Description
- Diese Erfindung betrifft ein Mittel zur Förderung von Knochenbildung, das ein Peptid oder Polypeptid umfasst, enthaltend die Aminosäuresequenz, bestehend aus ArgGlyAsp (nachfolgend bezeichnet als RGD-Sequenz), im Molekül. Weiterhin bezieht sich die Erfindung auf eine prophylaktische Verwendung für Knochenbrüche, die ein Mittel zur Förderung von Knochenbildung verwendet, umfassend das Peptid oder Polypeptid, enthaltend die RGD-Sequenz. Ebenfalls bezieht sich die Erfindung auf ein neues zyklisches Peptid, enthaltend die RGD-Sequenz. Und weiterhin bezieht sie sich auf ein Mittel zur Förderung von Knochenbildung, das eine Verbindung umfasst, dargestellt durch die allgemeinen Formeln (IX), (X) und (XII).
- Der Knochen ist aus dem äußeren Kortikalknochen und dem inneren Trabekularknochen aufgebaut. Die Funktion von Knochen im lebenden Körper ist eine vorgegebene Form als Skelett aufrechtzuerhalten und verschiedene anorganische Substanzen, wie Calcium, Phosphorsäure und dergleichen, zu speichern. Der Knochen kann offensichtlich als Gewebe mit geringer Variabilität auftreten, aber tatsächlich wird alter Knochen adsorbiert und stattdessen neuer Knochen gebildet. Dies wird in der Regel als Knochenneubildung bezeichnet. Die Knochenneubildung kann von einer Kopplung von Osteoklasten, die die Knochenadsorption kontrollieren, mit Osteoblasten, die die Knochenbildung kontrollieren, erreicht werden, da beide in erster Linie daran teilnehmen. Es wurde jüngst aufgeklärt, dass die Funktion von Osteoblasten nicht allein auf die Knochenbildung begrenzt ist, sondern diese bezieht sich auch auf die Differenzierung und Aktivierung von Osteoklasten, und Osteoblasten können als Kontrollzentrum in der zellularen Knochenrekonstruktion eine Rolle spielen.
- Diese Krankheiten, die im Allgemeinen als Knochenmetabolismuserkrankungen bezeichnet werden, können Osteoporose, Behceterkrankung, Osteomalazie, Hyperostose und Osteoperose umfassen. Von diesen ist die Osteoporose die am häufigsten entwickelte Erkrankung, und die Häufigkeit ihres Auftretens scheint sich mit der Seneszenz zu erhöhen, so dass die Diagnose und effektive Therapie hierfür ernsthaft erwünscht würde.
- Die Knochenmetabolismuserkrankungen bedeuten jene Erkrankungen, worin Knochenzellen spezifische metabolische Abnormitäten in irgendwelchen Knochengeweben haben. Die Erfinder haben ernsthafte Studien durchgeführt, um einen unterstützenden Faktor für die Knochenbildung unter Verwendung eines Kulturtests zu finden und haben schließlich diese Erfindung vervollständigt.
- Integrin kann in der Wechselwirkung zwischen Zellen und Zellen oder zwischen Zellen und extrazellulären Matrizen teilnehmen und spielt eine wichtige Rolle bei der Wundheilung, Entwicklung, Immunisierung, Hämostase oder Metastase. Die Integrin-Superfamilie ist eine α,β-Heterodimergruppe, die auf der Zelloberfläche gefunden wird und kann extrazelluläre Liganden und Zytoskelett kombinieren. Sämtliche Integrine sind Heterodimere und jede Untereinheit ist mit 90% extrazellulär und kann eine lange membranpermeable Domäne und kurze intrazelluläre Domäne aufweisen. Die extrazelluläre Domäne ist mit den extrazellulären Matrizen oder Liganden der Zelloberfläche gebunden, während die intrazelluläre Domäne mit Zytoskelettproteinen gebunden wird. Knochenmatrizen, wie Osteopontin, Knochensialoglycoproteine, Thrombospondin, Fibronektin und Vitronektin werden in Knochen gefunden und von sämtlichen Proteinen wurde festgestellt, dass sie die RGD-Sequenz aufweisen. Jüngst wurde bei Osteoklasten festgestellt, dass sie Integrin αVβ3 und α2β1 in der Zellenmembranoberfläche aufweisen (Davies J. et al., J. Cell Biol., Band 1, 109, S. 1817, 1989 und Zambonin Z. A. et al., Connect. Tissue Res., Band 20, S. 143, 1989). Aus diesen Tatsachen, dass Knochenabsorption durch Osteoklasten durch die Wirkung eines Antikörpers auf Integrin inhibiert werden kann (Davies J. et al., J. Cell Biol., Band 109, S. 1817, 1989), dass Knochenabsorption durch Rattenosteoklasten durch Synthese von GRGDSP-Peptid (GlyArgGly-AspSerPro) (Horton M. A. et al., Exp. Cell Res., Band 195, S. 368, 1991) und weiterhin das Echstatin, ein Protein abgeleitet vom Schlangenvenom und enthaltend die RGD-Sequenz, sowie eine plättchenaggreationsinhibierende Aktivität, ein synthetisches GdRGDSP-Peptid und ein zyklisches synthetisches GPenGRGDSPCA-Peptid Knochenabsorption durch Mausosteoklasten inhibieren kann und das GdRGDSP-Peptid die Bildung von Weinsäureresistenz und phosphatasepositiven multinuklearen Osteoklasten inhibieren kann (Gabri V. D. P. et al., J. Bone Miner. Res., Band 9, S. 1021, 1994), wird vorgeschlagen, dass die Wiedererkennung und Adhäsion von Knochenmatrizen durch Integrin und des diesbezüglichen Zytoskeletts bei der Entwicklung der Knochenabsorptionsfunktion von Osteoklasten in hohem Maße beteiligt ist.
- Es kann dann berücksichtigt werden, dass die Adhäsion von Zellmatrizen zwischen Osteoblasten und Knochenmatrizen durch den Adhäsionsmechanismus über Collagen und Fibronektin in den Knochenmatrizen und dem β1-Integrin der Osteoblasten verursacht werden würde. Auch wäre es im Adhäsionsmechanismus zwischen Heterozyten möglich, dass die Zelladhäsion von Osteoblasten mit Osteoklasten über Fibronektin durchgeführt werden kann, da sowohl β3-Integrin von Osteoklasten als auch β1-Integrin von Osteoblasten einen Rezeptor für Fibronektin darstellen kann. Jedoch wurde bislang nicht vorgeschlagen, dass die Disintegrinfamilie, einschließlich Echstatin oder Kistrin (William R. G. et al., Protein Science, Band 2, S. 1749, 1993) und das RGD-Peptid eine unterstützende Wirkung für die Knochenbildung zeigen.

- Es ist das Ziel dieser Erfindung, ein Mittel zur Förderung von Knochenbildung bereitzustellen sowie die Verwendung für die Knochenbildung.
- Die Knochenbildung kann durch Verabreichen eines Peptids oder Polypeptids enthaltend die RGD-Sequenz im Molekül, oder einem biologisch akzeptablen bzw. annehmbaren Salz hiervon an Patienten gefördert werden. Die Peptide oder Polypeptide, enthaltend die RGD-Sequenz im Molekül, können veranschaulichend umfassen: Kistrin, Echstatin, ein Peptid, dargestellt durch Gly-Arg-Gly-Asp-Ser (nachfolgend bezeichnet als GRGDS), eine Verbindung dargestellt durch die allgemeine Formel (I) worin R1, R2, R3, R4, R5 und R6 gleich oder verschieden sein können und jedes eines ist, ausgewählt aus der Gruppe, bestehend aus einem Wasserstoffatom; einer Alkylgruppe mit 1-8 Kohlenstoffatomen, gegebenenfalls substituiert mit einem, ausgewählt aus der Gruppe, bestehend aus einer Hydroxygruppe, einer Carboxygruppe, einer Cycloalkylgruppe mit 3-10 Kohlenstoffatomen, gegebenenfalls substituiert mit einer Hydroxygruppe, sowie einer Arylgruppe mit 6-12 Kohlenstoffatomen, gegebenenfalls substituiert mit einer Hydroxygruppe; einer Cycloalkylgruppe mit 3-10 Kohlenstoffatomen, gegebenenfalls substituiert mit einer Hydroxygruppe, und einer Arylgruppe mit 6-12 Kohlenstoffatomen, gegebenenfalls substituiert mit einer Hydroxygruppe, wobei R7 und R8 gleich oder verschieden sein können und jedes eine Gruppe darstellt, ausgewählt aus der Gruppe, bestehend aus einer Hydroxygruppe, einer Alkoxygruppe mit 1-8 Kohlenstoffatomen, einer Alkenyloxygruppe mit 2-12 Kohlenstoffatomen, einer Cycloalkyloxygruppe mit 3-10 Kohlenstoffatomen sowie einer Aryloxygruppe mit 6-12 Kohlenstoffatomen, und X stellt S oder SO2 dar, sowie eine Verbindung, dargestellt durch die allgemeine Formel (II) worin R9, R10, R11 und R12 gleich oder verschieden sein können und jeweils eine Gruppe darstellen, ausgewählt aus der Gruppe, bestehend aus einem Wasserstoffatom, einer Alkylgruppe mit 1-8 Kohlenstoffatomen, einer Cycloalkylgruppe mit 3-10 Kohlenstoffatomen und einer Arylgruppe mit 6-12 Kohlenstoffatomen, gegebenenfalls substituiert mit einer Hydroxygruppe, R13, R14 und R15 können gleich oder verschieden sein und stellen jeweils eine Gruppe dar, ausgewählt aus der Gruppe, bestehend aus einer Hydroxygruppe, einer Alkoxygruppe mit 1-8 Kohlenstoffatomen, einer Alkenyloxygruppe mit 2-12 Kohlenstoffatomen, einer Cycloalkylgruppe mit 3-10 Kohlenstoffatomen und einer Aryloxygruppe mit 6-12 Kohlenstoffatomen, und X stellt S oder SO dar.
- Weiterhin liefert diese Erfindung eine neue Verbindung, dargestellt durch die Formel (II).
- Diese Erfindung ist ebenfalls auf die Verwendung zur Förderung der Knochenbildung gerichtet, die umfasst: Verabreichen einer Verbindung an Patienten, dargestellt durch die allgemeine Formel (IX). worin R16 darstellt: -N(R20)2, -C(=NH)-NH2, -NH-C(=NH)-NH2 oder -CO-NH-C(=NH)-NH2 (worin R20 unabhängig ein Wasserstoffatom oder eine Alkylgruppe mit 1-4 Kohlenstoffatomen, gegebenenfalls mit einer Phenylgruppe substituiert, darstellt), R17 ein Wasserstoffatom oder eine Alkylgruppe mit 1-4 Kohlenstoffatomen, gegebenenfalls substituiert mit einer Phenylgruppe, darstellt, R18 ein Wasserstoffatom, eine Alkylgruppe mit 1-4 Kohlenstoffatomen, eine Phenylgruppe, gegebenenfalls substituiert mit einer Methoxygruppe oder -COR21 darstellt (worin R21 -OH, -NH2, -NH-(CH2)2-Phenyl, eine Alkoxygruppe mit 1-3 Kohlenstoffatomen, eine Benzyloxygruppe, Pro oder Aoc darstellt), R19 eine Alkylgruppe mit 1-5 Kohlenstoffatomen, gegebenenfalls mit einem Substituenten, ausgewählt aus der Gruppe, bestehend aus OH, -NH2, -CONH2, Cyclohexyl, Phenyl, Naphtyl, Indolyl oder Adamantyl, substituiert ist, eine Methylgruppe, substituiert mit -COOH und -NHCOOCH2-Phenylgruppe, eine Cyclohexylgruppe, gegebenenfalls substituiert mit einer Methoxygruppe oder eine Arylgruppe mit 6-10 Kohlenstoffatomen, gegebenenfalls substituiert mit einer Methoxygruppe, vorausgesetzt, dass R18 und R19 zusammen mit den Kohlenstoffatomen, mit denen sie verknüpft sind, Adamantyl, Naphtyl oder Fluorenyl bilden, Y stellt -NH-, -O- oder eine direkte Bindung dar, a stellt 1, 2 oder 3 dar, b stellt 1 oder 2 dar, c stellt 0 oder 1 dar und d stellt 0 oder 1 dar,
einer Verbindung, dargestellt durch die allgemeine Formel (X) worin R22 darstellt: -N(R23)2, -C(=NH)NH2, -NH-C(=NH)-NH2 oder -CO-NH-C(=NH)-NH2 (worin R23 unabhängig ein Wasserstoffatom oder eine Alkylgruppe mit 1-4 Koh lenstoffatomen darstellt, gegebenenfalls mit einer Phenylgruppe substituiert) und e stellt 2-6 dar,
oder
einer Verbindung der Formel (XII) oder ein biologisch akzeptables Salz hiervon. -
- Kistrin und Echstatin sind Proteine mit Molekulargewichten von etwa 7300 bzw. etwa 5400 und sie werden beispielsweise in Protein Science (1993) 2, 749 bis 1755, offenbart. Das GRGDS kann ohne Weiteres gemäß einer herkömmlichen Peptidsynthese hergestellt werden. Die Verbindungen der allgemeinen Formel (I) sind in dem US-Patent Nummer 5,384,309 offenbart und ihre repräsentativen Verbindungen können durch die Formeln (III) bis (VI) veranschaulicht werden.
- Die Verbindungen der allgemeinen Formel (II) sind neu und können gemäß dem Verfahren, wie beschrieben in Barker et al., J. Med. Chem., 1992, 35, 2040, hergestellt werden. Ihre repräsentativen Verbindungen können durch die Formeln (VII) und (VIII) veranschaulicht werden.
- Veranschaulichende Verbindungen der allgemeinen Formel (IX) sind nachfolgend gezeigt.

- Die Verbindungen der allgemeinen Formeln (IX), (X) und (XII) sind bekannt und können gemäß dem in der EP-0449079, der EP-A-0530505, der EP-A-0566919, der WO 95/14008, der EP-A-0528586 und der WO 93/19046 offenbarten Verfahren synthetisiert werden. Wenn Kistrin oder Echstatin einem Mensch als Mittel zur Förderung der Knochenbildung verabreicht wird, wird eine tägliche Dosis von 0,001 bis 100 μg/kg Körpergewicht, bevorzugt, 0,01 bis 10 μg/kg Körpergewicht verwendet. Im Falle des GRGDS wird eine tägliche Dosis von 0,001 bis 10 mg/kg Körpergewicht, bevorzugt 0,01 bis 1 mg/kg Körpergewicht verwendet. Im Falle der Verbindung, die durch die allgemeinen Formeln (I), (II), (IX), (X) oder (XII) dargestellt ist, wird eine tägliche Dosis von 0,001 bis 10 mg/kg Körpergewicht, bevorzugt 0,01 bis 1 mg/kg Körpergewicht verwendet.
- Das vorliegende Arzneimittel kann systemisch durch intravenöse Injektion, intramuskuläre Injektion, intraperitoneale Injektion, orale Verabreichung, parenterale Verabreichung, wie durch Zäpfchen, oder irgendein anderes Mittel im Stand der Technik verabreicht werden. Als pharmazeutische Zubereitungen können injizierbare oder orale pharmazeutische Zubereitungen beabsichtigt werden. Injizierbare pharmazeutische Zubereitungen können beispielsweise injizierbare Pulver umfassen. In diesem Fall können ein oder mehrere geeignete wasserlösliche Hilfsstoffe, wie Mannitol, Saccharose, Maltose, Glucose und Fructose zugegeben und in Wasser gelöst werden, und die Lösung wird portionsweise in Ampullen gefüllt und gefriergetrocknet und abgedichtet, um pharmazeutische Zubereitungen zu bilden. Orale pharmazeutische Zubereitungen können herkömmliche Tabletten, Kapseln, Granulate, feine Granulate, Pulver genauso wie enterisch beschichtete Zubereitungen umfassen.
- Bei der Behandlung eines Knochenbruchs kann dieses systemisch verabreicht werden oder lokal durch Injektion und anders gegeben werden. Für lokale Verabreichung kann ein Träger, enthaltend das vorliegende Arzneimittel, bevorzugter im Bereich nahe des gebruchenen Abschnitts implantiert werden. In diesem Fall können natürliche polymere Substanzen, wie Kollagen oder Fibrinkleber, synthetische polymere Substanzen, die in der Lage sind, im lebenden Körper dispergiert zu werden, wie polylaktierte Glycolsäure, als Träger verwendet werden. In der plastischen Chirurgie, kosmetischen Chirurgie, Knochentransplantation oder dentalen Transplantation kann das vorliegende Mittel durch Beschichten der Oberfläche des zu transplantierenden Knochens oder Zahns mit einer adhäsiven Substanz, wie Kollagenpaste oder Fibrinkleber, aufgebracht werden. Und es kann auf das Gewebe, den Knochen oder Alveolarknochen in dem Abschnitt aufgebracht werden, in dem der Knochen oder Zahn transplantiert werden soll. Bei der Transplantation von Knochen oder Zahn kann künstlicher Knochen oder künstlicher Zahn verwendet werden, der beispielsweise aus Metallen, Keramiken, Glas und anderen natürlichen oder künstlichen anorganischen Substanzen, wie Hydroxyapatit, gebildet wird. In diesem Fall kann es ebenfalls praktikabel sein, einen Kernabschnitt mit einem dichten Material zu bilden und einen Oberflächenabschnitt mit einem porösen Material, wie Hydroxyapatit, zu bilden und das vorliegende Mittel in den porösen Abschnitt penetrieren zu lassen. Auch ist es möglich, dass die Oberfläche des künstlichen Knochens, der aus einem dichteren Material aufgebaut ist, aufzurauen, um das vorliegende Mittel auf der Oberfläche zu halten.
- Beispiel: Synthesebeispiel der Verbindungen GH 4 und GH 5
- Die Verbindungen GH 4 und GH 5 werden gemäß dem Standard-FMOC-Verfahren, wie beschrieben in P.L. Barker et al., J. Med. Chem., 35, S. 2040-2048, 1992 hergestellt. FMOC-S-Trityl-Cystein, gebunden an Wangharz, wurde als Ausgangsmaterial verwendet und die FMOC-Aminosäure mit einer geeigneten Seitenkettenschutzgruppe, D-Aspartinsäure (O-t-Butyl); L-Aspartinsäure (O-t-Butyl); Glycin; L-Arginin-(N-2,2,5,7,8-pentamethylchroman-6-sulfonyl) wurden ihrerseits in 3 Mol-Portionen verwendet. Das FMOC am N-Ende wurde mit Piperidin (in Form einer 20%igen Lösung in Dimethylacetamid) entfernt und razemische 2-Brom-2-phenylessigsäure (4 Äquivalente), aktiviert mit Diisopropylcarbodiimid (2 Äquivalente), wurde zum freien N-Ende zugegeben. Die S-Tritylgruppe an der Cysteinseitenkette wurde mit einer Chlormethanlösung entfernt, enthaltend verdünnte Trifluoressigsäure (2%) und dann das resultierende Peptid durch Zugabe einer Lösung von Diisopropylethylamin (2 Äquivalente) in Dimethylacetamid cyclisiert. Entschützen und Aufteilen des cyclisierten Peptids wurde durch Behandeln mit Trifluoressigsäure, enthaltend Triethylsilan (2%), durchgeführt. Entfernung von Triethylfluoressigsäure ergab ein rohes Peptid, das dann unter Verwendung einer Umkehrphasen-HPLC-Säule [Vydac, C-18, eine 0,1%ige wässerige Lösung von Trifluoressigsäure, Acetonitril] mit linearem Gradienten (Acetonitril: 0-40%, 80 min) gereinigt wurde. Die in diesen Fraktionen enthaltenen Verbindungen, die nach etwa 22 Minuten bzw. etwa 24 Minuten erhalten wurden, wurden als GH 4 und GH 5 bezeichnet. Die Peptide wurden als weiße Pulver durch Gefriertrocknen in Wasser isoliert. Beide Verbindungen wurden durch den molekularen Ionenpeak (M+H)+ = 680,7 im Elektronensprühmassenspektrum detektiert. Beide Verbindungen zeigten offensichtlich verschiedene NMR-Spektren bei 0-10 ppm in Wasser bei pH 4-5.
- Von GH 4 und GH 5 wurde bestätigt, dass sie die Formel (VII) oder (VIII) aufweisen, aber es konnte nicht identifiziert werden, zu welcher diese jeweils gehörte.
- Herstellungsbeispiel 1
- Herstellung von Injektionen
- 10 μg Echstatin wurden in 1 ml Wasser gelöst und ein wasserlöslicher Hilfsstoff, Saccharose, wurde zugegeben, um bei 1,25-40 Gew/Vol.% zu liegen. Es wurde mittels eines 0,22 μm Filters (erhältlich von Milipore Inc.) steril filtriert. Es wurde in Portionen in Gefäße gegeben, gefriergetrocknet und abgedichtet, um Zubereitungen zu bilden.
- Die fördernde Wirkung der Knochenbildung durch das vorliegende Mittel wird anhand der nachfolgenden Testergebnisse veranschaulicht.
- Testergebnis 1
- Calvaria wurde aus einem Rattenfötus, der 20 Tage alt war, herausgeschnitten und osteoblastähnliche Zellen wurden durch enzymatische Behandlung präpariert. Die osteoblastähnlichen Zellen wurden in MEMα(+)-Medium, enthaltend 100 μl/ml Ascorbinsäure, 2 mM β-Glycerinphosphat und 10%igem fötalem Rinderserum inkubiert, um Knochentuberculum zu bilden. Für die Knochentuberculumbildung durch die primär kultivierten Osteoblasten unterstützte Kistrin und Echstatin die Calcifizierung bzw. Verkalkung und die Anzahl von Knochentuberculum bei 10-7 und 10-8 M. Eine geförderte alkalische Phosphataseaktivität wurde bei 10-8 M beobachtet.
- Cyclische Synthesepeptide, die Verbindungen der Formeln (IV), (VII), (VIII) und (V) förderten die Calcifizierung und die Anzahl von Knochentuberculum bei 10-6 und 10-8 M. Die Verbindung der Formel (III) unterstützte die Calcifizierung bei 10-8 und 10-10° M und die Anzahl von Knochentuberculum bei 10-6 M. Das GRGDS unterstützte die Calcifizierung bei 10-6 M, aber verhinderte die Calcifizierung bei 10-4 M vollständig.
- Testergebnis 2
- Knochentuberculumbildung durch Osteoblasten aus fötalem Rattencalvaria
- 1) Herstellung einer Enzymlösung
- 100 mg Collagenase (0,2%) und 50 mg Hyalurondiase (0,1%) wurden eingewogen und zu 50 ml fötalem rinderserumfreiem P12 Medium zugegeben und mit einem Rührer gerührt. Es wurde mit einem 0,22 μm Filter steril filtriert und dann angewendet.
- 2) Zubereitung und Kultivierung von Osteoblasten
- Föten, die 20 Tage alt waren (etwa 14 Föten), wurden einer schwangeren Ratte entnommen und in 70%igem Ethanol eingetaucht. Calvaria wurde herausgeschnitten durch Entfernen der Kopfhaut unter Verwendung gebogener Zangen und Scheren. Calvaria wurde ohne Entfernung des Periosteums entfernt und in einem fötalen rinderserumfreien Medium eingetaucht. Bindegewebe, das sich in Richtung nach vorne und hinten im zentralen Bereich bewegte und das weiche Gewebe umgab, wurde unter Verwendung eines Skalpells abgeschnitten. Sämtliche derart präparierte Knochenstücke wurden in ein 50 ml Zentrifugenröhrchen gegeben. 10 ml der Enzymlösung wurden zugegeben und das Röhrchen in einem Thermostat für 5 Minuten bei 37°C geschüttelt. Die behandelte Flüssigkeit mit dispergierten Zellen wurde gewonnen und bei 1200 U/min für 5 Minuten zentrifugiert, gefolgt von Entfernung des Überstands. 15 ml F12, 10%igem FCS-Medium, wurden zugegeben und die Zellen wurden in Vertiefungen gegeben und dann auf einer 10 cm Petrischale verteilt, die als Fraktion 1 definiert wurde. Nach Rückgewinnung der behandelten Lösung der Fraktion 1 wurden zu den Knochenstücken 10 ml der Enzymlösung zugegeben und in einem Thermostat für 10 Minuten bei 37°C geschüttelt. Nach Zentrifugation wurden die resultierenden Zellen als Fraktion 2 definiert. Daraufhin wurden die Zellen in diesen Fraktionen bis zur Fraktion 5 mit der ähnlichen Enzymbehandlung wie oben erhalten und die Zellen der Fraktionen 3-5 wurden in einem Kohlendioxidinkubator für 2-3 Tage unter Verwendung von F12- Medium, enthaltend 10% fötales Rinderserum, inkubiert. Nach Inkubation wurden die Zellen durch Behandlung mit 0,25% Trypsin (EDTA frei) zurückgewonnen. Nach Auszählen der Anzahl der Zellen wurden die Zellen in einer Inkubationsplatte (Nunc) mit vier Vertiefungen bei 1900 Zellen/300 μl (1000 Zellen/cm2) inokuliert und die Inkubation wurde für weitere 2 bis 3 Tage fortgesetzt. Nach Inkubation wurde sämtliche kultivierte Flüssigkeit entfernt, stattdessen 300 μl des Mediums, enthaltend die aktive Verbindung oder Dexamethason (10-8 M) zugegeben und dann die Inkubation für 14 Tage fortgesetzt. Für die Inkubation von diesem Punkt an wurde das MEMα(+)-Medium, enthaltend 100 μl/ml Ascorbinsäure, 2mM β-Glycerolphosphat und 10%iges fötales Rinderserum optimal zur Calcifizierung verwendet. Das Medium wurde alle zwei Tage ausgetauscht und die aktive Verbindung bei jedem Austausch zugegeben. Der Bereich in der calcifizierten Region, und die Anzahl von Knochentuberculum wurde als Index für die Knochentuberculumbildung verwendet. Nach Inkubation wurden die Zellen mit 10%iger Formalin/Phosphat gepufferter physiologischer Salzlösung für 60 Minuten fixiert und dann dreimal mit destilliertem Wasser gewaschen. Hiernach wurde Calciumphosphat in Knochentuberculum durch Behandlung mit einer 1 %igen Alizarinrotfärbung für 10 Minuten gefärbt und dreimal mit destilliertem Wasser gewaschen.
- Der Bereich des gefärbten Knochentuberculums wurde unter Verwendung von einer Nikonruzex-3U-Bildanalysevorrichtung gemessen, während die Anzahl des Knochentuberculums visuell unter dem Mikroskop gezählt wurde.
- 3) Alkalische Phosphataseaktivität
- Die in derselben Art und Weise wie oben in 2) inkubierten Zellen wurden zweimal mit 500 ml phosphatgepufferter physiologischer Salzlösung gewaschen und 300 ml des Eluierungsmittels (1% Triton X-100, 0,5 mM Magnesiumchlorid, 10mM Tris, pH-Wert 7,2) wurden zugegeben, und man ließ die Mischung für 3 Minuten auf Eis stehen. Diese wurde dann in ein Eppendorf-Röhrchen transferiert, mittels eines Homogenisators homogenisiert, und man ließ dies über Nacht bei 37°C in einem Thermostaten stehen, um alkalische Phosphatase zu eluieren. Daraufhin wurden Reste von Zellen und extrazellulärem Stroma durch Zentrifugieren bei 12.000 U/min für 10 Minuten entfernt, und 10 μl des Überstands wurden in ein anderes Röhrchen gegeben. 190 μl einer alkalischen Phosphatase Substratlösung (100 mM 2-Amino-2-methyl-1-propanol, 2 mM Magnesiumchlorid und 2mM Natrium-p-nitrophenylphosphat) wurden zugegeben, und man ließ in einem Thermostat für 10 Minuten bei 37°C reagieren. Die Reaktion wurde durch Zugabe von 800 μl einer 1N Natriumhydroxidlösung abgebrochen und die Absorption bei einer Wellenlänge von 405 nm gemessen.
- Die alkalische Phosphataseaktivität wurde gemäß der nachfolgenden Gleichung berechnet.
- Die Ergebnisse der Knochentuberculumbildungswirkung, die in primär kultivierten Osteoblasten beobachtet wurde, sind in Tabelle 1 gezeigt.
- In einer positiven Kontrolle wurden für die Dexamethasonverabreichungsgruppe bemerkbare Förderungseffekte in sämtlichen der Bereiche der calcinierten Region, der Anzahl an Knochentuberculum und der alkalischen Phosphataseaktivität, verglichen mit der nichtbehandelten Gruppe, beobachtet. Kistrin und Echstatin förderten die Calcifizierung und die Anzahl an Knochentuberculum bei 10-7 und 10-8 M, wohingegen sie die Calcifizierung bei 10-6 M vollständig verhinderten, aber es wurde nicht beobachtet, dass sie die Anzahl an Knochentuberculum beeinflussen. Auch wurde von diesen beobachtet, dass sie die alkalische Phosphataseaktivität bei 10-8 M fördern.
- Die Ergebnisse der Wirkung durch die Verbindungen der Formeln (III), (IV), GH 4, GH 5 und (V) sind in Tabelle 2 gezeigt.
- Mittelwert ± Standardabweichung (n = 4)
- Die Verbindungen der Formel (IV), GH 4, GH 5 und der Formel (V) förderten die Calcifizierung und die Anzahl an Knochentuberculum bei 10-6 und 10-8 M. Die Verbindungen der Formel (III), GH 4, GH 5 und der Formel (V) verhinderten vollständig die Calcifizierung bei 10-4 M, aber es wurde nicht beobachtet, dass sie die Anzahl an Knochentuberculum beeinflusste. Die Verbindung der Formel (III) förderte die Calcifizierung bei 10-8 und 10-10 M und förderte die Anzahl an Knochentuberculum bei 10-6 M. Das GRGDS fördert die Calcifizierung bei 10-6 M, aber verhinderte die Calcifizierung bei 10-4 M vollständig. Von der Anzahl an Knochentuberculum wurde beobachtet, dass er eine Tendenz zeigt, sich bei diesen Konzentrationen zu erhöhen.
- Kistrin und Verbindung Nr. 43 wurden unter Verwendung des oben beschriebenen Knochentuberculumbildungstests getestet. Die Ergebnisse sind in Tabelle 3 gezeigt.
- Verbindung Nr. 63 wurde unter Verwendung des oben beschriebenen Knochentuberculumbildungstests getestet. Die Ergebnisse sind in Tabelle 4 gezeigt.
- Wirkung der Erfindung
- Ein Mittel zur Förderung der Knochenbildung, das als einen aktiven Bestandteil ein Peptid oder Polypeptid mit der Aminosäuresequenz, zusammengesetzt aus ArgGly-Asp, im Molekül aufweist, wie Kistrin, Echstatin, ein Peptid, dargestellt durch Gly-Arg-Gly-Asp-Ser, eine Verbindung der obigen Formel (I) oder (II) oder eine Verbindung der Formel (IX), (X) oder (XII), kann dem menschlichen Körper verabreicht oder im Bereich nahe des gebrochenen Knochens implantiert werden, um wirksame Prophylaxe und Therapie von Knochenbrüchen zu erreichen.
Claims (13)
- Verbindung, dargestellt durch die nachfolgende Formel (II) worin R9, R10, R11 und R12 gleich oder verschieden sein können und jeweils eine Gruppe darstellen, ausgewählt aus der Gruppe, bestehend aus einem Wasserstoffatom, einer Alkylgruppe mit 1-8 Kohlenstoffatomen, einer Cycloalkylgruppe mit 3-10 Kohlenstoffatomen und einer Arylgruppe mit 6-12 Kohlenstoffatomen, gegebenenfalls substituiert mit einer Hydroxygruppe, R13, R14 und R15 stellen eine Gruppe dar, ausgewählt aus der Gruppe, bestehend aus einer Hydroxygruppe, einer Alkoxygruppe mit 1-8 Kohlenstoffatomen, einer Alkenyloxygruppe mit 2-12, einer Cyloalkylgruppe mit 3-10 Kohlenstoffatomen und einer Aryloxygruppe mit 6-12 Kohlenstoffatomen, und X stellt S oder SO dar.
- Verwendung einer Verbindung, ausgewählt aus der Gruppe, bestehend aus Kistrin, Echstatin, einem Peptid, dargestellt durch Gly-Arg-Gly-Asp-Ser, und einer Verbindung, dargestellt durch die allgemeine Formel (I), (II), (IX), (X) oder (XII) oder ein biologisch akzeptables Salz hiervon zur Herstellung eines Mittels zur Förderung der Knochenbildung.
- Verwendung von Kistrin zur Herstellung eines Mittels zur Förderung der Knochenbildung.
- Verwendung von Echstatin zur Herstellung eines Mittels zur Förderung der Knochenbildung.
- Verwendung eines Peptids, dargestellt durch Gly-Arg-Gly-Asp-Ser, zur Herstellung eines Mittels zur Förderung der Knochenbildung.
