene såsom opløsningsmiddel, temperatur og rækkefølgen for tilsæt-10 ningen indvirker kun i meget ringe omfang på isomerfor- holdet, som ligger i nærheden af det statistiske lsl-for-hold.

Fremstilling af udgangsmateriale.
h3c-p[n(ch3)2]2 I en trehalset 1000 ml kolbe med dråbetragt og mekanisk omrører anbringes 125 ml (1,9 mol) dimethylamin 20 i 400 ml vandfri diethylether, og under isafkøling tilsættes i løbet af 60 minutter 60 ml (0,40 mol) methyldi-chlorphosphan opløst i 200 ml vandfri ether. Efter omrøring i 2 timer ved stuetemperatur og i 1 time ved 50°C filtreres bundfaldet fra under beskyttende gas, eftervaskes 2 25 gange med 100 ml ether, og filtratet inddampes ved ca.
0,1 bar. Den tilbageblivende remanens destilleres hurtigt over ved 0,5 bar/124°C. En findestillation med en Vigreux-kolonne på 50 cm giver ved 64-65°C/65 mbar et udbytte på 36,6 g (66% af det teoretiske) af en farveløs 30 væske.
Analyse: Cl - <0,2% 31P-NMR (THF) i = 87 ppm 1H-NMR (CDC1J S = 1,23 ppm (d, 7Hz, P-CHJ S = 2,66 ppm (d, 7Hz, N(CH3)2 35 9

Eksempel a) 51-Tritylnucleosidet med formlen (III) (1 mmol) opløses i 6 ml vandfri chloroform under indifferent ni-trogenatmosfære, og hertil sættes 2 mmol H^CP[N(-CH3)2J2· Reaktionen er tilendebragt efter 12 timer ved stuetemperatur (omrøring) eller allerede efter 2 timer ved tilsætning af katalytiske mængder (på 0,1 mmol) collidinhydrochlorid. Derefter overføres opløsningen sammen med 100 ml methy-lenchlorid til en 250 ml skilletragt og rystes to gange med 50 ml mættet kogsaltopløsning (indeholdende 0,1 ml triethylamin). Den organiske fase tørres over vandfrit natriumsulfat og inddampes til et skum. Dette omrøres i 2 timer med 50 ml pentan. Resten suges fra, opløses i 2 ml diethylether og tildryppes langsomt til 50 ml godt 1 o omrørt pentan. Det fine bundfald filtreres og tørres, hvilket giver et udbytte på 85 til 95% af forbindelsen med den almene formel (IV) (se tabel 2 og 3).

Forbindelserne kan påvises direkte ved P-kernere- 20 sonansspektroskop! eller kan påvises efter oxidation med tert.butylhydroperoxid som phosphonsyreesteramider med den almene formel (VIII) (se tabellerne 4 og 5).

Indenfor P-NMR-spektret viser disse stoffer indtil 3% hydrolyseret produkt (nucleosidmethylphosphinat), 25 men ingen påviselig mængde af symmetrisk 3',3'-dinu- cleosidphosphonit. Herved vises overlegenheden i forhold til de hidtil kendte metoder, der konsekvent har leveret fra ca. 5 til ca. 10% af disse produkter. Som tørrede pulvere opbevares det dannede produkt ved -20°C, og der kan 30 ingen sønderdeling iagttages indenfor 1 måned.

Den yderligere rensning sker ved fældning således som ovenfor (se tabellerne 2 og 3).
Den under a) eller b) fremstillede forbindelse med 20 formlen IV tørres sammen med 0,20 mmol 3 *-O-benzoylthymidin og 0,80 mmol 1-H-benzotriazol i en rundbundet kolbe og opløses derpå i 1,0 ml tørt acetonitril. Indenfor et minut er reaktionen afsluttet, hvorved der opstår et meget luft- og syrelabilt phosphonit med formlen (VI), som, opløst i ace- 25
tonitnl eller tetrahydrofuran,direkte ved oxidation med 0,25 mmol vandfrit tert.butylhydroperoxid (ifølge H. Langhals, E. Fritz og J. Mergelsberg, Chem. Ber. 113. 3366 (1980)) overføres i phosphonatet med formlen (I), hvor Z
betyder oxygen, i et udbytte på 80 til 90%. (jvf. tabel 6).

Alternativt tilsættes 30 mg (0,95 mmol) svovl til 0,7 mmol af forbindelsen med formlen (VI) ved -20°C, idet der omrøres natten over ved stuetemperatur. For det meste er reaktionen allerede afsluttet efter nogle få timerDerefter tilsættes 20 ml chloroform, og den orga-35 niske fase rystes tre gange med vand. Efter tørring over

natriumsulfat og fjernelse af opløsningsmidlet fås et råprodukt, som renses ved chromatografi på kiselgel, og som giver et udbytte på 80 til 90% af forbindelsen med formlen (I) (jvf. tabel 6) .
Alternativt sættes 118 mg (1,5 mmol) sort selen 5 til 0,7 mmol af forbindelsen med formlen (VI), og der omrøres natten over. Efter oparbejdningen (gennemføres således som ovenfor anført) fås forbindelsen med formlen (I) i et udbytte på 60% (jvf. tabel 6) .
HPLC-analyse af reaktionsmroduktet (i tilfældet 10 Z = 0 ved sammenligning med autentisk reference ifølge P.O.P. T'so m.fl. Biochemistry 18, 5134 (1979)) viser et indhold på ca. 1% af 3' ,3'-phosphonaterne, men intet 5',5'-isomer.

Claims (8)
1. Fremgangsmåde til fremstilling af desoxyribo-nucleosidphosphonater med den almene formel
2. Fremgangsmåde ifølge krav 1, kendetegnet ved, at der anvendes forbindelser med formlerne (II)- (IV), hvori T betyder triphenylmethyl, p-anisoyldiphenylmethyl eller 5 di-(p-anisoyl)-phenylmethyl, B betyder 1-thyminyl, 1-(N-4-benzoylcytosinyl), 9-(N--6-benzoyladeninyl) eller 9-(N-2-isobutyroylguaninyl), og R betyder methyl, ethyl, phenyl eller benzyl.
3. Fremgangsmåde ifølge krav 1 eller 2, ken detegnet ved, at forbindelserne med formlen (II) og (III) omsættes ved en temperatur fra -80 til +100°C.
4. Fremgangsmåde ifølge krav 3, kendetegnet ved, at omsætningen finder sted ved -20°C til 0°C.
5. Fremgangsmåde ifølge et eller flere af de foregående krav, kendetegnet ved, at forbindelserne med formlerne (IV) og (V) omsættes ved en temperatur fra -20°C til +100°C.

6. Fremgangsmåde ifølge krav 5, kendeteg-20 net ved, at omsætningen finder sted ved stuetemperatur.
7. Fremgangsmåde ifølge et eller flere af de foregående krav, kendetegnet ved, at forbindelsen med formlen (VI) overføres oxidativt eller ved behandling med svovl eller selen i en forbindelse med formlen 25 (I) ved -80 til +100eC.
8. Fremgangsmåde ifølge krav 7, kendetegnet ved, at omsætningen finder sted ved en temperatur fra -20°C til stuetemperatur.
