Description
- Die vorliegende Erfindung betrifft ein metallisches enossales Implantat aus Titan oder einer Titanlegierung, das in einen humanen oder tierischen Knochen eingepflanzt wird, wobei das Implantat eine glatte oder rauhe Textur aufweist und wobei die Oberfläche mit mindestens einer ausgewählten organischen Verbindung behandelt wurde, die eine oder mehr Phosphonsäuregruppen [-P(O)(OH)2] oder ein pharmazeutisch unbedenkliches Salz davon trägt. Es hat sich gezeigt, dass eine solche chemisch modifizierte Oberfläche die Bindungsstärke zum Knochen überraschend verbessert. Implantate gemäss der vorliegenden Erfindung können als Prothesen in der Medizin, insbesondere in der Orthopädie, zum Ersetzen oder Verstärken gebrochener oder erkrankter Knochen und in der Zahnmedizin zum Verankern künstlicher Zähne und zum Verankern von am Knochen verankerten Hörprothesen verwendet werden.
- Implantate, die in der Medizin als Prothesen zum Ersetzen oder Verstärken gebrochener oder erkrankter Knochen oder als künstliche Zähne verwendet werden, sind bekannt. Diese Implantate müssen aus einem korrosionsfreien Material bestehen und mit dem umgebenden Gewebe verträglich sein, ohne immunologische Reaktionen hervorzurufen, die die Abstossung durch den Körper bewirken. Im Folgenden bezeichnen die Begriffe ”Oberfläche” oder „Kontaktfläche” die Implantatoberfläche aus Titan oder einer Titanlegierung, die noch nicht gemäss der vorliegenden Erfindung behandelt wurde, und der Begriff ”modifizierte Oberfläche” bezeichnet die Oberfläche, die gemäss der Erfindung behandelt wurde.
- Es ist bekannt, dass das Implantieren von Geräten in Form von Schrauben, Platten, Nägeln, Stiften und spezifisch geformten Teilen in die Skelettstruktur von Menschen und Tieren als künstliche Prothese ein Mittel zum dauerhaften Ersatz fehlender struktureller Teile oder als dauerhafte Verankerungsgeräte darstellt. In solchen Fallen, in denen das implantierte Gerät dauerhaft an der Knochenkontaktfläche befestigt bleiben soll, ist eine ausgezeichnete ”Osseointegration” erforderlich.
- Es ist allgemein bekannt, für solche Implantate Titanmetall oder Titanlegierungen zu verwenden. Bei sorgfältiger Herstellung zeigt das Titanimplantat mit seinem Oberflächenoxid Biokompatibilität in dem Sinne, dass es für die Knochenregenerierung passiv bleibt und nicht per se beeinträchtigende Reaktionen wie etwa Entzündungen oder die Bildung von Weichgewebe oder Einkapselungen hervorruft. Die Berührungsfläche, die zwischen dem Implantat und dem Knochengewebe entsteht, besteht normalerweise aus einer Proteinschicht von etwa 100 nm bis 1 μm Stärke, die verhindert, dass das Knochengewebe in direktem molekularen Kontakt mit dem Implantat steht.
- Der gegenwärtige Stand der Technik bei enossalen Implantaten basiert auf verschiedenen Ansätzen, beispielsweise (i) auf der Erzeugung einer geeigneten Rauhheit der Implantatoberfläche, die eine mechanische Verzahnung zwischen Knochen und Implantat ergibt, und/oder (ii) auf der Beschichtung der Implantatoberfläche aus Titan- oder Titanlegierung, z. B. mit einem künstlichen Hydroxyapatit, um den Heilungsprozess und den engen Kontakt zwischen Knochen und Implantat zu verbessern.
- Es ist bekannt, dass eine hohe Oberflächenrauheit an Titanimplantaten die mechanische Stabilität des Implantats im Knochengewebe erhöht. Mechanische Oberflächenbehandlung verändert deutlich die Topographie, während die Oberflächenchemie im Wesentlichen unverändert bleibt. Die Nachteile eines Implantats mit einer hohen Oberflächenrauheit bestehen darin, dass eine rein mechanische Verankerung sehr empfindlich gegenüber Mikrobewegungen ist, was zu einer Verschlechterung der mechanischen Verankerung führen kann, und dass die Zeit für die Ossseointegration des Implantats relativ lang ist.
- Das Beschichten des Implantats mit einem künstlichen Hydroxyapatit verringert die Osseointegrationszeit. Es ist jedoch sehr schwierig, wenn nicht gar unmöglich, auf lastentragenden Implantaten Hydroxyapatit-Beschichtungen mit langfristiger Stabilität herzustellen. Die Berührungsfläche zwischen der Beschichtung und dem Implantat wird oft unterbrochen, oder die Beschichtungen schälen sich ab.

US 5,733,564 schlägt vor, ausgewählte Bisphosphonate, die bekannte Arzneimittelsubstanzen sind, zur Förderung der Knochengewebebildung einzusetzen, indem Titanmetalloberflächen von Prothesen oder Implantaten mit diesen Verbindungen beschichtet werden. - Es wurde jetzt festgestellt, dass, wenn die Oberfläche eines enossalen Implantats aus Titan oder einer Titanlegierung mit mindestens einer ausgewählten organischen Verbindung wie im Nachfolgenden definiert, die wie im Nachfolgenden beschrieben eine oder mehr Phosphonsäuregruppen oder ein Derivat davon, vorzugsweise einen Ester, ein Amid oder ein Salz davon, trägt, behandelt wurde, die Oberfläche eine überraschend verbesserte Bindungsstärke zum Knochen und eine überraschend verkürzte Osseointegrationszeit im Vergleich zur unbehandelten Oberfläche zeigt und nicht die Nachteile aufweist, wie sie für Oberflächen mit einer Hydroxyapatit-Beschichtung bekannt sind.
- Die vorliegende Erfindung ist in den Ansprüchen definiert. Die vorliegende Erfindung betrifft insbesondere ein enossales Implantat, das in einen humanen oder tierischen Knochen eingepflanzt wird, wobei das Implantat eine aus Titan oder einer Titanlegierung hergestellte Oberfläche aufweist, wobei die Oberfläche eine glatte oder rauhe Textur aufweist, und das dadurch gekennzeichnet ist, dass die Oberfläche mit wenigstens einer der allgemeinen Formel (I)
A-[P(O)(OH)2]p (I)
A1 für einen Rest eines geradkettigen, verzweigten oder cyclischen, gesättigten oder ungesättigten Kohlenwasserstoffrestes mit n Kohlenstoffatomen steht, wobei der Rest durch Carboxyl substituiert sein kann und gegebenenfalls weiter durch ein oder mehrere Sauerstoff- und/oder Schwefel- und/oder Stickstoffatome mit p Phosphonsäuregruppen substituiert sein kann, wobei n für eine Zahl von 2 bis 70, vorzugsweise 2 bis 40, vorzugsweise 2 bis 22, steht und p für 3 bis 6 steht;
oder A für A2 steht und
A2 für einen Rest einer Aminosäure, einer Sequenz von Aminosäuren, eines Proteins oder eines Polypeptids steht, wobei die Reste A2 jeweils p Phosphonsäuregruppen tragen, wobei
p für 1 bis 6, vorzugsweise 1, 2, 3 oder 4, vorzugsweise 1, 2 oder 3 steht; oder die Verbindung der Formel (I) Methylendiphosphonsäure, Propan-1,3-diphosphonsäure oder ein Salz davon ist. - Die vorliegende Erfindung betrifft ferner ein Verfahren zur Herstellung des Implantats gemäss der vorliegenden Erfindung, das dadurch gekennzeichnet ist, dass die Oberfläche mit wenigstens einer organischen Verbindung der Formel (I) oder einem Salz davon behandelt wird.
- Es wird davon ausgegangen, dass die Phosphonatverbindungen wie im Vorliegenden definiert, insbesondere als Sauren oder Salze, eine kovalente Bindung mit der Oberfläche des Implantats bilden und somit die Osseointegrationseigenschaften der Oberfläche in einem bemerkenswerten und unerwarteten Mass verbessern. Die vorliegende Erfindung ist jedoch nicht an diese Erklärung gebunden.
- A1 steht vorzugsweise für einen gesättigten Kohlenwasserstoffrest der Formel -(CnH2n+2-p)-, wobei n 1 bis 70, vorzugsweise 1 bis 40, vorzugsweise 1–22 bedeutet. Bevorzugt wird die freie Säure oder die Salzform der Verbindung der Formel (I), vorzugsweise diejenige, in der das pharmazeutisch unbedenkliche Salz ein Alkalisalz, vorzugsweise Natrium- oder Kaliumsalz ist.
- Die metallische Oberfläche des enossalen Implantats, die gemäss der vorliegenden Erfindung behandelt werden soll, kann aus Titan oder einer Titanlegierung hergestellt sein, wie sie bekanntermassen für die Herstellung von enossalen Implantaten verwendet werden. Titanlegierungen können aus Titan und jedem anderen Metall hergestellt sein, das bekanntermassen eine Legierung mit Titan bildet, wie etwa Chrom, Niob, Tantal, Vanadium, Zirkonium, Aluminium, Kobalt, Nickel und/oder Edelstahle. Solche Titan-Metall-Legierungen zur Anfertigung von Implantaten sind an sich bekannt und sind beispielsweise in Breme et al., Metals as Biomaterials, S. 1–71 (1998) beschrieben.
- Implantate gemäss der Erfindung können in Form von Schrauben, Platten, Nägeln, Stiften und spezifisch geformten Teilen vorliegen und können als Prothesen in der Medizin, insbesondere in der Orthopädie, zum Ersetzen oder Verstärken gebrochener oder erkrankter Knochen und in der Zahnmedizin zum Verankern künstlicher Zähne und zum Verankern von am Knochen verankerten Hörprothesen in der Skelettstruktur von Menschen und Tieren verwendet werden. Die Fläche der Implantatoberfläche, die an das Körpergewebe bzw. die Knochen gebunden werden soll, kann eine glatte oder rauhe Oberflächentextur aufweisen. Solche Oberflächentexturen sind bekannt und können beispielsweise erzielt werden, indem die Oberfläche mechanisch und/oder mit Säuren und/oder elektrolytisch und/oder mit einem Glimmentladungsplasma und/oder Plasmaspritzen und/oder durch elektromaschinelle Bearbeitung bearbeitet werden. Solche Materialien und Verfahren wurden in verschiedenen Veröffentlichungen beschrieben, beispielsweise in B.-O. Aronsson et al., J. Biomed. Mater. Res. 35 (1997), S. 49 f.
- Beispiele für Verbindungen der Formel (I), in denen A für einen Rest eines gesättigten Kohlenwasserstoffs steht, sind Polyphosphonsäuren wie etwa Ethan-1,1,2-triphosphonsäure, Butan-1,1,4-triphosphonsäure, Pentan-1,1,5-triphosphonsäure, Pentan-2,2,5-triphosphonsäure, Hexan-2,2,6-triphosphonsäure, Pentan-1,1,5,5-tetraphosphonsäure, Heptan-1,4,4,7-tetraphosphonsäure oder Propari-1,1,3,3-tetraphosphonsäure.
- Beispiele für Verbindungen der Formel (I), in denen A für einen Rest eines Proteins bzw. eines Polypeptids in Form eines Transforming Growth Factor beta (TGF-β) steht, umfassen alle Mitglieder der Wachstumsfaktor-Superfamilie und insbesondere den TGF-β1, TGF-β2, TGF-β3, TGF-β4 und TGF-β5 wie beispielsweise in A. B. Roberts, M. B. Sporn, Handbook of Experimental Pharmacology, 95 (1990), S. 419–472 oder D. M. Kingsley, Genes and Development 8 (1994), S. 133–146 und Verweisen darin beschrieben, in denen die Peptidkette so modifiziert wurde, dass sie eine Alkylphosphansäuregruppe oder ein Salz davon enthält. In diesem Sinne stellt die Verbindung der Formel (I) einen Transforming Growth Factor beta (TGF-β) dar, wie er durch die Mitglieder der Wachstumsfaktor-Superfamilie definiert ist, vorzugsweise den TGF-β1, TGF-β2, TGF-β3, TGF-β4 und TGF-β5, wobei die Peptidkette jeweils so modifiziert wurde, dass sie wenigstens eine Alkylphosphonsäuregruppe oder ein Salz davon enthält.

- Beispiele für Verbindungen der Formel (I), in denen A für einen Rest eines Peptids steht, umfassen, sind aber nicht beschränkt auf RGD-enthaltende Peptide, RGDS-Peptide, GRGDS-Peptide, RGDV-Peptide, RGDE-Peptide und/oder RGDT-Peptide. Solche Peptide sind beispielsweise in Y. Hirano, J. Biomed. Materials Res., 25 (1991), S. 1523–1534 oder in
WO 98/52619 - Bevorzugte Verbindungen der Formel (I) sind diejenigen, die einen Rest A2 wie oben definiert, vorzugsweise einen Rest einer Aminosäure oder einer Sequenz einer Aminosäure bzw. eines Proteins oder eines Polypeptids, vorzugsweise einen Rest der Superfamilie des Transforming Growth Factor beta (TGF-β), vorzugsweise ein knochenbildendes Protein (BMP) enthalten.
- Folgende Schritte werden empfohlen, um das Implantat gemäss der vorliegenden Erfindung herzustellen, d. h. um die Oberfläche des Implantats mit mindestens einer Verbindung der Formel (I) oben zu behandeln. Das Implantat wird zunächst in einem Reinigungsbad gereinigt, um unerwünschte Moleküle bzw. Verunreinigungen von der Oberfläche zu entfernen. Vorzugsweise wird das Implantat zuerst mit einem Entfettungsmittel, beispielsweise mit einem organischen Lösungsmittel wie etwa Alkohol, Chloroform, und einem weiteren organischen Lösungsmittel und/oder einem anorganischen Reinigungsmittel wie etwa einer wässrigen Alkalilösung auf der Basis von Natriumhydroxid oder Kaliumhydroxid behandelt. Anschliessend wird das Implantat sorgfältig in reinem Wasser, vorzugsweise in destilliertem ultra-reinem Wasser, das vorzugsweise einen Leitwiderstand von mindestens 15 Mohm/cm aufweist, gespült. Nach dem Reinigen und Spülen wird das Implantat mit Stickstoffstrom oder im Trocken- oder Heissluftstrom getrocknet und unter geregelten Bedingungen gelagert. Alternativ kann das Implantat nach dem Entfetten ferner mit einem Glimmentladungsplasma zum Reinigen der Oberfläche behandelt werden. Die saubere Oberfläche des Implantats wird dann mit mindestens einer Verbindung der Formel (I) oder einem Salz davon behandelt, d. h. mit mindestens einer solchen Verbindung oder einer Mischung davon. Die Verbindung oder die Mischung der Verbindungen wird mit einem beliebigen geeigneten Mittel auf die Oberfläche des Implantats gebracht, wie etwa durch Aufbürsten, Sprühen, Tauchen oder Aufdampfen, einschliesslich Glimmentladungsplasma-assistierter Dampfabscheidung, Die Phosphonsäureverbindung oder das Salz davon wird vorzugsweise in einem polaren Lösungsmittel gelöst, sodass eine Lösung mit einer Konzentration zwischen etwa 1,0 × 10–5 mol/10 ml und 5 × 10–2 mol/10 ml, vorzugsweise zwischen etwa 5 × 10–4 mol/10 ml und 2,0 × 10–2 mol/10 ml, bezogen auf das Gewicht des Lösungsmittels, erzielt wird. Vorzugsweise ist die Konzentration so gehalten, dass eine teilweise oder vollständige (1% bis 100%, vorzugsweise 50% bis 100% einer) monomolekulare Schicht auf der Implantatoberfläche gebildet wird. Das bevorzugte Lösungsmittel ist reines destilliertes Wasser. Das Implantat wird über eine ausreichend lange Zeit, vorzugsweise einige Minuten bis einige Stunden lang, im Kontakt mit der Lösung belassen. Danach wird das Implantat sorgfältig mit reinem Wasser gespült und mit einem Reinverpackungsmaterial aus Kunststoff oder Metall verpackt, vorzugsweise in eine luftdichte Verpackung, die vorzugsweise luftentleert oder mit einem inerten Gas wie etwa Stickstoff oder einer inerten Flüssigkeit wie etwa reinem Wasser wie oben definiert gefüllt wird. Das reine Wasser kann anorganische Salze, vorzugsweise Alkalisalze wie etwa Alkalichloride, -sulfate, -phosphate, -phosphonate, vorzugsweise die Natrium und/oder Kaliumsalze, und/oder Verbindungen der Formel (I) oder ein Salz davon enthalten, vorzugsweise in einer Konzentration zwischen etwa 1,0 × 10–5 mol/10 ml und 5 × 10–2 mol/10 ml, vorzugsweise zwischen etwa 5 × 104 mol/10 ml und 2,0 × 10–2 mol/10 ml des Lösungsmittels, bei dem es sich vorzugsweise um destilliertes Wasser handelt.
- Analytische Untersuchungen, z. B. Röntgen-Photoelektronen-Spektroskopie (XPS) oder Kernresonanzspektroskopie (NMR), haben ergeben, dass bei dem Kontakt der Phosphonsäureverbindung der Formel (I) mit der Titanoberfläche des Implantats unmittelbare Adsorption stattfindet. Es wird eine starke Bindung zwischen der Oberfläche und der Phosphonsäureverbindung gebildet, sodass eine chemische Oberflächenmodifizierung erzielt wird. Es wurden mehrere verschiedene Alkanpolyphosphonsäuren wie oben genannt synthetisiert. Zahnimplantate, die mit diesen Verbindungen gemäss der vorliegenden Erfindung hergestellt wurden, haben ausgezeichnete Resultate gezeigt.
- Die Verbindungen nach der allgemeinen Formel (I), in denen p für 3 bis 6, vorzugsweise 3 oder 4, und n für 4 bis 70, vorzugsweise 4 bis 40, vorzugsweise 4 bis 22, die Salze oder Ester oder Amide davon stehen, sind neu. Beispiele solcher Verbindungen sind Butan-1,1,4-triphosphonsäure, Pentan-1,1,5-triphosphonsäure, Pentan-2,2,5-triphosphonsäure, Hexan-2,2,6-triphosphonsäure, Pentan-1,1,5,5-tetraphosphonsäure, Heptan-1,4,4,7-tetraphosphonsäure oder Nonan-1,5,5,9-tetraphosphonsäure.
- Die Verbindungen Butan-1,1,4-triphosphonsäurehexaisopropylester und Heptan-1,4,4,7-tetraphosphonsäureoctaisopropylester bzw. eine Mischung dieser Verbindungen werden gewonnen, indem ein Alkalimetall, vorzugsweise Natrium, Methylendiphosphonsäureester mit vier niedrigen Alkylen, vorzugsweise Methylendiphosphonsäuretetraisopropylester, mit mindestens einer stöchiometrischen Menge eines Dihalomethans, vorzugsweise Dibromomethan, in Gegenwart eines organischen Lösungsmittels ohne aktive Wasserstoffatome, vorzugsweise trockenes Hexan oder Benzen oder Toluen umsetzt.
- Die Reaktion erfolgt vorzugsweise bei einer Temperatur im Bereich zwischen 30° und 125°, vorzugsweise zwischen 40° und 110°, bis die Reaktion abgeschlossen ist, was im allgemeinen innerhalb einer Zeitspanne von 10 bis 48 Stunden, vorzugsweise von 18 bis 36 Stunden, der Fall ist.
- Dem Reaktionsprodukt wird dann das gereinigte Produkt aus Triisopropylphosphit zugesetzt, das mit Diisopropyl-3-bromopropan reagiert wurde. Die gewonnene Mischung der Verbindungen kann dann auf herkömmliche Weise, beispielsweise durch Säulenchromatographie getrennt werden.
- In analoger Weise werden durch das Umsetzen von 1,4-Dibromobutan in einem molaren Überschussverhältnis im Bereich 1:6 bis 1:0,5 mit Triisopropylphosphit überraschend die neuen Verbindungen Pentan-1,1,5-triphosphonsäurehexaisopropylester und Nonan-1,5,5,9-tetraphosphonsäureoctaisopropylester hergestellt. Ferner wurden in analoger Weise Pentan-2,2,5-triphosphonsäurehexaisopropylester und Hexan-2,2,6-triphosphonsäurehexaisopropylester gewonnen, indem gleiche Teile von 1,1-Diphosphonsäuretetraisopropylester mit 3-Bromopropylphosphonsäurediisopropylester umgesetzt wurden.
- Das Verfahren ist ferner dadurch gekennzeichnet, dass diese Produkte hydrolysiert werden, um die analogen Säuren herzustellen, indem sie in molarem Überschuss von HCl über eine Zeit zwischen 1 und 12 Stunden, vorzugsweise zwischen 1 und 6 Stunden, refluxiert werden. Die Verbindungen werden dann vorzugsweise im Vakuum über P2O5 getrocknet.
- Die folgenden Beispiele illustrieren die vorliegende Erfindung, schränken sie jedoch nicht ein.
- Beispiel 1
- Methylendiphosphonsäure wurde gemäss
US-Patentschrift 3,400,176 und B. A. Arbusov, Pure Appl. Chem. 9 (1967), S. 307–353 und Verweisen darin synthetisiert. Die Verbindung wurde mittels NMR-Spektroskopie (1H, 31P, 13C), massenspektroskopischer Elementanalyse und anhand ihres Schmelzpunkts charakterisiert. All diese Daten entsprechen der Literatur O. T. Quimby et al., J. of Organomet. Chem. 13, 199–207 (1968). - Aus Methylendiphosphonsäuretetraisopropylester wurde Propan-1,1,3,3-tetraphosphonsäure synthetisiert. Die Tetraphosphonsäure-Lösung wurde unter Vakuum konzentriert, über P2O5 im Vakuum getrocknet. Die NMR-Ergebnisse 1H, 31P, 13C (D2O) entsprechen den angegebenen Daten in der Literatur.
- In analoger Weise werden Propan-1,3-diphosphonsäure, Ethan-1,1,2-triphosphonsäure, Butan-1,1,4-triphosphonsäure, Pentan-1,1,5-triphosphonsäure, Pentan-2,2,5-triphosphonsäure, Hexan-2,2,6-triphosphonsäure, Pentan-1,1,5,5-tetraphosphonsäure, Heptan-1,4,4,7-tetraphosphonsäure oder Nonan-1,5,5,9-tetraphosphonsäure synthetisiert.
- Beispiel 2

- A) Eine Probe aus Titan in Form einer kreisrunden Platte mit einem Durchmesser von 14 mm und einer Stärke von 1 mm wird in herkömmlicher Weise hergestellt. Die Probenoberfläche wird nach Standardverfahren durch mechanisches Polieren mit Diamantpaste mit einer glatten Oberflächenrauhheit versehen. Mit Hilfe der Rasterkraftmikroskopie wurde die Oberflächenrauheit auf einen Srms-Wert von etwa 6 nm über eine Fläche der Oberfläche von 400 Quadratmikronen gemessen.
- B) Das wie in Kapitel A) oben hergestellte Implantat wird dann in eine wässrige Lösung aus (i) Methylendiphosphonsäure [1,5 × 10–3 mol pro 10 ml destillierten Wassers], (ii) Ethan-1,1,2-triphosphonsäure [1,2 × 10–3 mol pro 10 ml, in destilliertem Wasser], (iii) Propan-1,1,3,3-tetraphosphonsäure [6,2 × 10–4 mol pro 10 ml, in destilliertem Wasser] gegeben und dort 15 Minuten lang bei Raumtemperatur belassen. Das Implantat wird dann mit reinem Wasser gespült und getrocknet.
- Die nach den Zubereitungen B(i), B(ii) und B(iii) bearbeiteten Implantate werden mit Knochenbildungszellen, Osteoblasten, von Ratten beschichtet. Die Osteogenese wird als (I) die Zellvermehrung und (II) die Knochenproteinsynthese gemessen. Vergleichstestwerte sind für das unbehandelte Implantat angegeben. Die Ergebnisse sind in Tabelle 1 dargestellt. Analoge Ergebnisse werden bei allen oben genannten Phosphonsäuren sowohl auf glatter als auch auf rauher Oberfläche erzielt. Die Analyse mit XPS und ToF-SIMS deutete an, dass eine molekulare (Mono-)Schicht sowohl auf einer Titanoberfläche als auch auf einer TiO2-Oberfläche gebildet wurde. Tabelle 1

cpm = Zählungen pro Minute (counts per minute) aus radiomarkierten Proteinen. - Die Ergebnisse verdeutlichen die verbesserte Osteogenese der Implantate gemäss der vorliegenden Erfindung im Vergleich zu dem unbehandelten Implantat.
- Beispiel 3 (Referenz)
- Beispiel 2 wird mit dem Unterschied wiederholt, dass das Implantat mit Ethan-1,1,3-triphosphonsäure behandelt wird, die modifiziert wurde, indem der Aminoterminus eines Glycinmoleküls mit einer der Phosphonatgruppen verbunden wurde.
- Beispiel 4 (Referenz)
- Beispiel 2 wird mit dem Unterschied wiederholt, dass das Implantat mit Ethan-1,1,3-triphosphonsäure behandelt wird, die modifiziert wurde, indem der Aminoterminus eines GRGDS-zellbindenden Polypeptids mit einer der Phosphonatgruppen verbunden wurde.

- Beispiel 2 wird mit dem Unterschied wiederholt, dass es sich bei der Verbindung der Formel (I) um Ethan-1,1,3-triphosphonsäure handelt, die modifiziert wurde, indem der Aminoterminus (Methionin) eines humanen knochenbildenden Proteins Typ 2 (BMP-2) mit einer der Phosphonatgruppen verbunden wurde, was zu Testergebnissen führt, die analog zu denen in Tabelle 1 sind.
Claims (13)
- Enossales Implantat, das in einen humanen oder tierischen Knochen eingepflanzt wird, wobei das Implantat eine aus Titan oder einer Titanlegierung hergestellte Oberfläche aufweist, wobei die Oberfläche eine glatte oder rauhe Textur aufweist, dadurch gekennzeichnet, dass die Oberfläche mit wenigstens einer der allgemeinen Formel (I)
A-[P(O)(OH)2]p (I) - Implantat nach Anspruch 1, wobei A1 für einen gesättigten Kohlenwasserstoffrest der Formel -(CnH2n+2-p)- steht, wobei n 1 bis 70 bedeutet.
- Implantat nach Anspruch 1 oder 2, dadurch gekennzeichnet, dass es sich bei dem pharmazeutisch unbedenklichen Salz um ein Alkalisalz handelt.
- Implantat nach einem der Ansprüche 1–3, wobei es sich bei der Verbindung der Formel (I) um eine Polyphosphonsäure vorzugsweise ausgewählt aus Ethan-1,1,2-triphosphonsäure, Butan-1,1,4-triphosphonsäure, Pentan-1,1,5-triphosphonsäure, Pentan-2,2,5-triphosphonsäure, Hexan-2,2,6-triphosphonsäure, Pentan-1,1,5,5-tetraphosphonsäure, Heptan-1,4,4,7-tetraphosphonsäure und/oder Propan-1,1,3,3-tetraphosphonsäure oder ein Salz davon handelt.
- Implantat nach Anspruch 1, wobei es sich bei der Verbindung der Formel (I) um einen Transforming Growth Factor beta (TGF-β) wie durch die Mitglieder der Wachstumsfaktor-Superfamilie definiert handelt, wobei die Peptidkette jeweils so modifiziert wurde, dass sie wenigstens eine Alkylphosphonsäuregruppe oder ein Salz davon enthält.
- Implantat nach Anspruch 1, wobei es sich bei der Verbindung der Formel (I) um ein knochenbildendes Protein (Bone Morphogenic Protein, BMP) handelt, wobei die Peptidkette jeweils so modifiziert wurde, dass sie wenigstens eine Alkylphosphonsäuregruppe oder ein Satz davon enthält.
- Implantat nach Anspruch 1, wobei es sich bei der Verbindung der Formel (I) um 2-Amino-4,4-bis-(diethoxyphosphoryl)buttersäure, 2-Amino-5-(diethoxyphosphoryl)pentansäure und/oder 2-Amino-4-phosphonobuttersäure handelt.
- Implantat nach Anspruch 1, wobei es sich bei der Verbindung der Formel (I) um eine der zwanzig proteinogenen Aminosäuren handelt, wobei die Aminosäure so modifiziert wurde, dass sie wenigstens eine Alkylphosphonsäuregruppe oder ein Salz davon enthält.
- Implantat nach Anspruch 1, wobei es sich bei der Verbindung der Formel (I) um ein RGD-enthaltendes Peptid handelt, das so modifiziert wurde, dass es wenigstens eine Alkylphosphonsäuregruppe oder ein Salz davon enthält.
- Verfahren zur Herstellung des Implantats nach einem der Ansprüche 1–9, dadurch gekennzeichnet, dass die Oberfläche mit wenigstens einer organischen Verbindung der Formel (I) oder einem Salz oder einer Mischung davon behandelt wird.
- Implantate nach einem der Ansprüche 1–9 in Form von Schrauben, Platten, Nägeln und Stiften.
- Kunststoff- oder Metallverpackungsmaterial, das gegebenenfalls evakuiert oder mit einem inerten Gas oder einer inerten Flüssigkeit gefüllt ist, enthaltend ein Implantat nach Anspruch 11.
- Verpackungsmaterial nach Anspruch 12, dadurch gekennzeichnet, dass die Verpackung mit reinem Wasser gefüllt ist, das ein anorganisches Salz und/oder eine Verbindung der Formel (I) oder ein Salz davon enthält.
