Claims (32)
1. Verfahren zur Identifikation von Verbindungen mit dem
Potential, zur Behandlung von Neoplasmen eingesetzt zu werden, umfassend
den Nachweis der Cyclooxygenase-(COX)-Hemmwirkung der Verbindung sowie
den Nachweis der PDE5-Hemmwirkung der Verbindung,
wobei eine geringe COX-Hemmwirkung im Verhältnis zu einer hohen Hemmung der PDE5-Aktivität als Zeichen dafür gedeutet
wird, daß eine Verbindung das Potential besitzt, zur Behandlung von Neoplasmen eingesetzt zu werden.
2. Verfahren nach Anspruch 1, weiterhin umfassend
den Nachweis, ob die Verbindung das Wachstum von Tumorzellen in einer Kultur hemmt,
wobei die Hemmung des Wachstums von Tumorzellen als weiteres Zeichen dafür gedeutet wird, daß die Verbindung
das Potential besitzt, zur Behandlung von Neoplasmen eingesetzt zu werden.
3. Verfahren nach Anspruch 1, worin die COX-Hemmwirkung der Verbindung dadurch nachgewiesen wird, daß
die Verbindung mit gereinigter Cyclooxygenase in Berührung gebracht wird und
die Cyclooxygenase-Hemmung, falls eine solche eintritt,
gemessen wird,
wobei eine COX-Hemmwirkung von weniger als 25% bei einer Konzentration von etwa 100 &mgr;&Mgr; als Zeichen dafür gedeutet
wird, daß die Verbindung einer weiteren Prüfung auf das Potential, zur Behandlung von Neoplasmen eingesetzt zu
werden, unterzogen werden sollte.

4. Verfahren nach Anspruch 1, worin die COX-Heitunwirkung der
Verbindung dadurch nachgewiesen wird, daß
die Verbindung mit einer Zelle, die PGE-2 sezerniert, in
Berührung gebracht wird, und die Minderung der PGE-2-
Sekretion aus der Zelle, falls eine solche Minderung
eintritt, gemessen wird,
wobei eine Minderung der PGE-2-Sekretion in Korrelation mit
einer Minderung der Prostaglandinsynthetase-Aktivität steht.
5. Verfahren nach Anspruch 1, weiterhin umfassend
den Nachweis, ob die Verbindung in einer Tumorzelle den
Zelltod auslöst,
wobei ein Auslösen des Zelltods als weiteres Zeichen dafür gedeutet wird, daß die Verbindung das Potential besitzt,
zur Behandlung von Neoplasmen eingesetzt zu werden.
6. Verfahren nach Anspruch 5, weiterhin umfassend
den Nachweis, ob die Verbindung in einer Probe das Wachstum von Tumorzellen hemmt,
wobei die Hemmung des Wachstums von Tumorzellen als weiteres Zeichen dafür gedeutet wird, daß die Verbindung
dazu geeignet ist, zur Behandlung von Neoplasmen eingesetzt zu werden.
7. Verfahren nach Anspruch 5, worin die PDE-Aktivität dadurch nachgewiesen wird, daß
die Verbindung mit einer Zellkultur in Berührung gebracht und
die intrazelluläre zyklische GMP-Konzentration bestimmt wird,
wobei eine PDE-Hemmwirkung von mehr als 50% bei einer Konzentration von 10 &mgr;&Mgr; als Zeichen dafür gedeutet wird,
daß die Verbindung einer weiteren Prüfung auf das Potential, zur Behandlung von Neoplasmen eingesetzt zu
werden, unterzogen werden sollte.
8. Verfahren nach Anspruch 5, worin die PDErAktivität dadurch
nachgewiesen wird, daß
das Verhältnis zwischen der Konzentration von intrazellulärem zyklischem GMP und zyklischem AMP bestimmt
wird,
wobei eine mehr als dreifache Erhöhung des besagten Verhältnisses bei einer Konzentration von 10 &mgr;&Mgr; als Zeichen
dafür gedeutet wird, daß die Verbindung einer weiteren Prüfung auf das Potential, zur Behandlung von Neoplasmen
eingesetzt zu werden, unterzogen werden sollte.
9. Verfahren nach Anspruch 5, worin der Zelltod dadurch nachgewiesen wird, daß
die Verbindung mit einer Zellkultur in Berührung gebracht und
festgestellt wird, ob die Verbindung im Zytoplasma zu einer Erhöhung der Menge der fragmentierten DNS geführt hat,
wobei eine um mehr als zweifach erhöhte Stimulierung des Zelltods bei einer Konzentration von 100 um als weiteres
Zeichen dafür gedeutet wird, daß die Verbindung einer weiteren Prüfung auf das Potential, zur Behandlung von
Neoplasmen eingesetzt zu werden, unterzogen werden sollte.
10. Verfahren zur Auswahl von Verbindungen, die sich potentiell
zur Behandlung von Neoplasmen eignen, umfassend
den Nachweis der wachstumshemmenden Wirkung einer Verbindung,
den Nachweis der PDE5-Hemmwirkung der Verbindung sowie die Auswahl von Verbindungen, die eine wachstumshemmende
Wirkung sowie ein PDE5-Hemmwirkung aufweisen.
11. Verfahren nach Anspruch 10, weiterhin umfassend
die Identifikation von Verbindungen, bei denen der IC50-Wert für die wachstumshemmende Wirkung unter etwa 100 &mgr;&Mgr;
liegt, zur potentiellen Anwendung bei der Behandlung von Neoplasmen.
12. Verfahren nach Anspruch 10, weiterhin umfassend

den Nachweis, ob die Verbindung in einer Zelle den Zelltod auslöst, sowie
die Auswahl von Verbindungen, die den Zelltod auslösen.
13. Verfahren nach Anspruch 12, weiterhin umfassend
die Auswahl von Verbindungen, bei denen der EC50-Wert für
die apoptotische Aktivität unter etwa 100 &mgr;&Mgr; liegt.
14. Verfahren nach Anspruch 10, weiterhin umfassend
den Nachweis der COX-Hemmwirkung der Verbindung sowie die Auswahl von Verbindungen mit einer geringen COX-Hemmwirkung
im Verhältnis zur PDE5-Hemmwirkung.
15. Verfahren nach Anspruch 14, worin die COX-Hemmwirkung
dadurch nachgewiesen wird, daß
die Verbindung mit einer Cyclooxygenase in Berührung gebracht und
die Änderung.der Cyclooxygenase-Aktivität, falls eine
solche eintritt, gemessen wird,
wobei eine Minderung der Cyclooxygenase-Aktivität mit einer Minderung der Prostaglandinsynthetase-Aktivität in
Korrelation steht.
16. Verfahren nach Anspruch 14, worin die COX-Hemmwirkung der Verbindung dadurch nachgewiesen wird, daß
die Verbindung mit der Zelle, die PGE-2 sezerniert, in Berührung gebracht und
die Minderung der PGE-2-Sekretion aus der Zelle, falls eine solche eintritt, gemessen wird,
wobei eine Minderung der PGE-2-Sekretion mit einer Minderung der Prostaglandinsynthetase-Aktivität in
Korrelation steht.
17. Verfahren zur Identifikation von Verbindungen, die potentiell zur Verabreichung an Patienten geeignet sind, bei
denen eine prophylaktische oder chronische Behandlung von Neoplasmen erforderlich ist, umfassend
den Nachweis der COX-Hemmwirkung der Verbindungen,
den Nachweis der PDE5-Hemmwirkung der Verbindungen sowie die Identifikation dieser Verbindungen zur potentiellen
Anwendung in der Behandlung von Neoplasmen bei Patienten, bei denen eine solche Behandlung erforderlich ist, falls
die Verbindungen eine PDE5-Hemmwirkung aufweisen.
18. Verfahren nach Anspruch 17, worin die Selektivität der Verbindungen zur Hemmung des PDE5-Isoenzyms nachgewiesen
wird.
19. Verfahren nach Anspruch 17, weiterhin umfassend den Nachweis der wachstumshemmenden Wirkung der Verbindungen sowie die
Identifikation derjenigen Verbindungen mit einer Phosphodiasterase-Hemmwirkung, die wesentlich höher als die
COX-Hemmwirkung bei eine wesentliche wachstumshemmende Aktivität aufweisenden Konzentrationen ist.
20. Verfahren nach Anspruch 19, worin die wachstumshemmende Wirkung in einer Probe durch die Reduktion der Zahl der
Zellen nachgewiesen wird.
21. Verfahren nach Anspruch 17, worin die wachstumshemmende Wirkung durch den Grad bestimmt wird, in dem in einer Probe
der Zelltod ausgelöst wird.
22. Verfahren nach Anspruch 17, worin Verbindungen zwecks ihres potentiellen Einsatzes in der Behandlung von Neoplasmen bei
Patienten, bei denen eine solche Behandlung erforderlich ist, einer weiteren Identifikation unterzogen werden, falls
der Grad, in dem der Zelltod ausgelöst wird, den Grad, in dem die Nekrose ausgelöst wird, wesentlich überschreitet.
23. Verfahren zur Prüfung von Verbindungen mit dem Potential,
zur Behandlung von Neoplasmen eingesetzt zu werden, umfassend den Nachweis der PDE5-Hemmwirkung der Verbindung
sowie die Auswahl von Verbindungen mit einer Hemmwirkung, die einen vorbestimmten Wert überschreitet.
24. Verfahren nach Anspruch 23, weiterhin umfassend den Nachweis, ob die ausgewählten Verbindungen den Zelltod in
einer Zelle auslösen, sowie die weitere Auswahl von
Verbindungen mit einer den Zelltod auslösenden Wirkung, die einen vorbestimmten Wert überschreitet.
25. Verfahren nach Anspruch 24, weiterhin umfassend den Nachweis, ob die ausgewählten Verbindungen die
Prostaglandinsynthese hemmen, sowie die weitere Auswahl von Verbindungen mit einer Hemmwirkung, die unter einem
vorbestimmten Wert liegt.
26. Verfahren nach Anspruch 24, worin die PDE5-Aktivität durch Messung der intrazellulären zyklischen GMP und der zyklichen
AMP und durch den Nachweis, ob eine Erhöhung des Verhältnisses zwischen der zyklischen GMP und der zyklischen
AMP stattgefunden hat, bestimmt wird.
27. Verfahren zur Identifikation einer Verbindung mit dem Potential, zur Behandlung von Neoplasmen eingesetzt zu
werden, umfassend
den Nachweis der COX-1-Hemmwirkung der Verbindung sowie
den Nachweis der PDE5-Hemmwirkung der Verbindung, wobei eine geringe COX-1-Hemmwirkung und eine hohe Hemmung
der PDE5-Aktivität als Zeichen dafür gedeutet werden, daß eine Verbindung das Potential besitzt, zur Behandlung von
Neoplasmen eingesetzt zu werden.
28. Verfahren zur Identifikation einer Verbindung mit dem Potential, zur Behandlung von Neoplasmen eingesetzt zu
werden, umfassend
die Behandlung neoplastischer Zellen mit der zu prüfenden Verbindung bei einer Konzentration zwischen etwa 200 &mgr;&Mgr; bis
200 pM,
den Nachweis der intrazellulären Menge von cGMP in den behandelten Zellen sowie den Nachweis der intrazellulären
Menge von cAPM in den behandelten Zellen, wobei eine etwa dreifache oder noch höhere Steigerung des
Verhältnisses von cGMP zu cAMP in den behandelten Zellen im Vergleich zum Verhältnis von cGMP zu cAMP in unbehandelten
neoplastischen Zellen als Zeichen dafür gedeutet wird, daß die Verbindung das Potential besitzt, zur Behandlung von
Neoplasmen eingesetzt zu werden.

29. Verfahren nach Anspruch 28, worin eine intrazelluläre Menge von cGMP in den behandelten Zellen von mehr als 500 fmol/mg
Protein als Zeichen dafür gedeutet wird, daß die Verbindung das Potential besitzt, zur Behandlung von Neoplasmen
eingesetzt zu werden.
30. Verfahren nach Anspruch 28, worin eine intrazelluläre Menge von cAMP in den behandelten Zellen von weniger als 4000
fmol/mg Protein als Zeichen dafür gedeutet wird, daß die Verbindung das Potential besitzt, zur Behandlung von
Neoplasmen eingesetzt zu werden.
31. Verfahren nach Anspruch 30, worin eine intrazelluläre Menge von cAMP in den behandelten Zellen von weniger als 4000
fmol/mg Protein als Zeichen dafür gedeutet wird, daß die Verbindung das Potential besitzt, zur Behandlung von
Neoplasmen eingesetzt zu werden.
32. Verfahren zur Auswahl von Verbindungen, die potentiell dazu geeignet sind, zur Behandlung von Neoplasmen eingesetzt zu
werden, umfassend
den Vergleich der wachstumshemmenden Wirkung der Verbindung mit der PDE5-Hemmwirkung der Verbindung sowie
die Auswahl der Verbindungen, die eine wachstumshemmende Wirkung und eine PDE-5-Hemmwirkung aufweisen.
