Description
- Die vorliegende Erfindung betrifft neue Taxane, die als Mittel gegen Leukämie und Tumore brauchbar sind. Die Taxanfamilie der Terpene von denen Taxol ein Vertreter ist, hat in der biologischen und chemischen Wissenschaft beträchtliches Interesse auf sich gezogen. Taxol ist ein vielversprechendes Krebs-Chemotherapeutikum mit einem breiten Spektrum von antileukemischer und tumorhemmender Aktivität. Taxol hat eine 2'R, 3'S-Konfiguration und die folgende Strukturformel:
- worin Ac Acetyl bedeutet. Wegen dieser günstigen Aktivität wird Taxol gegenwärtig klinischen Versuchen in Frankreich und in den Vereinigten Staaten unterzogen.
- Colin et al. berichteten in US-A-4,814,470, daß Taxol-Derivate mit der nachfolgend angegebenen Strukturformel (2) eine Aktivität haben, die bedeutend größer als die des Taxols (1) ist.
- R' bedeutet Wasserstoff oder Acetyl, und einer von R" und R"' bedeutet Hydroxy und der andere davon bedeutet tert-Butoxy-carbonylamino und seine stereoisomeren Formen, sowie deren Gemische. Die Verbindung der Formel (2), in der R' wasserstoff, R" Hydroxy und R"' tert-Butoxy-carbonylamino in der 2'R, 3'S-Konfiguration ist, wird allgemein als Taxotere bezeichnet.
- Obgleich Taxol und Taxotere vielversprechende Chemotherapeutika sind, sind sie nicht universell wirksam. Demgemäß verbleibt ein Bedarf an weiteren therapeutischen Mitteln.
- Eine der Aufgaben der vorliegenden Erfindung ist daher die Schaffung neuer Taxan-Derivate, die wertvolle Antileukämie- und Antitumormittel sind.
- Die vorliegende Erfindung ist daher kurz gesagt gerichtet auf Taxan-Derivate der Formel:

- Base = NH&sub3;, (HOC&sub2;H&sub4;)&sub3;N, N(CH&sub3;)&sub3;, CH&sub3;N(C&sub2;H&sub4;OH)&sub2;, NH&sub2;(CH&sub2;)&sub6;NH&sub2;, N-Methylglucamin, NaOH oder KOH.
- Andere Aufgaben und Merkmale der Erfindung werden nachfolgend teils deutlich und teils ausgeführt.
- Erfindungsgemäß wurde gefunden, daß Verbindungen mit der Strukturformel (3) im allgemeinen und den Strukturformeln (4) und (5) im besonderen in vitro bemerkenswerte Eigenschaften zeigen und wertvolle Antileukämie- und Antitumormittel sind. Ihre biologische Aktivität wurde in vitro mit Tubulinproben nach den Verfahren von Parness et al., J.Cell Biology, 91:479-487 (1981) und menschlichen Krebszellstämmen bestimmt und ist vergleichbar mit der von Taxol und Taxoteren.
- Taxane mit den Formeln (4) und (5) und der 2'R, 3'S-Konfiguration können erhalten werden durch Umsetzung eines β-Lactams mit Metallalkoxiden, die den tetrazyklischen Taxan-Kern und an C-13 einen Metalloxid- Substituenten haben, unter Bildung von Verbindungen mit einem β-Amidoester-Substituenten an C-13. Die β-Lactame haben beispielsweise die folgende Strukturformel:

- β-Lactame (6) können aus leicht erhältlichem Ausgangsmaterialien hergestellt werden, wie durch das folgende Reaktionsschema erläutert wird:
- (a) LDA, -78ºC bis -50ºC;
- (b) LHMDS, THF, -78ºC bis 0ºC;
- (c) THF, -78ºC bis 25ºC, (2h); und
- (d) Triethylamin und ein Acylchlorid
- oder Alkylchloroformat (R&sub3; = Alkoxycarbonyl) (LDA ist Lithiumdiisopropylamid und LHMDS ist Lithiumhexamethyldisilazan).
- Die 3-Hydroxy-Schutzgruppe ist in dem obigen Reaktionsschema ist -SiR&sub5;, worin R&sub5; Trialkyl oder Triaryl, wie z. B. Triethyl ist. Das 3-Hydroxyl kann mit anderen Standard-Schutzgruppen geschützt werden, wie z. B. 1- Ethoxyethyl oder 2,2,2-Trichlorethoxymethyl. Weitere Hydroxy-Schutzgruppen und ihre Synthese sind zu finden in "Protective Groups in Organic Synthesis" von T.W. Greene, John Wiley & Sons, 1981.
- Die razemischen β-Lactame können vor dem Schutz in die reinen Enantiomeren durch Umkristallisation der entsprechenden 2-Methoxy-2-(Trifluoromethyl)phenylessigester getrennt werden. Die nachfolgend beschriebene Reaktion, bei der die β-Amidoester-Seitenkette angehängt wird, hat jedoch den Vorteil einer hohen Diastereoselektivität und erlaubt somit den Einsatz eines razemischen Gemisches aus Seitenketten-Vorprodukt.
- Die Metallalkoxide mit dem tetrazyklischen Taxankern und einem Metalloxid-Substituenten an C-13 haben die folgende Strukturformel:
- worin Z die Bedeutung von -OT&sub1; hat; T, Wasserstoff, Hydroxyl-Schutzgruppe oder -COT&sub2; ist; T&sub2; Wasserstoff, C&sub1;- C&sub6;-Alkyl, Alkenyl mit bis zu C&sub6;, Alkinyl mit bis zu C&sub6; oder monozyklisches Aryl ist; T&sub3; eine Hydroxy- Schutzgruppe ist und M ein Metall ist, das vorzugsweise aus der die Gruppe IA, die Gruppe IIA und die Übergangsmetalle umfassenden Gruppe ausgewählt ist und insbesondere Li, Mg, Na, K oder Ti ist.
- Metallalkoxide werden hergestellt durch Umsetzung eines Alkohols, der einen tetrazyklischen Taxankern und an C-13 eine Hydroxylgruppe hat, mit einer metallorganischen Verbindung in einem geeigneten Lösungmittel Vorzugsweise ist der Alkohol ein aeschütztes Baccatin III, insbesondere 7-O-Triethylsilyl- Baccatin III (das nach der Beschreibung von Greene et al. in JACS 110: 5917 (1988) oder auf anderen Wegen erhalten werden kann) oder 7,10-bis-O-Triethylsilyl-Baccatin III.
- Wie bei Greene et al. berichtet wird, wird 10- Deacetyl Baccatin III zu 7-O-Triethylsilyl-10-Deacetyl- Baccatin III gemäß dem folgenden Reaktionsschema umgesetzt:
- Nach den gemäß Bericht sorgfältig optimierten Bedingungen wird 10-Deacetyl-Baccatin III mit 20 Äquivalenten (C&sub2;H&sub5;)&sub3;SiCl bei 23ºC unter einer Argonatmosphäre 20 Stunden in Gegenwart von 50 ml Pyridin/mmol 10-Deacetyl- Baccatin III umgesetzt, wobei sich als Reaktionsprodukt nach Reinigung in 84-86% Ausbeute 7-Triethylsilyl-10- Deacetyl-Baccatin III (9a) ergibt. Das Reaktionsprodukt kann dann wahlweise mit fünf Äquivalenten CH&sub4;COCl und 25 ml Pyridin/mmol der Verbindung 9a bei 0ºC 48 Stunden unter einer Argonatmosphäre acetyliert werden, wobei sich in 86% Ausbeute 7-O-Triethylsylin-Baccatin III (9b) ergibt, siehe Greene et al. in JACS 110, 5917 auf 5918 (1988).
- Das 7-O-Triethylsilyl-Baccatin III (9b) wird mit einer metallorganischen Verbindung, etwa n-Butyllithium, in einem Lösungsmittel, wie etwa Tetrahydrofuran (THF), unter Bildung des Metallalkoxids 13-O-Lithium-7-O- Triethylsilyl-Baccatin III (10) umgesetzt, wie in dem folgenden Reaktionsschema gezeigt ist:
- Wie in dem folgenden Reaktionsschema gezeigt wird, reagiert 13-O-Lithium-7-O-Triethylsilyl-Baccatin III (10) mit β-Lactam (6), in dem R&sub2; Triethylsilyl ist, zu einem Zwischenprodukt, in dem die Hydroxylgruppen an C-7 und C- 2' mit einer Triethylsilylgruppe geschützt sind. Die Triethylsilylgruppen werden dann unter milden Bedingungen hydrolysiert, damit die Esterbindung oder die Taxan- Substituenten nicht beeinträchtigt werden. Pyridin

- Die Umsetzung des Alkohols zu dem Metallalkoxid und die Endsynthese des Taxan-Derivates können in dem gleichen Reaktionsbehälter erfolgen. Vorzugsweise wird das β-Lactam dem Reaktionsbehälter zugesetzt nachdem sich in ihm das Metallalkoxid gebildet hat.
- Die vorliegende Erfindung schafft auch pharmazeutische Zusammensetzungen, die eine Verbindung der Formel (3) im allgemeinen und Verbindungen der Formeln (4) und (5) im besonderen in Kombination mit einem oder mehreren pharmazeutisch zulässigen, reaktionsträgen oder physiologisch aktiven Verdünnungsoder Zusatzstoffen enthalten.
- Diese Zusammensetzungen können in irgendeiner für den vorgesehenen Verabreichungsweg geeigneten Form abgegeben werden. Der parenterale Weg, und insbesondere der intravenöse Weg ist der bevorzugte Verabreichungsweg.
- Die erfindungsgemäßen Zusammensetzungen für die parenterale Verabreichung können wässrige oder nichtwässrige sterile Lösungen, Suspensionen oder Emulsionen sein. Propylenglykol, pflanzliche Öle, insbesondere Olivenöl, und injektionsfähige organische Ester, z.B. Ethyloleat, können als Lösungsmittel oder -träger dienen. Diese Zusammensetzungen können auch Zusatzstoffe enthalten, insbesondere Netzmittel, Emulgatoren oder Dispersionsmittel. Die Sterilisierung kann in verschiedener Weise erfolgen, z. B. unter Benutzung eines bakteriologischen Filters, durch Einbringen von Sterilisiermitteln in die Zusammensetzung, durch Bestrahlung oder durch Erhitzung. Die Zusammensetzungen können auch in Form steriler fester Zusammensetzungen vorliegen, die in sterilem Wasser oder irgendeinem anderen injizierbaren sterilen Medium gelöst oder dispergiert werden können.
- Die Produkte der allgemeinen Formel (3) werden insbesondere zur Behandlung akuter Leukämien und voll ausgebildeter Tumore in täglichen Dosen verwendet, die auf dem intravenösen Weg (Perfusion) im allgemeinen zwischen 1 und 2 mg/kg für einen Erwachsenen betragen.
- Die Wasserlöslichkeit der Verbindungen der Formel (3) kann dadurch verbessert werden, daß man die Substituenten an C2' und/oder C7 modifiziert, indem man geeignete funktionelle Gruppen E&sub1; und E&sub2; einbaut. Zur erhöhten Wasserlöslichkeit können E&sub1; und E&sub2; unabhängig voneinander Wasserstoff oder -COGCOR¹ sein, worin G und R¹ die oben definierte Bedeutung haben. Die Darstellung von Verbindungen, in denen E&sub1; oder E&sub2; die Bedeutung von -COGCOR¹ hat, ist in US-A-4942184 angegeben.
- Die folgenden Beispiele erläutern die Erfindung. BEISPIEL 1
- Einer Lösung von 7-Triethylsilyl-Baccatin III (100 15 mg, 0,143 mmol) in 1 ml THF wurden bei -45ºC tropfenweise 0,087 ml einer 1,63 N-Lösung von n-BuLi in Hexan zugesetzt. Nach 0,5 Stunden bei -45ºC wurde dem Gemisch tropfenweise eine Lösung von cis-1-Benzoyl-3- triethylsilyloxy-4-(2-furyl)azetidin-2-on (266 mg, 0,715 mmol) in 1 ml THF zugesetzt. Die Lösung wurde auf 0ºC erwärmt und 1 Stunde auf dieser Temperatur gehalten, bevor 1 ml einer 10% Lösung von AcOH in THF zugesetzt wurde. Das Gemisch wurde zwischen gesättigtem, wässrigem NaHCO&sub3; und Ethylacetat/Hexan 60/40 verteilt. Die Verdampfung der organischen Schicht ergab einen Rückstand, der durch Filtration durch Kieselsäuregel gereinigt wurde und 143 mg eines Gemisches ergab, das aus (2'R,3'S)-2', 7-(bis)Triethylsilyl-3'despenyl-3'- (2-furyl)-taxol und eine kleine Menge des (2'S,3'R)- Isomeren enthielt.
- Einer Lösung von 142 mg des aus der vorgenannten Reaktion erhaltenen Gemisches in 6 ml Acetonitril und 0,3 ml Pyridin wurden bei 0ºC 0,9 ml wässriger, 48%iger HF zugesetzt. Das Gemisch wurde 3 Stunden bei 0ºC und dann 13 Stunden bei 25ºC gerührt und zwischen gesättigtem, wässrigem Natriumbicarbonat und Ethylacetat verteilt. Die Verdampfung der Etylacetat-Lösung ergab 115 mg eines Materials, das nach Reinigung durch Kurzwegchromatographie 98 mg (81%) 3'-Desphenyl-3'-(2- furyl)-taxol ergab, das aus Methanol/Wasser umkristallisiert wurde.

- Einer Lösung von 7-Triethylsilyl-Baccatin III(100 mg, 0,143 mmol) in 1 ml THF wurde bei -45ºC tropfenweise 0,087 ml einer 1,63 M-Lösung von n-BuLi in Hexan zugesetzt. Nach 0,5 h bei -45ºC wurde dem Gemisch tropfenweise eine Lösung von cis-1-(4-Benzoyl)-3- Triethylsilyloxy-4-(2-thienyl)azetidin-2-on (277 mg, 0,715 mmol) in 1 ml THF zugesetzt. Die Lösung wurde auf 0ºC erwärmt und 1 h auf dieser Temperatur gehalten, bevor 1 ml einer 10% Lösung von AcOH in THF zugesetzt wurde. Das Gemisch wurde zwischen gesättigter wässriger NaHCO&sub3; und Ethylacetat/Hexan 60/40 verteilt. Die Verdampfung der organischen Schicht ergab einen Rückstand, der nach Reigung durch Filtration durch Kieselgel 169 mg eines Gemisches ergab, das (2'R,3'S)-2',7-(Bis)triethylsilyl- 3'-Desphenyl-3'-(2-thienyl)taxol und eine kleine Menge des (2'S,3'R)-Isomeren enthielt.
- Einer Lösung von 169 mg des aus der vorgenannten Umsetzung erhaltenen Gemisches in 6 ml Acetonitril und 0,3 ml Pyridin wurden bei 0ºC 0,9 ml 48% wässrige HF zugesetzt. Das Gemisch wurde 3 Stunden bei 0ºC, dann 13 Stunden bei 25ºC gerührt und zwischen gesättigtem, wässrigem Natriumbicarbonat und Ethylacetat verteilt. Die Eindampfung der Ethylacetat-Lösung ergab 140 mg Material, das bei Reinigung durch Kurzwegchromatographie 93 mg (76%) 3'-Desphenyl-3'-(2-thienyl)taxol ergab, das aus Methanol/Wasser umkristallisiert wurde.

- Es wurden Tubulin-Bindungsversuche mit den Verbindungen (4) und (5) im wesentlichen nach den Angaben in Parness et al., J. Cell Biology 91: 479-487 (1981) durchgeführt und mit Taxol und Taxotere verglichen. Die Ergebnisse sind in Tabelle 1 angegeben. - IC&sub5;&sub0;-Daten wurden in vitro von einem menschlichen Krebszellenstamm (HCT 116), der von dem National Cancer Institute erhältlich ist, und einem gegen viele Arzneimittel resistenten Zellstamm (HCT/VM46), der gegen verschiedene hydrophobe Mittel einschließlich Taxol resistent ist, erhalten. Die Cytotoxizität wurde an menschlichen Dickdarmkrebszellen HCT 116 und HCT VM46 durch XTT geprüft (2, 3-bis(2-Methoxy-4-nitro-5- sulfophenyl)-5-[(phenylamino)carbonyl]-2H-tetrazoliumhydroxid). (Scudiero et al, "Beurteilung einer löslichen Tetrazolium/Formazan-Prüfung für das Zellwachstum und die Arzneimittelempfindlichkeit in Kultur unter Benutzung von menschlichen und anderen Tumorzellstämmen", Cancer Res. 48:4827-4833, 1988). Jeweils 4000 Zellen/Vertiefung wurden auf Mikrotiterplatten mit 96 Vertiefungen gebracht und 24 Stunden später wurden die Arzneimittel zugesetzt und es wurde seriell verdünnt. Die Zellen wurden 72 Stunden bei 37ºC incubiert, worauf der Tetrazolium- Farbstoff XTT zugesetzt wurde. Ein Dehydrogenase-Enzym reduziert in lebenden Zellen das XTT zu einer Form, die Licht von 450 nm absorbiert, das spektrophotometrisch quantitativ bestimmt werden kann. Je größer die Absorption, um so größer ist die Anzahl der lebenden Zellen. Die Ergebnisse werden ausgedrückt als IC&sub5;&sub0;-Wert, der die erforderliche Arzneimittelkonzentration ist, um das Zellwachstum (d. h. die Absorption bei 450 nm) auf 50% des Wachstum der unbehandelten Kontrollzellen zu verzögern. Die Ergebnisse sind in Tabelle 2 angegeben. Die kleineren Zahlen geben die größere Aktivität an. Tabelle 2 Verbindung Name/Formel Taxol Taxotere
- Im Hinblick auf das oben gesagte ist ersichtlich, daß die verschiedenen Ziele der Erfindung erreicht werden.
Claims (6)
1. Taxan-Derivat der Formel
worin
ist;
Z = -OT&sub1; ist;
T&sub1; = Wasserstoff, Wasserstoff-Schutzgruppe oder -COT&sub2; ist;
T&sub2; = H, C&sub1;-C&sub6;-Alkyl, Alkenyl mit bis zu C&sub6;, Alkinyl mit bis
zu C&sub6; oder monocyclisches Aryl ist;
R&sub3; = Benzoyl, substituiertes Benzoyl oder C&sub1;-C&sub6;-Alkoxycarbonyl ist,
Ac = Acetyl ist, und
E&sub1; und E&sub2; unabhängig voneinander unter Wasserstoff und
funktionellen Gruppen ausgewählt sind, die die Löslichkeit des Taxan-
Derivats in Wasser erhöhen, wobei die funktionellen Gruppen die
Formel -COGCOR¹ haben, worin
G = Ethylen, Propylen, -CH=CH-, 1,2-Cyclohexylen oder
1,2-Phenylen ist;
R¹ = OH-Base, NR²R³, OR³, SR³, OCH&sub2;CONR&sup4;R&sup5; oder OH ist;
R² = Wasserstoff oder Methyl ist;
R³ = (CH&sub2;)nNR&sup6;R&sup7; oder (CH&sub2;)nN R&sup6;R&sup7;R&sup8;X ist;
n = 1 bis 3;ist;
R&sup4; = Wasserstoff oder niederes Alkyl mit 1 bis 4 Kohlenstoff-
Atomen ist;
R&sup5; Wasserstoff, niederes Alkyl mit 1 bis 4
Kohlenstoff-Atomen, Benzyl, Hydroxyethyl, CH&sub2;CO&sub2;H oder Dimethylaminoethyl ist;
R&sup6; und R&sup7; = unter niederem Alkyl mit 1 oder 2 Kohlenstoff-
Atomen unabhängig ausgewählt
dem Stickstoff-Atom des NR&sup6;R&sup7; einen der folgenden Ringe
R&sup8; niederes Alkyl mit 1 oder 2 Kohlenstoff-Atomen oder Benzyl ist;
X = Halid ist; und
Base = NH&sub3;, (HOC&sub2;H&sub4;)&sub3;N, N(CH&sub3;)&sub3;, CH&sub3;N(C&sub2;H&sub4;OH)&sub2;, NH&sub2;(CH&sub2;)&sub6;NH&sub2;,
N-Methylglucamin, NaOH oder KOH ist.
2. Taxan-Derivat nach Anspruch 1, bei dem R&sub1;
ist und das Taxan die Konfiguration 2'R, 3'S hat.
3. Taxan-Derivat nach Anspruch 1, bei der R&sub1;
ist und E&sub1; gleich -COGCOR¹ ist, worin G und R¹ wie in Anspruch 1
definiert sind.
4. Taxan-Derivat nach Anspruch 1, in dem R&sub1;
ist und das Taxan-Derivat die Konfiguration 2'R, 3'S hat.
5. Taxan-Derivat nach Anspruch 1, in dem R&sub1;
ist und E&sub1; gleich -COGCOR ist, worin G und R¹ wie in Anspruch 1
definiert sind.
6. Pharmazeutische Zusammensetzung, die ein Taxan-Derivat
nach einem der Ansprüche 1 bis 5 und ein oder mehrere
pharmakologisch zulässige, inerte oder physiologisch aktive Verdünnungsmittel
oder Hilfsstoffe enthält.
