Description
- Diese Erfindung betrifft einen chirurgischen Klebstoff.
- Matsuda et al. US-Patente 4,740,534 und 4,994,542 zeigen chirurgische Klebstoffe, die NCO-terminierte hydrophile Urethanprepolymere enthalten, die von organischen Polyisocyanaten und hydrophilen Polyetherpolyolen abgeleitet sind. Das US-PAtent 4,804,691 zeigt chirurgische Klebstoffe, die NCO-terminierte Urethanprepolyrnere enthalten, die von aromatischen Polyisocyanaten und Polyesterpolyolen abgeleitet sind, die durch ringöffnende Polymerisierung von ε- Caprolacton oder Lactid erhalten sind.
- Solche Klebstoffe haben jedoch gewisse Probleme, da die erhaltenen Polymeren, die durch Härtung von Prepolymeren gebildet sind, in lebenden Körpern für lange Zeit nach Erholung der operierten Teile unzersetzt bleiben.
- Die vorliegende Erfindung liefert einen chirurgischen Klebstoff, der sich leicht innerhalb lebender Körper nach Erholung der operierten Teile zersetzen kann.
- Diese Erfindung liefert weiter einen Klebstoff für die Chirurgie, der verbesserte Härtbarkeit hat.
- Weiterhin liefert die Erfindung einen chirurgischen Klebstoff von geringerer Toxizität für Gewebe.
- Die vorliegende Erfindung liefert einen chirurgischen Klebstoff, umfassend wenigstens ein NCO-terminiertes hydrophiles Urethanprepolymer, das abgeleitet ist von (a) einem organischen Polyisocyanat und (b) einer Polyolkomponente, umfassend wenigstens ein Polyester-Polyol, das von einer Dicarbonsäure stammt, die durch die allgemeine Formel

- dargestellt ist, worin m = 0 oder 1, A = -CH&sub2;- oder eine Elektronen-anziehende Gruppe, dargestellt durch die Formel -R-CO- oder
- sind, worin R eine zweiwertige Kohlenwasserstoffgruppe ist, die 1 bis 8 Kohlenstoffatome enthält, R' eine zweiwertige Kohlenwasserstoffgruppe oder eine Halogen-substituierte Kohlenwasserstoffgruppe ist, die 1 bis 20, vorzugsweise 1 bis 8, Kohlenstoffatome enthält, X ein Halogenatom oder eine Nitro- oder Cyanogruppe ist, Y ein Wasserstoffatom, ein Halogenatom oder eine Nitrogruppe oder Cyanogruppe ist und n = 0, 1 oder 2 ist.
- Zu geeigneten zweiwertigen Kohlenwassertsoffgruppen R gehören solche, die 1 bis 8 Kohlenstoffatome (vorzugsweise 1 bis 6 Kohlenstoffatome) enthalten, z.B. Alkylengruppen, wie die Methylen-, Ethylen-, Propylen-, Butylen-, Hexylen- und Octylengruppen; Alkenylengruppen, wie die Ethenylen-, Propenylen- und Butenylengruppen; Alkinylengruppen, wie die Acetylengruppen, Cycloalkylengruppen, wie Cyclohexylengruppen, Arylengruppen, wie die Phenylen- und Tolylengruppen. Unter diesen sind Alkylengruppen bevorzugt, wie die Methylen- und Ethylengruppen.
- Zu geeigneten zweiwertigen Kohlenwasserstoffgruppen R' gehören solche, die 1 bis 20 Kohlenstoffatome, (vorzugsweise 1 bis 6 Kohlenstoffatome) enthalten, z.B. die gleichen wie oben für R erwähnt und zweiwertige C9-C20 Kohlenwasserstoffgruppen, wie die Decylen-, Dodecylen-, Tridecylen- und Octadecylengruppen sowie diese zweiwertigen Kohlenwasserstoffgruppen, die z.B. mit einem oder mehreren Halogenatomen (wie Fluor, Chlor und/oder Brom) substituiert sind, Fluor-substituierte Alkylengruppen, wie -(CH&sub2;)n'-, -(CF&sub2;)m'-, (worin n' 0, 1 oder 2 ist und m' eine ganze Zahl von wenigstens 1 ist). Unter diesen sind Alkylengruppen bevorzugt, wie die Methylen- und Ethylengruppen. Beispiele von Halogenatomen für X und Y sind Fluor-, Chlor- und Bromatome. Bevorzugte Substituenten X und Y sind Halogenatome, insbesondere Fluoratome. n ist eine ganze Zahl von 0, 1 oder 2, vorzugsweise 0 oder 1.
- Illustrativ für geeignete Dicarbonsäuren, die durch die allgemeine Formel (1) dargestellt sind, sind Oxalsäure und Malonsäure; alpha-Ketocarbonsäuren, wie Oxalomilchsäure, 2-Ketoglutarsäure und alpha-Ketoadipinsäure; Fluor-haltige Dicarbonsäuren, die durch die Formel:
- HOOC-(CH&sub2;)n'-(CF&sub2;)m'-(CH&sub2;)n'-
- dargestellt sind, (worin n' 0, 1 oder 2 ist und m' eine ganze Zahl von wenigstens 1, vorzugsweise 1 bis 8 ist) und dergleichen. Unter diesen Dicarbonsäuren sind alpha-Ketocarbonsäuren bevorzugt, insbesondere 2-Ketoglutarsäure.
- Bei der Herstellung der Polyesterpolyole, die für die Herstellung der Prepolymeren in dieser Erfindung verwendet werden, können diese Dicarbonsäuren der Formel (1) in Verbindung mit einer oder mehreren anderen Carbonsäuren verwendet werden. Zu geeigneten anderen Carbonsäuren gehören Dicarbonsäuren, z.B. aliphatische oder aromatische Dicarbonsäuren, (wie Glutar-, Adipin-, Pimelin-, Suberin-, Azelain-, Sebacin-, Fumar-, Malein-, Phthal- und Terephthalsäure) und Ester-bildende Derivate davon [Anhydride, niedere Alkylester, (wie Methyl-, Ethyl- und Propylester) und Halogenide, (wie Chloride)], z.B. Malein- und Phthalsäureanhydrid, Dimethylterepthalat und dergleichen; und Hydroxyl-haltige Carbonsäuren, z.B. aliphatische oder aromatische Hydroxycarbonsäuren, wie Glykol-, Milch-, Wein-, Äpfel- und Salicylsäure und Hydroxyketocarbonsäuren der Formel
- HO-(CH2)n-CO-COOH,
- (worin n eine ganze Zahl von wenigstens 1 ist), wie Hydroxybrenztraubensäure sowie Ester-bildende Derivate davon, z.B. Lactone, wie ε-Caprolacton und Lactide, wie Glykolid und Lactid.
- Polyesterpolyole können durch Umsetzung der sauren Komponente, die diese Carbonsäure der Formel (1) enthält und gegebenenfalls mit einer oder mehreren anderen Carbonsäuren oder Ester-bildenden Derivaten davon mit der Alkoholkomponente, die eines oder mehrere Polyole und/oder Alkylenoxide enthält, hergestellt werden. Zu geeigneten Polyolen gehören niedrigmolekulare Polyole, einschließlich von aliphatischen, cycloaliphatischen und aromatischen, z.B. zweiwertige Alkohole, wie Ethylenglykol, Propylenglykol, 1,2-, 1,3- und 1,4- Butandiol, 1,6-Hexandiol, Neopentylglykol, Diethylenglykol, Cyclohexandiol, Bis- (hydroxymethyl)-cyclohexan, Bis-(hydroxyethyl)-benzol, hydriertes Bisphenol A und hydriertes Bisphenol F; dreiwertige Alkohole, wie Glycerin, Triemethylolpropan, Trimethylolethan, 1,2,4-Butantriol und 1,2,6-Hexantriol; mehrwertige Alkohole, die 4 bis 8 oder mehr Hydroxylgruppen enthalten, wie Pentaerythrit, Diglycerin, alpha-Methylglucosid, Sorbit, Xylit, Mannit, Glucose, Fructose und Saccharose; Silanol-terminierte Polysiloxane, hochmolukulare Polyole, z.B. Polyetherpolyole, (wie diejenigen, die unten beschrieben sind) und Polyesterpolyole (diejenigen, die von diesen Polyolen und den oben erwähnten anderen Carbonsäuren stammen); sowie zwei oder mehr von diesen Polyolen. Unter diesen Polyolen sind niedrigmolekulare Polyole bevorzugt, (wie Mono-, Di- und Triethylen- und -propylenglykole und 1,4-Butandiol) und Polyetherpolyole, (insbesondere hydrophile). Illustrativ für geeignete Alkylenoxide (im folgenden als AO bezeichnet) sind Ethylenoxid (im folgenden als EO bezeichnet), Propylenoxid (im folgenden als PO bezeichnet), 1,2-, 2,3-, 1,3- und 1,4-Butylenoxide, Styroloxid und dergleichen sowie Kombinationen von zwei oder mehr davon, z.B. Kombinationen von EO mit anderen AO (wie PO). Unter diesen ist EO bevorzugt.
- Bei der Umsetzung der Säurekomponente mit der Alkoholkomponente kann das Äquivalenzverhältnis der Alkoholkomponente zur Säurekomponente in weitem Umfang variieren, wie 1 bis 10 oder mehr, vorzugsweise 1,5 bis 4.
- Die Umsetzung der Säurekomponente mit der Alkoholkomponente kann nach jeder bekannten Methode durchgeführt werden, z.B. 1) durch Dehydratisierungspolykondensation einer Carbonsäure mit einem Polyol, 2) durch Addition eines AO an eine Carbonsäure, 3) durch dehydrohalogenierende Polykondensation eines Carbonsäurehalogenids mit einem Polyol, 4) durch Esteraustauschreaktion eines niederen Carbonsäureesters mit einem Polyol, 5) durch Ringöffnungsreaktion eines Lactons oder Lactids, 6) durch Reaktion eines Carbonsäureanhydrids mit einem AO in Gegenwart eines Polyols usw.
- Polyesterpolyole, die von dieser Dicarbonsäure der Formel (1) abgeleitet sind, haben ein Äquivalenzgewicht von gewöhnlich 20 bis 1 000, vorzugsweise 50 bis 500.
- Bei der Herstellung der Prepolymeren in dieser Erfindung können Polyesterpolyole, die von dieser Dicarbonsäure der Formel (1) abgeleitet sind, in Verbindung mit einem oder mehreren anderen Polyolen verwendet werden, z.B. niedermolekularen Polyolen, wie oben erwähnt und hochmolekularen Polyolen, wie Polyetherpolyole und andere Polyesterpolyole [diejenigen, die von diesen Polyolen (niedrigmolekularen Polyolen und/oder Polyetherpolyolen) und den oben erwähnten anderen Carbonsäuren abgeleitet sind] sowie zwei oder mehr dieser Polyole.
- Geeignete Polyetherpolyole sind Addukte von einem oder mehreren AOS, (wie EO, PO, 1,2-, 2,3-, 1,3- und 1,4-Butylenoxiden, Styroloxiden, Epichlorhydrin und Kombinationen von zwei oder mehr davon) mit einer oder mehreren Verbindungen, die wenigstens zwei aktive Wasserstoffatome enthalten, wie mehrwertige Alkohole, mehrwertige Phenole, Amine, Polycarbonsäuren, Phosphorsäuren und dergleichen. Zu geeigneten Beispielen von mehrwertigen Alkoholen gehören zweiwertige Alkohole, wie Ethylenglykol, Propylenglykol, 1,3- und 1,4-Butandiole, 1,6-Hexandiol, Neopentylglykol, Diethylenglykol, Bis-(hydroxymethyl)-cyclohexan, Bis-(hydroxyethyl)-benzol, hydriertes Bisphenyol A, hydriertes Bisphenol F, Polytetramethylenglykole, Polyesterdiole und Silanol-terminierte Polysiloxxane; dreiwertige Alkohole, wie Glycerin, Trimethylolpropan, Trimethyolethan, 1,2,3-Butantriol und 1,2,6-Hexantriol und Polyestertriole und mehrwertige Alkohole, die 4 bis 8 oder mehr Hydroxylgruppen, wie Pentaerythrit, Diglycerin, alpha-Methylglucosid, Sorbit, Xylit, Mannit, Glucose, Fructose, Saccharose und dergleichen. Beispielhaft für geeignete mehrwertige Phenole sind ein- und mehrkernige Phenole, wie Hydrochinon, Brenzcatechin, Resorzin, Pyrogallol und Bisphenole (Bisphenol A, Bisphenol F, Bisphenol B, Bisphenol S und dergleichen) sowie Phenol-Formaldehyd-Kondensationsprodukte. Zu geeigneten Aminen gehören Ammoniak, Alkanolamine, wie Mono-, Di- und Triethanolamin, Isopropanolamine und dergleichen; aliphatische, aromatische, araliphatische und alicyclische Monoamine, wie C&sub1;-C&sub2;&sub0;-Alkylamine [Methyl-, Ethyl-, Isopropyl-, Butyl-, Octyl- und Laurylamin und dergleichen], Anilin, Toluidin, Naphthylamine, Benzylamin, Cyclohexylamin und dergleichen, aliphatische, aromatische, alicyclische und araliphatische Polyamine, wie C&sub2;-C&sub6;-Alkylendiamine [Ethylendiamin, Hexamethylendiamin und dergleichen], Diethylentriamin, Triethylentetramin, Tolylendiamine, Phenylendiamine, Xylylendiamine, Diethyltolylendiamine, Methylendianiline, Diphenyletherdiamine, Isophorondiamin, Cyclohexylendiamine, Dicyclohexylmethandiamine und dergleichen und heterocyclische Polyamine, wie Piperazin, N-Aminoethylpiperazin und andere heterocyclische Polyamine, wie sie in der japanischen Patentpublikation Nr. 21044/1 980 gezeigt sind. Illustrativ fur geeignete Polycarbonsäuren sind die oben erwähnten, wie Adipin und Phthalsäuren. Zu Beispielen von Phosphorsäuren gehören Ortho-, Meta-, Pyro- und Polyphosphorsäuren, phosphorige Säure und saure Ester von diesen Säuren. Unter diesen aktive Wasserstoffatome enthaltenden Verbindungen sind mehrwertige Alkohole bevorzugt, insbesondere zweiwertige und dreiwertige Alkohole (wie Ethylenglykol und Propylenglykol). Unter den AOs sind EO und Kombinationen davon mit anderen AOs (insbesondere PO bevorzugt). Die Addition von AO (EO oder Kombinationen davon mit anderen AOs) an aktiven Wasserstoff-enthaltende Verbindungen kann in üblicher Weise durchgeführt werden, mit oder ohne Katalysatoren, (wie alkalischen Katalysatoren, Aminkatalysatoren und sauren Katalysatoren) unter normalem oder erhöhtem Druck in einer einzigen Stufe oder mehrstufig. Die Addition von EO und anderen Aos kann durch statistische Addition, Blockaddition oder Kombination davon durchgeführt werden (z.B. statistische Addition gefolgt von Blockaddition). Bevorzugt ist die statistische Addition.
- Polyetherpolyole haben ein Äquivalentgewicht (Molekulargewicht pro Hydroxygruppe) von gewöhnlich 100 bis 5.000, vorzugsweise 200 bis 3.000. Polyetherpolyole mit Äquivalentgewichten von höher als 5.000 sind zu viskos, um als chirurgische Klebstoffe verwendet zu werden, während Äquivalentgewichte von weniger als 100 zu einem Mangel an Biegsamkeit führen, die für chirurgische Klebstoffe nötig ist.
- Vorzugsweise wird ein oder mehrere hydrophile Polyetherpolyole in Kombination mit diesem Polyesterpolyol verwendet, das von dieser Dicarbonsäure der Formel (1) stammt oder/und bei der Herstellung dieses Polyesterpolyols als wenigstens ein Teil der Polyolkomponente dafür. Der Oxyethylengehalt solcher hydrophiler Polyetherpolyole ist gewöhnlich wenigstens 30 %, vorzugsweise 50 bis 90 Gew.-%. Polyetherpolyole mit einem Oxyethylengehalt von weniger als 30 Gew.-% haben unzureichende hydrophile Natur, geringe Reaktivität mit Körperflüssigkeiten, was zur verminderten Härtungsgeschwindigkeit führt und schlechte Bindekraft mit wasserreichem Gewebe und liefern gehärtete Produkte von schlechter Zersetzbarkeit.
- Illustrative Beispiele von Polyesterpolyolen sind Kondensationsprodukte von zweiwertigen und/oder dreiwertigen Alkoholen (Ethylenglykol, Propylenglykol, 1,3- und 1,4-Butandiole, 1,6-Hexandiol, Neopentylglykol, Diethylenglykol, Glycerin, Trimethylolpropan und dergleichen) und/oder Polyetherpolyolen, (wie die oben beschriebenen) mit Dicarbonsäuren (aliphatischen oder aromatischen Dicarbonsäuren, wie Glutar-, Adipin-, Sebacin-, Fumar-, Malein-, Phthal- und Terephthalsäure) oder Ester-bildenden Derivaten davon (Anhydriden und niederen Alkylestern, wie Malein- und Phthalsäureanhydrid, Dimethylterepthalat und dergleichen), Ringöffnungs-Polymerisationsprodukte von Lactonen, (wie ε-Lacton).
- Unter diesen Polyolen sind Polyetherpolyole gegenüber Polyesterpolyolen bevorzugt.
- Im allgemeinen umfaßt die Polyolkomponente (b), die zur Herstellung der NCO- terminierten Urethanprepolymeren benutzt wird, wenigstens 1 0 %, vorzugsweise wenigstens 20 % dieser Polyesterpolyole, die von dieser Dicarbonsäure der Formel (1) stammen, 0 bis 90 % an anderen hochmolekularen Polyolen (Polyetherpolyolen und/oder anderen Polyesterpolyolen) und 0 bis 20 %, vorzugsweise 0 bis 10 % an niedermolekularen Polyolen, bezogen auf das Gesamtgewicht der Polyole. Im Falle von Polyesterpolyolen, die von dieser Dicarbonsäure der Formel (1) stammen, die frei von Oxyethylengruppen sind, ist die Menge dieser Polyesterpolyole gewöhnlich 10 bis 70 %, vorzugsweise 20 bis 50 %, und hydrophile Polyetherpolyole werden in einer Menge von im allgemeinen 30 bis 90 %, vorzugsweise 50 bis 80 % verwendet. Die Polyolkomponente (b) kann einen Oxyethylengehalt (Durchschnitt) von gewöhnlich wenigstens 30 %, vorzugsweise 50 bis 90 Gew.-% und ein Äquivalentgewicht (Durchschnitt) von gewöhnlich 100 bis 5.000, vorzugsweise 200 bis 3.000 und gewöhnlich 2 bis 8 Hydroxylgruppen, vorzugsweise 2 bis 4 Hydroxylgruppen aufweisen.
- Unter diesen Polyolen sind Polyetherpolyole gegenüber Polyesterpolyolen bevorzugt.
- Zu organischen Polyisocyanaten, die zur Herstellung der NCO-terminierten hydrophilen Urethanprepolymeren benutzt werden, die in dem chirurgischen Klebstoff gemäß der vorliegenden Erfindung verwendet werden, gehören fluorhaltige oder fluorfreie und Kombinationen von diesen.
- Zu geeigneten F(fluor)-haltigen Polyisocyanaten gehören F-haltige aliphatische Polyisocyanate und F-haltige cycloaliphatische Polyisocyanate und Gemische von zwei oder mehr von diesen.
- Zu geeigneten F-haltigen aliphatischen Polyisocyanaten gehören z.B. F-haltige Diisocyanate, die durch die Formeln (2) und (3) dargestellt sind:
- OCN-Rf-NCO (2)
- und
- OCN-CH&sub2;-Rf-CH&sub2;-NCO (3).
- In den Formeln (2) und (3) ist Rf eine Perfluoralkylengruppe, die gewöhnlich 1 bis 20, vorzugsweise 1 bis 10 Kohlenstoffatome und 0 bis 10, vorzugsweise 0 bis 4 Etherbindungen enthält. Speziell können erwähnt werden
- -(CF&sub2;)k-, -CF&sub2;CF&sub2;OCF&sub2;CF&sub2;-, -CF&sub2;CF&sub2;O(CF&sub2;)k', OCF&sub2;CF&sub2;-,
- CF(CF&sub3;)O(CF&sub2;)k'OCF(CF&sub3;)-, und
- -CF&sub2;CF&sub2;OCF&sub2;CF(CF&sub3;)O(CF&sub2;)k'OCF(CF&sub3;)CF&sub2;OCF&sub2;CF&sub2;-, worin k 1 bis 20, vorzugsweise 1 bis 10, k'1 bis 5, vorzugsweise 1 bis 2 sind. Unter diesen sind -(CF&sub2;)k- bevorzugt. F-haltige Polyisocyanate können gemäß den Methoden erzeugt werden, die in J. Macromol. Sci. Phys., B1, 831 ('67) und dem offengelegten japanischen Patent Nr. 108055/1982 beschrieben sind.
- Illustrative Beispiele von F-haltigen cycloaliphatischen Polyisocyanaten sind fluorierte cycloaliphatische Polyisocyanate, die 4 bis 1 5 Kohlenstoffatome enthalten, (ausgenommen diejenigen in den NCO-Gruppen), wie F-haltige Isophorondiisocyanate, fluorierte hydrierte Xylylendiisocyanate, fluorierte hydrierte 4,4'- Diphenylmethandiisocyanate und fluorierte trans-Cyclohexan-1,4-diisocyanate.
- Unter diesen Polyisocyanaten sind F-haltige aliphatische Polyisocyanate bevorzugt, insbesondere F-haltige Diisocyanate der Formel (3). Am meisten bevorzugt ist 2,2,3,3,4,4,5,5-Octafluorhexamethylendiisocyanat (im folgenden als FHDI bezeichnet).
- Zu geeigneten Beispielen von fluorfreien Polyisocyanaten gehören aromatische Polyisocyanate, die 6 bis 20 Kohlenstoffatome enthalten [ausgenommen Kohlenstoffatome in NCO-Gruppen], wie o-, m- und p-Phenylendiisocyanate [im folgenden als PDI bezeichnet], 2,4- und 2,6-Tolylendiisocyanate [TDI], Diphenylmethan-2,4'- und -4,4'-Diisocyanate [MDI], Naphthalin-1,5-diisocyanat, Triphenylmethan-4,4'4"-Triisocyanat, Polymethylen-polyphenylenpolyisocyanate [PAPI], die durch Phosgenierung von Anilin-Formaldehyd-Kondensationsprodukten, m- und p-Isocyanato-Phenylsulfonylisocyanat und dergleichen erhalten sind; aliphatische Polyisocyanate, die 2 bis 18 Kohlenstoffatome enthalten, wie Ethylendiisocyanat, Tetramethylendiisocyanat, Hexamethylendiisocyanat (im folgenden als HDI bezeichnet), Dodecamethylendiisocyanat, 1,6,11-Undecandiisocyanat, 2,2,4-Trimethylhexandiisocyanat, Lysinesterdiisocyanat, Lysinestertriisocyanat, 2,6-Diisocyanato-Methylcaproat, Bis-(2-isocyanatoethyl)-fumarat, Bis-(2-isocyanatoethyl)-carbonat, 2-Isocyanatoethyl-2,6-diisocyanatohexanoat und dergleichen; alicyclische Polyisocyanate, die 4 bis 15 Kohlenstoffatome enthalten, wie Isophorondiisocyanat, Dicyclohexylmethandiisocyanate, Cyclohexylendiisocyanate, Methylcyclohexylendiisocyanate, Bis-(2-isocyanatoethyl)-4-cyclohexen-1,2- dicarboxylat und dergleichen; araliphatische Polyestercyanate, die 8 bis 15 Kohlenstoffatome enthalten, wie Xylylendiisocyanate, Diethylbenzoldiisocyanate, und dergleichen; und modifizierte Polyisocyanate dieser Polyisocyanate, die Urethan-, Carbodiimid-, Allophanat-, Harnstoff-, Biuret-, Urethdion-, Urethimin-, Isocyanurat- und/oder Oxazolidon-Gruppen enthalten, wie Urethan-modifiziertes TDI, Carbodiimid-modifiziertes MDI Urethan-modifiziertes MDI und dergleichen; sowie Gemische von zwei oder mehr davon. Unter diesen fluorfreien Polyisocyanaten sind aromatische Polyisocyanate bevorzugt (vorzugsweise Diisocyanate), insbesondere PDI, TDI (einschließlich 2,4- und 2,6-Isomere, Gemische davon und rohes TDI), MDI (einschlieolich 4,4'- und 2,4'-Isomere, Gemische davon und rohes MDI oder PAPI), und modifizierte Polyisocyanate, die Urethan-, Carbodiimid-, Allophanat-, Harnstoff-, Biuret- und/oder Isocyanurat-Gruppen enthalten, die vom PDI, TDI und/oder MDI abgeleitet sind. Das am meisten bevorzugte ist p-PDI, hinsichtlich der geringen Toxizität.
- Unter diesen organischen Polyisocyanaten sind F-haltige Polyisocyanate und Kombinationen davon mit 50 % oder weniger, insbesondere 20 % oder weniger an F-freiem Polyisocyanaten bevorzugt, bezogen auf das Gesamtgewicht der Polyisocyanate.
- Bei der Umsetzung von wenigstens einem Polyisocyanat (a) mit der Polyolkomponente (b), welche diesen Polyesterpolyol enthält, der von einer Dicarbonsäure der allgemeinen Formel (1) abgeleitet ist, um NCO-terminierte hydrophile Urethanprepolymere zu bilden ist das Verhältnis von NCO/OH im allgemeinen 1,5 oder mehr, vorzugsweise 1,6 bis 5, noch bevorzugter 1,7 bis 3.
- Die Umsetzung von (a) mit (b) zur Bildung der Prepolymeren kann in üblicher Weise durchgeführt werden, unter den üblichen Bedingungen (z.B. bei einer Temperatur zwischen 50 ºC und 100 ºC). Die Reaktion kann in Gegenwart eines Katalysators durchgeführt werden. Prepolymere können in jeder Reihenfolge gebildet werden, z.B. 1) durch Umsetzung von (a) mit (b); 2) durch Umsetzung von (a) mit einem Teil von (b), z.B. entweder dem Polyesterpolyol oder einem Polyetherpolyol und/oder niedrigmolekularem Polyol), gefolgt von Umsetzung des erhaltenen intermediären Prepolymeren mit dem Rest von (b) [z.B. entweder einem Polyetherpolyol und/oder einem niedermolekularen Polyol oder diesem Polyesterpolyol] oder 3) durch Mischen eines Prepolymeren dieses Polyesterpolyols mit einem Prepolymeren eines Polyetherpolyols und/oder niedrigmolekularem Polyol. Unter diesen ist 1) bevorzugt. In dem Fall, wo zwei oder mehr Polyisocyanate (z.B. F-haltige und F-freie) miteinander verwendet werden, können diese Polyestercyanate in jeder Reihenfolge entsprechend umgesetzt werden. Es ist bevorzugt, F-freie Polyisocyanate bei frühen Stufen der Prepolymerherstellung umzusetzen, um F-haltige Polyisocyanat-terminierte Prepolymere zu erhalten.
- Die NCO-Gehalte der NCO-terminierten hydrophilen Prepolymeren sind gewöhnlich 1 bis 10 %, vorzugsweise 2 bis 8 Gew.-%. Prepolymere mit einem NCO- Gehalt von weniger als 1 Gew.- % sind von schlechter Reaktivität und ergeben eine Verminderung der Härtungsgeschwindigkeit und unzureichende Bindekraft an Gewebe. Höhere NCO-Gehalte als 10 Gew.-% führen zu spröden gehärteten Harzen von schlechter Biegsamkeit, die nicht der Bewegung des lebenden Organismus folgend deformierbar sind.
- Klebstoffe der vorliegenden Erfindung können erforderlichenfalls physiologisch aktive Materialien enthalten [wie Mittel, welche das zentrale Nervensystem beeinflussen, antiallergische Mittel, kardiovaskulare Mittel, Mittel, welche die Atmungsorgane beeinflussen, Mittel, welche die Verdauungsorgane beeinflussen, Hormonpräparate, Mittel, welche den Metabolismus beeinflussen, Antitumormittel, antibiotische Präparationen, Chemoterapeutika, Antimikrobenmittel, Lokalanästhetika, Antihistamine, Antiphlogistika, Adstringentien, Vitamine, Antifungusmittel, Anästhetika für das periphere Nervensystem, Vasodilatoren, rohe Arzneimittelessenzen, Tinkturen, rohe Arzneimittelpulver, Hypotensivmittel, und dergleichen], Füllstoffe [z.B. Ruß, Metalloxide, wie rotes Eisenoxid und Titandioxid, Silikate, wie Kalziumsilikat und Natriumsilikat, Acrylharzpulver, verschiedene keramische Pulver und dergleichen], Weichmacher, [wie DBP (Dibutylphosphat), DOP (Dioctylphosphat), TCP (Tricresylphosphat), Tributoxyethylphosphate und andere Ester von verschiedenen Arten], Stabilisatoren, wie Trimethyldihydrochinon, Phenyl-beta-naphthylamin, p-Isopropoxydiphenylamin, Diphenyl-p-phenylendiamin und dergleichen. Diese Zusätze können in einer Menge von gewöhnlich 0 bis 20 %, vorzugsweise 0 bis 5 %, bezogen auf das Gewicht des Klebstoffs gemäß der Erfindung verwendet werden.
- Die NCO-terminierten Prepolymeren können rasch in Gegenwart von Spurenmengen von Wasser polymerisieren, wie Feuchtigkeit in der Luft und führen zur Bildung von festen Membranen. Demgemäß ist es notwendig, sie wasserfrei als diese Hauptkomponenten zu verwenden und auch die anderen Mischungszusätze, und es ist bevorzugt, Luft während der Erzeugung der Klebstoffe auszuschließen. So erhaltene Klebstoff können für eine lange Zeitspanne in Iuftdichten Gefäßen, wie einer Ampulle gelagert werden.
- Bei der Anwendung der Klebstoffe der vorliegenden Erfindung in der Chirurgie gehören zu geeigneten Anwendungsmethoden diejenigen unter Verwendung von Bürsten, Pinzetten, Applikatoren, speziell entwickelten Spachteln oder Spritzen oder dergleichen und diejenigen, welche die Sprühbeschichtung unter Verwendung von inerten Gasen umfassen, wie Freone, Stickstoff oder dergleichen. Das Binden der Gewebe kann z.B. erreicht werden durch direkte Beschichtungsarbeitsweisen, indem man einfach den Klebstoff auf die Gewebe aufträgt; durch Abdeckungsbeschichtungsarbeitsweisen, wo man als Hilfe für die Blutstillung oder die Anastomose dünne Folien oder Maschenware aus Polyestern (wie Dacron), oxidierter Zellulose, Kollagen, Polyurethane oder dergleichen verwendet, baumwollähnliche Materialien oder Fragmente von Geweben, wie Venen, Muskulatur oder Muskelmembran oder dergleichen [worin diese Materialien auf die befallene Stelle aufgebracht werden, gefolgt von Aufschichten von Klebstoff darauf] oder durch Abdichtungsarbeitsweisen für genähte Teile, wobei die Nähte teilweise angebracht werden, gefolgt von Aufbringen des Klebstoffs zum Abdichten der verbleibenden Verbindungsteile. Die Klebstoffe der vorliegenden Erfindung können nicht nur für Gewebsadhäsion benutzt werden, sondern auch als Beschichtung, Embolus oder Dichtungsmaterialien bei der kardiovaskulären Chirurgie über direkte Beschichtung oder Injektion durch Katheder. Klebstoffe dieser Erfindung können in Form von Klebstoffolien oder unter Verwendung von chirurgischen Instrumenten aufgebracht werden, wie in der EP-A-0 390 481 beschrieben.
- Zu annehmbaren Geweben gehören z.B. Vaskulargefäße, Herz, Lunge, Luftröhre, Speiseröhre, Magen, Nieren, Zwölffingerdarm, Dünndarm, Dickdarm, Rektum, Leber, Pankreas, Milz, Nerven, Haut und dergleichen.
- Chirurgische Klebstoffe gemäß dieser Erfindung, die NCO-terminiertes hydrophi- les Urethanprepolymer enthalten, das von (a) einem organischen Polyisocyanat und (b) einer Polyolkomponente stammt, die einen Polyesterpolyol aufweist, der von einer Dicarbonsäure der Formel (1) stammt, sind dazu befähigt, innerhalb lebender Körper innerhalb weniger Wochen im Falle der leicht zersetzbaren zersetzt zu werden und innerhalb von mehreren Monaten im Falle der langsam zersetzbaren. So können die Klebstoffe der Erfindung bei lebenden Körper sicher angewandt werden, da sie sich innerhalb einer gewissen Zeitspanne nach der Erholung der operierten Teile zersetzen ohne daß die erhaltenen Polymeren für eine lange Zeitspanne verbleiben.
- Zusätzlich zur Zersetzbarkeit haben die chirurgischen Klebstoffe der vorliegenden Erfindung auch hinreichend hohe Härtungsgeschwindigkeit, ausreichende Bindekraft für Gewebe und Flexibilität, die sie befähigt, gemäß der Bewegung der Gewebe deformiert zu werden.
- Insbesondere Klebstoffe der vorliegenden Erfindung, die ein Prepolymeres aufweisen, das erhalten ist durch Umsetzung von (a) mit (b) bei einem NCO/OH- Verhältnis von mehr als 1,5 können verbesserte Härtbarkeit und verbesserte Bindekraft für das Gewebe entwickeln.
- Daneben sind die Klebstoffe dieser Erfindung, die ein Prepolymeres aufweisen, das unter Verwendung eines F-haltigen Polyisocyanats als (a) hergestellt ist von geringer Toxizität fegen Gewebe und bewirken keine oder wenig toxikolische Probleme.
- Demgemäß haben die Klebstoffe der vorliegendne Erfindung die Wirkungen, die Sicherheit und den Schutz bei medizinischen Behandlungen beträchtlich zu verbessern, einschließlich von chirurgischen Operationen. Man kann bemerkenswerte Verbesserungen in der medizinischen Technologie erzielen, wie die Verkürzung der Operationszeit, Blutstillung, Verhinderung des Ausleckens von Enzymen aus inneren Organen oder dergleichen, die Verhinderung der Occlusion von sehr kleinen Blutgefäßen und Nervenanastomose sowie die vorläufige Fixierung vor dem Nähen, und sie gewährleisten die Bindung durch Kombination von Adhäsion mit Nähen. Überdies kann die Erfindung hohe Zuverlässigkeit und hohe Wirksamkeit nicht nur bei der Operation sondern bei der medizinischen Behandlung allgemein liefern, z.B. das Verbinden von Schnittwunden oder Cultingteilen, die Klebstoffbehandlung in der Zahnchirurgie, bei Heilmitteln durch gesteuerte Freisetzung von Arzneimitteln in Kombination mit physiologisch aktiven Materialien.
- Nach der allgemeinen Beschreibung der Erfindung kann ein vollständigeres Verständnis unter Bezugnahme auf gewisse spezifische Beispiele erhalten werden, die lediglich zur Erläuterung einbezogen sind und nicht beschränken sollen, wenn nichts anderes angegeben ist.
- In den folgenden Beispielen sind Teile und Prozent immer Gewichtsteile bzw. Gewichtsprozent. Im folgenden bedeutet PEO Polyethylenoxid, PPO bedeutet Polypropylenoxid, PEG bedeutet Polyethylenglykol und PPG bedeutet Polypropylenglykol.
- Ein Gemisch von 2-Ketoglutarsäure (1 Mol), Diethylenglykol (3 Mol) und wasserfreiem Aluminiumchlorid (0,001 Mol) wurde gerührt, und die Entwässerungskondensationsreaktion wurde unter Erhitzen bei 90 ºC für 3 Stunden lang und dann bei 130 ºC für 2 Stunden lang und schließlich bei 150 ºC für 2 Stunden lang durchgeführt. Das Reaktionsprodukt wurde durch Auflösen in Chloroform, gefolgt von Flüssig-Trennungsbehandlung (zweimal mit Wasser, dann mit einer wäßrigen Lösung von Natriumhydrogencarbonat und schließlich mit einer gesättigten wäßrigen Lösung von Natriumchlorid) und dann Entfernen von Chloroform zur Erzielung eines Polyesterpolyols (Polyol A-1) gereinigt.
- Ein Reaktionsprodukt von 1,4-Butandiol (3 Mol) mit metallischem Natrium wurde in Tetrahydrofuranlösungsmittel unter Rühren eingemischt und Malonsäuredichlorid (1 Mol) wurde tropfenweise zugegeben. Aus dem Reaktionsprodukt wurde anorganisches Salz abfiltriert und das Lösungsmittel entfernt, um ein Polyesterpolyol (Polyol A-2) zu erhalten.
- Ein Gemisch von HOOC-(CF&sub2;)&sub4;-COOH (1 Mol), Tripropylenglykol (3 Mol) und wasserfreiem Aluminiumchiorid (0,001 Mol) wurde gerührt, und die Entwässerungskondensationsreaktion wurde in der gleichen Weise wie in Herstellungsbeispiel 1 durchgeführt, um ein Polyesterpolyol (Polyol A-3) zu erhalten.
- Zu einem Gemisch von Oxalsäure (1 Mol) mit Lithiumhexafluorphosphat (0,01 Mol) wurde nach und nach Ethylenoxid (1 Mol) gegeben und bei 120 ºC umgesetzt. Nach beendigter Reaktion wurde das erhaltene Produkt neutralisiert und gereinigt, um ein Polyesterpolyol zu erhalten (Polyol A-4).
- Ein Gemisch von 90 Teilen eines Polyetherpolyols (PEO-PPO statistisches Copolymeres mit einem Durchschnittsmolekulargewicht von 3.000 und einem Oxyethylengehalt von 80 %) mit 10 Teilen Polyol A-1 wurde unter vermindertem Druck entwässert, und dann wurde FHDI in solcher Menge zugegeben, daß man ein NCO/OH-Verhältnis von 2/1 hatte, und es wurde bei einer Temperatur von 80 ºC für 8 Stunden lang umgesetzt, um ein NCO-terminiertes Urethanprepolymer zu erhalten, das der chirurgische Klebstoff I der vorliegenden Erfindung ist.
- Ein Gemisch von 80 Teilen eines Polyetherpolyols (PEO-PPO statistisches Copolymeres mit einem Durchschnittsmolekulargewicht von 4.000 und einem Oxyethylengehalt von 60 %) mit 20 Teilen Polyol A-2 wurde unter vermindertem Druck entwässert, und dann wurde p-PDI in solcher Menge zugegeben, daß man ein NCO/OH-Verhältnis von 2/1 hatte, und es wurde bei einer Temperatur von 80 ºC für 8 Stunden lang umgesetzt, um ein NCO-terminiertes Urethanprepolymer zu erhalten, das der chirurgische Klebstoff II der vorliegenden Erfindung ist.
- Ein Gemisch von 50 Teilen eines PEG mit einem Durchschnittsmolekulargewicht von 1.000 mit 50 Teilen Polyol A-3 wurde unter vermindertem Druck entwässert, und dann wurde TDI in solcher Menge zugegeben, daß man ein NCO/OH- Verhältnis von 2/1 hatte, und es wurde bei einer Temperatur von 80 ºC für 8 Stunden lang umgesetzt, um ein NCO-terminiertes Urethanprepolymer zu erhalten, das der chirurgische Klebstoff III der vorliegenden Erfindung ist.
- Ein Gemisch von 50 Teilen eines PPG mit einem Durchschnittsmolekulargewicht von 3.000 mit 50 Teilen Polyol A-1 wurde unter vermindertem Druck entwässert, und dann wurde FHDI in solcher Menge zugegeben, daß man ein NCO/OH- Verhältnis von 2/1 hatte, und es wurde bei einer Temperatur von 80 ºC für 8 Stunden lang umgesetzt, um ein NCO-terminiertes Urethanprepolymer zu erhalten, das der chirurgische Klebstoff IV der vorliegenden Erfindung ist.
- Polyol A-4 wurde unter vermindertem Druck entwässert, und dann wurde TDI in solcher Menge zugegeben, daß man ein NCO/OH-Verhältnis von 2/1 hatte, und es wurde bei einer Temperatur von 80 ºC für 8 Stunden lang umgesetzt, um ein NCO-terminiertes Urethanprepolymer zu erhalten, das der chirurgische Klebstoff V der vorliegenden Erfindung ist.
- Ein Gemisch von 90 Teilen eines Polyetherpolyols (PEO-PPO statistisches Copolymeres mit einem Durchschnittsmolekulargewicht von 3.000 und einem Oxyethylengehalt von 80 %) mit 10 Teilen eines PPG mit einem Durchschnittsmolekulargewicht von 200 wurde unter vermindertem Druck entwässert, und dann wurde FHDI in solcher Menge zugegeben, daß man ein NCO/OH-Verhältnis von 2/1 hatte, und es wurde bei einer Temperatur von 80 ºC für 8 Stunden lang umgesetzt, um ein NCO-terminiertes Urethanprepolymer zu erhalten, das der chirurgische Vergleichsklebstoff VI ist.
- Ein Gemisch von 90 Teilen eines Polyetherpolyols (PEO-PPO statistisches Copolymeres mit einem Durchschnittsmolekulargewicht von 3.000 und einem Oxyethylengehalt von 80 %) mit 10 Teilen eines Polycaprolactonpolyols mit einem Durchschnittsmolekulargewicht von 520 wurde unter vermindertem Druck entwässert, und dann wurde TDI in solcher Menge zugegeben, daß man ein NCO/OH-Verhältnis von 2/1 hatte, und es wurde bei einer Temperatur von 80 ºC für 8 Stunden lang umgesetzt, um ein NCO-terminiertes Urethanprepolymer zu erhalten, das der chirurgische Vergleichsklebstoff VII Erfindung ist.
- Eine Polymilchsäure mit einem Durchschnittsmolekulargewicht von 2.900 wurde unter vermindertem Druck entwässert, und dann wurde TDI in solcher Menge zugegeben, daß man ein NCO/OH-Verhältnis von 2/1 hatte, und es wurde bei einer Temperatur von 80 ºC für 8 Stunden lang umgesetzt, um ein NCO-terminiertes Urethanprepolymer zu erhalten, das der chirurgische Vergleichsklebstoff Vlll ist.
- Jeder der chirurgischen Klebstoffe I bis VIII wurde auf einen Film aus fluorhaltigem Harz aufgeschichtet, um eine chirurgische Klebefolie zu bilden.
- Die Leber eines Hundes wurde mit der Pinzette gemäß einer imaginären Schneidlinie von etwa 2 cm Länge eingeklemmt und mit der Innenseite der Pinzette längs geschnitten. Auf alles über diesen Abschnitt wurde jede chirurgische Klebfolie gepreßt und nach 1 0 Minuten wurde die Folie aus Fluorharz davon entfernt, und der Klebstoff wurde auf den befallenen Teil überführt.
- Die chirurgischen Klebstoffe I bis VII erzielten die vollständige Abdichtung und Blutstillung ohne irgendwelche Schwierigkeiten bei der Behandlung und in der Blutstillung während der chirurgische Klebstoff VIII von Vergleichsbeispiel 3 hochgradig kristallin und wachsig war und Schwierigkeiten bei der Behandlung und bei der Blutstillung ergab.
- Methanol wurde zu jedem der chirurgischen Klebstoffe I bis VIII zugegeben und damit bei 50 ºC für 48 Stunden umgesetzt, um die endständigen NCO-Gruppen des Prepolymeren mit Methanol zu blockieren. Zu jedem mit Methanol geblockten Prepolymeren wurde ein Phosphatpuffer (pH = 6,8) zugefügt und die Zersetzbarkeit wurde bei 37 ºC durch Messung der Molekulargewichtsverteilung durch Gelpermeationschromatographie (GPC) nach 3 Tagen und nach 10 Tagen bestimmt. Die Ergebnisse waren wie folgt
- Chirurigische Klebstoffe I, II und IV:
- Die Zersetzung wurde mit GPC nach 10 Tagen beobachtet. (Es wurde durch NMR bestätigt, daß die Zersetzung im Polyesterpolyolteil auftrat).
- Chirurgische Klebstoffe III und V:
- Der Polyesterpolyolteil wurde vollständig nach 3 Tagen zersetzt.
- Chirurgische Klebstoffe VI, VII und VIII:
- Keine Zersetzung wurde nach 10 Tagen beobachtet.
- Jede der chirurgischen Klebstoffe I bis VIII wurde in einer kleinen Menge auf eine Oberfläche der Leber einer Maus aufgeschichtet, um den befallenen Teil abzudichten. Nach 3 Monaten wurde jede Maus seziert, und die Zersetzbarkeit wurde geprüft.
- Es wurde beobachtet, daß die chirurgischen Klebstoffe I, II und IV beträchtlich zersetzt waren, und die Adhäsionskraft mit dem Gewebe verloren hatten, obwohl die ursprüngliche Form beibehalten wurde, und die Zersetzung teilweise fortgeschritten war. Bezüglich der chirurgischen Klebstoffe III und V wurde eine stärkere Zersetzung beobachtet im Vergleich zu den obigen drei Klebstoffen. Andererseits war praktisch keine Zersetzung bei den chirurgischen Klebstoffen VI und VII zu bemerken. Der chirurgische Klebstoff VIII war wachsig und konnte dem Test nicht unterzogen werden.
Claims (10)
1. Chirurgischer Klebstoff, umfassend wenigstens ein NCO-terminiertes
hydrophiles Urethanprepolymer, das abgeleitet ist von (a) einem
organischen Polyisocyanat, enthaltend ein fluorhaltiges Polyisocyanat und/oder
ein aromatisches Polyisocyanat, und (b) einer Polyolkomponente,
umfassend wenigstens ein Polyester-Polyol das von einer Dicarbonsäure
stammt, die durch die allgemeine Formel
HOOC-(A)M-COOH (1)
dargestellt ist, worin m = 0 oder 1, A = -CH&sub2;- oder eine Elektronen
anziehende Gruppe, dargestellt durch die Formel -R-CO- oder
sind, worin R eine zweiwertige Kohlenwasserstoffgruppe ist, die 1 bis 8
Kohlenstoffatome enthält, R' eine zweiwertige Kohlenwasserstoffgruppe
oder eine Halogen-substituierte Kohlenwasserstoffgruppe ist, die 1 bis 20
Kohlenstoffatome enthält, X ein Halogenatom oder eine Nitro- oder
Cyanogruppe ist, Y ein Wasserstoffatom, ein Halogenatom oder eine
Nitro- oder Cyanogruppe ist und n = 0, 1 oder 2 ist.
2. Klebstoff nach Anspruch 1, dadurch gekerinzeichnet, daß die
Dicarbonsäure wenigstens eine Dicarbonsäure ist ausgewählt aus der Gruppe
bestehend aus Oxalsäure, Alpha-keto-Carbonsäuren und fluorhaltigen
Dicarbonsäuren der Formel: HOOC-(CH&sub2;)n, (CF&sub2;)m,-(CH&sub2;)n,-COOH, worin
n' = 0, 1 oder 2 und m' eine ganze Zahl von wenigstens 1 sind.
3. Klebstoff nach Anspruch 1, dadurch gekennzeichnet, das die
Dicarbonsäure 2-Ketoglutarsäure umfaßt.
4. Klebstoff nach irgendeinem der Ansprüche 1 bis 3, dadurch
gekennzeichnet, daß die Polyolkomponente wenigstens 10 Gew.-% des
Polyesterpolyols und 0 bis 90 Gew.-% eines hydrophilen Polyethers umfaßt.
5. Klebstoff nach irgendeinem der Ansprüche 1 bis 4 dadurch
gekennzeichnet, daß das Polyesterpolyol und/oder das hydrophile Polyetherpolyol
Oxyethylengruppen enthält.
6. Klebstoff nach irgendeinem der Ansprüche 1 bis 5, dadurch
gekennzeichnet, daß die Polyolkomponente einen Oxyethylengehalt von 30 bis 90
Gew.-% hat.
7 Klebstoff nach irgendeinem der Ansprüche 1 bis 6, dadurch
gekennzeichnet, daß die Polyolkomponente ein Äquivalentgewicht von 200 bis 3000
hat.
8. Klebstoff nach irgendeinem der Ansprüche 1 bis 7, dadurch
gekennzeichnet, daß das Polyisocyanat wenigstens ein Polyisocyanat enthält
ausgewählt aus der Gruppe bestehend aus fluorhaltigen Diisocyanaten der
Formel (2) oder (3)
OCN-Rf-NCO (2)
OCN-CH&sub2;-Rf-CH&sub2;-NCO (3)
worin Rf eine Perfluoralkylengruppe ist, die 1 bis 20 Kohlenstoffatome
und 0 bis 10 Etherbindungen enthält.
9. Klebstoff nach irgendeinem der Ansprüche 1 bis 8, dadurch
gekennzeichnet, daß das Prepolymere erhalten ist durch Umsetzen dieses
Polyisocyanats mit der Polyolkomponente in solcher Menge, daß sich ein NCO/OH
Verhältnis von 1,7 bis 3 ergibt.
10. Klebstoff nach irgendeinem der Ansprüche 1 bis 9, dadurch
gekennzeichnet, daß er bis zu 20 Gew.-% von wenigsten einem Zusatz enthält
ausgewählt aus der Gruppe bestehend aus Ruß, Metalloxiden, Silikaten,
Acrylharzpulvern, Keramikpulvern, Erweichungsmitteln, Stabilisatoren und
physiologisch aktiven Materialien.
