Description
- Die Erfindung betrifft ein biologisches Verfahren zur Herstellung eines α-Hydroxyamids oder einer α-Hydroxysäure.
- Bekannte biologische Verfahren zur Herstellung eines α- Hydroxyamids umfassen ein Verfahren, das eine Umsetzung von Lactonitril, Hydroxyacetonitril, α-Hydroxymethylthiobutyronitril usw. mit einem Mikroorganismus der zum Stamm Bacillus, Bacteridium, Micrococcus oder Brevibacterium gehört, umfaßt, unter Erhalt eines entsprechenden Amids, wie in JP-B-62-21519 (entspricht dem US Patent 4 001 081) offenbart (der Begriff "JP-B", wie er hier verwendet wird, bedeutet eine "geprüfte japanische Patentanmeldung"). Es wird außerdem in Grant, D.J.W., Antonie van Leeuwenhoek; J. Microbiol. Serol, Bd. 39, S. 273 (1973) berichtet, daß die Hydrolyse von Lactonitril durch die Wirkung eines Mikroorganismus des Stammes Corynebacterium unter Erzeugung von Milchsäure die Anhäufung von Lactamid als Zwischenprodukt mit sich bringt.
- Bekannte biologische Verfahren zur Herstellung einer a- Hydroxysäure umfassen ein Verfahren, um Glykolsäure, Milchsäure, α-Hydroxyisobuttersäure usw. aus dem entsprechenden α-Hydroxynitril unter Wirkung eines Mikroorganismus des Stammes Corynebacterium zu erhalten (siehe JP-A-61-56086, wobei der Begriff "JP-A", wie er hier verwendet wird, eine "ungeprüfte, offengelegte japanische Patentanmeldung" bedeutet), ein Verfahren, um Milchsäure, Glykolsäure usw. aus dem entsprechenden α-Hydroxynitril unter Verwendung eines Mikroorganismus des Stammes Bacillus, Bacteridium, Micrococcus oder Brevibacterium (siehe JP-B-58- 15120, entspricht dem US Patent 3 940 316), ein Verfahten, um Milchsäure, α-Hydroxyisobuttersäure, Mandelsäure, a- Hydroxybuttersäure, α-Hydroxyvaleriansäure, α-Hydroxy-a- phenylpropionsäure, α-Hydroxy-a-(p-isobutylphenyl)propionsäure usw. aus dem entsprechenden α-Hydroxynitril unter Verwendung eines Mikroorganismus des Stammes Pseudomonas, Arthrobacter, Aspergillus, Penicillium, Cochliobolus oder Fusanum zu erhalten, (siehe JP-A-63- 222696) und ein Verfahren, um α-Hydroxy-β,β-dimethyl-α- butyrolacton aus dem entsprechenden α-Hydroxynitril unter Verwendung eines Mikroorganismus des Stammes Arthrobacter, Aspergillus, Bacillus, Bacteridium, Brevibacterium, Cochliobolus, Corynebacterium, Micrococcus, Nocardia, Penicillium, Pseudomonas oder Fusanum zu erhalten (siehe JP- A-64-10996).
- Es ist jedoch bekannt, daß ein α-Hydroxynitril mehr oder weniger teilweise in das entsprechende Aldehyd und in Cyanwasserstoff in einem polaren Lösungsmittel dissoziiert wird, wie es von V. Okano et al., in J. Am. Chem. Soc., Bd. 98, S. 4201 (1976) gelehrt wird. Da ein Aldehyd im allgemeinen an Proteine gebunden ist, um Enzyme zu inaktivieren, wie es von G.E. Means et al. in Chemical Modification of Proteins, S. 125, Holden-Day (1971) beschrieben wird, ist die enzymatische Hydratisierung oder Hydrolyse eines α-Hydroxynitrils mit dem Problem verbunden, daß das Enzym innerhalb kurzer Zeit inaktiviert wird. Es ist daher schwierig, ein a-Hydroxyamid oder eine α-Hydroxysäure in hoher Konzentration mit hoher Produktivität zu erhalten.
- Um das oben beschriebene Problem, das mit konventionellen Verfahren zur Herstellung eines α-Hydroxyamids oder einer a- Hydroxysäure verbunden ist, zu überwinden, führten die Erfinder umfassende Untersuchungen durch und haben als Ergebnis festgestellt, daß in Gegenwart eines Sulfitions, eines Disulfitions oder eines Dithionitions in dem Reaktionssystem die Aktivität und Stabilität der Enzyme deutlich verbessert, und haben das oben beschriebene Problem gelöst.
- Diese Erfindung stellt ein biologisches Verfahren zur Herstellung eines α-Hydroxyamids oder einer α-Hydroxysäure zur Verfügung, dargestellt durch die Formel (III):

- worin R eine substituierte oder unsubstituierte Alkylgruppe, eine substituierte oder unsubstituierte Alkenylgruppe, eine substituierte oder unsubstituierte Cycloalkylgruppe, eine substituierte oder unsubstituierte Alkoxygruppe, eine substituierte oder unsubstituierte Arylgruppe, eine substituierte oder unsubstituierte Aryloxygruppe oder eine substituierte oder unsubstituierte und gesättigte oder ungesättigte, heterocyclische Gruppe darstellt, und X eine Amidogruppe oder eine Carboxylgruppe darstellt, umfassend die Reaktion eines α-Hydroxynitrils, dargestellt durch die Formel (I):

- worin R wie oben definiert ist, oder einer Mischung aus einem Aldehyd, dargestellt durch die Formel (II):
- R - CRO (II)
- worin R wie oben definiert ist, und Cyanwasserstoff mit einem Mikroorganismus, der ein solches Amid oder eine solche Säure aus dem entsprechenden α-Hydroxynitril herstellen kann, wobei das Reaktionssystem ein Sulfition, ein Disulfition oder ein Dithionition enthält.
- Ein Sulfition, ein Disulfition oder ein Dithionition, das zu dem Reaktionssystem gegeben wird, wird z.B. als Natriumsulfit, Natriumbisulfit, Natriumdithionit, Kaliumsulfit, Kaliumbisulfit, Kaliumdithionit, Ammoniumsulfit, Ammoniumbisulfit oder Ammoniumdithionit zugeführt.
- Es wird angenommen, daß der folgende Mechanismus für die Wirkung dieser Ionen verantwortlich ist. Ein Sulfition, ein Disulfition oder ein Dithionition haben die Eigenschaften mit einem Aldehyd einen Komplex zu bilden. Dass heisst diese Ionen reagieren schnell mit einem Aldehyd, das bei der Dissozuerung eines α-Hydroxynitrils in einem polaren Lösungsmittel freigesetzt wird, unter Bildung eines Komplexes, wodurch eine geringe Konzentration an freiem Aldehyd in dem Reaktionssystem beibehalten werden kann. Der gebildete Komplex geht mit einem Proton oder einem Cyanwasserstoff eine nudeophile Reaktion ein, wobei reversibel ein Aldehyd oder ein α-Hydroxynitril freigesetzt wird.
- Gemäß der Erfindung kann die Hydrierung oder die Hydrolyse eines Nitrils unter Beibehaltung einer niedrigen Aldehydkonzentration in dem Reaktionssystem durch die Verbindung dieser Wirkungen durchgeführt werden, so daß die hemmende Wirkung des Aldehyds auf das Enzym minimiert wird, und die Umsetzung kann für einen längeren Zeitraum auf eine stabile Art und Weise, ohne eine drastische Inaktivierung des Enzyms zu verursachen, dauern, wodurch es möglich wird, das hergestellte Amid oder die hergestellte Säure in hoher Konzentration anzuhäufen. Es ist jedoch immer noch unklar, ob der Stabilisationsmechanismus des Enzyms durch ein Sulfition, ein Disulfition oder ein Dithionition nur der Verringerung der Konzentration an freiem Aldehyd in dem Reaktionssystem zuzuschreiben ist.
- Im Hinblick auf den oben beschriebenen Tatbestand, daß ein Aldehyd im allgemeinen die Eigenschaften hat, an Proteine zu binden, um ein Enzym zu inaktivieren, scheint die hemmende Wirkung auf die Enzymaktivität durch ein Sulfition, ein Disulfition oder ein Dithionition auf alle biologischen Reaktionen, die ein Aldehyd beinhalten, zuzutreffen. Mit anderen Worten ist gemäß der Erfindung der Mikroorganismus, der für die Herstellung eines Amids oder einer Säure aus einem α-Hydroxynitril verwendet werden kann, nicht besonders begrenzt, solange der Mikroorganismus ein solches Amid oder eine solche Säure aus dem entsprechenden α-Hydroxynitril herstellen kann. Das α-Hydroxynitrilsubstrat ist ebenfalls nicht besonders begrenzt, solange es ein Dissoziationsgleichgewicht mit einem Aldehyd in dem Reaktionssystem erreichen kann.
- Mikroorganismen, die durch Hydrolyse ein α-Hydroxynitril in die entsprechende Säure umwandeln können, umfassen diejenigen, die zu dem Stamm Pseudomonas, Alcaligenes, Acinetobacter, Caseobacter, Corynebacterium, Brevibacterium, Nocardia, Rhodococcus, Arthrobacter, Bacillus, Aureobacterium, Enterobacter, Escherichia, Micrococcus, Streptomyces, Flavobacterium, Aeromonas, Mycoplana, Cellulomonas, Erwinia, Candida, Bacteridium, Aspergillus, Penicillium, Cochliobolus, Fusanum oder Rhodopseudomonas gehören.
- Besondere Arten dieser Mikroorganismen sind Pseudomonas sp. BC13-2 (FERM BP-3319), Pseudomonas sp. BC15-2 (FERM BP-3320), Pseudomonas sp. SK13 (FERM BP-3325), Pseudomonas sp. - Außerdem sind die Mikroorganismen, die in JP-B-58-15120 (entspricht US Patent 3 940 316, JP-A-61-56086, JP-A-63- 222696, und JP-A-64-10996) und JP-A-2-84198 (entspricht EP 0 348 901A2) offenbart sind, auch geeignet.
- Mikroorganismen, die durch Hydratisierung ein α- Hydroxynitril in das entsprechende Amid umwandeln können, umfassen diejenigen, die zu dem Stamm Rhodococcus, Corynebacterium, Pseudomonas, Arthrobacter, Alcaligenes, Bacillus, Bacteridium, Micrococcus, Brevibacterium oder Nocardia gehören.
- Besondere Arten dieser Mikroorganismen sind Rhodococcus rhodochrous ATCC 33278, Rhodococcus erythropolis IFO 12320, Corynebacterium nitrilophilus ATCC 21419, Pseudomonas sp. SK87 (FERM P-11311), Arthrobacter sp. HR1 (FERM BP-3323) und Alcaligenes sp. BC16-2 (FERM BP-3321). Einzelheiten dieser Mikroorganismen sind in der japanischen Patentanmeldung Hei- 2-148724 aufgeführt. Außerdem sind die Mikroorganismen, die in JP-B-62-21519 (entspricht dem US Patent 4 001 081) offenbart sind, auch geeignet.
- Von den oben erwähnten Stämmen sind Pseudomonas sp. BC13-2, BC15-2, SK13, SK31 und SK87, Alcaligenes sp. BC12-2, BC20, BC35-2 und BC16-2, Acinetobacter sp. BC9-2, Caseobacter sp. BC4 und BC23, Rhodococcus sp. SK70, SK92 und HR11, Arthrobacter sp. SK103, HR1 und HR4, Enterobacter sp. SK12, und Flavobacteriums sp. SK150 neue Stämme, die von den Erfindern aus dem Boden isoliert wurden und bei dem Fermentation Reserach Institute, Agency of Industrial Science & Technology, Tsukuba, Japan unter den jeweiligen Hinterlegungsnummern (FERM Nummern) hinterlegt wurden. Morphologische und physiologische Eigenschaften dieser neuen Stämme sind unten beschrieben. All die anderen Mikroorganismen sind bekannt unter den jeweiligen oben aufgeführten Hinterlegungsnummern und erhältlich von American Type Culture Collection, Rockville, Md. - Das α-Hydroxynitril, das als ein Substrat in dieser Erfindung verwendet werden kann, wird durch die Formel (I) dargestellt, worin R eine substituierte oder unsubstituierte Alkylgruppe, eine substituierte oder unsubstituierte Alkenylgruppe, eine substituierte oder unsubstituierte Cycloalkylgruppe, eine substituierte oder unsubstituierte Alkoxygruppe, eine substituierte oder unsubstituierte Arylgruppe, eine substituierte oder unsubstituierte Aryloxygruppe oder eine substituierte oder unsubstituierte und gesättigte oder ungesättigte heterocyclische Gruppe darstellt. Das α-Hydroxynitril der Formel (I) setzt ein Aldehyd und ein Cyanwasserstoff in polaren Lösungsmitteln wie Wasser und Puffern, frei, und das so freigesetzte Aldehyd bildt mit einem Sulfition, einem Disulfition oder einem Dithionition einen Komplex.
- In der Formel (I) umfaßt die heterocyclische Gruppe solche, die mindestens eines von einem Stickstoffatom, Sauerstoffatom und Schwefelatom als Heteroatom enthalten. Substituenten der heterocyclischen Gruppe umfassen eine Alkylgruppe, eine Alkoxygruppe, eine Acylgruppe, eine Arylgruppe, eine Aryloxygruppe, ein Halogenatom, z.B. Chlor und Brom, eine Hydroxylgruppe, eine Aminogruppe, eine Nitrogruppe und eine Thiolgruppe.
- Besondere Beispiele des Hydroxynitrils der Formel (I) sind Lactonitril, α-Hydroxy-n-propionitril, α-Hydroxy-n-butyronitril, α-Hydroxyisobutyronitril, α-Hydroxy-n-hexylnitril, α-Hydroxy-n-heptylnitril, α-Hydroxy-n-octylnitril, α,γ- Dihydroxy-β,β-dimethylbutyronitril, Acroleincyanhydrin, Methacrylaldehydcyanhydrin, 3-Chlorolactonitril, 4-Methylthio-α-hydroxybutyronitril, α-Hydroxy-α-phenylpropionitril und substituierte Verbindungen davon. Beispiele des α- Hydroxynitrils der Formel (I) mit einer aromatischen oder heterocyclischen Gruppe sind, Mandelsäurenitril 2- Thiophencarboxyaldehydcyanhydrin, 2- Pyridincarboxyaldehydcyanhydrin, 2- Pyrrolcarboxyaldehydcyanhydrin, 2-Furaldehydcyanhydrin, 2- Napthylaldehydcyanhydrin und substituierte Verbindungen davon.
- Wenn ein Mikroorganismus mit einer stereospezifischen Nitrilhydratase oder -hydrolase in der biologischen Reaktion verwendet wird, kann das gesamte Produkt, ein α-Hydroxyamid oder eine α-Hydroxysäure, leicht in eine der optisch aktiven Verbindungen umgewandelt werden. In der Erfindung kann deshalb ein α-Hydroxyamid oder eine α-Hydroxysäure mit sehr guten Vorteilen gegenüber konventionellen Verfahren, die optische Auflösung und/oder Racemisierung umfassen, stereospezifisch erhalten werden.
- Die Hydratisierung oder Hydrolyse eines α-Hydroxynitrils der Formel (I) kann durch Kontaktieren des α-Hydroxynitrils oder einer Mischung eines Aldehyds der Formel (II) und Cyanwasserstoff mit mikrobischen Zellen oder behandelten mikrobischen Zellen (z.B. gebrochenen mikrobischen Zellen, einem rohen oder gereinigten Enzym, imobilisierten mikrobischen Zellen oder einem imobilisierten Enzym) eines Mikroorganismus in einem wäßrigen Medium z.B. Wasser oder einem Puffer bewirkt werden. Ein Sulfition, ein Disulfition oder eine Dithionition werden gewöhnlich in einer Konzentration von ungefähr 1 mM bis etwa 1000 mM zu dem Reaktionssystem gegeben.
- Die Substratkonzentration in dem Reaktionssystem bewegt sich gewöhnlich in den Bereichen von 0,1 bis 10 Gew.-%, bevorzugt von 0,2 bis 5,0 Gew.-%, bezogen auf die Konzentration des α- Hydroxynitrils. Die Konzentrationen des Aldehyds und des Cyanwasserstoffs in dem Reaktionssystem liegen gewöhnlich in dem Bereich von 0,1 bis 10 Gew.-%, bevorzugt von 0,2 bis 5 Gew.-%, bzw. 0,1 bis 1,0 Gew.-%, bevorzugt 0,1 bis 0,5 Gew. %. Ein Mikroorganismus wird in einer Menge von 0,01 bis 5,0 Gew.-%, auf Trockenbasis, bezogen auf das Substrat verwendet. Die Reaktion wird gewöhnlich bei einer Temperatur von dem Gefrierpunkt bis 50ºC bevorzugt von 10 bis 30ºC, für einen Zeitraum von 0,1 bis 100 Stunden durchgeführt.
- Hat das α-Hydroxynitril in dem verwendeten wäßrigen Medium eine besonders niedrige Löslichkeit, wird empfohlen, zwischen 0,1 bis 5,0 Gew.-% eines geeigneten oberflächenaktiven Stoffes, z.B. Triton X-100 (Polyethylenglycol-p-isooctylphenylether) und Tween 60 (Polyoxyethylensorbitanmonostearat) oder eines Hilfslösemittels, z.B. Methanol, Ethanol und Dimethylsulfoxid zu dem Reaktionssystem zu geben, um eine effiziente Reaktion zu erreichen.
- Folglich wird das α-Hydroxynitril durch die Hydratisierung oder die hydrolytische Aktivität des Mikroorganismus in das entsprechende Amid oder in die entsprechende Säure umgewandelt und das Produkt reichert sich in dem Reaktionssystem ein. Nach der Entfernung der mikrobischen Zellen und der anderen unlöslichen Substanzen aus dem Reaktionsgemisch kann das Produkt durch bekannte Verfahren, wie Konzentration, lonenaustausch, elektrische Dialyse, Extraktion und Kristallisation isoliert werden.
- Die Erfindung wird im folgenden unter Bezugnahme auf Beispiele ausführlich veranschaulicht, aber das soll nicht heißen, daß die Erfindung auf diese begrenzt ist. Alle Prozentangaben sind Gewichtsprozente, sofern nicht anderweitig angegeben.
- Alkaligenes sp. BC35-2 (FERM Nr. 11265) wurde in 0,8 ml eines 50 mM Phosphorsäurepuffers (pH = 7,5), der 14 mM Mandelsäurenitril und eine vorgeschriebene Menge Natriumsulfit, die in der nachfolgenden Tabelle 1 aufgeführt ist, enthält, suspendiert, um eine Zellsuspension mit einer optischen Dichte von 9 bei 630 nm (OD&sub6;&sub3;&sub0;) bereitzustellen. Die Zellsuspension wurde bei 30ºC für 4 min inkubiert und anschließend zentrifugiert, um die mikrobischen Zellen zu entfernen. Als Vergleich wurde dieselbe Reaktion durchgeführt, außer daß eine Substratlösung, die kein Natriumsulfit enthält, verwendet wurde.
- Der abgetrennte Überstand wurde mittels Flüssigchromatographie unter Verwendung einer Säule "SHODEX ODS F511A, die von Showadenko K.K., Japan hergestellt wird" untersucht, um die hergestellte Mandelsäure zu bestimmen. Außerdem wurde die optische Reinheit der Mandelsäure unter Verwendung einer Säule "CHIRALPAC WH, die von Daicel Chemical Industries, Ltd., Japan hergestellt wird" bestimmt. Die erhaltenen Ergebnisse sind in Tabelle 1 gezeigt. Tabelle 1
- Das gleiche Verfahren wie in Beispiel 1 wurde verfolgt, außer daß Natriumsulfit durch Natriumdisulfit ersetzt wurde. Die erhaltenen Ergebnisse sind nachstehend in Tabelle 2 gezeigt. Tabelle 2
- Jeder der Stämme Alcaligenes sp. BC35-2 (FERM Nr. 11265), Pseudomonas sp. BC13-2 (FERM Nr. 11266), Caseobacter sp. BC4 (FERM Nr. 11260), Acinetobacter sp. BC9-2 (FERM Nr. 11262), Nocardia asteroides IFO 3384, Bacillus subtilis ATCC 21697, Brevibacterium acetylicium IAM 1790, Aureobacterium testaceum IAM 1561 und Alcaligenes fecalis ATCC 8750 wurde in 0,8 ml eines 50 mM Phosphorsäurepuffers (pH = 7,5), der 14 mM Mandelsäurenitril oder dessen Derivate enthält, wie nachstehend in Tabelle 3 gezeigt, und 100 mM Natriumsulfit suspendiert, um eine Zellsuspension mit einer OD&sub6;&sub3;&sub0; von 5 bis 25 herzustellen. Nach der Inkubation für einen Zeitraum von 4 bis 20 min bei 30ºC wurden die mikrobischen Zellen durch Zentrifugieren entfernt. Im Überstand wurde die Menge an hergestellter Mandelsäure oder an dessen Derivat auf die gleiche Art und Weise, wie in Beispiel 1 beschrieben, bestimmt.
- Als Vergleich wurde das gleiche Verfahren befolgt, außer daß kein Natriumsulfit verwendet wurde.

- Alcaligenes sp. BC35-2 (FERM Nr. 11265) wurde in 200 ml eines 50 mM Phosphorsäurepuffers bei einer Konzentration von OD&sub6;&sub3;&sub0; = 25 suspendiert. Die Zellsuspension wurde in zwei Hälften geteilt, und 100 mM Natriumsulfit wurden zu einer Hälfte gegeben. Zu jeder Hälfte wurden 0,48 g Mandelsäurenitril bei 15ºC unter Rühren gegeben. Nachdem bestätigt war, daß das Mandelsäurenitril verbraucht und in Mandelsäure umgewandelt war, wurden 0,48 g Mandelsäure auf die gleiche Weise zugegeben. Der Reaktionsvorgang und die Auffüllung mit Mandelsäurenitril wurden wiederholt, bis die Hydrolysereaktion aufhörte. Die nachfolgende Tabelle 4 zeigt die Menge der letzendlich erzeugten Mandelsäure. - Mikrobische Zellen des Rhodococcus sp. HT40-6 (FERM Nr. 11774) wurden in 1,5 ml eines 20 mM Phosphorsäurepuffers (pH = 7,5), der 14 mM Mandelsäurenitril in Anwesenheit oder Abwesenheit von 100 mM Natriumsulfit oder Natriumdisulfit bei einer Konzentration von OD&sub6;&sub3;&sub0; = 15 enthält, suspendiert und diese Suspension wurde für 6 Stunden bei 30ºC geschüttelt. Nach Vollendung der Reaktion wurden die mikrobischen Zellen mittels Zentrifugieren entfernt und die Menge des Mandelamids in dem Überstand wurde mittels Flüssigchromatographie (Säule: SHODEX ODS F511A, Träger: 0,2 M H&sub3;PO&sub4;/Acetonitril = 4:1, Monitor: 208 nm) bestimmt. Außerdem wurde die optische Reinheit des Mandelamids unter Verwendung einer Säule für optische Auflösung (CHIRALCEL CA-1, hergestellt von Daicel Chemical Industries, Ltd., Japan, Träger: 100% Ethanol) bestimmt. Die erhaltenen Ergebnisse sind unten in Tabelle 5 gezeigt. Tabelle 5
- Brevibacterium acetylicium IAM 1790 wurde für 24 Stunden bei 30 ºC in einem Reagenzglas (Durchmesser: 22 mm), das 10 ml eines Mediums, das 10 g/l Glukose, 5 g/l Polypepton, 5 g/l Hefeextrakt und 3 g/l Malzextrakt (pH = 7) enthält, prekultiviert. Die Keimkultur wurde in einem Erlenmeyerkolben, der ein Volumen von 500 ml hat, in 100 ml eines Mediums, das die folgende Zusammensetzung hat, auf eine Endkonzentration von 1% inokuliert und für 72 Stunden bei 30ºC kultiviert. Die Kultur wurde zentrifugiert und die gesammelten mikrobischen Zellen wurden zweimal mit 5 mM Phosphorsäurepuffer (pH = 7) gewaschen, um 0,3 g mikrobische Zellen pro Kultur zu erhalten.
- Glycerin 20 g
- Hefeextrakt 3 g
- Dichlorpantoylnitril 0,3 g
- K&sub2;HOP&sub4; 3 g
- MgCl&sub2; 0,2 g
- CaCl&sub2; 40 mg
- MnSC&sub4; 4H&sub2;O 4 mg
- FeCl&sub3; 7H&sub2;O 0,7 mg
- ZnSO&sub4; 7H&sub2;O 0,1 mg
- destilliertes Wasser 1000 ml pH = 7,2
- Zu 10 ml eines 50 mM Phosphorsäurepuffers (pH = 7), der 10 ml Pantoylnitril als Substrat und eine vorbeschriebene Menge Natriumsulfit, Natriumdisulfit oder Natriumdithionit enthält, wurden 0,15 mg/ml der oben hergestellten mikrobischen Zellen gegeben, und das System konnte für 15 min bei 30ºC reagieren. Nach Abschluß der Reaktion wurde das Reaktionssystem zentrifugiert, um die mikrobischen Zellen zu entfernen, und der flüssige Überstand wurde mittels Hochdruckflüssigchromatographie unter Verwendung einer SHODEX ODS F511A-Säule analysiert. Die erhaltenen Ergebnisse sind in der nachfolgenden Tabelle 6 gezeigt. Tabelle 6
- Pseudomonas sp. BC12-2 (FERM P-11263) wurde auf die gleiche Art und Weise wie in Beispiel 6 beschrieben kultiviert, außer daß das Lösungsmittel 0,3 g/l Benzonitril als Induzierer enthält, und kein Dichlorpantoylonitril zugegeben wurde. Die Zellen wurden geerntet und wie in Beispiel 6 beschrieben gewaschen. Zu den 20 ml der Zellsuspension (OD&sub6;&sub3;&sub0; = 30) in 50 mM Phosphorsäurepuffer (pH = 7,0) wurden Cyanwasserstoff (15 mM), Benzaldehyd (15 mM) und Natriumbisulfit (100 mM) gegeben, und die Mischung konnte bei 30ºC für 6 Stunden reagieren. Nach der Reaktion wurden die mikrobischen Zellen entfernt und der Überstand wie in Beispiel 1 beschrieben untersucht.
- Als Ergebnis wurde festgestellt, daß 13 mM Mandelsäure hergestellt wurde und die optische Reinheit des Produktes 98 % betrug.
Claims (7)
1. Verfahren zur biologischen Herstellung eines a-
Hydroxyamids oder einer α-Hydroxysäure, dargestellt
durch die Formel (III):
R - - X (III)
worin R eine substituierte oder unsubstituierte
Alkylgruppe, eine substituierte oder unsubstituierte
Alkenylgruppe, eine substituierte oder unsubstituierte
Cycloalkylgruppe, eine substituierte oder
unsubstituierte Alkoxygruppe, eine substituierte oder
unsubstituierte Arylgruppe, eine substituierte oder
unsubstituierte Aryloxygruppe oder eine substituierte
oder unsubstituierte und gesättigte oder ungesättigte,
heterocyclische Gruppe darstellt, und X eine Amidogruppe
oder eine Carboxylgruppe darstellt, umfassend die
Reaktion eines α-Hydroxynitrils, dargestellt durch die
Formel (I):
R - - CN (I)
worin R wie oben definiert ist, oder einer Mischung
eines Aldehyds, dargestellt durch die Formel (II):
R - CRO (II)
worin R wie oben definiert ist, mit einem
Cyanwasserstoff mit einem Mikroorganismus, der ein
solches Amid oder eine solche Säure aus dem
entsprechenden α-Hydroxynitril herstellen kann, wobei
das Reaktionssystem ein Sulfition, ein Disulfition oder
ein Dithionition enthält.
2. Verfahren gemäß Anspruch 1, wobei das α-Hydroxyamid oder
die α-Hydroxysäure eine optisch aktive Verbindung
darstellt.
3. Verfahren gemäß Anspruch 1 oder 2, zur Herstellung einer
α-Hydroxysäure, wobei der Mikroorganismus aus
Mikroorganismen ausgewählt wird, die zu dem Stamm
Pseudomonas, Alcaligenes, Acinetobacter, Caseobacter,
Corynebacterium, Brevibacterium, Nocardia, Rhodococcus,
Arthrobacter, Bacillus, Aureobacterium, Enterobacter,
Escherichia, Micrococcus, Streptomyces, Flavobacterium,
Aeromonas, Mycoplana, Cellulomonas, Erwinia, Candida,
Bacteridium, Aspergillus, Penicillium, Cochliobolus,
Fusanum oder Rhodopseudomonas, gehören.
4. Verfahren gemäß Anspruch 3, wobei der Mikroorganismus
ausgewählt wird aus Mikroorganismen mit den
Eigenschaften von Pseudomonas sp. BC13-2 (FERM PB-3319),
Pseudomonas sp. BC15-2 (FERM BP-3320), Pseudomonas sp.
SK13 (FERM BP-3325), Pseudomonas synxanta JAM 12356,
Alcaligenes sp. BC35-2 (FERM BP-3318), Acinetobacter sp.
BC9-2 (FERM BP-3317), Caseobacter sp. BC4 (FERM BP-
3316), Corynebacterium nitrilophilus ATCC 21419,
Brevibacterium acetylicum IAM 1790, Brevibacterium
helvolum ATCC 11822, Nocardia sp. N-775 (FERM BP-961),
Nocardia asteroides IFO 3384, Nocardia calcarea KCC

5. Verfahren gemäß Anspruch 1 oder 2, zur Herstellung eines
α-Hydroxyamids, wobei der Mikroorganismus aus
Mikroorganismen ausgewählt wird, die zu dem Stamm
Rhodococcus, Corynebacterium, Pseudomonas, Arthrobacter,
Alcaligenes, Bacillus, Bacteridium, Micrococcus,
Brevibacterium oder Nocardia gehören.
6. Verfahren gemäß Anspruch 5, wobei der Mikroorganismus
ausgewählt wird aus Mikroorganismen mit den
Eigenschaften von Rhodococcus rhodochrous ATCC 33278,
Rhodococcus erythropolis IFC 12320, Corynebacterium
nitrilophilus ATCC 21419, Arthrobacter sp. HRI (FERM BP-
3323) und Alcaligenes sp. BC16-2 (FERM BP-3321).
7. Verfahren nach Anspruch 1 oder 2, wobei der
Mikroorganismus ausgewählt wird aus Mikroorganismen mit
den Eigenschaften von Pseudomonas sp. BC13-2, BC15-2 und
SK13, Alcaligenes sp. BC35-2 und BC16-2, Acinetobacter
sp. BC9-2, Caseobacter sp. BC4, Rhodococcus sp. SK92,
Arthrobacter sp. HR1 und Enterobacter sp. SK12.
