Description
- Die vorliegende Erfindung betrifft neue Inhibitoren der Sexualsteroidaktivität wie Antiöstrogen-Verbindungen, die wirksame antagonistische Fähigkeiten besitzen, während ihnen im Wesentlichen agonistische Wirkungen fehlen. Insbesondere betreffen bestimmte bevorzugte Ausführungsformen der Erfindung bestimmte tricyclische Verbindungen, die hohe Affinität zu Östrogenrezeptoren besitzen, die aber solche Rezeptoren nicht aktivieren und/oder die die Produktion von Sexualsteroiden oder deren Vorstufen hemmen.
- Während der Behandlung von bestimmten Sexualsteroid-abhängigen Krankheiten ist es wichtig, bestimmte Sexualsteroid-induzierte Wirkungen stark zu reduzieren oder, wenn möglich, zu eliminieren. Zu diesem Zweck ist es wünschenswert, sowohl Rezeptorstellen zu blockieren, die durch Sexualsteroide stimuliert werden, als auch die Menge an Sexualsteroiden zu reduzieren, die für die Wirkung an diesen Stellen verfügbar ist. Zum Beispiel könnte eine alternative oder gleichzeitige Therapie zur Verabreichung von Antiöstrogenen Versuche zur Blockierung der Östrogen-Produktion umfassen (z. B. mittels Ovariektomie), so dass eine geringere Menge davon zur Verfügung steht, um Rezeptorstellen zu aktivieren. Verfahren im Stand dar Technik zur Blockierung der Östrogenproduktion hemmen die östrogeninduzierten Funktionen jedoch nicht ausreichend. Tatsächlich ist es möglich, dass selbst bei vollständigem Fehlen von Sexualsteroiden einige Rezeptoren aktiviert werden können. (vgl. Simard und Labrie, "Keoxifene shows pure antiestrogenic activity in pituitary gonadotrophs", Mol. Cell. Endocrinol., 39 (1985), S. 141-144, insbesondere Seite 144).
- Daher lassen sich mit Antagonisten von Sexualsteroiden größere therapeutische Erfolge erzielen als mit einer Therapie, die nur die Sexualsteroid- Produktion hemmt. Im Stand der Technik beschriebene Antagonisten besitzen jedoch oft nicht ausreichende Affinität zu Rezeptoren und einige können selbst als Agonisten wirken und in unerwünschter Weise genau die Rezeptoren aktivieren, die sie vor Aktivierung abschirmen sollen, obwohl sie an Rezeptoren binden können.
- Daher gibt es im Stand der Technik einen Bedarf für Antiöstrogene, die Östrogenrezeptoren mit geringer oder keiner agonistischen Wirkung wirksam blockieren. In Wakeling und Bowler, "Steroidal Pure Antiestrogens", J. Endocrinol. 112 (1987), R7-R10 ist ausgeführt, dass ein Steroidderivat als ein Antiöstrogen wirkt, aber eine bestimmte Östrogenaktivität zeigt. Die Nettowirksamkeit einer Verbindung setzt sich sowohl aus ihren agonistischen (unerwünschten) als auch ihre antagonistischen (erwünschten) Aktivitäten zusammen.
- In dem US-Patent 4,094,994 ist offenbart, dass die Verwendung von bestimmten Antiöstrogenen bestimmte menschliche Brusttumorzellen hemmen kann.
- H. Mouridson et al., Cancer Treatm. Rev., 5 (1978), S. 131-141, offenbaren, dass Tamoxifen, ein Antiöstrogen, bei ungefähr 30 Prozent der behandelten weiblichen Patienten eine Remission von fortgeschrittenem Brustkrebs bewirkt.
- Zur Behandlung von Brustkrebs ist auch die kombinierte Verwendung des Antiöstrogens Tamoxifen und eines Luteinisierungshormon-freisetzenden Hormonagonisten, Buserelin, bekannt (vgl. zum Beispiel: Klijn et al., J. Steroid Biochem., 420 (1984), Nr. 6B, S. 1381). Die objektive Remission derartiger Krebserkrankungen ist jedoch unakzeptabel niedrig.
- Es hat sich herausgestellt, dass bestimmte 7α- substituierte Derivate von Östradiol, zum Beispiel eine 7α-(CH&sub2;)&sub1;&sub0;CONMeBu-Substitution, Antiöstrogen- Aktivität besitzt (Bowler et al., (1985); Eur. Patent-Anmeldung 0138504; Wakeling und Bowler, J. Steroid Biochem., 30 (1988), S. 141-147). Vgl. auch US-Patent 4,659,516. Die Substitution (CH&sub2;)&sub9;SOC&sub5;H&sub6;F&sub5; ist auch bei bestimmten Verbindungen verwendet worden (Wakeling et al., Cancer Res., 51 (1991), S. 3867-3873).
- Bestimmte -(CH&sub2;)&sub1;&sub0;CONMeBu-substituierte Verbindungen werden auch in dem US-Patent 4,732,912 (vgl. z. B. Beispiel 5 und 16) offenbart (vgl. auch EP-Pat. Nr. 166 509, EP-Pat. Nr. 124 369, EP-Pat. Nr. 160 508, EP-Pat. Nr. 163 416, US-Pat. Nr. 4,760,061, US-Pat. Nr. 4,751,240 sowie Wakeling, A. E. und Bowler, J., J. Endocrinol., 112 (1987), R7-R10).
- Östradiol-Derivate, die einen Carboxyalkyl-Substituenten in 7α-Position besitzen, behielten ihre Affinität zum Östrogenrezeptor, wenn sie über ihre Carboxy-Gruppe an Agarose oder Polyacrylamidharz zur affinitätschromatographischen Reinigung des Östrogenrezeptors gebunden wurden (Bucourt et al., J. Biol. Chem., 253 (1978), S. 8221).
- Einige Steroidderivate, wie 16-Methylen-Östradiol und 16-Methylen-Östron, sind als Inhibitoren der 17β-Hydroxysteroid-Dehydrogenaseaktivität beschrieben worden (Thomas et al., J. Biol. Chem., 258 (1983), S. 11500).
- Bestimmte nicht-steroidartige Verbindungen, denen antiandrogene Wirkung zugeschrieben wird, sind von Furr et al., J. Endocrinol., 113 (1987), R7-R9 beschrieben.
- Das US-Pat. Nr. 4,659,695 betrifft ein Verfahren zur Behandlung von Prostata-Krebs für empfängliche männliche Tiere einschließlich Menschen, deren Hoden-Hormonsekretionen durch chirurgische oder chemische Mittel, z. B. Verwendung eines LHRH-Agonisten, z. B. [D-Trp&sup6;, Des-Gly-NH&sub2;¹&sup0;]-LHRH-Ethylamid, blockiert sind. Die Behandlung umfasst die Verabreichung eines Antiandrogens, z. B. Flutamid zusammen mit mindestens einem Inhibitor der Sexualsteroidbiosynthese, z. B. Aminoglutethimid und/oder Ketoconazol (vgl. auch PCT/US 85/01454 (Internationale Veröffentlichungsnummer WO 86/01105) bezüglich einer Kombinationstherapie zur Behandlung von hormonabhängigem Krebs.
- Das US-Pat. Nr. 4,472,382 betrifft ein Verfahren zur Behandlung von Prostatakrebs unter Verwendung einer Kombination eines Antiandrogens und eines LHRH-Agonisten.
- Das US-Pat. Nr. 4,386,080 betrifft neue Amid- Derivate, insbesondere neue Acylanilide, die Antiandrogeneigenschaften besitzen.
- In dem Französischen Patent 2528434 und in Jordan und Koch, "Regulation of Prolactin Synthesis in vitro by estrogenic and antiestrogenic derivatives of estradiol and Estrone", Endocrinology, 124 (4) (1989), S. 1717-1725, werden Antiöstrogen- Wirkungen für bestimmte 11β-substituierte Östradiol-Derivate beschrieben.
- In den US-Patenten Nr. 3,995,060, 4,161,540 und 4,139,638 ist offenbart, dass bestimmte 4'-substituierte und 3'-,4'-disubstituierte Anilide Antiandrogen-Eigenschaften besitzen.
- Seit mehreren Jahren haben Forscher versucht, Verbindungen zu entwickeln, die Androgen- und/oder Östrogenbildung wirksam hemmen können, ohne schädliche Wirkungen auf gesunde Gewebe auszuüben. Insbesondere ist die Hemmung der 17β-Hydroxysteroid- Dehydrogenase, die an der Biosynthese von Testosteron, Androst-5-en-3β,17β-diol und Östradiol beteiligt ist, von einigen Forschern untersucht worden. Einige mit Affinitätsmarkern versehenen Inhibitoren der Östradiol-17β-Dehydrogenase der menschlichen Plazenta sind beschrieben worden (C. C. Chin und J. C. Warren, J. Biol. Chem., 250 (1975), S. 7682- 7686; Y. M. Bhatnagar et al., J. Biol. Chem., 253 (1978), S. 811-815; C. C. Chin et al., J. Biol. Chem., 255 (1980), S. 3660-3664; J. L. Thomas und R. C. Strickler, J. Biol. Chem., 258 (1983), S. 1587-1590).
- B. Tobias et al., J. Biol. Chem., 257 (1982), S. 2783-2786 beziehungsweise R. J. Auchus und D. E. Covey, Biochemistry, 25 (1986), S. 7295-7300 offenbaren die Verwendung von 17β-propinylsubstituiertem Progestin und propinylsubstituiertem 3-Hydroxy-14,15-secoestra-1,3,5(10)-trien-17-on als Inhibitoren der 17β-Östradiol-Dehydrogenase.
- Thomas, J. L. et al., J. Biol. Chem., 258 (1983), S. 11500 haben beschrieben, dass 16-Methylen- Östradiol und 16-Methylen-Östron Inhibitoren der 17β-Hydroxysteroid-Dehydrogenaseaktivität sind.
- Die im Stand der Technik beschriebenen Verfahren konnten die Sexualsteroidsynthese nicht vollständig hemmen, obwohl sie keine unerwünschten Nebenwirkungen aufweisen.
- Von Angerer et al. diskutieren andere Antiöstrogene in "1-(aminoalkyl)-2-phenylindoles als Novel Pure Estrogen Antagonists", J. Med. Chem., 33 (1990), S. 2635-2640. In dem US-Patent 4,094,994 wird festgestellt, dass durch die Verwendung von bestimmten Antiöstrogenen bestimmte menschliche Brusttumorzellen gehemmt werden (vgl. auch DE 38 21 148).
- A. Saeed et al., J. Med. Chem., 33 (1990), S. 3210 -3216, und A. P. Sharma et al., J. Med. Chem. 33 (1990), S. 3216-3222 und S. 3222-3229 haben die Synthese und biologische Aktivitäten von 2,3- Diaryl-2H-1-benzopyran-Analoga als Antiöstrogene beschrieben, die folgende molekulare Struktur haben:
- N. Durani et al., J. Med. Chem., 32 (1989), S. 1700 -1707 beschreiben die Synthese und biologische Aktivitäten von Benzofuran und Triarylfuran-Analoga als Antiöstrogene.
- Das europäische Gegenstück der prioritätsbegründenden Anmeldungen 07/377 010 und 07/265 150 wurde am 9. Mai 1990 als Europäische Anmeldung Nr. 0367576 veröffentlicht. Der europäische Recherchenbericht für diesen Fall offenbarte die folgenden Publikationen:
- In dem EP-Patent Nr. 305 242 beziehen sich Nique et al. auf die Synthese und die Verwendung von 17- Acyl-Steroiden als Arzneimittel. Der Recherchenbericht hob Seite 7, Verbindung I'C, hervor.
- In dem EP-Patent Nr. 280 618 beziehen sich Nique et al. auf 7-substituierte 19-nor-Steroide als Arzneimittel. Der Recherchenbericht hob die Beispiele 2, 3, Seiten 22, 23 und die Ansprüche hervor.
- In dem DE-Patent Nr. 32 42 894 A1 beziehen sich Neef et al. auf 17α-substituiertes Equilenin für die Hemmung der Progesteron-Biosynthese und Kontrolle der Fertilität.
- In dem US-Patent Nr. 2,875,199 diskutiert Cella, J. A. 17-carboxylierte Östradiole zur Senkung der Serumkonzentration von Cholesterin.
- Blickenstaff et al. (Steroids, Bd. 46, Nr. 4 und 5, Seite 889-902) beschreiben die Synthese von 16- und 17-substituierten Östradiolen, die zur Kopplung an Vinblastinarten geeignet sind.
- Andere im Recherchenbericht aufgefundenen Referenzen wurden vorstehend diskutiert.
- Eine Aufgabe der vorliegenden Erfindung ist die Bereitstellung von Verfahren zur Hemmung der Sexualsteroidaktivität. Solche Verfahren können zur Behandlung von Krankheiten, die mit Sexualsteroiden im Zusammenhang stehen, geeignet sein.
- Eine weitere Aufgabe der Erfindung ist die Bereitstellung eines reinen Antiöstrogens zur therapeutischen Verwendung.
- Eine weitere Aufgabe der Erfindung ist die Bereitstellung von Zusammensetzungen, die die Sexualsteroidsynthese, insbesondere die Östrogensynthese, hemmen können.
- Eine weitere Aufgabe ist die Bereitstellung eines Antiöstrogens, das gute Affinität zu Östrogenrezeptoren aufweist, jedoch im Wesentlichen keine unerwünschten agonistischen Aktivitäten bezüglich dieser Rezeptoren hat und im Wesentlichen keine hormonale Aktivität aufweist.
- Eine weitere Aufgabe der Erfindung ist die Bereitstellung einer therapeutischen, antiöstrogenen Zusammensetzung, die zur Behandlung von Krankheiten, die mit Östrogenen im Zusammenhang stehen, geeignet ist. Diese Krankheiten umfassen, ohne darauf beschränkt zu sein, Brustkrebs, Uteruskrebs, Eierstockkrebs, Endometriose, Uterusfibrom, vorzeitige Pubertät und gutartige Prostatahyperplasie.
- Erfindungsgemäß wird eine Verbindung der Molekülstruktur:
- bereitgestellt, wobei die gepunktete Linie eine optionale Doppelbindung darstellt;
- wobei R&sup5; und R&sup6; unabhängig voneinander ein Wasserstoff-Atom, eine Hydroxyl-Gruppe oder ein Rest ist, der in vivo in eine Hydroxyl-Gruppe umgewandelt wird;
- wobei R¹&sup0;&sup0; ein zweiwertiger Rest ist, der zwischen dem L- und dem B-Ring einen Abstand durch 4-10 dazwischenliegenden Atome erzeugt, ausgewählt aus gerad- oder verzweigtkettigen Alkylen-, Alkenylen- und Alkinylen-Gruppen und Phenylen-Gruppen und Phenylenoxyalkylen-Gruppen;
- wobei L ein zweiwertiger oder dreiwertiger polarer Rest ist, ausgewählt aus -CO-, -SO-, -CON< , -N< und -SON< ;
- wobei G' entweder fehlt oder ausgewählt ist aus einem Wasserstoff-Atom, einem C&sub1;- bis C&sub5;-Kohlenwasserstoff, einem gesättigten oder ungesättigten C&sub5;- bis C&sub7;-Cycloalkyl-Rest, einem zweiwertigen Rest, der G² mit L unter Bildung eines 5- bis 7- gliedrigen heterocylischen Ringes verbindet und halogensubstituierten Derivaten davon;
- wobei G² entweder fehlt oder ausgewählt ist aus einem Wasserstoff-Atom, einem Q- bis C&sub5;- Kohlenwasserstoff, einem substituierten oder unsubstituierten C&sub5;- bis C&sub7;-Cycloalkyl-Rest, einem zweiwertigen Rest, der G¹ und L unter Bildung eines 5- bis 7-gliedrigen heterocylischen Ringes verbindet und halogensubstituierten Derivaten davon; und
- wobei G³ ein C&sub1;- bis C&sub3;-Kohlenwasserstoff ist.
- G³ ist vorzugsweise ausgewählt aus der Gruppe bestehend aus einer Methyl-, Ethyl- und Propyl- Gruppe. In einer anderen bevorzugten Ausführungsform sind L, G¹ und G² zusammen -NCxH2x oder -NCx- 1H2x-2O (wobei X eine ganze Zahl von 4 bis 6 ist).
- In einer weiteren Ausführungsform der vorliegenden Erfindung wird eine Verbindung mit der Molekülstruktur:
- bereitgestellt, wobei R&sup9; eine Methyl-, Ethyl-, Propyl-, Ethenyl- oder Ethinyl-Gruppe, insbesondere eine Verbindung mit der Molekülstruktur
- ist.
- In einer anderen bevorzugten Ausführungsform wird eine Verbindung mit einer der folgenden Molekülstrukturen:
- bereitgestellt,
- wobei R&sup9; eine Methyl-, Ethyl-, Propyl-, Ethenyl- oder Ethinyl-Gruppe ist;
- 7-Hydroxy-3-(4'hydroxyphenyl)-4-methyl-2-(4"-(2'''- piperidino-ethoxy)phenyl)-2H-benzopyran (EM 343);
- wobei R&sup5; und R&sup6; unabhängig voneinander ein Wasserstoff-Atom, eine Hydroxyl-Gruppe oder ein Rest sind, der in vivo in eine Hydroxyl-Gruppe umgewandelt wird; oder
- 7-Hydroxy-3-(4'-hydroxyphenyl)-4-methyl-2-(6"- piperidinohexyl)-2H-benzopyran (EM 721).
- Ferner wird eine pharmazeutische Zusammensetzung bereitgestellt, die einen pharmazeutisch akzeptablen Träger oder ein Verdünnungsmittel und eine therapeutisch wirksame Menge einer der vorstehend definierten, erfindungsgemäßen Verbindungen umfasst.
- Fig. 1 zeigt einen Vergleich der Inhibitoraktivität bei steigenden Konzentrationen von EM 343 ( - ) und EM 312 (o-o) auf das Wachstum von menschlichen 17β-Östradiol-stimulierten ZR-75-1-Brustkrebszellen. Die entsprechenden IC&sub5;&sub0;-Werte werden für EM 343 mit 2,55 · 10&supmin;¹&sup0; M und für EM 312 mit 8,43 · 10&supmin;¹&sup0; M berechnet, was eine 3-fach höhere Aktivität für EM 343 zeigt.
- Bei systemischer Verabreichung können die erfindungsgemäßen Pharmaka zur Behandlung von Brustkrebs, Uteruskrebs, Eierstockkrebs, Endometriosis, Uterusfibrom, vorzeitiger Pubertät und gutartiger Prostatahyperplasie verwendet werden.
- Bei erfindungsgemäßer Verabreichung von Inhibitoren der Sexualsteroidaktivität werden diese vorzugsweise in einer Dosierung von etwa 1 mg bis etwa 2000 mg des Wirkstoffes (z. B. Sexualsteroidaktivitätsinhibitors) pro Tag pro 50 kg Körpergewicht, am stärksten bevorzugt in einer Dosierung von etwa 10 mg bis etwa 100 mg pro Tag pro 50 kg Körpergewicht verabreicht.
- Pharmazeutische Zusammensetzungen umfassen therapeutisch wirksame Mengen eines oder mehrerer der vorstehend diskutierten Inhibitoren der Sexualsteroid-Aktivität (einschließlich Antiöstrogenen), wobei ein pharmazeutisch akzeptables Verdünnungsmittel oder ein pharmazeutisch akzeptabler Träger zusammen mit dem/den Wirkstoff(en) enthalten ist. In Abhängigkeit von der Art der Verabreichung der pharmazeutischen Zusammensetzung kann das Verdünnungsmittel oder der Träger gemäß den bekannten Verfahren unterschiedlich sein.
- Eine zur oralen Verabreichung geeignete Zusammensetzung kann vorzugsweise mindestens einen Inhibitor der Sexualsteroid-Aktivität enthalten, wobei die Gesamtkonzentration aller dieser Inhibitoren in der pharmazeutischen Zusammensetzung etwa 1% bis etwa 95% (bezogen auf das Gewicht) der Zusammensetzung, vorzugsweise etwa 5% bis etwa 20% beträgt. Die Zusammensetzung enthält ferner vorzugsweise ein pharmazeutisch verträgliches Verdünnungsmittel, zum Beispiel Stärke oder Lactose mit oder ohne Tartrazin.
- Bei der Herstellung einer parentalen Injektion wird einem Träger, der vorzugsweise ausgewählt ist aus der Gruppe bestehend aus physiologischer Kochsalzlösung, Wasser, wässrigem Ethanol, wässrigem Dimethylsulfoxid und Öl ein Inhibitor der Sexualsteroid-Aktivität vorzugsweise in einer. Konzentration von etwa 1 mg/ml bis etwa 100 mg/ml (vorzugsweise etwa 2 mg/ml bis etwa 10 mg/ml) zugegeben.
- Eine zur parentalen Verabreichung geeignete Zusammensetzung enthält erfindungsgemäß vorzugsweise einen Träger und ein erfindungsgemäßes Antiöstrogen in einer Konzentration, die ausreicht, um etwa 1 mg bis etwa 1000 mg (vorzugsweise 5 bis 50 mg) des Antiöstrogens pro 50 kg Körpergewicht pro Tag einzuführen. Der Volumenfluss variiert selbstverständlich mit der Konzentration, bei der die pharmazeutische Zusammensetzung verabreicht wird.
- In frühen Behandlungsstadien werden vorzugsweise gelegentlich Blutproben entnommen und die Dosierung wird im Bedarfsfall geändert, um Gesamt-Serumkonzentrationen der Wirkstoffe von etwa 0,2 ug/ml bis 10 ug/ml aufrechtzuerhalten.
- In bestimmten anderen Ausführungsformen kann die pharmazeutische Zusammensetzung gemäß den bekannten Verfahren im Hinblick einer verzögerten Freisetzung formuliert werden. Diese Formulierungen zur verzögerten Freisetzung werden vorzugsweise in geeigneter Weise zur oralen, intramuskulären oder subkutanen Verabreichung zubereitet.
- Bevorzugte erfindungsgemäße Verbindungen enthalten
- N-n-Butyl-N-methyl-11-(7'-hydroxy-3'-(4"-hydroxyphe -nyl)-4'-methyl-2H-benzopyran-2-yl)undecanamid (EM 467)
- und 7-Hydroxy-3-(4'-hydroxyphenyl)-4-methyl-2-2-(6"- pipe-ridino)hexyl-2H-benzopyran (EM 721)
- Die folgende erfindungsgemäße Verbindung:
- wurde synthetisiert und hinsichtlich ihrer Fähigkeit, das Wachstum der menschlichen Brustkrebs- Zell-Linie ZR-75-1 zu hemmen, getestet. Die Ergebnisse sind in Fig. 1 dargestellt, wie nachstehend ausführlicher diskutiert.
- Die Synthese ist in Schema A (Seite 22 hierin) und Schema 33 (Seite 30 hierin) beschrieben. Die Synthese der Verbindung 23 ist in Schema A beschrieben, wobei die Ausgangsmaterialien und Reagenzien von Aldrich Chemical Company Inc. (Milwaukee, Wis.) bezogen wurden. Demgemäß wurde Salzsäure 1 (20,0 g; 0,1 mol) tropfenweise zu Methanol (60 ml) bei Raumtemperatur und unter Rühren hinzugegeben. Die Lösung wurde unter Rückfluss 1 h erhitzt. Das Lösungsmittel wurde unter vermindertem Druck entfernt und das resultierende Öl wurde in Ethylacetat gelöst. Die organische Lösung wurde mit wässriger, gesättigter Natriumbicarbonatlösung gewaschen. Die organische Lösung wurde getrocknet (MgSO&sub4;) und das Lösungsmittel wurde unter vermindertem Druck entfernt. Der Rückstand wurde mittels Chromatographie an Kieselgel (Ethylacetat : Hexane; 1 : 9) gereinigt, wobei die Verbindung 2 (Schema A) (18,0 g; 92 %) erhalten wurde.
- Der vorstehend erhaltene Ester 2 (21,2 g; 0,108 mol) und das Nitril 3 (erhältlich von Aldrich Chemical Company Inc., Milwaukee, Wis.)(17,5 ml; 0,129 mol) wurden in Benzol (750 ml) gelöst. Die Lösung wurde unter Rückfluss erhitzt und etwas Benzol (100 ml) wurde mit einem Dean Stark-Apparat entfernt. Die Lösung ließ man herunterkühlen; dann wurde Natriumethoxid (9,2 g; 0,135 mol) hinzugefügt. Das erhaltene Gemisch wurde 18 h unter Rückfluss erhitzt; es wurde mit 1 N wässriger Salzsäurelösung gewaschen. Die organische Lösung wurde getrocknet (MgSO&sub4;) und das Lösungsmittel wurde unter vermindertem Druck entfernt. Der Rückstand wurde mittels Chromatographie an Kieselgel gereinigt (Ethylacetat : Hexane; 1 : 4), wobei Verbindung 4 (23 g; 69 %) erhalten wurde. - Eine Lösung des Ketons 4 (37,8 g; 0,12 mol) in Essigsäure (400 ml) und in konzentrierter, wässriger Salzsäurelösung (200 ml) wurde 3 h bei 90 ºC gehalten. Das Reaktionsgemisch wurde mit konzentrierter, wässriger Natriumhydroxidlösung neutralisiert und wurde dann mit Ethylacetat extrahiert. Der organische Extrakt wurde getrocknet (MgSO&sub4;) und das Lösungsmittel wurde unter vermindertem Druck entfernt. Der Rückstand wurde mittels Chromatographie an Kieselgel (Ethylacetat : Hexane; 1 : 9) gereinigt, wobei die Verbindungen 5 (17,0 g; 49%) und 6 (6,6 g; 20%) erhalten wurden.
- Einem Gemisch der Ketone 5 (17,0 g; 59,4 mmol) und 6 (6,6 g; 24,3 mmol) wurde Pyridinhydrochlorid (90 g) zugegeben. Das Gemisch wurde 20 min bei 220ºC erhitzt. Eine wässrige 1 N Salzsäurelösung (250 ml) wurde zugegeben und das resultierende Gemisch wurde mehrmals mit Ethylacetat extrahiert. Der organische Extrakt wurde getrocknet (MgSO&sub4;) und das Lösungsmittel wurde unter vermindertem Druck entfernt. Der Rückstand wurde mittels Chromatographie an Kieselgel (Ethylacetat : Hexane; 3 : 7) gereinigt, wobei Verbindung 23 (14,1 g, 69%) erhalten wurde.
- Die weitere Synthese von EM 343 ist nachstehend in Schema 33 (Seite 30 hierin) beschrieben.
- Einem Gemisch des Triphenols 23 (14,1 g; 57,8 mmol) in 3,4-Dihydro-2H-pyran (200 ml) wurde vorsichtig p-Toluolsulfonsäure-Monohydrat (2,0 g) bei 0ºC unter starkem Rühren zugegeben. Das Reaktionsgemisch wurde eine weitere 1 h bei 0ºC gerührt. Ether (300 ml) wurde zugegeben und die Lösung wurde mit wässriger 1 N Natriumhydroxidlösung gewaschen. Der organische Extrakt wurde getrocknet (MgSO&sub4;) und das Lösungsmittel wurde unter vermindertem Druck entfernt. Der Rückstand wurde mittels Chromatographie an Kieselgel (Ethylacetat : Hexane; 1 : 9) gereinigt, wobei Verbindung 24 (23,4 g; 98%) erhalten wurde. - Einem Gemisch des Diethers 24 (24,4 g; 59,2 mmol) und des Aldehyds (OHCC&sub6;H&sub4;OH) (7,6 g; 62,18 mmol) in trockenem Benzol (750 ml) wurde Piperidin (500 ul) zugegeben. Die Lösung wurde 48 h unter Rückfluss erhitzt und Wasser wurde mit einem Dean Stark- Apparat kontinuierlich entfernt. Das Lösungsmittel wurde unter vermindertem Druck entfernt und der Rückstand wurde mittels Chromatographie an Kieselgel (Ethylacetat : Hexane; 1 : 9) gereinigt, wobei die Verbindungen 25 (R = H) (14,3 g; 47%) und 26 (R = H) (8,4 g; 27%) erhalten wurden. Letztere Verbindung kann durch Erhitzen mit Natriumacetat in Methanol zu Verbindung 25 (R = H) umgewandelt werden.

- Ein Gemisch der Verbindung 25 (R = H) (1,90 g; 3,8 mmol), 1-(2-Chlorethyl)piperidinhydrochlorid Cl(CH&sub2;)&sub2;NC&sub5;H&sub1;&sub0;. HCl (1,18 g; 6,5 mmol) und Natriumcarbonat (0,97 g; 9,1 mmol) in Aceton (100 ml) wurde 48 h unter Rückfluss und unter Rühren gehalten. Das Präzipitat wurde abfiltriert und gründlich mit Aceton gewaschen. Das Lösungsmittel wurde unter vermindertem Druck entfernt und der Rückstand wurde mittels Chromatographie an Kieselgel (Hexane : Aceton; 7 : 3 + ein paar Tropfen Triethanolamin) gereinigt, wobei Verbindung 25 (R = (CH&sub2;)&sub2;NC&sub5;H&sub1;&sub0;) (1,57 g; 66%) erhalten wurde. Einer Lösung der Verbindung 25 (R = (CH&sub2;)&sub2;NC&sub5;H&sub1;&sub0;) (90 mg; 143 umol) in Ether (30 ml) wurde Methylmagnesiumjodid (3,0 M Lösung in Ether; 1,2 ml; 3,6 mmol) bei 0ºC und unter Rühren zugegeben. Das Gemisch wurde weitere 3 h bei Raumtemperatur gerührt und dann mit gesättigter Ammoniumchloridlösung gewaschen. Die organische Lösung wurde getrocknet (MgSO&sub4;) und das Lösungsmittel unter vermindertem Druck entfernt. Der Rückstand wurde schnell durch Kieselgel (Ethylacetat : Aceton; 1 : 1) filtriert, wobei Verbindung 27 (R = (CH&sub2;)&sub2;NC&sub5;H&sub1;&sub0;, Rc = CH&sub3;) (90 mg; 97%) erhalten wurde, die direkt in der nächsten Reaktion verwendet wurde.
- Eine Lösung von Verbindung 27 (R = (CH&sub2;)&sub2;NC&sub5;H&sub1;&sub0;, Rc = CH&sub3;) (90 mg; 139 umol) in einem Gemisch aus Essigsäure (60 ml) und Wasser (6 ml) wurde 10 min bei 100ºC gehalten. Das Lösungsmittel wurde unter vermindertem Druck entfernt und der Rückstand wurde mittels Kieselgel-Chromatographie (Ethylacetat : Aceton; 3 : 2) gereinigt, wobei Verbindung 28 (EM 343, R = (CH&sub2;)&sub2;NC&sub5;H&sub1;&sub0;, Rc = CH&sub3;) (40 mg; 62%) erhalten wurde. - Das Produkt EM 343 wurde dann für Wirksamkeitsstudien zubereitet, indem die humane Brustkrebszelllinie ZR-75-1 verwendet wurde.
- ZR-75-1-Zellen (83. Passage) wurden von der American Type Culture Collection (Rockville, MD) erhalten und routinemäßig in phenolrotfreiem RPMI 1640- Medium, supplementiert mit 1 nM E&sub2;, 2 mM L- Glutamin, 1 mM Natriumpyruvat, 15 mM N-2- Hydroxyethyl-piperazin-N'-2-ethansulfonsäure, 100 IU Penicillin/ml, 100 ug Streptomycin/ml und 10% (v/v) fötalem Rinderserum (Hyclone, Logan, UT), in einer feuchten Atmosphäre von 95% Luft und 5% CO&sub2; bei 37ºC kultiviert. Alle Medien und Medienzutaten wurden von Sigma bezogen. Zellen wurden wöchentlich durch Behandlung mit einer Pankreas-Lösung, enthaltend 0,02% EDTA (w/v), subkultiviert. Für die hier beschriebenen Experimente wurden Zellkulturen der Passagen 89 bis 94 verwendet.
- Zellen wurden in ihrer logarithmischen Wachstumsphase geerntet, kurz zentrifugiert und in RPMI 1640-Medium resuspendiert. Dann wurden die Zellen jeweils 3-mal auf LIMBRO-Plastik-Kulturplatten mit 24 Vertiefungen (2 cm²/Loch) plattiert. Da die Plattierungsdichte die Hormonwirkung auf das Wachstum der ZR-75-1-Zellen beeinflusst, wurden die Zellen in einer Dichte von 1 · 10&sup4; Zellen/Vertiefung plattiert. Nach 72 h wurde das Medium durch frisches Medium mit identischer Zusammensetzung ersetzt, das zusätzlich die auf der X-Achse von Fig. 1 gezeigten Konzentrationen an Steroiden und/oder Inhibitoren (z. B. EM 312 oder EM 343) enthielt. Kontrollkulturen erhielten nur das Vehikel Ethanol. Dann ließ man die Zellen 10 Tage bei 37ºC wachsen, wobei alle 2 Tage ein Wechsel der Medien (identischer Zusammensetzung) erfolgte. In Abwesenheit von Inhibitoren haben die Zellen in 0,1 nM Östradiol- (E&sub2;)-enthaltendem Medium eine Verdopplungszeit von ungefähr 48 h.
- Nach der E&sub2;- und/oder Antiöstrogen-Behandlung wurden die Zellen durch Zugabe von 0,5 ml einer Pankreatinlösung (Sigma) 5-10 min bei 37ºC geerntet, bevor 0,5 ml RPMI 1640-Medium, enthaltend 5 % Dextran-beschichtete Aktivkohle-fötales Rinderserum, zugegeben wurde, um die Enzymwirkung zu blockieren. Die Zellzahl (0,10 ml-Aliquote) wurde durch Messung des DNA-Gehaltes, wie bereits beschrieben, (Simard et al., Endocrinology, 126 (1990), S. 3223-3231) bestimmt.
- Wie aus Fig. 1 ersichtlich, hemmte EM 343 das Zellwachstum bei einer niedrigen Konzentration sehr stark. Eine halb-maximale Wirkung trat bei einer Konzentration von 2,55 · 10&supmin;¹&sup0; M auf. Ohne an eine Theorie gebunden zu sein, ist davon auszugehen, dass die B-Ring-Alkyl-Substitution, die in bestimmten bevorzugten Ausführungsformen der Erfindung genutzt wird (z. B. die Methyl-Substitution von EM 343), die Wirksamkeit gegenüber Verbindungen, denen eine solche Substitution fehlt, verstärkt. Ein nahes Analogon der erfindungsgemäßen Verbindungen
- das nach Saeed et al. (J. Med. Chem., 33 (1990), S. 3210-3216) und Sharma et al. (J. Med. Chem. 33 (1990), S. 3216-3222 und S. 3222-3229) Antiöstrogenaktivität aufweisen soll, zeigte beispielsweise bei vergleichenden Untersuchungen in dem Labor schlechtere Ergebnisse auf das Wachstum von menschlichen ZR-75-1-Brustkrebszellen, wobei der IC&sub5;&sub0;-Wert von EM 312 mit 8,43 · 10&supmin;¹ M 3-mal höher (Fig. 1) war.
- Ohne an eine Theorie gebunden zu sein, ist davon auszugehen, dass die hier diskutierten Diphenylethylen- und Diphenylethenylenkerne zu einer verstärkten Affinität zum Östrogenrezeptor beitragen. Die Ethenylen-Variante mit der vorhandenen optionalen Doppelbindung wird bevorzugt, da es einen geschlossenen dritten Ring gibt, der in seinen Seiten die Ethenyl-Doppelbindung und eine Seite einer der Phenylgruppen umfasst. Der geschlossenen dritte Ring wird durch den "B"-Ring der nachstehenden Formel XXI
- veranschaulicht.
- Der G³-Substituent von Formel XXI ist vorzugsweise ein C&sub1;-C&sub3;-Kohlenwasserstoff, wie zum Beispiel eine Methyl-, Ethyl- oder Propyl-Gruppe. Im Allgemeinen ist diese Substitutionsstelle, d. h. das Atom, das G³ von Formel XXI aufnimmt, das Atom, welches sowohl (A) ein Atom vom A-Ring entfernt ist als auch (B) eines von den zwei Atomen, die, wenn vorhanden, die optionale Doppelbindung bilden. Im Gegensatz dazu ist die bevorzugte Seitenkette (z. B. R¹&sup0;&sup0;LG¹G² von Formel XXI) vorzugsweise an dem Atom substituiert, welches sowohl (A) ein Atom von der optionalen Pi-Bindung entfernt als auch (B) ein Atom von Z entfernt ist, wie in Formel XXI gezeigt. Es ist davon auszugehen, dass eine niedrigere Kohlenwasserstoff-Substitution an dem entsprechenden von Formel XXI die Wirksamkeit eines Inhibitors wesentlich verstärkt.
- Während sowohl die A- als auch die 17-Ringe unsubstituiert sein können, umfassen bestimmte Ausführungsformen Hydroxysubstitution an einem oder beiden der A- und D-Ringe (besonders an den 3- und/oder 12-Positionen der Formel XXI) oder einen Rest, der nach Verabreichung der pharmazeutischen Verbindung an einen Patienten, zu einer Hydroxyl- Gruppe in vivo umgewandelt werden kann. Solche Substitutionen umfassen zum Beispiel Methoxy-, Ethoxy- oder Ester-Gruppe.
- Nachstehend sind mehrere bevorzugte Verbindungen zur Verwendung in der erfindungsgemäßen pharmazeutischen Zusammensetzung aufgezeigt, die der Struktur
- entsprechen. Beispiel 33 Schema 33
- Die Verbindung 26 (R = H, Rc = H) wurde mit Natrium-borhydrid in Ethanol reduziert und ein Gemisch der resultierenden Verbindung (300 mg; 0,6 mmol), 4-(2-Chlorethyl)morpholinhydrochlorid (267 mg; 1,4 mmol), Zäsiumcarbonat (978 mg; 3,0 mmol) und Kaliumjodid (100 mg; 0,6 mmol) in N,N-Dimethylformamid (10 ml) wurde 1 h bei 90ºC und unter Rühren gehalten. Wasser wurde zugegeben und das resultierende Gemisch wurde mehrmals mit einem Gemisch aus Ether und Ethylacetat (1 : 1) extrahiert. Durch Trocknen (MgSO&sub4;) und Entfernen des Lösungsmittels wurde ein wachsartiges Material erhalten, das mittels Chromatographie an Kieselgel (Hexane : Ethylacetat, 3 : 7 + ein paar Tropfen Triethylamin) gereinigt wurde, wobei das dehydratisierte Derivat der Verbindung 27 (R = (CH&sub2;)&sub2;NC&sub4;H&sub8;O, Rc = H) (153 mg; 41%) erhalten wurde.
- Eine Lösung der vorstehend erhaltenen Verbindung (153 mg; 249 umol) in einem Gemisch aus Essigsäure (60 ml) und Wasser (6 ml) wurde 10 min bei 100ºC gehalten. Das Lösungsmittel wurde unter vermindertem Druck entfernt und der Rückstand wurde mittels Chromatographie an Kieselgel (Ethylacetat : Aceton; 3 : 1) gereinigt, wobei Verbindung 28 (EM 349, R = (CH&sub2;)&sub2;NC&sub4;H&sub8;O, Rc = H) (100 mg; 90%) erhalten wurde. - Die Verbindung 26 (R = H, Rc = H) wurde auf diese Weise mit Natriumborhydrid in Ethanol reduziert und ein Gemisch der resultierenden Verbindung (300 mg; 0,6 mmol), Chloressigsäure, Piperidylamid (242 mg; 1,5 mmol) und Zäsiumcarbonat (978 mg; 3,0 mmol) in N,N-Dimethylformamid (10 ml) wurde 1 h bei 90ºC und unter Rühren gehalten. Wasser wurde zugegeben und das resultierende Gemisch wurde mehrmals mit Ether extrahiert. Durch Trocknen (MgSO&sub4;) und Entfernen des Lösungsmittels unter vermindertem Druck wurde ein gummiartiger Rückstand erhalten, der mittels Chromatographie an Kieselgel (Hexane : Ethylacetat; 1 : 1) gereinigt wurde, wobei Verbindung 27 (R = CH&sub2;CONC&sub5;H&sub1;&sub0;, Rc = H) (127 mg; 34%) erhalten wurde.
- Eine Lösung der vorstehenden Verbindung (127 mg; 203 umol) in einem Gemisch aus Essigsäure (10 ml) und Wasser (1 ml) wurde 10 min bei 100ºC gehalten. Das Lösungsmittel wurde unter vermindertem Druck entfernt und der Rückstand wurde mittels Chromatographie an Kieselgel (Ethylacetat : Hexane; 1 : 1) gereinigt, wobei Verbindung 28 (EM 350, R = CH&sub2;CONC&sub5;H&sub1;&sub0;, Rc = H) (43 mg; 46%) erhalten wurde. - Alle diese Verbindungen wurden mit Hilfe des folgenden Verfahrens hergestellt. Wenn jedoch n größer als 1 war, wurde während der Kopplungsreaktionen dem Reaktionsgemischen Kaliumjodid zugegeben.
- Die Verbindung 26 (R = H, Rc = H) wurde demgemäß mit Natriumborhydrid in Ethanol reduziert und ein Gemisch der resultierenden Verbindung (418 mg; 0,84 mmol), N-Methyl,N-butylchloracetamid (342 mg; 2,09 mMol) und Zäsiumcarbonat (1,36 g; 4,18 mMol) in N,N-Dimethylformamid (20 ml) wurde 12 h bei 90ºC unter Rühren aufbewahrt. Wasser wurde zugegeben und das resultierende Gemisch wurde mehrmals mit Ether extrahiert. Durch Trocknen (MgSO&sub4;) und Entfernen des Lösungsmittels unter vermindertem Druck wurde ein gummiartiger Rückstand erhalten, der mittels Chromatographie an Kieselgel (Hexane. Ethylacetat; 1 : 1) gereinigt wurde, wobei Verbindung 27 (R = CH&sub2;CONMeBu, Rc = H) (276 mg; 53%) erhalten wurde.
- Eine Lösung der vorstehenden Verbindung (138 mg; 220 umol) in einem Gemisch aus Essigsäure (10 ml) und Wasser (1 ml) wurde 10 min bei Raumtemperatur gehalten. Das Lösungsmittel wurde unter vermindertem Druck entfernt und der Rückstand wurde mittels Chromatographie an Kieselgel (Ethylacetat : Hexane; 1 : 1) gereinigt, wobei Verbindung 28 (EM 357, R = CH&sub2;CONMeBu, Rc = H) (38 mg; 37%) erhalten wurde. - Ein Gemisch der Verbindung 24 (2,52 g; 6,12 mmol), de s Aldehyds 29 (n = 7, Ra = Me, Rb = Bu) (1,00 g; 4,08 mmol) und Piperidins (500 ul) in Benzol (170 ml) wurde 48 h unter Rückfluss mittels eines Dean Stark-Apparates erhitzt. Das Lösungsmittel wurde unter vermindertem Druck entfernt und das resultierende Öl wurde mittels Chromatographie an Kieselgel (Hexane : Ethylacetat; 7 : 3) gereinigt, wobei das Chalcon 30 (n = 7, Ra = Me, Rb = Bu) (620 mg; 77% korrigierter Ertrag) und das nicht umgesetzte Ausgangsmaterial 24 (2,00 g) erhalten wurden.
- Einer Lösung des Chalcons 30 (n = 7, Ra = Me, Rb = Bu) (469 mg; 0,73 mmol) in Ethanol (30 ml) wurde Natriumborhydrid (34 mg; 0,89 mmol) bei Raumtemperatur unter Rühren zugegeben. Das Reaktionsgemisch wurde weitere 12 h bei Raumtemperatur gerührt. Das Lösungsmittel wurde unter vermindertem Druck entfernt. Der ölige Rückstand wurde in Ethylacetat aufgenommen und mehrmals mit wässriger, gesättigter Ammoniumchloridlösung gewaschen. Dir organische Extrakt wurde getrocknet (MgSO&sub4;) und das Lösungsmittel wurde unter vermindertem Druck entfernt. Der Rückstand wurde mittels Chromatographie an Kieselgel (Hexane : Ethylacetat; 4 : 1) gereinigt, wobei das Chromen 31 (n = 7, Ra = Me, Rb = Bu, Rc = H) (300 mg; 66%) erhalten wurde.
- Eine Lösung der Verbindung 31 (n = 7, Ra = Me, Rb, = Bu, Rc = H) (300 mg; 482 umol) in einem Gemisch aus Essigsäure (30 ml) und Wasser (3 ml) wurde 3 0 min bei 100ºC gehalten. Das Lösungsmittel wurde unter vermindertem Druck entfernt und der Rückstand wurde mittels Chromatographie an Kieselgel (Ethylacetat : Hexane; 1 : 4) gereinigt, wobei Verbindung 32 (EM 345, n = 7, Ra = Me, Rb = Bu, Rc = H) (104 mg; 48%) erhalten wurde. - Einer Lösung des Amids 31 (n = 10, Ra = Me, R = Bu, Rc = H) (100 mg; 0,15 mmol) in Tetrahydrofuran (10 ml) wurde eine Lösung von Lithiumaluminiumhydrid (1 M in Tetrahydrofuran; 0,42 ml; 0,42 mmol) unter Rückfluss und unter Rühren zugegeben. Das resultierende Gemisch würde weitere 48 h unter Rückfluss gehalten. Eine 2 N wässrige Natriumhydroxidlösung wurde zu dem Reaktionsgemisch zugegeben und die wässrige Schicht wurde mehrmals mit Ethylacetat extrahiert. Die vereinigten organischen Extrakte wurden getrocknet (MgSO&sub4;) und das Lösungsmittel wurde unter vermindertem Druck entfernt. Der Rückstand wurde mittels Chromatographie an Kieselgel (Hexane : Aceton; 1 : 1) gereinigt, wobei das Amin 33 (n = 10, Ra = Me, Rb = Bu, Rc = H) (60 mg; 62%) erhalten wurde.
- Eine Lösung der vorstehenden Verbindung (60 mg; 93 umol) und Pyridin-p-Toluolsulfonat (46 mg; 185 umol) in Methanol (10 ml) wurde 12 h unter Rückfluss gehalten. Das Lösungsmittel wurde unter vermindertem Druck entfernt und der Rückstand wurde mittels Chromatographie an Kieselgel (Hexane : Aceton; 3 : 2) gereinigt, wobei das Chromen 34 erhalten wurde (EM 371, n = 10, Ra = Me, Rb = Bu, Rc = H) (30 mg; 67%) (δ NMR; 300 MHz; Lösungsmittel: CD&sub3;OD; Standard: TMS) 0,93 (3H; t; J 7,5 Hz; N-CH&sub2;- CH&sub2;-CH&sub2;-CH&sub3;). 1,15-1,85 (24H; m; N-CH&sub2;-CH&sub2;-CH&sub2;-CH&sub3; und O-CH-(CH&sub2;)&sub1;&sub0;-CH&sub2;-N) 2,24 (3H; s; N-CH&sub3;) 2,35- 2,45 (4H; m; CH&sub2;&submin;N-CH&sub2;) 5,19 (1H; dd; J 2,5 Hz, 10 Hz; O-CH-CH&sub2;) 6,26 (1H; d; J 2,5 Hz; CH Phenyl) 6,33 (1H; dd; J 2,5 Hz, 8,5 Hz; CH Phenyl) 6,59 (1H; s; HC=C) 6,78 (2H; d; 8,5 Hz; OH Phenyl) 6,88 (1H; d; J 8 Hz; CH Phenyl) 7,30 (2H; d; J 8,5 Hz; CH Phenyl). Claims (12)
1. Verbindung der Molekülstruktur
worin sind:
die gestrichelte Linie eine optionale Doppelbindung;
R&sup5; und R&sup6; unabhängig Wasserstoff, Hydroxyl oder ein Teil, der in vivo in
Hydroxyl überfuhr wird;
R¹&sup0;&sup0; ein zweiwertiger Teil, der L in Bezug auf den B-Ring durch 4 bis 10
Zwischenatome auf Distanz bringt, ausgewählt aus geradkettigen oder verzweigten
Alkylen-, Alkenylen- und Alkinylen-Gruppen und Phenylen-Gruppen sowie
Phenylenoxyalkylen-Gruppen;
L ein zweiwertiger oder dreiwertiger polarer Teil ausgewählt aus CO-, -SO-, -
CONC< , -N< und -SON< ;
worin G¹ entweder fehlt oder ausgewählt ist aus Wasserstoff; einem C&sub1; bis C&sub5;-
Kohlenwasserstoff, einem gesättigten oder ungesättigten C&sub5;- bis C&sub7;-Cycloalkyl, einem
zweiwertigen Teil, der G² und L unter Bildung eines 5- bis 7-gliedrigen heterocyclischen
Ringes verbindet, sowie halogensubstituierten Derivaten der Vorgenannten;
worin G² entweder fehlt oder ausgewählt ist aus Wasserstoff, einem C&sub1; bis C&sub5;-
Kohlenwasserstoff, einem substituierten oder nichtsubstituierten C&sub5;- bis C&sub7;-Cycloalkyl,
einem zweiwertigen Teil, der G¹ und L unter Bildung eines 5- bis 7-gliedrigen
heterocyclischen Ringes verbindet, sowie halogensubstituierten Derivaten der
Vorgenannten;
worin G³ ein C&sub1;- bis C&sub3;-Kohlenwasserstoff ist.
2. Verbindung nach Anspruch 1, worin G³ ausgewählt ist aus Methyl, Ethyl und
Propyl.
3. Verbindung nach Anspruch 1, worin L, G¹ und G² gemeinsam -NCxH2x oder
-NCx-1H2x-2O sind (worin x eine ganze Zahl von 4 bis 6 ist).
4. Verbindung mit der Molekülstruktur
worin R&sup9; Methyl, Ethyl, Propyl, Ethenyl oder Ethinyl ist.
