Description
- Die vorliegende Erfindung betrifft das Gebiet der organischen Synthese. Im besonderen betrifft sie ein Verfahren zur Herstellung von 3-Oxo-2-pentyl-1-cyclopenten-alkylacetaten, Verbindungen, die als zwischenverbindungen für die Herstellung von Jasmonderivaten geeignet sind, welche mit geschätzten Dufteigenschaften ausgestattet sind. Es ist nämlich bekannt, dass die katalytische Hydrierung des 3-Oxo-2-pentyl-1-cyclopenten-methylacetats, beispielsweise mit Hilfe von Pd/C in Anwesenheit von Aluminiummethylat [DE-OS 2'162'820], zur Bildung des cis-Methyldihydrojasmonats führt, dessen blumige Note das Interesse der Parfumeure weckte.
- Der Stand der Technik berichtet über zahlreiche Wege zur Synthese des 3-Oxo-2-pentyl-1-cyclopenten-methylacetats. Nur einer von ihnen greift in seiner letzten Stufe auf ein acyclisches Denvat zurück [JP 58 118'536; Chem. Abstr. 1983, 99, 175481 d], die übrigen besitzen als Schlüsselzwischenverbindungen cyclische Vorläufer. Entsprechend der Beschaffenheit dieser Vorläufer können die bekannten Synthesen in vier unterschiedliche Kategorien eingeteilt werden:
- a. über ein gamma-Lakton durch Umlagerung mit Polyphosphorsäure [NL69 18'228; Chem. Abstr. 1971, 109 953d];
- b. über das Methyldihydrojasmonat durch Bromierung und Dehydrobromierung [NL70 02'279; Chem. Abstr. 1972, 3462g] oder durch anodische Oxidation des Enolacetat-Derivates [J Am. Chem. Soc. 1975, 97, 6144];
- c. über das 2-Pentyl-1,3-cyclopentandion durch Reaktion mit Dimethylmalonat [siehe beispielsweise DE-OS 2 008 833; Chem. Abstr. 1970, 73, 109 363d]
- d. über das 2-Pentyl-2-cyclopenten-1-on durch radikalische Addition von Methylhydroxyacetat, gefolgt von Wasserentzug [Parf. Cosm. Sav. France 1972, 2(8), 356]; durch Umsetzung mit Methyldiazoacetat [Perfum. Essent. Oil Rec. 1969, 267; JP 70 00862] oder durch Addition von Methylbromacetat [J. Org. Chem. 1974, 39, 2637] oder von Methyllithiumacetat, gefolgt von einer Oxidation mit Chromsäure.
- Keine der angeführten Synthesen, zum grössten Teil akademischer Natur, hat bisher eine industrielle Anwendung in grossem Massstab gefunden.
- Wir haben nunmehr gefunden, dass es möglich ist, das 3- oxo-2-pentyl-1-cyclopenten-alkylacetat auf einfache und wirksame Weise durch Isomerisierung eines Epoxyesters der Formel
- worin R für einen niederen, geradekettigen oder verzweigten C&sub1;-C&sub6;- Alkylrest steht und die Wellenlinie eine C-C-Bindung mit cis- oder trans-Konfiguration definiert, mit Hilfe eines saueren Isomerisierungsmittels zu synthetisieren.
- Die Erfindung hat dementsprechend dieses Verfahren zum Gegenstand, welches durch die Beschreibung in den Ansprüchen definiert ist.
- Als saures Isomerisierungsmittel kann man eine anorganische oder organische Protonsäure oder eine Säure vom sogenannten Lewis-Typus verwenden. Unter den aktiven Säuren kann man beispielsweise die Chlorwasserstoffsäure, Schwefelsäure, Phosphorsäure, p- Toluolsulfonsäure oder auch BF&sub3;, SnCl&sub4;, oder TiCl&sub4; anführen. Saure Ionenaustauschharze können ebenfalls geeignet sein.
- Die Isomerisation findet unter milden Reaktionsbedingungen statt, was das Verfahren aus industrieller Sicht besonders attraktiv macht. Tatsächlich haben sich Temperaturen in der Grössenordnung von 5-30 ºC als vollständig zufriedenstellend zur Darstellung des gewünschten Produktes mit ausgezeichneten Ausbeuten von gleich oder mehr als 90 Gew.% erwiesen.
- Die im erfindungsgemässen Verfahren als Ausgangsverbindungen verwendeten Epoxyester der Formel (I) sind Verbindungen mit neuer Struktur, und im Hinblick darauf bilden sie ebenfalls einen der Gegenstände der vorliegenden Erfindung.
- Sie können ausgehend vom 2,3-Epoxy-2-pentyl-1-cyclopentanon, einer in der Literatur beschriebenen Verbindung [siehe beispielsweise Synthesis, 1975, 104), mit Hilfe eines Verfahrens, welches durch eine Wittig-Reaktion charakterisiert ist, insbesondere unter Anwendung eines von Wadsworth und Emmons [siehe J. Amer. Chem. Soc. 1961, 83, 1733] ausgearbeiteten Verfahrens gemäss dem nachfolgenden Schema:
- hergestellt werden.
- Die Umsetzung erfolgt durch Addition eines Dialkylphosphonoalkylacetats, beispielsweise des Dimethylphosphonomethylacetats, was erlaubt, das 2,3-Epoxy-2-pentyl-1-cyclopentyliden-methylacetat zu erhalten. Die Umsetzung erfolgt analog zu den in der Literatur beschriebenen Methoden, die speziell angewendeten Bedingungen werden nachfolgend dargestellt.
- Die Erfindung wird in detaillierterer Weise mit Hilfe der nachfolgenden Beispiele dargestellt, worin die Temperaturen in Grad Celsius angegeben sind und die Abkürzungen die im Stand der Technik übliche Bedeutung besitzen.
- 20 ml 70%iges Wasserstoffperoxid in Wasser (0,43 Mol), gefolgt von 1,12 g (0,02 Mol) Kaliumhydroxid in Plätzchenform wurden bei 10º unter Rühren zu einer Lösung von 60,8 g (0,36 Mol) 3- Oxo-2-pentyl-1-cyclopenten in 100 ml Methanol hinzugefügt. Während der Zugabe stieg die Temperatur bis 25º, und das Reaktionsgemisch wurde während 1,5 Stunden bei diesem Temperaturwert belassen. Eine neue Fraktion von gleichem Volumen von Wasserstoffperoxid, beziehungsweise KOH, wurde in zwei Portionen dem Reaktionsgemisch zugesetzt, was den Anteil der Umwandlung nach 4 Stunden der Reaktion auf ca. 75% bringt.
- 10 g (0,07 Mol) K&sub2;CO&sub3; wurden anschliessend innerhalb von 2 Stunden in zwei Portionen dem erhaltenen Gemisch hinzugefügt, und das Gemisch wurde während weiterer zwei Stunden unter Rühren bei 25º belassen. Anschliessend extrahierte man mit CH&sub2;Cl&sub2;, und die vereinigten organischen Extrakte wurden mit Wasser gewaschen, über Na&sub2;SO&sub4; getrocknet und konzentriert, um einen Rückstand zu ergeben, welcher durch Destillation bei vermindertem Druck 41 g (Ausb. 61%) des gewünschten Epoxyketons in Form einer farblosen Flüssigkeit ergab. - Zu einer Suspension von Natriumhydrid (7,3 g; 0,15 Mol einer 50%igen Dispersion in Öl), welche vorgangig mit Petrolether (30-50) gewaschen wurde, in 200 ml Tetrahydrofuran, fügte man tropfenweise bei 10-20º 27,6 g (0,15 Mol) Trimethylphosphonoacetat in 200 ml Tetrahydrofuran (THF) hinzu. 15 Minuten nach Beendigung der Zugabe fügte man zum Reaktionsgemisch bei 11- eine Lösung von 10,2 g (0,056 Mol) des gemäss dem obigen Abschnitt (a) hergestellten Epoxyketons in 150 ml THF hinzu. Das Gemisch wurde unter Rühren während 16 Stunden bei 25º gehalten, danach wurde es in 500 ml Wasser eingetragen und mit Ether extrahiert. Die vereinigten organischen Extrakte wurden mit einer gesättigten wässrigen NaCl-Lösung gewaschen, über Na&sub2;SO&sub4; getrocknet und konzentriert. Der erhaltene Rückstand wurde destilliert, um eine Fraktion des gewünschten Epoxyesters beim Siedepunkt 92-103º/4 Pa zu ergeben (11,9 g; Ausb. 93%; Isomerenanteil trans/cis 94:5).

- MS: 224(M&spplus;,1), 208(2), 195(4), 167(100), 151(14), 139(24), 126(19), 107(20), 91(24), 79(40), 67(15), 55(20), 41(20).
- 0,1 ml (1,9 Inmol) konzentrierter Schwefelsäure wurden unter Rühren bei 4º zu einer Lösung des im obigen Abschnitt (b) hergestellten Epoxyesters (8,6 g; Reinheit 79%, 30 Mmol) in 50 ml Ether hinzugefügt. Das Gemisch wurde gerührt, während die Temperatur allmählich bis auf ca. 30º anstieg. Nachdem man es bei dieser Temperatur während 1 Stunde belassen hat, wurde das Gemisch mit Ether verdünnt, mit einer gesättigten wässrigen NaHCO&sub3;- und NaCl-Lösung gewaschen, danach über Na&sub2;SO&sub4; getrocknet und konzentriert, um 7,7 g Rückstand zu ergeben. Der letztere lieferte durch Destillation im Kugelrohr (0,04 mbar, Temp.= 110º) 6,97 g (Reinheit 93%; Ausb. 95%) 3-Oxo-2-pentyl- 1-cyclopenten-methylacetat, mit folgenden analytischen Daten:

- ¹H-NMR (360 MHz, CDCl&sub3;) : 0,88(t, J=7, 3H); 1,2-1,4(m, 6H) ; 2,18(t, J7, 2H); 2,41(m, 2H); 2,62(m, 2H) δ ppm
- ¹³C-NMR (90,5 MHz, CDCl&sub3;) : 209,2(s), 169,6(s), 163,5(s), 143,3(s), 52,3(q), 36,6(t), 34,4(t), 31,8(t), 29,7(t), 28,0(t), 23,2(t), 22,5(t), 13,0(q) δ ppm
- MS : 224(M+, 0,5), 206(2), 168(14), 151(100), 135(12), 121(17), 109(55), 108(47), 93(33), 79(42), 67(13), 55(19), 41(28).
Claims (7)
1. Verfahren zur Herstellung eines
3-Oxo-2-pentyl-1-cyclopentenalkylacetats, dadurch gekennzeichnet, dass man einen Epoxyester der Formel
worin R für einen niederen, geradkettigen oder verzweigten C&sub1;-C&sub6;-Alkylrest
steht und die Wellenlinie eine C-C-Bindung mit cis- oder trans-
Konfiguration definiert, mit Hilfe eines saueren Isomerisierungsmittels
isomerisiert.
2. Verfahren gemäss Patentanspruch 1, dadurch gekennzeichnet, dass
man als Ausgangsverbindung der Formel (1) 2,3-Epoxy-2-pentyl-1-
cyclopentyliden-methylacetat verwendet und man 3-Oxo-2-pentyl-1-
cyclopenten-methylacetat erhält.
3. Verfahren gemäss Patentanspruch 1, dadurch gekennzeichnet, dass
das sauere Isomerisierungsmittel eine anorganische oder organische
Protonsäure ist, eine sogenannte Lewis-Säure oder ein saueres
Ionenaustauschharz.
4. Verfahren gemäss Patentanspruch 3, dadurch gekennzeichnet, dass
man als Protonsäure Schwefelsäure verwendet.
5. Verfahren gemäss Patentanspruch 1, dadurch gekennzeichnet, dass
die Isomerisierungsreaktion bei einer Temperatur von 5 bis 30ºC
stattfindet.
6. Epoxyester der Formel
worin R für einen niederen, geradkettigen oder verzweigten C&sub1;-C&sub6;Alkylrest
steht und die Wellenlinie eine C-C-Bindung mit cis- oder trans-
Konfiguration definiert.
7. 2,3-Epoxy-2-pentyl-1-cyclopentyliden-methylacetat als Epoxyester
gemäss Patentanspruch 6.
